MPEG was modified with 1,1′-carbonyldiimidazole, then the activated MPEG reacted with primary amino groups of chitosan. Synthesize the graft copolymer of chitosan and polyethylene glycol in two steps. The structure of the copolymer was characterized by FT-IR and 1H-NMR. It agrees with the PEG content of classical stealth nanoparticles materials. The X-ray diffraction and DSC analysis proved that the crystallinity of the copolymer increased. It is a promising material for the stealth nanoparticles. It is a potential new carrier for the drug delivery systems of long-circulation and solid carcinoma.  相似文献

本研究旨在构建单纯疱疹病毒2型(herpes simplex virus type 2,HSV-2)衣壳支架蛋白ICP35的原核表达载体,并分析其在HSV-2增殖中的表达特性。以HSV-2基因组DNA为模板,采用聚合酶链反应(polymerase chain reaction,PCR)扩增HSV-2 ICP35的编码基因UL26.5,并克隆至原核表达载体pET-32a(+),转化大肠埃希菌BL21(DE3)进行诱导表达,将纯化的重组ICP35免疫家兔后获得抗体,采用免疫荧光检测ICP35在HSV-2增殖中的表达特性。结果显示,HSV-2 UL26.5基因的PCR产物大小约为1 065 bp,原核表达重组蛋白的相对分子质量约为6.3×104,免疫家兔的血清抗体可特异性识别HSV-2 ICP35。免疫荧光检测结果显示,HSV-2 ICP35可在感染后8 h出现于细胞核周围,12 h表达量进一步增加并向细胞核内聚集,16 h后在细胞核、细胞质内均可检测到聚集成斑点状的衣壳支架蛋白。结果表明,HSV-2 ICP35原核表达载体的成功构建及对其在HSV-2增殖中表达特性的分析,将为研究UL26.5基因的功能、蛋白相互作用、病毒与宿主的相互作用及筛选药物靶标等奠定基础。  相似文献

Extracellular ATP (eATP) plays essential roles in plant growth, development, and stress tolerance. Extracellular ATP-regulated stomatal movement of Arabidopsis thaliana has been reported. Here, ATP was found to promote stomatal opening of Vicia faba in a dose-dependent manner. Three weakly hydrolysable ATP analogs (adenosine 5′-O-(3-thio) triphosphate (ATPγS), 3′-O-(4-benzoyl) benzoyl adenosine 5′-triphosphate (Bz-ATP) and 2-methylthio-adenosine 5′-triphosphate (2meATP)) showed similar effects, indicating that ATP acts as a signal molecule rather than an energy charger. ADP promoted stomatal opening, while AMP and adenosine did not affect stomatal movement. An ATP-promoted stomatal opening was blocked by the NADPH oxidase inhibitor diphenylene iodonium (DPI), the reductant dithiothreitol (DTT) or the Ca2+ channel blockers GdCl3 and LaCl3. A hyperpolarization-activated Ca2+ channel was detected in plasma membrane of guard cell protoplast. Extracellular ATP and weakly hydrolyzable ATP analogs activated this Ca2+ channel significantly. Extracellular ATP-promoted Ca2+ channel activation was markedly inhibited by DPI or DTT. These results indicated that eATP may promote stomatal opening via reactive oxygen species that regulate guard cell plasma membrane Ca2+ channels.  相似文献

玉米干旱胁迫相关突变体在发掘玉米耐旱关键基因研究中具有重要利用价值。在玉米自交系综31的田间扩繁过程中,发现一个玉米干旱胁迫敏感的自然突变体,该突变体在轻度干旱条件下叶片发生卷曲,严重干旱时叶尖变黄,衰老坏死。遗传分析表明突变性状受1对主效单基因控制,表现为隐性遗传,将突变基因命名为DS。利用B73与突变体ds组配F2分离群体,以干旱条件下叶片是否卷曲为指标,将DS基因初定位在第3号染色体SSR标记umc1772和umc2158之间,物理距离为5 Mb。以上研究结果为该基因的克隆及功能分析奠定了基础。  相似文献

目的:研究担载血管内皮生长因子(VEGF)的乳液法电纺纤维膜的亲水性能、外观形态和机械性能,纤维膜中VEGF的包封率和体外释放动力学,为评价其能否应用于血管再生领域的研究奠定基础。方法:将VEGF水溶液通过W/O乳液法制备成缓释VEGF的生物可降解的丙交酯-乙交酯共聚物(PLGA)静电纺丝纤维膜,对该纤维膜的接触角、外观形态、机械性能进行表征,Elisa法测定该纤维膜的体外14天的释放行为,分别观察纤维膜释放0天、7天、14天后的电镜图。结果:加入VEGF后,纤维膜的接触角由140.0°减小到136.1°,亲水性增强,具有类似细胞外基质(ECMs)网状结构和良好的力学性能,纤维膜第1天的突释不超过载药量的50%,电镜图下显示纤维膜释放1周时纤维发生断裂。结论:通过乳液法制备的担载VEGF的电纺纤维膜具有良好的物理性能,能够持续缓释VEGF,可作为血管再生的组织工程支架进行深入研究。  相似文献

Chemerin是2007年新确认的一种脂肪因子,其主要功能受体为ChemR23。近期研究发现chemerin可能是联系肥胖、糖尿病及动脉粥样硬化的潜在因子,有望为糖尿病及其血管并发症的预防及治疗提供新的靶点。然而,chemerin及其受体ChemR23参与糖尿病及其大血管病变的具体机制尚不明确。本文将就目前研究中chemerin及其受体ChemR23与糖尿病及其大血管病变的关系作一综述,并从免疫及炎症反应、氧化应激、自噬、糖脂代谢和胰岛素抵抗等方面,分析chemerin分别对巨噬细胞、血管内皮细胞、脂肪细胞及骨骼肌细胞的影响,从而进一步阐述chemerin及其受体ChemR23参与糖尿病及其大血管病变的具体生物学机制。  相似文献

Copy number variations (CNVs) represent a substantial source of structural variants in mammals and contribute to both normal phenotypic variability and disease susceptibility. Although low-resolution CNV maps are produced in many domestic animals, and several reports have been published about the CNVs of porcine genome, the differences between Chinese and western pigs still remain to be elucidated. In this study, we used Porcine SNP60 BeadChip and PennCNV algorithm to perform a genome-wide CNV detection in 302 individuals from six Chinese indigenous breeds (Tongcheng, Laiwu, Luchuan, Bama, Wuzhishan and Ningxiang pigs), three western breeds (Yorkshire, Landrace and Duroc) and one hybrid (Tongcheng×Duroc). A total of 348 CNV Regions (CNVRs) across genome were identified, covering 150.49 Mb of the pig genome or 6.14% of the autosomal genome sequence. In these CNVRs, 213 CNVRs were found to exist only in the six Chinese indigenous breeds, and 60 CNVRs only in the three western breeds. The characters of CNVs in four Chinese normal size breeds (Luchuan, Tongcheng and Laiwu pigs) and two minipig breeds (Bama and Wuzhishan pigs) were also analyzed in this study. Functional annotation suggested that these CNVRs possess a great variety of molecular function and may play important roles in phenotypic and production traits between Chinese and western breeds. Our results are important complementary to the CNV map in pig genome, which provide new information about the diversity of Chinese and western pig breeds, and facilitate further research on porcine genome CNVs.  相似文献

通过对NK58S和NK58F这一对光敏核不育水稻等位突变系的AFLP分析,比较了AFLP,RAPD及RFLP检测DNA多态性的相对效率。结果表明,这三种分子标记的DNA多态性检出效率依次为AFLP>RAPD>RFLP;找出了水稻AFLP分析的最适反应条件;通过AFLP和集群混合分析(Bulked segregating analysis,BSA),筛选出了一批与水稻光敏核不育(PGMS)基因连锁的多态性AFLP产物,已完成了对4个多态性AFLP产物的克隆,Southern杂交证明其中2个为单拷贝顺序,另外2个为低拷贝顺序。对上述三种分子标记各自的优缺点及它们在DNA多态性检测中的适用之处进行了分析探讨。  相似文献

An Arabidopsis cell death mutation locus was mapped to chromosome 2 between lGS1 and mi421. The YAC clone ends, CIC9A3R, CIC11C7L, CIC2G5R and RFLP marker CDs3 within this interval, were used to probe TAMU BAC library and 31 BAC clones were obtained. A BAC contig encompassing the mutation locus, which consists of T6P5, T7M23, T12A21, T8L6 and T18A18, was identified by Southern hybridization with the BAC ends as probes. 11 CAPS and 12 STS markers were developed in this region. These results will facilitate map-based cloning of the genes and sequencing of the genomic DNA in this region.  相似文献
