Investigations on enamel microstructure provided new data for the debate on hippopotamid origin. Observations indicated a diversity of patterns relevant to phylogenetic inferences. Within Hippopotamoidea, the distribution of these patterns seems to be in favour of a hippopotamid origin within the Palaeogene African anthracotheres. Enamel microcharacters therefore prove to be particularly relevant for future phylogenetic analysis of the superfamily, and have implications for our understanding of ecological transitions within hippopotamoids at the end of the Miocene. Indeed, unlike equids or bovids, which developed grass feeding thanks to their hypsodont molars, hippopotamoids may have had another way to exploit this resource. The combination of inter‐row sheets, which appeared early in the evolutionary history of the group, and the increased thickness of radial enamel could have eased the consumption of highly abrasive graminoids. © 2014 The Linnean Society of London 相似文献

In Tris-washed Photosystem-II particles we are able to induce an EPR signal in the dark by addition of an iridium salt (K2IrCl6). This signal is attributed to signal IIs (slow) (D+) and the redox titration gives an Em value of 760 mV for the couple . On the basis of our previous studies on the equilibrium between D+Z and DZ+ (K = 104) (Boussac, A. and Etienne, A.L. (1982) Biochem. Biophys. Res. Commun. 109, 1200–1205), we therefore attribute a value of 1 V for the Em of the couple. A second effect of K2IrCl6 is to modify the spectral characteristics of signal II. We conclude that K2IrCl6 is able to change the environment of the species from which signal IIs and signal IIf originate. 相似文献

SliK, a K+ channel encoded by the Streptomyces KcsA gene, was expressed, purified, and reconstituted in liposomes. A concentrative 86Rb+ flux assay was used to assess the ion transport properties of SliK. SliK-mediated ionic flux shows strong selectivity for K+ over Na+ and is inhibited by micromolar concentrations of Ba2+, mirroring the basic permeation characteristic of eukaryotic K+ channels studied by electrophysiological methods. 86Rb+ uptake kinetics and equilibrium measurements also demonstrate that the purified protein is fully active. 相似文献

Pollen material of the two anemophilous plants of known allergic significance Dactylis glomerata (Poaceae) and Betula verrucosa (Betulaceae) was collected at several places (in Sweden and France) and exposed to natural or experimental conditions. This material was analyzed with Electron Probe X Microanalysis (EPMA) (System ORTEC 5000) attached to a Scanning Electron Microscope (Jeol JMS 35), or with EDAX, a 10000 X-Ray Analyser, Link System, connected to a SEM-microscope (Jeol JSM 820). It is clear that the environment has an effect on such a co-inductor or adjuvant of allergenicity as K. The possible presence of pollutant inorganic (mineral) particles on the pollen grain surface may play a role in the allergenic activity. Glucokinase is the predominant hexokinase expressed in hepatocytes and pancreatic β-cells, with a pivotal role in regulating glucose-stimulated insulin secretion, illustrated by glucokinase gene mutations causing monogenic diabetes and congenital hyperinsulinemic hypoglycemia. A complex tissue-specific network of mechanisms regulates this enzyme, and a major unanswered question in glucokinase biology is how post-translational modifications control the function of the enzyme. Here, we show that the pancreatic isoform of human glucokinase is SUMOylated in vitro, using recombinant enzymes, and in insulin-secreting model cells. Three N-terminal lysines unique for the pancreatic isoform (Lys-12/Lys-13 and/or Lys-15) may represent one SUMOylation site, with an additional site (Lys-346) common for the pancreatic and the liver isoform. SUMO-1 and E2 overexpression stabilized preferentially the wild-type human pancreatic enzyme in MIN6 β-cells, and SUMOylation increased the catalytic activity of recombinant human glucokinase in vitro and also of glucokinase in target cells. Small ubiquitin-like modifier conjugation represents a novel form of post-translational modification of the enzyme, and it may have an important regulatory function in pancreatic β-cells. 相似文献

Background
The bovine spongiform encephalopathy (BSE) epidemic and the emergence of a new human variant of Creutzfeldt-Jakob Disease (vCJD) have led to profound changes in the production and trade of agricultural goods. The rapid tests currently approved for BSE monitoring in slaughtered cattle are all based on the detection of the disease related isoform of the prion protein, PrPd, in brain tissue and consequently are only suitable for post-mortem diagnosis. Objectives: In instances such as assessing the health of breeding stock for export purposes where post-mortem testing is not an option, there is a demand for an ante-mortem test based on a matrix or body fluid that would permit easy access and repeated sampling. Pyruvate dehydrogenase (PDH) E1α is a key component in the PDH complex which catalyzes the oxidative decarboxylation of pyruvate
to acetyl-CoA. Defects in the gene coding for PDH E1α (PDHA1) are associated with a variety of clinical symptoms, often of
a severe character. In the present study, the segregation of three polymorphic CA repeats located in PDHA1 was followed in
the 40 CEPH reference pedigrees. Using these data, multipoint linkage analysis was carried out, refining the genetic location
of PDHA1. The 16-point map presented locates PDHA1 in an approximately 3-cM interval between DXS999 and DXS365 with odds of
more than 1000 : 1. From known physical localizations of the flanking marker loci, PDHA1 could be regionally assigned to Xp22.1-p22.2.
The information provided should be of value in clinical settings.
