以J3、IS1112、IS1113和ERIC为引物对17组(56株)来自中国、日本和菲律宾的水稻白叶枯病菌(Xanthomonas oryzae pv.oryzae)代表菌株和它们的单细胞系进行IS-PCR和Rep-PCR扩增以及田间致病性鉴定,研究母株和单细胞系之间的遗传和致病性变异.结果表明:1)母株与单细胞系属于同一分子谱型的比率依次为:52.9%(J3)、23.5%(IS1112)、29.4%(IS1113)和35.3%(ERIC); 2)综合4个引物的PCR扩增结果,采用UPGMA聚类,母株与其全部单细胞系相似率在90%以上的组群占29.4%,差异率达到30%~41%的组群占参试组群的52.9%;以带位相似率80%为簇界,参试菌可以划分为15簇,来自中国、日本和菲律宾的9个组的母株和单细胞系聚合在同一簇中,占参试菌的52.9%,说明三国病菌在遗传上存在高度相关性;三国病菌都存在簇群差异,以菲律宾茵系变异最大,如Pxo79和Pxo112母株和单细胞系各分在不同的3簇中,Pxo79、Pxo86、Pxo99和Pxo112等菌系的母株和单细胞系之间的差异率达到了41%;结果证明:母株是"由遗传上有不同程度差异的细胞"组成的混合体.3)田问致病性测定表明,母株与其单细胞系毒力不同的有15组,占88.2%,说明母株是由毒力不同的单细胞系组成的混合体,但病原菌的分子谱型以及遗传分簇与致病型之间没有相关性.  相似文献

The seedlings of wheat were treated by salt-stress (SS, molar ratio of NaCl: Na2SO4 = 1: 1) and alkali-stress (AS, molar ratio of NaHCO3: Na2CO3 = 1: 1). Relative growth rate (RGR), leaf area, and water content decreased with increasing salinity, and the extents of the reduction under AS were greater than those under SS. The contents of photosynthetic pigments did not decrease under SS, but increased at low salinity. On the contrary, the contents of photosynthetic pigments decreased sharply under AS with increasing salinity. Under SS, the changes of net photosynthetic rate (P N), stomatal conductance (g s), and transpiration rate (E) were similar and all varied in a single-peak curve with increasing salinity, and they were lower than those of control only at salinity over 150 mM. Under AS, P N, g s, and E decreased sharply with rising salinity. The decrease of g s might cause the obvious decreases of E and intercellular CO2 concentration, and the increase of water use efficiency under both stresses. The Na+ content and Na+/K+ ratio in shoot increased and the K+ content in shoot decreased under both stresses, and the changing extents under AS were greater than those under SS. Thus SS and AS are two distinctive stresses with different characters; the destructive effects of AS on the growth and photosynthesis of wheat are more severe than those under SS. High pH is the key feature of the AS that is different from SS. The buffer capacity is essentially the measure of high pH action on plant. The deposition of mineral elements and the intracellular unbalance of Na+ and K+ caused by the high pH at AS might be the reason of the decrease of P N and g s and of the destruction of photosynthetic pigments.  相似文献

There are many dynamic optimization problems in the real world, whose convergence and searching ability is cautiously desired, obviously different from static optimization cases. This requires an optimization algorithm adaptively seek the changing optima over dynamic environments, instead of only finding the global optimal solution in the static environment. This paper proposes a novel comprehensive learning artificial bee colony optimizer (CLABC) for optimization in dynamic environments problems, which employs a pool of optimal foraging strategies to balance the exploration and exploitation tradeoff. The main motive of CLABC is to enrich artificial bee foraging behaviors in the ABC model by combining Powell’s pattern search method, life-cycle, and crossover-based social learning strategy. The proposed CLABC is a more bee-colony-realistic model that the bee can reproduce and die dynamically throughout the foraging process and population size varies as the algorithm runs. The experiments for evaluating CLABC are conducted on the dynamic moving peak benchmarks. Furthermore, the proposed algorithm is applied to a real-world application of dynamic RFID network optimization. Statistical analysis of all these cases highlights the significant performance improvement due to the beneficial combination and demonstrates the performance superiority of the proposed algorithm.  相似文献

以重庆市合川土地整理项目区为例,对项目区整理前(2003年)、后(2007年)农业生态系统功能的变化进行量化描述,将工程设计和影子价格法相结合对干扰功能进行了量化,并采用成本效益分析法进一步解释生态系统功能的经济学意义,分析了工程影响下区域农业生态系统服务价值的变化.结果表明:与土地整理之前相比,研究区农业生态系统服务价值在整理后有所改善,其中增幅最大的是养分循环功能,变化最小的是土壤保持功能;在土地整理项目中,农田水利、田间道路和土地平整工程带来的生态服务价值改变主要体现为干扰能的变化.项目建成后第7年到第35年,研究区成本效益率处于高速增长期,之后趋于稳定,表现为较弱的生态压力和较少的生态服务价值损失,有利于区域生态环境的改善.  相似文献

Four structurally diverse complexes, [Cd(dppz)(bdoa)]n (1), [Zn(dppz)(bdoa)(H2O)]n (2), [Fe(dppz)2(bdoa)]n·2nH2O (3), and [Co2(dppz)2(bdoa)2(H2O)]n·3nH2O (4), where H2bdoa = benzene-1,4-dioxyacetic acid and dppz = dipyrido[3,2-a:2′,3′-c]phenazine, have been hydrothermally synthesized. Compounds 1-4 feature chain structures. There exist π-π interactions in the structures of 1, 2 and 4. Two neighboring chains of 1 are linked through the π-π interactions into a double chain supramolecular structure. The chains of 2 and 4 are further extended by the π-π interactions to form 3D and 2D supramolecular structures, respectively. The structural differences among such complexes show that the transition metals have important influences on their structures. The photoluminescent property of complex 2 and the magnetic property of complex 4 have also been investigated.  相似文献

Reaction of copper(I) chloride and bromide, CuX (X = Cl or Br), with triphenylarsine in acetonitrile solution has resulted in two adducts, respectively, of 2:3:1, [(Ph3As)2Cu(μ-Cl)2Cu(AsPh3)(NCMe)] (1), and 1:1:2 stoichiometry, [(Ph3As)(MeCN)Cu(μ-Br)2Cu(NCMe)(AsPh3)] · 2MeCN (2), characterized by elemental analysis, IR, ESI MS and NMR spectroscopy, and room temperature single crystal X-ray structure determinations. The environments of the two four-coordinate copper(I) atoms in 1 are different, being As2Cu(μ-Cl)2 and As,NCu(μ-Cl)2. (2) is also binuclear, being a centrosymmetric dimer with the two four-coordinate, symmetry-related copper atoms having As,NCu(μ-Br)2-environments.  相似文献

近年来稻米的香味品质受到消费市场的格外重视。水稻OsBADH2基因是影响稻米香味的重要基因,选择优质粳稻品种,针对该基因进行基因编辑,有望创制出香味品质优异的粳稻材料。本研究通过分析OsBADH2基因外显子序列的保守性,选择了3个特异性靶点,分别构建了U6启动子驱动的包含不同靶点的gRNA表达盒,并将其连入植物基因编辑表达载体。利用基因枪介导的瞬时转化体系,对非香型粳稻品种"吉粳88"进行遗传转化,T_0代共获得847株再生材料。经高分辨率溶解曲线(High Resolution Melting,HRM)分析和测序验证,24株T_0代材料的OsBADH2基因编辑靶点发生了不同类型的编辑。在T_1代材料中获得了7类不含转基因成分,且OsBADH2基因靶点纯合的基因编辑后代材料。本研究结果表明基于基因枪介导的瞬时转化体系,转化再生过程中省略了抗性筛选过程,并辅以高效的HRM检测,能够在T_1代即获得位点发生纯合编辑且无转基因成分的基因编辑后代材料。本研究获得的OsBADH2基因编辑的"吉粳88"后代材料也为培育香味品质改良的粳稻品种提供了新材料。  相似文献

以长白落叶松(Larix olgensis)未成熟合子胚为外植体诱导胚性愈伤组织, 通过调节影响体胚发生的营养物质和植物生长调节剂配比, 进行愈伤组织的胚性恢复与保持以及体胚发生再生体系的优化。结果表明: 不同无性系之间胚性愈伤组织诱导率差异显著, 胚性愈伤组织在S+0.2 mg·L -1NAA+0.5 mg·L -1BA+0.5 mg·L -1KT+0.5 g·L -1谷氨酰胺+0.5 g·L -1水解酪蛋白+30 g·L -1蔗糖及3.0 g·L -1植物凝胶培养条件下, 可以恢复胚性并长久保持。在S+20 mg·L -1ABA+60 g·L -1PEG4000+60 g·L -1蔗糖及3.0 g·L -1植物凝胶条件下分化培养6周, 体胚发生率可达100%。将正常发育的体胚先在WPM+ 6 mg·L -1间苯三酚+1.0 g·L -1活性炭+3.0 mg·L -1VB1+20 g·L -1蔗糖及3.0 g·L -1植物凝胶条件下培养2周, 再转接至B5+ 0.4 mg·L -1NAA+1.0 mg·L -1IBA+0.5 mg·L -1GA3+2.0 mg·L -1VB1+1.0 g·L -1活性炭+20 g·L -1蔗糖及3.0 g·L -1植物凝胶条件下培养2周, 可见子叶舒展、下胚轴伸长且根系正常的体胚苗。该研究建立了长白落叶松胚性愈伤组织胚性恢复与保持方法, 并进一步优化了体胚发生的植株再生体系, 为林木资源快速繁育和遗传改良奠定了基础。  相似文献
