Clinical transcranial Doppler assessment of cerebral vasomotor reactivity (CVMR) uses linear regression of cerebral blood flow velocity (CBFV) vs. end-tidal CO(2) (Pet(CO(2))) under steady-state conditions. However, the cerebral blood flow (CBF)-Pet(CO(2)) relationship is nonlinear, even for moderate changes in CO(2). Moreover, CBF is increased by increases in arterial blood pressure (ABP) during hypercapnia. We used a modified rebreathing protocol to estimate CVMR during transient breath-by-breath changes in CBFV and Pet(CO(2)). Ten healthy subjects (6 men) performed 15 s of hyperventilation followed by 5 min of rebreathing, with supplemental O(2) to maintain arterial oxygen saturation constant. To minimize effects of changes in ABP on CVMR estimation, cerebrovascular conductance index (CVCi) was calculated. CBFV-Pet(CO(2)) and CVCi-Pet(CO(2)) relationships were quantified by both linear and nonlinear logistic regression. In three subjects, muscle sympathetic nerve activity was recorded. From hyperventilation to rebreathing, robust changes occurred in Pet(CO(2)) (20-61 Torr), CBFV (-44 to +104% of baseline), CVCi (-39 to +64%), and ABP (-19 to +23%) (all P < 0.01). Muscle sympathetic nerve activity increased by 446% during hypercapnia. The linear regression slope of CVCi vs. Pet(CO(2)) was less steep than that of CBFV (3 vs. 5%/Torr; P = 0.01). Logistic regression of CBF-Pet(CO(2)) (r(2) = 0.97) and CVCi-Pet(CO(2)) (r(2) = 0.93) was superior to linear regression (r(2) = 0.91, r(2) = 0.85; P = 0.01). CVMR was maximal (6-8%/Torr) for Pet(CO(2)) of 40-50 Torr. In conclusion, CBFV and CVCi responses to transient changes in Pet(CO(2)) can be described by a nonlinear logistic function, indicating that CVMR estimation varies within the range from hypocapnia to hypercapnia. Furthermore, quantification of the CVCi-Pet(CO(2)) relationship may minimize the effects of changes in ABP on the estimation of CVMR. The method developed provides insight into CVMR under transient breath-by-breath changes in CO(2).  相似文献

Ecological traps are serious, anthropogenic threats to animal populations. However, in certain cases it is difficult to determine whether they really act in the expected manner. This applies to the harmful effects of beech timber stacked in forests on the endangered saproxylic beetle Rosalia longicorn Rosalia alpina, which have been mentioned in numerous scientific articles, conservation action plans and similar publications. The aim of this paper is to determine whether beech timber stacks meet the criteria of an ecological trap for the Rosalia longicorn. Two basic criteria of such a trap are analysed: the attractiveness of timber stacks and the impossibility of complete larval development. The results show that beech timber stacks are highly attractive to Rosalia longicorn imagines. Moreover, the time during which the timber is stacked is shown to be significantly shorter than the species’ larval development period. These results suggest that timber stacks can be treated as operative ecological traps for the Rosalia longicorn, even though the extent of their influence on the demographic parameters of this beetle’s population has not been estimated. Forest management practices, i.e. increasing amounts and shifts in timing of wood storage, could intensify this threat.  相似文献

The purpose of this study was to compare the pro-antioxidant status in healthy men exposed to muscle-damaging resistance exercise, and to investigate the practical application of Loverro's coefficient (P/A ratio) to evaluate the presence of oxidative stress. Twenty-eight healthy men were assigned to two groups performed multi-joint (M) or single-joint (S) resistance exercise. The activities of superoxide dismutase (SOD), glutathione peroxidase (GPx) and catalase (CAT) as well as the concentration of lipid peroxidation products (TBARS) in blood were evaluated. The P/A ratio was calculated from the mean values of erythrocyte TBARS, SOD, CAT and GPx. Creatine kinase (CK) activity was used as a marker of muscle damage. The applied resistance exercises triggered off the changes in pro-antioxidant ratio towards peroxidation which was proved by significant increase in erythrocyte TBARS concentration in M (+25%) and S (+27%) groups. Plasma TBARS increased only after multi-joint resistance exercise and correlated with erythrocyte P/A ratio (r = 0.536, P < 0.01). The multi-joint exercise caused decrease in SOD activity by 28% whereas the single-joint resistance exercise elevated enzyme activity by 20%. Activities of the other antioxidant enzymes changed simultaneously i.e. CAT activity increased by 14%-16% immediately after exercise, and GPx activity declined by 18%-34% during recovery in M and S groups. Even though, all erythrocyte parameters significantly changed following multi-joint and single-joint resistance exercises, the assessment of pro-antioxidant ratio showed the considerable increase in P/A only in M group. In summary, an analysis of pro- and antioxidant parameters showed significant changes in response to muscle-damaging exercise and demonstrated the practical application of P/A ratio to evaluate the risk of oxidative stress in athletes.  相似文献

Questions: How is seedling regeneration of woody species of semi‐deciduous rain forests affected by (a) historical management for combinations of logging, arboricide treatment or no treatment, (b) forest community type and (c) environmental gradients of topography, light and soil nutrients? Location: Budongo Forest Reserve, Uganda. Methods: Seedling regeneration patterns of trees and shrubs in relation to environmental factors and historical management types were studied using 32 0.5‐ha plots laid out in transects along a topographic gradient. We compared seedling species diversity, composition and distribution patterns along topographic gradients and within types of historical management regimes and forest communities to test whether environmental factors contributed to differences in species composition of seedlings. Results: A total of 85 624 woody seedlings representing 237 species and 46 families were recorded in this rain forest. Cynometra alexandri C.H. Wright and Lasiodiscus mildbraedii Engl. had high seedling densities and were widely distributed throughout the plots. The most species‐rich families were Euphorbiaceae, Fabaceae, Rubiaceae, Meliaceae, Moraceae and Rutaceae. Only total seedling density was significantly different between sites with different historical management, with densities highest in logged, intermediate in logged/arboricided and lowest in the nature reserve. Forest communities differed significantly in terms of seedling diversity and density. Seedling composition differed significantly between transects and forest communities, but not between topographic positions or historical management types. Both Chao‐Jaccard and Chao‐Sørensen abundance‐based similarity estimators were relatively high in the plot, forest community and in terms of historical management levels, corroborating the lack of significant differences in species richness within these groups. The measured environmental variables explained 59.4% of variance in seedling species distributions, with the three most important being soil organic matter, total soil titanium and leaf area index (LAI). Total seedling density was positively correlated with LAI. Differences in diversity of >2.0 cm dbh plants (juveniles and adults) also explained variations in seedling species diversity. Conclusions: The seedling bank is the major route for regeneration in this semi‐deciduous tropical rain forest, with the wide distribution of many species suggesting that these species regenerate continuously. Seedling diversity, density and distribution are largely a function of adult diversity, historical management type and environmental gradients in factors such as soil nutrient content and LAI. The species richness of seedlings was higher in soils both rich in titanium and with low exchangeable cations, as well as in logged areas that were more open and had a low LAI.  相似文献

Quantifying the Trade in Cycads ( Encephalartos Species) in the Traditional Medicine Markets of Johannesburg and Durban, South Africa. Cycads have been used for traditional purposes for centuries, mainly as a source of starch during droughts and famines. In South Africa, Encephalartos species are traded for traditional medicine (TM) in local TM markets. The 2009 IUCN Red Data assessment for South African flora lists 78% of the 37 indigenous Encephalartos species as “Threatened”, with three species already “Extinct in the Wild” (EW). South African cycads face an extinction crisis, primarily due to collecting for the horticultural trade. The trade in Encephalartos for TM also has a significant impact on several species, and is largely understudied. In this paper, the trade in Encephalartos for TM was quantified in South Africa’s two largest TM markets, Faraday in Johannesburg and Warwick in Durban. Harvesting of South African Encephalartos for TM involves the removal of bark strips and/or whole stem sections. Encephalartos species were sold at 26% and 13% of the stalls at Faraday and Warwick, respectively, with an estimated 9.0 metric tons traded at Warwick in 2009. Stem samples purchased in the markets were assigned to stem diameter size classes using a size class chart, and regression analysis was used to validate the allocations. Most harvesting was from sub-adult and adult cycads, and it appears that bark strips are removed from large arborescent plants, whereas smaller individuals and subterranean-stemmed species are harvested by removing the entire plant. There is generally a positive relationship between stem diameter and leaf base length as well as stem diameter and pith radius. The former can be used as a predictor of stem diameter size class for market samples that prove difficult to assign to diameter classes using the chart. Overall, this is the first study to quantify the trade in Encephalartos for TM in South Africa with reference to the size classes of the plants traded.  相似文献

Baeyer-Villiger monooxygenases catalyze oxidations that are of interest for biocatalytic applications. Among these enzymes, phenylacetone monooxygenase (PAMO) from Thermobifida fusca is the only protein showing remarkable stability. While related enzymes often present a broad substrate scope, PAMO accepts only a limited number of substrates. Due to the absence of a substrate in the elucidated crystal structure of PAMO, the substrate binding site of this protein has not yet been defined. In this study, a structural model of cyclopentanone monooxygenase, which acts on a broad range of compounds, has been prepared and compared with the structure of PAMO. This revealed 15 amino acid positions in the active site of PAMO that may account for its relatively narrow substrate specificity. We designed and analyzed 30 single and multiple mutants in order to verify the role of these positions. Extensive substrate screening revealed several mutants that displayed increased activity and altered regio- or enantioselectivity in Baeyer-Villiger reactions and sulfoxidations. Further substrate profiling resulted in the identification of mutants with improved catalytic properties toward synthetically attractive compounds. Moreover, the thermostability of the mutants was not compromised in comparison to that of the wild-type enzyme. Our data demonstrate that the positions identified within the active site of PAMO, namely, V54, I67, Q152, and A435, contribute to the substrate specificity of this enzyme. These findings will aid in more dedicated and effective redesign of PAMO and related monooxygenases toward an expanded substrate scope.  相似文献

Large quantities of plants are traded annually in South Africa's traditional medicine or ‘muthi’ markets. A resource in high demand in the Faraday (Johannesburg) and Warwick (Durban) markets is uMavumbuka, a root holoparasite usually identified as either Hydnora africana Thunb. or Sarcophyte sanguinea Sparrm. subsp. sanguinea. However, rhizomes regularly observed in Faraday between 1994 and 2008 did not resemble either species, thereby suggesting that a third, and undocumented, species was being harvested. This was confirmed when the rhizomes were identified as H. abyssinica A.Br. by an American parasitic plant expert. An ethno-ecological study was initiated to verify its occurrence in selected muthi markets. The study further aimed to investigate the distribution of H. abyssinica through trader interviews, host species localities and some previously misidentified herbarium specimens. The study revealed that H. abyssinica was the only uMavumbuka species present in Faraday and Warwick in 2009. Furthermore, the rhizomes were being harvested in KwaZulu-Natal—an area not previously known to be part of its distribution range. Re-evaluated herbarium vouchers and recent photographs taken in the Kruger National Park have confirmed that H. abyssinica occurs in KwaZulu-Natal, Limpopo, Mpumalanga, Gauteng and Swaziland and hence eastern southern Africa. Fragments of Acacia xanthophloea Benth. roots were identified on 93% of the samples that had host roots attached, and we suspect that H. abyssinica follows the distribution of A. xanthophloea in suitable habitats north from KwaZulu-Natal and adjoining the South African border with Swaziland and Mozambique. Acacia karroo Hayne and A. grandicornuta Gerstner have also been positively identified as host species in South Africa from herbarium records. Plant harvesters in the markets cited the common names of several other species that uMavumbuka “grows under” that may be identified as hosts to H. abyssinica in the future. The collection of specimens in areas identified by the harvesters and in areas of suitable habitat is important to verify the occurrence, distribution and habitat of H. abyssinica in eastern southern Africa.  相似文献
