Understory light is known as one of the most important environmental predictors of growth response of woody species. Hence, the primary objective of most forest management practices is optimizing that resource for understory seedlings. In this study, growth responses of Quercus castaneifolia seedlings from five different provenances from west to east of the Hyrcanian forest were investigated along their ecological gradients (from high to low precipitation). An experimental design was executed under controlled conditions at eight different irradiance levels (10, 20, 30, 40, 50, 60, 70 and 100 % full irradiance). Results showed that the irradiance is probably the most important determinant of variation in seedling characteristics. Among all investigated variables, variability in seedling size was affected significantly by provenance, while seedling morphology and their architectural response was affected by different levels of irradiance in a curvilinear manner. The biggest changes were observed at lowest irradiance levels (10–20 %) while at higher irradiance (70–100 %) the curves flatten. It was shown that, unlike at low irradiance levels, there is little capacity in seedling morphology to acclimatize with high irradiance intensity. Attaining maximal biomass varies across provenances and irradiance gradient. The highest biomass for the five provenances could be ranked as follows: 20–60 % and 50–60 % for the wetter and drier provenances, respectively. These results demonstrated that the light requirement increases from wetter to drier provenances, with a negative relationship between light requirement and precipitation regime. Different responses to irradiance levels may be the result of genetic adaptation to the ecological conditions prevailing in native habitat, especially precipitation regime.  相似文献

The excitation of surface plasmon polariton (SPP) at interface of a metal and an ambichiral sculptured thin film was theoretically investigated in the Kretschmann configuration using the transfer matrix method. The dependence of SPP modes for a P polarization plane wave on the incident angle of light and the angle of rise of nanocolumns of ambichiral dielectric medium was reported. We found that multiple SPP modes are excited at the interface of metal and ambichiral dielectric medium. The results of phase speed as a function of pitch showed only that a SPP mode can be excited at all pitches.  相似文献

Cytomegalovirus (CMV) infection represents a vital complication after Hematopoietic Stem Cell Transplantation (HSCT). We screened the entire CMV proteome to visualize the humoral target epitope-focus profile in serum after HSCT. IgG profiling from four patient groups (donor and/or recipient +/− for CMV) was performed at 6, 12 and 24 months after HSCT using microarray slides containing 17174 of 15mer-peptides overlapping by 4 aa covering 214 proteins from CMV. Data were analyzed using maSigPro, PAM and the ‘exclusive recognition analysis (ERA)’ to identify unique CMV epitope responses for each patient group. The ‘exclusive recognition analysis’ of serum epitope patterns segregated best 12 months after HSCT for the D+/R+ group (versus D−/R−). Epitopes were derived from UL123 (IE1), UL99 (pp28), UL32 (pp150), this changed at 24 months to 2 strongly recognized peptides provided from UL123 and UL100. Strongly (IgG) recognized CMV targets elicited also robust cytokine production in T-cells from patients after HSCT defined by intracellular cytokine staining (IL-2, TNF, IFN and IL-17). High-content peptide microarrays allow epitope profiling of entire viral proteomes; this approach can be useful to map relevant targets for diagnostics and therapy in patients with well defined clinical endpoints. Peptide microarray analysis visualizes the breadth of B-cell immune reconstitution after HSCT and provides a useful tool to gauge immune reconstitution.  相似文献

The present work examined morphological and physiological aspects of osmoregulation in some differently adapted populations of Typha domingensis. Six populations of T. domingensis were grown in waterlogged conditions under four levels of nickel (0, 50, 100 and 1500 mg/kg of Ni). The Ni tolerance was explored on the basis of different characteristics of shoots. Along with the increasing level of Ni stress, all populations showed an increment of proline and glycinebetaine content, low photosynthetic and transpiration rates and decreased chlorophyll content. Analyzed morphological parameters revealed possible adaptations to deal with Ni stress. The less tolerant Sahianwala population produced more aerenchyma, less accumulation of osmolytes and reduced shoot fresh weight under Ni stress than the tolerant ones. Our results suggested that T. domingensis has the capacity to survive under Ni stress and could be used for the absorption of toxic metal ions from industrially polluted water.  相似文献

The chromosome complements of human/mouse hybrid cell lines of mouse 3T3-4E and RAG parentage have been analyzed using chromosome banding methods. Three lines that were susceptible to lytic infection with poliovirus contained eleven to seventeen human chromosomes, including chromosome 19. Polio-resistant sublines of these contained no chromosome 19 and showed no other consistent change in the complement of human chromosomes. Human chromosome 19 therefore is essential for polio-sensitivity. Since polio sensitivity was correlated with receptor activity in these lines, we conclude that chromosome 19 carries the structural gene for the poliovirus receptor. Sensitivity to echo-7 and Rhino-1A viruses could not be related to the presence of a specific human chromosome.  相似文献

A cultured soybean cell line, SB-1 was used to evaluate the initial interaction between the soybean cells andRhizobium japonicum. Co-culturing ofR. japonicum with SB-1 cells in suspension resulted in strain-specific polar attachment. This attachment can be inhibited by galactose and antibodies raised against seed soybean agglutinin (SBA). A lectin was purified from SB-1 cells which shares properties with SBA in terms of immunological reactivity, sugar binding activity, polypeptide molecular weight and peptide maps. When the SB-1 cells were co-cultured withR. japonicum for three weeks in solid agar medium, histological staining revealed bacterial penetration into certain SB-1 cells. Furthermore, there were focal regions of cells with prominent nuclei representing actively proliferating regions. These observations are analogous to that ofin vivo nodule initiation in soybean roots.  相似文献

Summary Malignant epithelial tumor cells were isolated and cultured from ten human mammary specimens of cancerous origin. The 260F9 monoclonal antibody (MAB) bound to frozen sections of all of the human breast tumors tested and to primary cultured cells from the tumors. Cultured cells from all ten breast tumors were sensitive to the clonal inhibitory effects of immunotoxin 260F9 MAB-recombinant ricin A chain. At an immunotoxin concentration of 200 ng/ml (about 1 nM), inhibition of colony formation was >99% for all ten tumors.  相似文献

Prostate cancer (PCa) is one of the most prevalent non-drug delivery system cutaneous malignancies. Undoubtedly, introducing novel treatment options to achieve higher therapeutic index will be worthwhile. In this study, we report for the first time, a novel targeted self-assembled based on PEG-PLA nanoparticles (PEG-PLA NPs) containing galbanic acid (GBA) and docetaxel, which was targeted using ((S)-2-(3-((S)-5-amino-1-carboxypentyl) ureido) pentanedioic acid (ACUPA), a small molecule inhibitor targeting prostate-specific membrane antigen (PSMA), in prostate cancer cell line. The prepared NPs were characterized by different analytical methods. The MTT assay was used to compare the anti-proliferation of drugs-loaded PEG-PLA NPs and ACUPA-PEG-PLA against LNCaP (PSMA+) and PC3 (PSMA) cells. PEG-PLA NPs with an average size of 130–140 nm had an enhanced release of GBA and docetaxel at pH 5.5 compared with pH 7.5. Spectrofluorometric analysis suggested that ACUPA-modified PEG-PLA could effectively enhance the drug uptake in PSMA+ prostate cancer cells. Cytotoxicity studies showed that the targeted NPs loaded with different concentrations of GBA and fixed concentration of docetaxel (4 nM) have shown higher toxicity (IC50 30 ± 3 µM) than both free GBA (80 ± 4.5 µM) and nontargeted NPs (IC50 40 ± 4.6 µM) in LNCaP cells. Collectively, these findings suggest that ACUPA-conjugated PEG-PLA nanosystem containing GBA and docetaxel is a viable delivery carrier for various cancer-targeting PSMA that suffer from short circulation half-life and limited therapeutic efficacy.  相似文献
