S100 proteins are low-molecular-weight calcium-binding proteins of the EF-hand superfamily and appear to be involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. More than 10 members of the S100 protein family have been described from human sources so far. We have now isolated a YAC clone from human chromosome 1q21, on which 9 different genes coding for S100 calcium-binding proteins could be localized. Moreover, we have mapped the gene coding for S100P to human chromosome 4p16 and thereby completed the chromosomal assignments of all known human S100 genes. The clustered organization of S100 genes in the 1q21 region allows us to introduce a new logical nomenclature for these genes, which is based on the physical arrangement on the chromosome. The new nomenclature should facilitate and further the understanding of this protein family and be easily expandable to other species.  相似文献

Two phototoxic plants of the Asteraceae family were studied in relation to species of phytophagous insects for which they are hosts:Argyrotaenia velutinana Wlk. feeding onChrysanthemum leucanthemum L. andChlorochlamys chloroleucaria (Guenée) colonizingRudbeckia hirta L. The toxicity of these two plants is related to the presence of acetylenes and thiophenes that induce a light-mediated production of deleterious singlet oxygen and other free radicals (phototoxicity). Results showed that females ofA. velutinana laid their eggs preferentially in the shade and the larvae adopted hiding behaviors, such as bending of ligulate corollas and silk spinning to build opaque shelters. By avoiding direct exposure to the sun, both behaviors may reduce phototoxicity associated with ingested plant materials. Furthermore, larvae ofC. chloroleucaria demonstrated a preference in the field for pollen, which constitutes a nonphototoxic tissue of their host plant. Experimental alterations of these specific behaviors induced important biological consequences for larvae of both insects such as mortality or reduction of larval growth rate. These results reinforce the idea that behavior may constitute an efficient adaptation to avoid phototoxicity.  相似文献

The restriction enzymes AluI, BamHI, BglII, EcoRI, HindIII, and SalI have been used to characterize and map a new cauliflower mosaic virus strain (Cabb-S). These fragments have been ordered by examining their overlapping regions after double enzymatic digestion. The single SalI cleavage site was chosen as the point of origin. Summary Increasing concentrations of dimethylsulfoxide (DMSO) strip tobacco mosaic virus (TMV) stepwise from the 3 end. The RNA tail increases in length up to 2,000 nucleotides (nu) reaching a region of very strong protein-RNA affinity. Thereafter, uncoating occurs from the other end and produces a second RNA tail 500 nu long. Further stripping of TMV proceeds from both ends, the long tail increasing in length up to 4,000 nu and the short one increasing more moderately and remaining below 2,000 nu. The region of strongest protein-RNA affinity is located between 4,000 and 5,000 nu away from the 3 end.Using the same conditions as for in vitro TMV reassembly, it is possible to recoat the RNA tails with viral protein preferentially in the 5 direction.The advantages of DMSO in studies of TMV protein-RNA interactions are discussed.  相似文献

Summary The effect of aging on the morphology and function of the thyroid gland of the cream hamster was studied by light and electron microscopy coupled with autoradiography or histochemistry.Morphologically, aging induces an accumulation of lysosomal dense bodies and a loss of the phagocytosis of colloid droplets after stimulation with TSH. Iodine uptake and organification occur normally and thyroglobulin synthesis, estimated by autoradiography with 3H-leucine, is not different from that observed in young animals. The basal T4 and T3 plasma levels are lower in the old animals. A low iodine diet administered for several months prevents the age related accumulation of lysosomal dense bodies. In the genus Schizosaccharomyces intracellular osidases and nitrite and nitrate reductases are revealed; particularly all the species possessing invertase, -glucosidase and -galactosidase. These characters underline the homogeneity of the genus. On the basis of osidases, nitrite and nitrate reductases results, 2 groups can be distinguished in this genus.  相似文献

Summary A method to obtain amber mutations in ribosomal protein genes is described. It relies on the P1-mediated localized mutagenesis (Hong and Ames, 1971) and on the fact that the recipient strain contains (a) an efficient but genetically unstable suppressor, (b) a particular thermoinducible prophage which kills suppressor hosts at 42° C. Exposure of these bacteria to the high temperature yields frequent suppressor-free derivatives while none will be found if the strain carries an amber mutation in an essential gene. Eleven mutants have been isolated by this method, of which at least six appear to carry amber mutations. All of them map close to, and to the right of spcA, in a region which codes mostly for ribosomal proteins. Three mutants were studied biochemically; all three show defective ribosomal assembly in vivo upon loss of suppression.  相似文献

The absolute configurations of heteroxanthin ((3S,5S,6S,3′R)- 7′,8′-didehydro-5,6-dihydro-β,β-carotene-3,5,3′,6′-tetrol) ex Euglena gracilis and of diadinoxanthin ((3S,5R,6S,3′R)-5,6-epoxy-7′,8′-didehydro-5,6-dihydro-β,β-carotene-3,3′-diol) from the same source have been established by chemical reactions, hydrogen bonding studies, 1H NMR and CD. Two previously unknown carotenoids (artefacts?) from Trollius europaeus, assigned the structures (3S,5S,6S,3′S,5′R,6′R)-6,7-didehydro-5,6,5′,6′-tetrahydro-β,β -carotene-3,5,6,3′,5′-pentol and its 5R epimer, served as useful models.  相似文献

Auxin-induced variations of transmembrane potential difference have been shown to be a useful tool for analyzing hormone sensitivity in tobacco protoplasts. Using this technique, we demonstrated that protoplasts derived from wild-type, an auxin-resistant mutant and Agrobacterium-rhizogenes transformed plants differed widely in the sensitivity of their electrical response to naphthalene acetic acid. We have used different antibodies, raised to auxin binding proteins (ABP) from maize coleoptiles, or to the axr1 gene product (ABP1), to test whether changes in auxin sensitivity can be correlated with the presence of tobacco proteins immunologically related to this ABP. Titrations indicated that 0.4 nM anti-ABP IgG inhibited 50% of the auxin-specific response of wild-type protoplasts, whereas 0.04 nM or 4 nM anti-ABP IgG were necessary to inhibit the response of mutant and transformed protoplasts, respectively, to the same extent. On wild-type protoplasts, blocking part of the immunoreactive sites with anti-ABP antibodies resulted in a decrease in auxin sensitivity of the electrical response (0.4 nM anti-ABP IgG inducing a 10–fold decrease), whereas addition of maize ABP increased this auxin sensitivity (1 pM ABP1 raised the sensitivity more than 1000–fold). The results obtained suggest that the auxin sensitivity detected by our assay system correlates with the amount of tobacco proteins immunologically related to the axr1 gene product from maize. A hypothesis accounting for the presence of these proteins at the external surface of tobacco protoplasts and for the effects of hetero-logous maize ABP on auxin sensitivity is proposed.  相似文献
