The effect of the mitochondrial pyruvate transport inhibitors, α-cyanocinnamate and α-cyano-4-hydroxycinnamate, on the regulation of the pyruvate dehydrogenase multienzyme complex was investigated in the isolated perfused rat heart. Metabolic flux through pyruvate dehydrogenase was monitored by measuring 14CO2 production from [1-14C]pyruvate infused into the heart. A stepwise increase in the concentration of the inhibitor in the influent perfusate effected a stepwise reduction of the flux through the enzyme complex at all pyruvate concentrations tested. However, the magnitude of the α-cyanocinnamate-insensitive flux through pyruvate dehydrogenase increased markedly as the infused pyruvate concentration was elevated. The inhibition of pyruvate decarboxylation in the heart was nearly completely reversed following cessation of the inhibitor infusion. α-Cyanocinnamate was nearly 10 times more potent than α-cyano-4-hydroxycinnamate as an inhibitor of the flux through pyruvate dehydrogenase. Maximally inhibiting levels of α-cyano-4-hydroxycinnamate caused an increase in the ratio of the active form of pyruvate dehydrogenase to the total extractable enzyme complex from a value of 0.5 at 1 mm infused pyruvate (in the absence of the inhibitor) to a value of near unity. This result indicated that the intramitochondrial pyruvate concentration was severely depleted by the infusion of the inhibitor and that the enzyme complex was interconverted to its active form under these conditions. Removal of the inhibitor from the perfusion medium again lowered the ratio of the active/total pyruvate dehydrogenase to near its original level of 0.5 and restored the original flux through the enzyme complex indicating that mitochondrial pyruvate transport has been restored. The results of this study indicate that α-cyanocinnamate and its derivatives are effective inhibitors of pyruvate transport in the perfused heart and that carrier-mediated pyruvate transport can be an important parameter in the regulation of the activation state and the metabolic flux through the pyruvate dehydrogenase multienzyme complex in the heart.  相似文献

Since the discovery in 1971 of opiate receptors and later of the opiate-like peptides, there has been widespread interest in determining their exact localization, number and kinds, nature, and physiological and pharmacological functions. Between 1971 and 1978, vast amounts of research investigated these problem areas, but in 1979 alone the literature on the opiate peptides nearly doubled. Twenty-four male albino rats were given daily intraperitoneal injections of vasoactive intestinal polypeptide (VIP), motilin, human gastrin I (1–17) or the diluent control vehicle at a dose of 100 μg/kg for four consecutive days and food intake, water intake, body weight, and running wheel activity were determined every 24 hours. Animals injected with motilin or human gastrin I (1–17) exhibited decreased food intake relative to those injected with VIP or diluent, which did not differ from each other, although food intake increased reliably over days. The mean water consumption followed the same pattern as that of food intake. As expected from the above results, VIP produced weight gains as compared with rats injected with motilin or gastrin but not reliably more than after diluent. A reliable effect of trials for weight gain was the greatest on day three. Running wheel activity was not affected by injections of human gastrin I (1–17), motilin, or diluent but was reliably decreased by VIP. No significant differences existed across days. Although the results indicate that GI peptides may affect behavior when injected systemically and that like other peptides they have multiple effects, caution is urged in the interpretation of behavioral results at this time.  相似文献

Summary Scale formation in Cyprinodon variegatus was found to be initiated at about 26 to 30 days after hatching. Ultrastructural investigation revealed that within 4 to 6 h in the first-formed scales the marginal cells begin to flatten and differentiate into osteogenic cells, which later change to osteoblasts and fibroblasts. These cells are separated from the surrounding epithelial cells by a basal lamina. The osteoid is formed by the marginal and osteogenic cells; the osseous layer by the osteoblasts; and the fibrillary plate by the fibroblasts.The osteoid is formed within 2 to 3 h after the initiation of the scale, and within 20 to 24 h the osseous layer is formed. Hydroxyapatite crystals are deposited in the matrix of the osseous layer without apparent association with collagen fibers. No matrix vesicles or dense bodies are evident at the sites of calcification. The fibrillary plate arises 18 to 20 h after the initiation of the scale. It is also partially calcified, but not before the third week of scale formation. The crystals develop almost exclusively between the collagen fibers at the extreme edge of the calcifying front, but solid calcification of the fibers results with further growth of the crystals. The fibroblasts appear to participate in calcification of the fibrillary plate.Contribution No. 332, Belle W. Partially albinic Mystromys albicaudatus were examined to determine if the condition in these animals was homologous with the Chediak-Higashi syndrome. Tissues and cells from partially albinic and normal Mystromys albicaudatus were studied by light and electron microscopy. No evidence of cytoplasmic granule enlargement, which is characteristic of the Chediak-Higashi syndrome, was detected in the cells of the partially albinic rats when compared to controls. It was concluded that the inherited condition of partial albinism of Mystromys albicaudatus was not homologous with the inherited partially albinic disease known as the Chediak-Higashi syndrome.  相似文献

Thiobacillus ferrooxidans became significantly more tolerant to mercury stress after culturing in media of increasing mercury(II) concentrations. When mercuric chloride was added to the growth medium, the resistant organisms were found to volatilize elemental mercury (Hg0).T. ferrooxidans may be an important factor in the natural mercury cycle, since the environments whereT. ferrooxidans is found typically contain elevated levels of heavy metals, including mercury.  相似文献

Cell densities and respiratory rates of astrocytes from neonatal rat brain grown in primary culture were determined after 20–30 days in vitro. Cells grown in flasks reached lower densities (g DNA/cm2) and higher protein: DNA ratios than cells grown in petri dishes. Respiratory rates were lower for cells grown in flasks compared to cells grown in dishes. The pH of the medium in flasks fell below 6.9 between feedings while the pH of the medium in dishes remained at about 7.2. Cells grown in dishes with the medium pH adjusted to 6.8 also showed lower final cell densities, higher protein: DNA ratios, and lower respiratory rates, compared to cells grown under similar conditions at pH 7.5. Intermediate values of each parameter were found in cells grown at pH 7.5 for one week and then at 6.8 for 20 days. We conclude that the effects of ambient pH account for the differences in growth characteristics and respiratory rates of astrocytes grown in dishes versus those grown in flasks.  相似文献

Spoils samples collected from a coal strip mine in southeastern Montana were examined for populations and activities of iron- and sulfur-oxidizing bacteria. Spoils examined were of three types: (a) acidic pyrite-rich waste coal, (b) oxidation halo material, and (c) alkaline material, which was the most widespread type. Bacterial numbers, sulfur oxidation, and14CO2 uptake activity declined to low levels in the summer when spoils were dry. Even in wetter spring months pyritic spoils contained relatively low numbers of acidophilic iron- and sulfur-oxidizing bacteria, probably indicative of water stress since the same spoils incubated with excess water or dilute mineral salts showed considerably greater bacterial numbers and activity. Certain wells in coal and spoils aquifers contained substantial populations of iron-oxidizing acidophilic bacteria. However, these wells were always of alkaline or neutral pH, indicating that bacterial pyrite oxidation occurred where groundwaters contacted either replaced spoils or coal that contained pyrite or other metal sulfides. Bacterial activity may contribute to trace metal and sulfate leaching in the area.  相似文献
