以异源四倍体体细胞杂种为父本杂交培育三倍体柑桔植株的研究

以 3个柑桔原生质体融合而来的四倍体体细胞杂种为父本 ,与二倍体单胚性种柚子 (Citrusgrandis)以及单多胚混合型品种“华农本地早”桔 (C.reticulata)有性杂交 ,授粉后 90 d,发现种子干瘪 ,大部分种子的胚败育。将干瘪种子在 MT附加 1mg/L GA3 或 50 0 mg/L麦芽浸出物的培养基中 ,经培养抢救 ,有 2 5.6%的种子萌发成苗或继续进行胚的生长 ,后者进一步诱导能形成丛芽 ,经试管嫁接或诱导生根形成完整植株。共获得 6个组合 73棵完整植株 ,染色体数检查表明 ,2 0株为三倍体 (2 n=3x=2 7) ,32株为二倍体 (2 n=2 x=18) ,8株为非整倍体 ,其它 13株还有待于进一步检查。     

Ring- opening polymerization of ɛ - Caprolactone by bacterial protease

Summary The ring- opening polymerization of - caprolactone catalyzed by a neutral protease from Bacillus subtilis has been studied in acetone, heptane solvents or in the absence of the solvent at 50°C. The lower oligomeric product containing mainly cyclic pentamer and tetramer was identified after 10 days in heptane.     

Combinatorial interplay of promoter elements constitutes the minimal determinants for light and developmental control of gene expression in Arabidopsis.

Higher plants are able to integrate environmental and endogenous signals to regulate gene expression for optimal development. To define the minimal sequence requirement sufficient to integrate light and developmental signals in controlling promoter activity, we carried out a systematic analysis of the roles of four well-conserved 'light-responsive elements (LREs)' common to many nuclear-encoded photosynthetic genes. A gain-of-function assay using basal promoter-reporter fusions in stable transgenic Arabidopsis was employed to demonstrate that pairwise combinations of the LREs, but not the individual elements alone, can confer light-inducible expression to the reporter gene independently of the basal promoter context and the light-triggered morphological changes. The activity of the synthetic promoters with the paired LREs can be modulated at least by the phytochrome system. Further, those synthetic light-regulated promoters confer a photosynthetic cell-specific expression pattern and respond to the chloroplast development state. Our data suggest that distinct combinatorial interactions of LREs can serve as minimal autonomous promoter determinants which integrate light and developmental signals and modulate promoter activity.     

腥黑粉菌属一新种

本文报道了从意大利进口兔尾草(Lagurus ovatus L.)上截获的腥黑粉菌属一新种——兔尾草腥黑粉菌(Tilletia laguri G.M.Zhang,G.X.Lin & J.R.Deng),同时讨论了该种与近似种的区别,文中对其形态特征有拉丁文和汉文描述,并附有图片。主模式标本保存在深圳动植物检疫局真菌标本室,等模式标本分别保存在德国蒂宾根大学范基黑粉菌标本馆(HUV)、中国科学院微生物研究所真菌标本室和华南农业大学植保系真菌标本室。     

噬菌体φHAU3的cos位点及其与寄主相互作用的功能位点attP和attB的定位

ＨＡＵ３是寄主范围很广的放线菌噬菌体。Ｓｏｕｔｈｅｒｎ杂交实验表明,ＨＡＵ３可以整合到吸水链霉菌应城变种１０－２２和变铅青链霉菌６６的突变体ＺＸ１的染色体中,形成溶原,其溶原菌自发释放ＨＡＵ３,不受热激和紫外线照射的诱导。通过比较ＨＡＵ３衍生噬粒ｐＩＪ８３００的ＤＮＡ酶切片段在加热前、后电泳带谱的区别,将ＨＡＵ３的ｃｏｓ位点在ｐＩＪ８３００的图谱上得到了定位。还利用Ｓｏｕｔｈｅｒｎ杂交的方法定位了ＨＡＵ３与宿主形成溶原时附着位点（ａｔｔＰ）,并利用脉冲电泳技术定位了在变铅青链霉菌ＺＸ７和吸水链霉菌应城变种１０－２２中形成溶原的附着位点（ａｔｔＢ）。这些信息均有利于以ＨＡＵ３为基础的载体的发展和优化。     

Repeated polyketide synthase modules involved in the biosynthesis of a heptaene macrolide by Streptomyces sp. FR-008

Genes for biosynthesis of a Streptomyces sp. FR-008 heptaene macrolide antibiotic with antifungal and mosquito larvicidal activity were cloned in Escherichia coli using heterologous DNA probes. The cloned genes were implicated in heptaene biosynthiesis by gene replacement. The FR-008 antibiotic contains a 38-membered, poiyketide-derived macrolide ring. Southern hybridization using probes encoding domains of the type i modular erythromycin polyketide synthase (PKS) showed that the Streptomyces sp. FR-008 PKS gene cluster contains repeated sequences spanning c. 105 kb of contiguous DNA; assuming c. 5 kb for each PKS module, this is in striking agreement with the expectation for the 21-step condensation process required for synthesis of the FR-008 carbon chain. The methods developed for transformation and gene replacement in Streptomyces sp. FR-008 make it possible to genetically manipulate polyene macrolide production, and may later lead to the biosynthesis of novel polyene macrolides.     

Familial unbalanced translocation t(8;15)(p23.3;q11) with uniparental disomy in Angelman syndrome

A 29-year-old male with Angelman syndrome and an unbalanced reciprocal translocation, 45,XY,-8,-15,+der(8),t(8;15)(p23.3;q11)pat, was evaluated with DNA studies. These showed the underlying mechanism to be paternal uniparental disomy. This is the second case reported of Angelman syndrome that has resulted from a familial unbalanced reciprocal translocation.     

橐吾属的起源、演化与地理分布

橐吾属Ligularia Cass．是菊科千里光族款冬亚族的一个大属。在款冬亚族中本属与大吾风草属 Farfugium Lindl．亲缘关系最近,但进化程度较高。本属包括6组,11系129种。所有种类均分布在 亚洲,仅2种扩散至欧洲。在东亚地区有119种,占该属总种数的96％。高度集中在横断山区的有4组、 6系67种,其中61种为特有种,占该属总组数的66％,总系数的54．5％,总种数的52％。这个事实 表明了横断山区是该属的多度中心和多样化中心。通过性状分析,伞房组伞房系Sect．Corymbosae, Ser．Calthifoliae叶肾形,具掌状叶脉,头状花序大而少,排列呈伞房状,总苞半球形,被认为是该属的 原始类群。原始种齿叶橐吾L. dentata和鹿蹄橐吾L.hodgsonii的分布区从我国四川东部经过湖北、湖 南、安徽、福建等省至日本。这个分布格局与近缘属大吴风草属Farfugium一致。 根据共同起源原理,这两个属的祖先极有可能就发生在这一地区。因此我们推测东亚地区从中国四 川东部至日本这一地区是本属的发源地,然而根据地质历史和现代分布,作者认为中国中部(包括四川 东部)是本属的初始起源地。该属起源后,基本上沿亚洲南缘的山地扩散,少数种类向东北至亚洲东北部。本属起源时间至少不晚于中白垩纪。
