A rational attempt to prepare FmocHis(piTrt)OH regiospecifically gave in fact the well-known tau-trityl isomer, and experiments with model systems indicate that the prospects for access to pi-trityl histidine derivatives, which would be of great value for the racemization-free synthesis of histidine-containing peptides, are poor.     

NEUROCHEMISTRY OF THE SPINAL CORD IN BRITISH SADDLEBACK PIGLETS AFFECTED WITH CONGENITAL TREMOR, TYPE A-IV, A SECOND FORM OF HEREDITARY CEREBROSPINAL HYPOMYELINOGENESIS

Congenital tremor, type A-IV is an inherited abnormality of British Saddleback piglets characterized by the presence of poorly myelinated axons throughout the CNS. This is reflected by a general lowering of the spinal cord lipid content to about half control values. In particular, cerebroside and plasmalogen levels are markedly reduced. Changes in the fatty acid composition of total lipid extracts and two isolated cerebroside fractions suggest that there is a metabolic defect in which fatty acid chain elongation, desaturation and hydroxylation are sub-optimal. Cholesterol esters accumulate, make up about 30 per cent of total sterols and contain more than 90 per cent of C16 and C18 fatty acids. These may have arisen through the breakdown of cerebrosides containing abnormal proportions of these acids. Slightly increased levels of acid hydrolase activity appear to confirm that abnormal or immature myelin is disposed of by enhanced macrophagic activity. Differences between this and type A-III, the sex-linked inherited form of cerebrospinal hypomyelinogenesis are discussed and comparisons made with two forms of murine leuko-dystrophy.     

The destruction of spores of Bacillus subtilis by the combined effects of hydrogen peroxide and ultraviolet light

Ultraviolet light irradiation of bacterial spores in the presence of hydrogen peroxide has been shown to produce synergistic kills when compared with ultraviolet light (u.v.) and hydrogen peroxide used sequentially. This use in combination has been patented for the commercial sterilization of packaging before filling with UHT-processed products. Previous results have shown that lamps producing u.v. light with a maximum output at about 254 nm were extremely effective. Results obtained using a Synchrotron radiation source to produce a narrow band of irradiation now shows that the greatest kill of spores of Bacillus subtilis in the presence of hydrogen peroxide is obtained with radiation at ˜270 nm. Such results suggest that the action of the u.v. light is not directly on the spore DNA but may be related to the production of free hydroxyl radicals from hydrogen peroxide.     

1H NMR studies on bovine cyclophilin: preliminary structural characterization of this specific cyclosporin A binding protein

High-field 1H NMR spectroscopy has been used to study the conformation of the cytosolic cyclosporin A binding protein cyclophilin. For the drug-free form of cyclophilin, spectral editing methods in conjunction with a pH titration were used to identify all four His residues present in the protein, and two-dimensional COSY and RELAY spectroscopy was used to elucidate the scalar connectivities in the aromatic and upfield methyl regions of the spectrum. From these scalar connectivities, it was possible to distinguish between inter- and intraresidue dipolar interactions within the aromatic and upfield methyl regions of cyclophilin in the NOESY spectrum. The results of this analysis showed extensive interresidue cross-relaxation among and between these latter spectral regions indicative of the proximal relationships of several of these residues and the presence of a hydrophobic core within cyclophilin.     

A simple test for macromolecular heterogeneity in the analytical ultracentrifuge

A simple check for the presence of heterogeneity in a macromolecular system is proposed, employing comparison of Rayleigh sedimentation-equilibrium patterns for two solutions of the same fringe concentration but differing absolute concentrations. The method is illustrated by application to a bronchial glycoprotein from a cystic-fibrosis patient.
