High efficiency, long-term clinical expression of cottontail rabbit papillomavirus (CRPV) DNA in rabbit skin following particle-mediated DNA transfer.

The ability of skin to support long lasting expression of genes delivered with a particle-mediated system was evaluated in rabbits inoculated with cottontail rabbit papillomavirus (CRPV) DNA. The optimal delivery force for maximal gene expression in rabbit skin was determined in transient beta-galactosidase assays. Forty-five sites in four rabbits were then inoculated at 350-400 p.s.i. with CRPV DNA. All sites (100%) formed papillomas with multiple papillomas at most sites. These results support the feasibility of using a particle-mediated delivery system for gene therapy and suggest that some papillomavirus features, such an origin of replication, may be well suited for use in vectors to target long term expression to skin.     

豌豆外源凝集素基因的克隆及序列分析

从豌豆幼叶分离基因组ＤＮＡ,设计特异引物,用聚合酶链式反应方法扩增出豌豆外源凝集素基因并克隆到Ｅ．ｃｏｌｉ质粒ｐＢｌｕｅｓｃｒｉｐｔＳＫ（＋）的ＥｃｏＲＶ位点。进一步亚克隆至ｐＵＣ１９。序列分析表明,克隆到的片段大小为８３２ｂｐ,包含了豌豆外源凝集素基因完整的编码序列。该基因无内含子,同报道的已知序列相比,其核苷酸序列及推测的氨基酸序列的同源率分别为９９．６％和９８．９％。     

华南胡椒化学成分的研究

Visualization of Golgia apparatus as an intracellular calcium store by laser scanning confocal microscope

Using laser scanning confocal microscopy,we have found that the in cells loaded with fluo-3/AM,highest intracellular Ca^2 in the perinuclear region is associated with the Golgi apparatus.The spatiotemporal subcellular distribution of Ca^2 in living human fibroblasts exposing to calcium-free medium in response to agonists has been investigated.PDGF,which releases Ca^2 from intracellular stores by inositol(1,4,5)-trisphosphate pathway ,produced a biphasic transient rise in intracellular calcium.The initial rise was resulted from a direct release of calcium from the golgi apparatus.Calcium could be also released from and reaccumulated into the Golgi apparatus by the stimulation of thapsigargin,an inhibitor of the Ca^2 transport ATPase of intracellular calcium store,Permeablizing the plasma membrane by 10μM digitonin resulted in the calcium release from the Golgi apparatus and depletion of the internal calcium store.These results suggest that the Golgi apparatus plays a role in Ca^2 regulation in signal transduction.     

新的生物杀虫剂螨虫素对棉花、蔬菜害虫的毒力评价及应用技术研究

本文通过生物杀虫剂螨虫素对棉花、蔬菜害虫的毒力评价及田间应用研究表明,它对棉朱砂叶螨和菜青虫两种害虫杀伤力最强,其中LC90分别为0．078ppm和0．013ppm。兼有胃毒、触杀作用,残效期较长,分别达13天和9天。田间小区试验认为防治菜青虫和茄朱砂叶螨以2ppm浓度为宜。防治适期为卵孵化初期或低龄幼(若)虫（螨)期,其防效能达90%以上。该药特点用药量低、效果好、无公害、值得推广。     

玉米根冠细胞的脱落及其对根与根际关系的影响

以光学与电子显微镜技术观察了玉米幼苗根冠细胞脱落进程与胞间关系的变化。发现随着外沿细胞的脱离,固有的胞间连丝渐次伸展、孔径扩展,进而断裂为半连丝。半连丝内侧端仍与质膜保持固有的连接;外侧端向壁外伸展,直接与根际介质沟通,从而在根冠共质体与根际质外体之间建立了动态性的单向共质联系。以成列囊泡在半连丝中存在与由断口端溢出、半连丝呈现活跃ＡＴＰ酶活性、外源不透膜示踪物向胞内原生质掺入以及原生质的穿壁等实例证明,半连丝仍具生理活性,可作为根冠共质体与根际介质间物质与信息交流的直接通道。     

精子介导鱼类基因转移和聚合酶链反应检测技术

金鱼精子与美洲大绵ｗｅｉ的抗冻蛋白基因一起保温３０分钟后,再与卵子受精,共获得１４５尾成鱼和若干胚胎。从胚胎和成鱼中提取ＤＮＡ经聚合酶链反应（ＰＣＲ法）扩增和Ｓｏｕｔｈｅｒｎ ｂｌｏｔ分子杂交表明,外源的抗冻蛋白基因进入了部分受体鱼的染色体组内。测定了４５尾一年龄实验鱼中,有１２尾显示出明确的杂交带,阳性率为２６％。
