采用19,22,25,28和31℃5个温度对竹织叶野螟Algedonia coclesalis Walker各虫态(龄)发育起点温度和有效积温进行测定。结果表明,竹织叶野螟的在19~31℃范围内均能正常生长发育,尤其是28~31℃范围最适宜于竹织叶野螟的生长发育。卵、1龄幼虫、2龄幼虫、3龄幼虫、4龄幼虫、5龄幼虫、6龄幼虫、7龄幼虫、蛹、成虫及世代的发育起点温度分别为6.63,12.51,11.18,10.93,10.05,8.01,6.80,5.78,6.20,7.81和8.33℃,有效积温分别为124.19,64.54,72.59,82.08,93.46,136.84,155.42,201.06,211.55,111.49和1235.50日.度。  相似文献

The genes of the mitochondrial and cytosolic malate dehydrogenase (mMDH and cMDH) of Phytophthora infestans were cloned and overexpressed in Escherichia coli as active enzymes. The catalytic properties of these proteins were determined: both enzymes have a similar specific activity. In addition, the natural mitochondrial isoenzyme was semi-purified from mycelia and its catalytic properties determined: the recombinant mitochondrial isoform behaved as the natural enzyme. A phylogenetic analysis indicated that mMDH, present in all stramenopiles studied, can be useful to study the relationships between these organisms. MDH with the conserved domain MDH_cytoplasmic_cytosolic is absent in some stramenopiles as well as in fungi. This enzyme seems to be less related within the stramenopile group. The Phytophthora cMDHs have an insertion of six amino acids that is also present in the stramenopile cMDHs studied, with the exception of Thalassiosira pseudonana cMDH, and is absent in other known eukaryotic cMDHs.  相似文献

为了探索抗肿瘤药物体外筛选中磺酰罗丹明B(SRB)法和四甲基偶氮唑盐(MTT)比色法实验结果的相互验证作用,作者以PC-3、BGC-823、Bcap-37细胞系为研究对象,利用SRB法和MTT比色法对供试化合物行体外抑制活性考察,并用SPSS12.0对两种筛选方法得到的实验结果进行差异性分析。结果表明,SRB法和MTT比色法实验结果相关性好,可以相互验证实验结果的准确性和可靠性。  相似文献

In our previous work, we described the defensive potential of a wound- and methyl jasmonate-inducible 87 kDa tomato cystatin and its accumulation in a crystalline form. Here, we report the immunolocalization of this cysteine proteinase inhibitor in tomato leaf blade. Methyl jasmonate treated wild type plants showed accumulation of crystalline structures that were specifically and strongly stained with polyclonal antibodies against tomato cystatin. Crystalline cystatin was found in palisade and spongy parenchyma cells and immuno-gold electron microscopy analysis revealed that the crystals were compartmentalized in the cytosol. The same pattern in localization of cystatin was observed in transgenic tomato plants superexpressing prosystemin transgene. Our data showing the accumulation of cystatin in response to methyl jasmonate and in response to a overproduction of a wound signal corroborate the protective role of this inhibitor.  相似文献

采用世界粮农组织(FAO)和国际动物遗传学会(ISAG)推荐的27个微卫星,对久仰香猪、剑白香猪、从江香猪、环江香猪、黑香猪(贵州省种猪场)、五指山猪和滇南小耳猪等我国7小型猪种(类群)及杜洛克、长白和大白等3个外来猪种的群内遗传变异性和群间遗传差异进行了分析。结果表明,7个小型猪品种(类群)均有较高的群内遗传变异,但久仰、剑白、从江和环江4个香猪类群的群内遗传变异(平均多态信息含量(PIC)为0.61~0.64)显著低于其他3个小型猪种(平均PIC为0.80~0.84)。久仰、剑白、从江和环江4个香猪类群彼此间的遗传差异较小(奈氏标准遗传距离为0.12~0.22),但它们与其他3个小型猪种有较大的遗传差异(奈氏标准遗传距离为1.61~1.96),与3个外来猪种的遗传差异更大(奈氏标准遗传距离为1.99~3.30)。通过聚类分析,可将这些猪种清晰地分为3大类,久仰、剑白、从江和环江4个香猪类群紧密地聚为一类,其他3个小型猪种聚为一类,3个外来猪种聚为另一类。这一结果基本符合这些猪种的地理分布和品种来源。

为了研究红曲霉聚酮体途径,考察和比较了4种不同的转化方法以建立有效的红曲霉遗传转化系统。以潮霉素作为抗性筛选标记,pBC-Hygro作为转化载体,用基于原生质体的传统转化和电击转化、基于萌发孢子的电击转化以及REMI技术转化红曲霉。发现基于萌发孢子的电击转化由于转化率极低而不适于红曲霉转化。基于原生质体的传统转化和电击转化尽管每微克DNA分别能获得135个转化子和125个转化子,但因转化子稳定性差也适合红曲霉转化的转化。应用REMI技术,转化率提高约20倍,每微克DNA 2500个转化子,70%~75%的转化子的稳定,非常适合于红曲霉的转化。   相似文献

Tolerance to the effects of drought and subsequent recovery after a rainfall appear to be critical for plants in the karst regions of southwestern China, which are characterized by frequent but temporary drought events. This study investigated the effects of drought intensity and repetition on photosynthesis and photoprotection mechanisms of karst plants during successive cycles of drought and subsequent recovery. Leaf water potential, gas exchange, chlorophyll fluorescence and several associated metabolic processes were studied in six plant species, including Pyracantha fortuneana (PF), Rosa cymosa (RC), Broussonetia papyrifera (BP), Cinnamomum bodinieri (CB), Platycarya longipes (PL) and Pteroceltis tatarinowii (PT) during three cycles of drought treatments at four different intensities. The four treatments were: well‐watered, mild drought, moderate drought and severe drought, each followed by rewatering events. We found that limitations to CO2 diffusion accounted for photosynthetic declines under mild and moderate drought treatments, while metabolic limitations dominated the response to severe drought. Repetition of drought did not intensify the impairment of photosynthetic metabolism regardless of drought intensity in the six species studied. Repetition of severe drought delayed the photosynthetic recoveries in PF, RC and CB after rewatering. Repetition of drought increased thermal dissipation in PF, CB and BP, as well as superoxide dismutase (EC 1.15.1.1) activity in RC and CB. Enhanced photosynthetic performance, measured as increased intrinsic water use efficiency, photosynthetic performance per unit of photosynthetic pigment, maintenance of high thermal dissipation and high ratios of carotenoids to chlorophylls, was observed during the rewatering periods. This enhanced photosynthetic performance allowed for the complete recovery of the six karst species from successive intermittent drought events.  相似文献

Humans have five members of the well conserved RecQ helicase family: RecQ1, Bloom syndrome protein (BLM), Werner syndrome protein (WRN), RecQ4, and RecQ5, which are all known for their roles in maintaining genome stability. BLM, WRN, and RecQ4 are associated with premature aging and cancer predisposition. Of the three, RecQ4's biological and cellular roles have been least thoroughly characterized. Here we tested the helicase activity of purified human RecQ4 on various substrates. Consistent with recent results, we detected ATP-dependent RecQ4 unwinding of forked duplexes. However, our results provide the first evidence that human RecQ4's unwinding is independent of strand annealing, and that it does not require the presence of excess ssDNA. Moreover, we demonstrate that a point mutation of the conserved lysine in the Walker A motif abolished helicase activity, implying that not the N-terminal portion, but the helicase domain is solely responsible for the enzyme's unwinding activity. In addition, we demonstrate a novel stimulation of RecQ4's helicase activity by replication protein A, similar to that of RecQ1, BLM, WRN, and RecQ5. Together, these data indicate that specific biochemical activities and protein partners of RecQ4 are conserved with those of the other RecQ helicases.  相似文献
