A technique based on progressive fumigation was used to reduce soil microbial biodiversity, and the effects of such reductions upon the stability of key soil processes were measured. Mineral soil samples from a grassland were fumigated with chloroform for up to 24 h and then incubated for 5 months to allow recolonisation by surviving organisms. The diversity of cultivable and non‐cultivable bacteria, protozoa and nematodes was progressively reduced by increasing fumigation times, as was the number of trophic groups, phyla within trophic groups, and taxa within phyla. Total microbial biomass was similar within fumigated soils, but lower than for unfumigated soil. There was no direct relationship between biodiversity and function. Some broad‐scale functional parameters increased as biodiversity decreased, e.g. thymidine incorporation, growth on added nutrients, and the decomposition rate of plant residues. Other more specific parameters decreased as biodiversity decreased, e.g. nitrification, denitrification and methane oxidation. Thus specific functional parameters may be a more sensitive indicator of environmental change than general parameters. Although fumigation reduced soil microbial biodiversity, there was evidence to suggest that it selected for organisms with particular physiological characteristics. The consequences of this for interpreting biodiversity – function relationships are discussed. The stability of the resulting communities to perturbation was further examined by imposing a transient (brief heating to 40°C) or a persistent (addition of CuSO4) stress. Decomposition of grass residues was determined on three occasions after such perturbations. The soils clearly demonstrated resilience to the transient stress; decomposition rates were initially depressed by the stress and recovered over time. Resilience was reduced in the soils with decreasing biodiversity. Soils were not resilient to the persistent stress, there was no recovery in decomposition rate over time, but the soils with the highest biodiversity were more resistant to the stress than soils with impaired biodiversity. The study of functional stability under applied perturbation is a powerful means of examining the effects of biodiversity.  相似文献

The molecular mechanism via which keratinocyte differentiation assembles multiple layers of cells (stratification) is poorly understood. We describe here a novel function of the Rho family member RhoE as a regulator of epidermal morphogenesis. RhoE protein levels are specifically and transiently up-regulated upon keratinocyte differentiation. RhoE up-regulation requires the activity of Rho kinase (ROCK) I, suggesting that both RhoE and ROCKI are important during keratinocyte differentiation. RhoE overexpression results in a striking enlargement of cell size and the number of stratified cells. In contrast, RhoE depletion induces hyperproliferation and delays initiation of keratinocyte differentiation. Interestingly, up-regulation of RhoE protein is seen primarily in basal, undifferentiated cells, in which commitment to differentiation and stratification takes place. RhoE activation in basal cells negatively modulates integrin adhesion, thereby facilitating detachment from the substratum and migration to form suprabasal layers. Thus, RhoE integrates two processes essential for keratinocyte differentiation and stratification: regulation of proliferative status and integrin adhesion.  相似文献

Summary Treating soluble wastewaters anaerobically in UASB-reactors, the development of sludge-granules can be expected, provided that there are optimal growing conditions. Two types of granules were obtained, both with high specific activities and excellent settling properties. Type 1 mainly consists of short fragments of multicellular filaments of a bacterium showing much resemblance with a recently isolated acetate-degrading methanogenic bacterium (Zehnder et al, 1980). Type 2 is mainly made up by long filaments of presumably the same bacterium. The development of anaerobic bulking sludge must be prevented.  相似文献

Thermodynamic data that the reductive dechlorination of 3-chlorobenzoate is exergonic have led to the hypothesis that this reaction yields biologically useful energy. This hypothesis was tested with strain DCB-1, a dehalogenating bacterium. The organism was grown under strictly anaerobic conditions in vitamin-amended mineral medium with formate plus acetate as electron donor and 3-chlorobenzoate as electron acceptor. The cell yield increased stoichiometrically to the amount of 3-chlorobenzoate dechlorinated. No growth was observed in the absence of 3-chlorobenzoate, or when 3-chlorobenzoate was replaced by benzoate. To obtain further evidence on that energy is derived from dechlorination, 3-chlorobenzoate was added to starved cells. This amendment resulted in an increase in the ATP level of the cells at 10 nmol per mg protein versus 3 nmol per mg protein in non-amended controls. These data indicate that the reductive dehalogenation of chlorinated aromatic compounds can be coupled to a novel type of chemotrophy.  相似文献

The Gibbs free energy of formation of chlorinated aliphatic compounds was estimated with Mavrovouniotis' group contribution method. The group contribution of chlorine was estimated from the scarce data available on chlorinated aliphatics in the literature, and found to vary somewhat according to the position of chlorine in the molecule. The resulting estimates of the Gibbs free energy of formation of chlorinated aliphatic compounds indicate that both reductive dechlorination and aerobic mineralization of these compounds can yield sufficient energy to sustain microbial growth.  相似文献

New series of pyrazoles 4a – c and pyrazolopyrimidines 5a – f had been constructed. The newly synthesized compounds were assessed for their antimicrobial activity towards E. coli and P. aeruginosa (gram –ve bacteria), B. subtilis and S. aureus (gram +ve bacteria) and A. flavus and C. albicans (representative of fungi). The pyrazolylpyrimidine-2,4-dione derivative 5b is the most active candidate against B. subtilis (MIC=60 μg/mL) and P. aeruginosa (MIC=45 μg/mL). Regarding antifungal potential, compound 5f was the most effective against A. flavus (MIC=33 μg/mL). Similarly, compound 5c displayed strong antifungal activity towards C. Albicans (MIC=36 μg/mL) in reference to amphotericin B (MIC=60 μg/mL). Finally, the novel compounds had been docked inside dihydropteroate synthase (DHPS) to suggest the binding mode of these compounds.  相似文献
