The recent explosive adaptive radiation of cichlids in the great lakes of
Africa has attracted the attention of both morphologists and molecular
biologists. To decipher the phylogenetic relationships among the various
taxa within the family Cichlidae is a prerequisite for answering some
fundamental questions about the nature of the speciation process. In the
present study, we used the random amplification of polymorphic DNA (RAPD)
technique to obtain sequence differences between selected cichlid species.
We then designed specific primers based on these sequences and used them to
amplify template DNA from a large number of species by the polymerase chain
reaction (PCR). We sequenced the amplified products and searched the
sequences for indels and shared substitutions. We identified a number of
such characters at three loci-- DXTU1, DXTU2, and DXTU3--and used them for
phylogenetic and cladistic analysis of the relationships among the various
cichlid groups. Our studies assign an outgroup position to Neotropical
cichlids in relation to African cichlids, provide evidence for a
sister-group relationship of tilapiines to the haplochromines, group
Cyphotilapia frontosa with the lamprologines of Lake Tanganyika, place
Astatoreochromis alluaudi to an outgroup position with respect to other
haplochromines of Lakes Victoria and Malawi, and provide additional support
for the monophyly of the remaining Lake Victoria haplochromines and the
Lake Malawi haplochromines. The described approach holds great promise for
further resolution of cichlid phylogeny.

A procedure for regeneration of somatic embryogenesis from witloof chicory leaf has been developed. Explants were taken from the distal third part of leaf vein and cultured on Murashige and Skoog medium (MS) containing 100 mg/1 casein hydrolysate 1.3 M 2,4-D, and 1.3 M kinetin. A pale yellowisl nodular callus was formed after 4 weeks which was maintained in the same medium for 8–12 weeks with one change to a fresh medium every 4 weeks. Callus was then suspended in the same medium without agar for 4–6 weeks with one change to a fresh medium every 2 weeks. Embryo-like structures appeared upon transfer to liquid MS containing 1.8 M benzyladenine. Embryo germination was accomplished in half strength MS medium with or without 1 g/l activated charcoal.Abbreviations AC

Abstract The mating behavior of cat flea, Ctenocepholides felis (Bouche) was studied on an artificial feeding device. Male and female can mate repeatedly with same partner or Merent ones. In the situation of male: female ratio of 1 :5, each mating lasted an average of 6.6 min, with a mean interval between matings at 2.5 min., compared to 11. 1 min and 12.1 min respectively in a cell with 5 males and 1 female. As many as 48 mating events were observed for one male during an 8 h period. One female mated 27 times in 7 h with 5 males in the same cell. Newly emerged males and females can not mate before blood meal and about 24 h blood feeding is rewuired for successful mating. Newly emerged males can not mate with fed females (fed for 48 h), but fed males can mate with newly emerged females who are feeding the blood. Significantly more male contacts and male-male mating attempts were observed after the paper treated with female extract was introduced into the cell. The paper contacts and mating attempts were 16.75–32.25 times and 15.75–31.38 times, respectively, on average during a period of 20 min when different doses (FE) of extract were provided. 相似文献

Cabbage looper hemolymph induced rapid germination and outgrowth of spores of Bacillus popilliae. Spores germinated within sporangia but outgrowth occurred from free released spores as well as from spores retained in sporangia. With 37°C, an alkaline pH, and tyrosinase, outgrowth resulted in 1 hr. Of six strains of milky disease bacteria tested, hemolymph mediated germination and outgrowth of only those which are infective perorally for European chafer larvae, indicating a potential use as a screening tool to assess virulence for the chafer. 相似文献

Activity of proline dehydrogenase and pyrroline-5-carboxylate reductase was greatest after 5 and 7 days germination in green and etiolated cotyledons respectively of pumpkin (Cucurbita moschata Poir. cv. Dickinson Field). The ratio of pyrroline-5-carboxylate reductase to proline dehydrogenase activity was constant throughout germination. Both enzymes were purified 30-fold but the ratio pyrroline-5-carboxylate reductase—proline dehydrogenase activity was constant throughout purification. However, this ratio decreased with storage, especially in purified preparations. Both enzymes were stable at high temperature and the ratio pyrroline-5-carboxylate reductase—proline dehydrogenase remained unchanged on heating. Proline dehydrogenase and pyrroline-5-carboxylate reductase were inhibited by sodium bisulfite and cysteine. ATP, ADP and NADP caused inhibition of both enzymes. Proline dehydrogenase utilized NAD but not NADP. Pyrroline-5-carboxylate reductase had a 2.5-fold greater activity with NADH than NADPH. Most of the data presented suggest that proline dehydrogenase and pyrroline-5-carboxylate reductase activities occur on the same protein molecule. 相似文献

Aspergillus niger was found capable of rapidly converting about 97% of the sugar from brewery spent grain liquor to fungal mass. The yield of dry mycelium, based on the sugar consumed, was approximately 57%. Glutamate dehydrogenase from pumpkin (Cucurbita moschata Pior. cultivar Dickinson Field) cotyledons was found in both soluble and particulate fractions with the bulk of the activity in the soluble fraction. Both enzymes used NAD(H) and NADP(H) but NAD(H) was favored. The enzymes were classified as glutamate-NAD oxidoreductase, deaminating (EC 1.4.1.3). Both enzymes were heat stable, had a pH optimum for reductive amination of 8.0, and were inhibited by high concentrations of NH4+ or α-ketoglutarate. The soluble enzyme was more sensitive to NH4+ inhibition and was activated by metal ions after ammonium sulfate fractionation while the solubilized particulate enzyme was not. Inhibition by ethylenediaminetetraacetate was restored by several divalent ions and inhibition by p-hydroxymercuribenzoate was reversed by glutathione. Particulate glutamate dehydrogenase showed a greater activity with NADP. The molecular weights of the enzymes are 250,000. Separation of the enzymes by disc gel electrophoresis showed that during germination the soluble isoenzymes increased from 1 to 7 in number, while only one particulate isoenzyme was found at any time. This particulate isoenzyme was identical with one of the soluble isoenzymes. A procedure for the determination of hydrogen sulfide in fermentation broths containing up to 100 mug of SO(2) per ml is described. The method involves the sparging of H(2)S from the broth into a cadmium hydroxide absorption solution, the formation of methylene blue from the absorbed sulfide, and the measuring of this color spectrophotometrically. The use of cadmium hydroxide instead of zinc acetate, the common absorbent, substantially reduced the interference of SO(2) with the analysis. 相似文献
