Evolutionary adaptation affects demographic resilience to climate change but few studies have attempted to project changes in selective pressures or quantify impacts of trait responses on population dynamics and extinction risk. We used a novel individual-based model to explore potential evolutionary changes in migration timing and the consequences for population persistence in sockeye salmon Oncorhynchus nerka in the Fraser River, Canada, under scenarios of future climate warming. Adult sockeye salmon are highly sensitive to increases in water temperature during their arduous upriver migration, raising concerns about the fate of these ecologically, culturally, and commercially important fish in a warmer future. Our results suggest that evolution of upriver migration timing could allow these salmon to avoid increasingly frequent stressful temperatures, with the odds of population persistence increasing in proportion to the trait heritability and phenotypic variance. With a simulated 2°C increase in average summer river temperatures by 2100, adult migration timing from the ocean to the river advanced by ~10 days when the heritability was 0.5, while the risk of quasi-extinction was only 17% of that faced by populations with zero evolutionary potential (i.e., heritability fixed at zero). The rates of evolution required to maintain persistence under simulated scenarios of moderate to rapid warming are plausible based on estimated heritabilities and rates of microevolution of timing traits in salmon and related species, although further empirical work is required to assess potential genetic and ecophysiological constraints on phenological adaptation. These results highlight the benefits to salmon management of maintaining evolutionary potential within populations, in addition to conserving key habitats and minimizing additional stressors where possible, as a means to build resilience to ongoing climate change. More generally, they demonstrate the importance and feasibility of considering evolutionary processes, in addition to ecology and demography, when projecting population responses to environmental change. 相似文献

Nef proteins of primate lentiviruses promote viral replication, virion infectivity, and evasion of antiviral immune responses by modulating signal transduction pathways and downregulating expression of receptors at the cell surface that are important for efficient antigen-specific responses, such as CD4, CD28, T-cell antigen receptor, and class I and class II major histocompatibility complex. Here we show that Nef proteins from diverse groups of primate lentiviruses which do not require the chemokine receptor CXCR4 for entry into target cells strongly downmodulate the cell surface expression of CXCR4. In contrast, all human immunodeficiency virus type 1 (HIV-1) and the majority of HIV-2 Nef proteins tested did not have such strong effects. SIVmac239 Nef strongly inhibited lymphocyte migration to CXCR4 ligand, the chemokine stromal derived factor 1 (SDF-1). SIVmac239 Nef downregulated CXCR4 by accelerating the rate of its endocytosis. Downmodulation of CXCR4 was abolished by mutations that disrupt the constitutively strong AP-2 clathrin adaptor binding element located in the N-terminal region of the Nef molecule, suggesting that Nef accelerates CXCR4 endocytosis via an AP-2-dependent pathway. Together, these results point to CXCR4 as playing an important role in simian immunodeficiency virus and possibly also HIV-2 persistence in vivo that is unrelated to viral entry into target cells. We speculate that Nef targets CXCR4 to disrupt ordered trafficking of infected leukocytes between local microenvironments in order to facilitate their dissemination and/or impair the antiviral immune response. 相似文献

High densities of habitat modifiers can dramatically alter the structure of ecosystems. Whereas spawning sockeye salmon (Oncorhynchus nerka) dig nests that cover over 2 m2 and are at least 20 cm deep, and can spawn at high densities, relatively little attention has been devoted to investigating the impacts of this disturbance. We hypothesized that this temporally and spatially predictable bioturbation has large impacts on the coastal aquatic habitats used by sockeye. We experimentally investigated the impacts of disturbance caused by spawning sockeye in two streams and two lakes in Alaska by excluding salmon from 2.25 m2 plots where they traditionally spawn. We sampled exclusions and control plots before, during, and after spawning. During sockeye spawning, fine sediment accumulated in areas where sockeye were excluded from spawning. In addition, sockeye spawning significantly decreased algal biomass by 80% compared to exclusion plots. We found mixed effects of spawning on the invertebrate assemblage. Tricladida and Chironomidae densities increased by 3x in exclusion plots relative to control plots in one creek site. However, for most taxa and sites, invertebrate densities declined substantially as spawning progressed, regardless of experimental treatment. DNA ligase IV (LigIV) deficiency was identified as the molecular basis for a severe form of combined immunodeficiency in two microcephalic siblings with cellular radiosensitivity. In one patient the diagnosis was made directly after birth, allowing analysis of the role of LigIV in the development of specific immune cells. Absolute numbers of B cells were reduced 100-fold and alphabeta T cells 10-fold, whereas gammadelta T cells were normal. Spectratyping of all three cell populations showed a diverse repertoire, but sequencing of IgH V(D)J junctions revealed shorter CDR3 regions due to more extensive nucleotide deletions among D and J elements and fewer N nucleotide insertions. Clonal restriction of IgG-expressing, but not IgM-expressing, B cells and the lack of primary and secondary lymph node follicles indicated impaired class switch recombination. Observations in the older sibling showed that this rudimentary immune system was able to mount specific responses to infection. However, partial Ab responses and extensive amplification of gammadelta T cells could not prevent a life-threatening course of viral and bacterial infections, the development of an EBV-induced lymphoma, and immune dysregulation reflected by severe autoimmune cytopenia. Non-native brook trout (Salvelinus fontinalis) were eradicated from alpine Bighorn Lake, Alberta, Canada, to test whether strong cascading trophic interactions (CTI) can
occur in oligotrophic, high seston C:P, species-poor lakes. Fishless alpine Pipit Lake was used as a reference ecosystem.
Bighorn Lake zooplankton biomass increased from 0.14:1 relative to Pipit Lake before fish removal began in 1997 to 0.6:1 afterwards
due to an increase in the abundance of adult cyclopoid copepods beginning in 1997 and the reappearance of Daphnia middendorffiana in 1998. Following the reappearance of Daphnia, Bighorn Lake total phytoplankton biomass fell from 64:1 relative to Pipit Lake to 0.9:1. Over the same periods Bighorn Lake:Pipit
Lake chlorophyll-a ratios declined from 2.4:1 to 1.6:1, although the decrease was not statistically significant. Mid-summer Secchi disc depth
in Bighorn Lake increased from 3.1 m before manipulation to 9.2 m, the maximum depth of the lake, in 2001 and 2002. Increased
transparency was most likely due to increased filtration of suspended inorganic particles from the water column by higher
abundances of large zooplankton. Post-manipulation increases in dissolved inorganic nitrogen (DIN), DIN:total dissolved phosphorus
(TDP) ratio and declines in TDP in Bighorn Lake were not attributable to ecosystem manipulation, similar changes were observed
in reference Pipit Lake. We conclude that strong pelagic CTI, expressed as change in total phytoplankton biomass and largely
mediated by Daphnia, can occur in oligotrophic, high seston C:P, species-poor ecosystems. However, strong CTI responses in phytoplankton biomass

Natural communities are highly complex and dynamic over time, with populations structured by numerous abiotic and biotic forces acting through direct and indirect pathways. Multispecies Autoregressive (MAR) modeling can be used to partition effects of variables that are interrelated and temporally autocorrelated in time series from natural systems. Here we address two main questions in applying MAR models to community time series. First, what is the effect of observation scale on interpretation of community dynamics? We used a 10‐year weekly planktonic time series from Lake Washington to construct multiple “biweekly” and “monthly” data sets, and compared resulting community interaction models. Direct abiotic effects and intraspecific autocorrelation were apparent using all data sets. Biotic interactions were more apparent using biweekly and monthly data, indicating that time lags longer than one week were necessary to detect numerical response to interspecific interactions. Second, we examined effects of dropping the winter months from our analyses to simulate the common practice of sampling only during the “growing season” in long‐term ecological studies. We found that biotic interactions remained similarly characterized in models using only non‐winter months, but that the importance of seasonal physical factors nearly disappeared in non‐winter models. Exclusion of winter data in sampling designs may therefore allow us to characterize biotic interactions, although it may not help us understand populations’ relationships to seasonal abiotic variables. The models supported many previous findings from experimental and qualitative investigations of Lake Washington community interactions, implying that MARs provided plausible characterizations of community dynamics, but some previously unconsidered relationships did emerge, such as the importance of cryptomonads and picoplankton for zooplankton growth. We conclude that explicit consideration of time lags in biotic response is necessary to understand relative importance of abiotic and biotic factors, and that sampling regime can therefore strongly influence our interpretations of community dynamics. 相似文献

The European Union has introduced transmissible spongiform encephalopathy (TSE) resistance breeding programmes for several sheep breeds to cope with the genetic susceptibility to Scrapie infections. Due to the different allele frequencies among breeds, strong selection for ARR alleles is associated with a loss of genetic diversity in small populations and in larger populations with unfavourable ARR allele frequencies. To ensure maintenance of genetic diversity, an adhoc cryopreservation programme was initiated employing epididymal sperm from 109 rams representing 16 different breeds within one breeding season. Epididymal semen was chosen for this adhoc programme because time consuming training of rams for ejaculated semen collection via an artificial vagina was not possible. Prior to freezing, average sperm motility was 79.7% and acrosome integrity was 93.7%. After freezing, these levels were decreased to 60.5 and 72.8%, respectively. An insemination trial using frozen-thawed epididymal semen resulted in a lambing rate of 87.5%. Results show that this semen preservation method is robust and efficient and associated with high fertility. It may also be useful for other animal species. 相似文献
