Lactoferrin (LTF), a multifunctional glycoprotein of the transferrin family mainly found in exotic secretions in mammals, is an important defense molecule against not only microbial invasion but also tumors. It folds into two globular domains (N- and C-lobes) each containing an iron-binding site. The cationic antimicrobial peptide in N-lobe is known to exert anti-tumor effect via a non-receptor-mediated pathway. However, whether LTF C-lobe also contributes to its anti-tumor activity remains to be investigated. In this study, a human LTF fragment (amino acid residues 343–682) covering the C-lobe was expressed with a histidine tag in E. coli and the purified polypeptide refolded through a series of buffer changing procedure. The resultant recombinant protein caused significant growth arrest of breast carcinoma cells MDA-MB-231 in a dose- and time-dependent manner, evidently via induction of apoptosis of the cell. Our data suggest a positive role for the C-lobe of human LTF in controlling tumors in vitro. 相似文献

Virus particles of approximately 740–760 nm in length and 13 nm in diameter were observed from a diseased Nicotiana tabacum (tobacco) plant in Sichuan Province, China. The complete genomic sequence of the virus isolate XC1 was determined to contain 9659 nucleotides without 3′ terminal poly(A) tail. XC1 has a genome typical of members of the genus Potyvirus, encoding a large polyprotein of 3075 amino acids. Putative proteolytic cleavage sites and a number of well characterized functional motifs were identified by sequence comparisons with those of known potyviruses. Sequence comparison revealed that XC1 shared the highest level of nucleotide sequence identity (76.5%) with Wild tomato mosaic virus (WTMV). Phylogenetic analysis showed that XC1 was closely related to the WTMV Guangdong isolate with an identity of 94.3% between CP gene sequence of the two viruses. We thus named XC1 WTMV‐XC‐1 as a novel isolate of WTMV. The full sequence of WTMV‐XC‐1 may serve as a basis for future investigations on the gene diversity of WTMV. 相似文献

In plants, the first line of defense against abiotic stress is in their roots. If the soil surrounding the plant root is healthy and biologically diverse, the plant will have a higher chance to survive in stressful conditions. Different plant species have unique adaptations when exposed to a variety of abiotic stress conditions. None of the responses are identical, even though plants have become adapted to the exact same environment. Mangrove plants have developed complex morphological, anatomical, physiological, and molecular adaptations allowing survival and success in their high-stress habitat. This review briefly depicts adaptive strategies of mangrove roots with respect to anatomy, physiology, biochemistry and also the major advances recently made at the genetic and genomic levels. Results drawn from the different studies on mangrove roots have further indicated that specific patterns of gene expression might contribute to adaptive evolution of mangroves under high salinity. We also review crucial ecological contributions provided by mangrove root communities to the ecosystem including marine fauna.1000.
为了解毛竹(Phyllostachys edulis)种下类型的遗传关系,对花龟竹(P.edulis‘Mira’)、斑毛竹(‘Porphyrosticta’)、安吉锦毛竹(‘Anjiensis’)、麻衣竹(‘Exaurita’)、黄皮毛竹(‘Holochrysa’)、绿槽龟甲竹(‘Lücaoguijiazhu’)、孝丰紫筋毛竹(‘Purpureosulcata’)等7个新近发表的种下类型进行了AFLP分子标记分析。结果表明,采用筛选的8对引物组合共扩增了1728条条带,多态性条带百分率达98.77%,检测到多态性位点881个。Nei’s遗传多样性指数(H)为0.2736,Shannon信息指数(I)为0.4291。这丰富了毛竹所有23个种下类型的分子亲缘关系研究数据。 相似文献
