Human TRP-1 Has Tyrosine Hydroxylase but no DOPA Oxidase Activity

Human TRP-1 has been immunopurified from normal human melanocytes cultured from black neonatal subjects and used to investigate the catalytic function of TRP-1 for the two substrates, L-tyrosine and L-DOPA. Immunopurified TRP-1 did not demonstrate DOPA staining on SDS/PAGE nor DOPA oxidase (DO) activity with either routine or modified assays. The purified TRP-1 also demonstrated no tyrosine hydroxylase (TH) activity using the routine Pomerantz assay. However, there was apparent TH activity exhibited by immunopurified TRP-1 under conditions with low tyrosine concentration (≤0.8 μCi/ml of ³H-tyrosine), prolonged incubation time (i.e., overnight) and in the absence of the cofactor L-DOPA. Using these latter specific conditions, TH activity was also detected in cell lysates from a tyrosinase-negative albino melanocyte line which exhibited no TH activity with the routine Pomerantz assay. In addition, TH activity under low substrate assay conditions was not exhibited in a melanocyte line derived from a TRP-1 deficient, Brown albino individual. However, the absence of TH in this Brown albino cell line could be compensated for by the addition of L-DOPA to the assay. These results suggested that TRP-1 has some tyrosine hydroxylase but no DOPA oxidase activity. We propose that one function of TRP-1 is to modulate tyrosinase activity by making DOPA available as a cofactor to perpetuate the initial steps in melanogenesis.     

高脂血症兔球结膜微循环的改变

本实验用胆固醇粉喂家兔,造成实验性高脂血症,活体观察球结膜微循环的形态、流态及血管周围状态等１５项指标,并用球结膜微循环综合定量评价方法计算了综合积分值。处死后取球结膜进行组织学检查。结果表明：高脂血症家兔球结膜微循环有明显改变,主要为微血管形态异常、血流减慢、红细胞聚集、白微栓、静脉壁上有白色斑块等改变。球结膜微循环综合积分值明显增加,高于实验前。球结膜组织学检查发现上皮层下有泡沫细胞、血管壁增厚、有空泡、静脉内有血栓。虹膜睫状体内除有多数泡沫细胞外,还有胆固醇的菱形结晶。上述改变表明,高脂血症兔球结膜微循环有明显改变。血液有高凝、高聚倾向。黑茶中的茶色素有抗凝、抑制血小板聚集、促进纤溶等作用,口服黑茶可改善高脂血症兔球结膜微循环障碍。     

油菜菌核病危害损失的初步研究

本文就油菜菌核病病情、发病期及发病部位与油菜产量损失的相互关系进行了模似研究。结果表明：病斑绕茎度是影响产量的主要因素,其大小与产量损失成正相关,而病斑纵向长度与产量损失无明显相关性;发病期不同产量差异显著,以初花期发病产量损失最重,发病期与产量损失线性相关,可利用线性回归方程对产量损失进行预测;主茎发病部位愈近基部,枝条发病部位愈趋于主茎,单株产量损失率愈高。     

醋酸钙复盐对微生物生长的影响

应用四醋酸钙对黑曲霉、黄曲霉、米曲霉、链格孢霉等霉菌和大肠杆菌、枯草杆菌、绿脓杆菌、金黄色葡萄球菌等细菌生长影响作了研究,并将四醋酸钙与醋酸钙、苯甲酸钠对上述微生物的生长抑制作用加以比较。0.4％四醋酸钙对上述细菌的生长有明显的抑制作用。对上述霉菌生长亦有一定的抑制作用。当其与苯甲酸钠协同使用时,能更明显地抑制微生物生长。实验结果表明四醋酸钙很可能是一种安全性高、成本低的新型食品防腐剂。     

黑曲霉S4生淀粉糖化酶纯化组分的性质及其动力学常数

生淀粉糖化酶是可以水解生淀粉的葡萄糖糖化酶(Ec3.2.1.3,α-1.4-糖苷酶),它是近代工业中极为重要的酶类之一。虽然对生淀粉糖化酶可水解淀粉中所有的D-糖苷键已经清楚,由单糖逆合成寡糖的动力学研究也比较深入,但有关它对各种常用生淀粉材料的水解动力学以及各生淀粉糖化酶组分之间的相互关系的研究却很少。本研究通过在不同温度和pH下,生淀粉糖化酶的三个组分对寡糖和各种生淀粉作用的动力学常数,以及在不同pH值下它们对各生淀粉     

大熊猫消化道消化吸收区段游离面的扫描电镜观察

对大熊猫消化道的消化、吸收区段作扫描电镜观察后表明：（１）胃粘膜上皮细胞排列疏松,细胞表面具微绒毛,（２）十二指肠绒毛呈指状,表面凸凹不乎;绒毛表面具丰富的微绒毛,微绒毛表面粗糙,末端膨大。（３）直肠段具丰富的绒毛结构,表面不平滑,具颗粒状物质,但无微绒毛存在：直肠腺丰富。     

双裂解法提取血清HBV DNA技术的建立

为进行ＰＣＲ实验建立了一种两步裂解血清ＨＢＶ提取其ＤＮＡ的方法。该法首先用ＳＤＳ及蛋白酶消化裂解病毒,随后再进行碱裂解,操作较酚提取法大为简化,而ＰＣＲ实验进行的两法的灵敏性比较,说明双裂解法至少不低于酚法。     

Nonlethal sec71-1 and sec72-1 mutations eliminate proteins associated with the Sec63p-BiP complex from S. cerevisiae.

The sec71-1 and sec72-1 mutations were identified by a genetic assay that monitored membrane protein integration into the endoplasmic reticulum (ER) membrane of the yeast Saccharomyces cerevisiae. The mutations inhibited integration of various chimeric membrane proteins and translocation of a subset of water soluble proteins. In this paper we show that SEC71 encodes the 31.5-kDa transmembrane glycoprotein (p31.5) and SEC72 encodes the 23-kDa protein (p23) of the Sec63p-BiP complex. SEC71 is therefore identical to SEC66 (HSS1), which was previously shown to encode p31.5. DNA sequence analyses reveal that sec71-1 cells contain a nonsense mutation that removes approximately two-thirds of the cytoplasmic C-terminal domain of p31.5. The sec72-1 mutation shifts the reading frame of the gene encoding p23. Unexpectedly, the sec71-1 mutant lacks p31.5 and p23. Neither mutation is lethal, although sec71-1 cells exhibit a growth defect at 37 degrees C. These results show that p31.5 and p23 are important for the trafficking of a subset of proteins to the ER membrane.     

论晚二叠世的“克氏蛤”

系统评述目前世界上出现的晚二叠世克氏蛤类标本,并描述采自贵州,云南藏东的有关化石１属３种。其中１新属１新种,右壳足丝凹口特征在克氏蛤类研究中具有重要意义。晚二叠世的Ｃｌａｒａｉｏｉｄｅｓ ｇｅｎ．ｎｏｖ．以足丝凹口内端明显向腹方或后腹方扩展膨大成足丝凹湾而不同于早三叠世的Ｃｌａｒａｉａ．后者似由前者演化而来,它们在二叠－三叠系界线附近发生替代,可能与古生代末的绝灭事件相关。
