The adult subventricular zone (SVZ) is a highly organized microenvironment established during the first postnatal days when radial glia cells begin to transform into type B‐cells and ependymal cells, all of which will form regenerative units, pinwheels, along the lateral wall of the lateral ventricle. Here, we identify p73, a p53 homologue, as a critical factor controlling both cell‐type specification and structural organization of the developing mouse SVZ. We describe that p73 deficiency halts the transition of the radial glia into ependymal cells, leading to the emergence of immature cells with abnormal identities in the ventricle and resulting in loss of the ventricular integrity. p73‐deficient ependymal cells have noticeably impaired ciliogenesis and they fail to organize into pinwheels, disrupting SVZ niche structure and function. Therefore, p73 is essential for appropriate ependymal cell maturation and the establishment of the neurogenic niche architecture. Accordingly, lack of p73 results in impaired neurogenesis. Moreover, p73 is required for translational planar cell polarity establishment, since p73 deficiency results in profound defects in cilia organization in individual cells and in intercellular patch orientation. Thus, our data reveal a completely new function of p73, independent of p53, in the neurogenic architecture of the SVZ of rodent brain and in the establishment of ependymal planar cell polarity with important implications in neurogenesis. © 2015 Wiley Periodicals, Inc. A new antibiotic, designated LL-A491, was isolated by butanol extraction and crystallization from beers of an unidentified streptomycete, Lederle culture A491. LL-A491 was primarily active against gram-positive bacteria and was related to monazomycin. A tentative molecular formula of C72±2 H144±8 O25±1 N for the antibiotic, based on analyses of the crystalline hydrochloride, picrate, and picrolonate salts, is considerably larger than that proposed for monazomycin, from which LL-A491 was not differentiated by paper chromatography. Analysis of the amorphous polyacetate ester suggested 15 to 16 acetylable groups. Upon hydrolysis, LL-A491 liberated ammonia and a reducing sugar which appeared to be mannose. LL-A491 was dextrorotatory, [α]D25 + 14°, and exhibited only end absorption in the ultraviolet region.  相似文献

Duplication of sex chromosomes was studied in bone marrow cells from adult rats and in short-term cultures of rat fetus cells. Results obtained indicate that: a) G2 period takes 4 hours in cells from fetuses and 3 hours in bone marrow cells, b) S period lasts 7 hours in bone marrow cells and about 20 hours in short-term cultured cells from fetuses, c) In cells from female fetuses one X-chromosome is the last to start DNA synthesis and also one of the X's is the last to end replication, d) In cells from female adult rats both X-chromosomes start and finish DNA synthesis early, e) In both line of cells the Y-chromosome was the last to begin and the last to finish DNA synthesis. — Causes which can account for the differences between the two line of cells are discussed.  相似文献

Cassava, bean and maize leaves were fed with14CO2 in light and the primary products of photosynthesis identified 5 and 10 seconds after assimilation. In maize, approximately three quarters of the labelled carbon was incorporated in C4 acids, in beans about two thirds in PGA, and in cassava approximately 40–60% in C4 acids with 30–50% in PGA. These data indicate that cassava possesses the C4 photosynthetic cycle, however due to the lack of typical Kranz anatomy appreciable carbon assimilation takes place directly through the Calvin-Benson-Bassham cycle.  相似文献

Treatment of human promyelocytic leukemia cells U937 with phorbol 12-myristate 13-acetate (TPA) induces them to differentiate into monocytic cells [Harris, P., & Ralph, P. (1985) J. Leukocyte Biol. 37, 407-422]. Here we investigated the effects of TPA on interleukin 1 gene expression and the possible role of protein kinase C (PKC) in this process. Addition of TPA to serum-starved U937 cells induced the expression of the interleukin 1 beta (IL-1 beta) gene. This effect was apparent as early as 2 h and peaked at 24 h in the presence of 5 X 10(-8) M TPA. Higher concentrations of TPA, which partially or totally depleted protein kinase C levels in the cells (10(-9)-2 X 10(-5) M), had an inhibitory effect on IL-1 beta mRNA expression. Cell-permeable 1,2-dioctanoyl-sn-glycerol (diC8), a diacylglycerol that activates PKC in intact cells and cell-free systems, did not mimic the effect of TPA on the IL-1 beta mRNA induction. To determine the protein kinase C isozymes present in the control and TPA- (5 X 10(-8) M) treated U937 cells, we prepared antipeptide antibodies that specifically recognize the alpha, beta, and gamma isoforms of protein kinase C in rat brain cytosol and U937 cell extracts. In "control" U937 cells, 30% of PKC alpha was particulate, and PKC beta was cytosolic, while there was no detectable PKC gamma.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The relationship between the apparently random chromosomal changes found in aneuploidy and the genetic instability driving the progression of cancer is not clear. We report a test of the hypothesis that aneuploid chromosomal abnormalities might be selected to preserve cell-survival genes during loss of heterozygosity (LOH) formations which eliminate tumor suppressor genes. The LOHs and structurally abnormal chromosomes present in the aneuploid LoVo (colon), A549 (lung), SUIT-2 (pancreas), and LN-18 (glioma) cancer cell lines were identified by single nucleotide polymorphisms (SNPs) and Spectral Karyotyping (SKY). The Mann-Whitney U and chi square tests were used to evaluate possible differences in chromosome numbers and abnormalities between the cell lines, with two-tailed P values of <0.01 being considered significant. The cell lines differed significantly in chromosome numbers and frequency of structurally abnormal chromosomes. The SNP analysis revealed that each cell line contained at least a haploid set of somatic chromosomes, consistent with our hypothesis that cell-survival genes are widely scattered throughout the genome. Further, over 90% of the chromosomal abnormalities seemed to be selected, often after LOH formation, for gene-dosage compensation or to provide heterozygosity for specific chromosomal regions. These results suggest that the chromosomal changes of aneuploidy are not random, but may be selected to provide gene-dosage compensation and/or retain functional alleles of cell-survival genes during LOH formation.  相似文献

It is well‐known that sodium improves the performance of Cu2ZnSnS4 (CZTS) devices, yet the mechanism of the enhancement is still not fully understood. This work aims to present a unified account of the relationships between grain boundaries in CZTS, sodium content at these boundaries, non‐radiative recombination, and surfactant effects that produce large microstructural changes. Using temperature‐dependent photoluminescence measurements, it is demonstrated that samples containing dramatically different grain sizes display identical radiative and non‐radiative decay characteristics when sufficient sodium is present in the film. It is also shown that the sodium concentration needed to efficiently passivate non‐radiative defects is significantly less that the quantity needed to obtain micrometer‐sized CZTS grains. Finally, the high densities of donor‐acceptor pairs that are observed in CZTS films appear to reside within the grains themselves, rather than at grain boundaries.  相似文献

The development of cancer is a complex, multistage process during which a normal cell undergoes genetic changes that result in phenotypic alterations and in the acquisition of the ability to invade other sites. Inductively coupled plasma optical emission spectroscopy was used to estimate the contents of Al, Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Na, P, Pb, and Zn in healthy kidney and renal cell carcinoma (RCC), and significant differences were found for all elements. Along with the progression of the malignant disease, a progressive decrease of Cd and K was observed. In fact, for Cd, the concentration in stage T4 was 263.9 times lower than in stage T1, and for K, the concentration in stage T4 was 1.73 times lower than in stage T1. Progressive accumulation was detected for P, Pb, and Zn in stage T4. For P, the concentration in stage T4 was 11.1 times higher than in stage T1; for Pb, the concentration in stage T4 was 232.7 times higher than in T1; and for Zn, the concentration in T4 was 8.452 times higher than in T1. This study highlights the marked differences in the concentrations of selected trace metals in different malignant tumor stages. These findings indicate that some trace metals may play important roles in the pathogenesis of RCC.  相似文献
