We report the chemically determined sequence of most of the polypeptide chain of the coat protein of tomato bushy stunt virus. Peptide locations have been determined by comparison with the high-resolution electron density map from X-ray crystallographic analysis as well as by conventional chemical overlaps. Three small gaps remain in the 387-residue sequence. Positively charged side-chains are concentrated in the N-terminal part of the polypeptide (the R domain) as well as on inward-facing surfaces of the S domain. There is homology of S-domain sequences with structurally corresponding residues in southern bean mosaic virus. 相似文献

Wie aus Elektrophorese- und spektralphotometrischen Untersuchungen hervorgeht, liegt der kationische Farbstoff Chrysoidin G, je nach dem pH-Wert der wäßrigen Farblösungen, als I-, II-, III- und IV-wertiges Kation und elektroneutrales Farbbasenmolekül vor. Von physiologischer Bedeutung ist nur das I-wertige Kation und das Farbbasenmolekül. Die Unabhängigkeit der Absorptionsmaxima wäßriger Farbstofflösungen mit konstantem pH-Wert von der Farbstoffkonzentration deutet darauf hin, daß Chrysoidin keine Assoziate bildet. In organischen Lösungsmitteln ergibt Chrysoidin G je nach dem Grad der Polarität des Solvens und dem pH-Wert der wäßrigen Phase bei Ausschüttelungs-versuchen unterschiedliche Absorptionskurven. Natriumnucleinat bedingt eine negative Metachromasie; die jeweilige Lage des Maximums wird von der Natriumnucleinatkonzentration bestimmt. Rutin übt keinen wahrnehmbaren Einfluß auf das Absorptionsspektrum aus. Nach einer Vitalfärbung von Oberepidermiszellen der Schuppenblätter von Allium cepa mit Chrysoidin G zeigen das diffus gefärbte Plasma und die darin auftretenden gelben Kugeln übereinstimmende Absorptionsspektren mit einem breiten Bandenmaximum bei ? 420 nm. Der lebende Zellkern färbt sich nicht. Der gefärbte volle Zellsaft der Unterepidermis besitzt ein Maximum bei ? 448 nm. Aus der Lage der Absorptionsmaxima und dem Verlauf der Absorptionskurven kann geschlossen werden, daß die Färbung des lebenden Plasmas auf eine Anreicherung des einwertigen Kations und des Farbbasenmoleküls in polaren Lipoiden beruht, während es sich bei der Färbung des fixierten Zellkerns um eine Bindung des Chrysoidins an Nucleinsäuren handelt. Die Vitalfärbung des vollen Zellsaftes mit Chrysoidin G ist nicht auf den Gehalt der Vakuolen an Flavonolen zurückzuführen, sondern hängt vermutlich vom pH-Wert des Zellsaftes ab. 相似文献

Measurements of the rates of oxidation of various compounds by a fluorescent Pseudomonas indicated that metabolism of 2,4-xylenol was initiated by oxidation of the methyl group para to the hydroxyl group. 2. 4-Hydroxy-3-methylbenzoic acid was isolated as the product of oxidation of 2,4-xylenol by cells inhibited with alphaalpha'-bipyridyl. 3. 4-Hydroxyisophthalic acid accumulated at low oxygen concentrations when either 2,4-xylenol or 4-hydroxy-3-methylbenzoic acid was oxidized by cells grown with 2,4-xylenol. 4. When supplemented with NADH, but not with NADPH, cell extracts oxidized 4-hydroxy-3-methylbenzoic acid readily. 2-Hydroxy-5-methylbenzoic acid was not oxidized. 5. Both 4-hydroxyisophthalic acid and p-hydroxybenzoic acid were oxidized to beta-oxoadipic acid by cell extracts supplemented with either NADH or NADPH. 4,5-Dihydroxyisophthalic acid was not oxidized. 6. Summary 1. Studies of the broad-leafed turtle grass,Thalassia testudinum
König, have revealed a diverse range of fungal infestation different in generic composition and dynamics of attack from that found on submerged wood. Certain of the fungi, notably the AscomyceteLindra thalassiae, initiate considerable degradation of leaf tissue and show a developmental cycle in nature related to the physiological state of the host plant.2. Use of fungal-cellulose mats as a trapping substrate has been extremely effective for discernment of ecologically significant shifts in nematode concentrations, especially those of the omnivorous species,Metoncholaimus scissus.3. Patterns of activity ofM. scissus, as well as those of various foliicolous nematodes, suggest that loci of organic material, such as fungal infested leaves and decaying plant tissue, significantly affect biological activity of these animals.4. Laboratory analysis of degraded cotton cellulose filters show a striking incidence of fungal reproduction of the ascomycetous fungusLulworthia, along with development of a considerable associated nematode fauna, especially species ofViscosia (V. macramphida) andLeptolaimus (L. plectoides). Successional patterns in nematode development are noted with continued degradation of the cotton cellulose matrix.

Studien über marine Pilz-Nematoden-Assoziationen und Pflanzendegradation
Kurzfassung Untersuchungen am SeegrasThalassia testudinum König haben ergeben, daß sich hier Pilzinfektionen hinsichtlich der Komposition der beteiligten Gattungen und der Dynamik des Befalls von den am untergetauchten Holz festgestellten Infektionen unterscheiden. Bestimmte Pilze, insbesondere der AscomycetLindra thalassiae, leiten eine erhebliche Degradation des Blattgewebes ein und zeigen einen Entwicklungszyklus, welcher in Beziehung steht zum physiologischen Zustand der Wirtspflanze. Die Anwendung von Pilz-Zellulose-Matten als Einfangsubstrat war außerordentlich erfolgreich für das Erkennen ökologisch signifikanter Verschiebungen in den Nematodenkonzentrationen, insbesondere bei der omnivoren ArtMetoncholaimus scissus. Die Aktivitätsmuster vonM. scissus — ebenso wie die verschiedener foliicolöser Nematoden — deuten darauf hin, daß pilzinfizierte und zerfallende Pflanzenteile in entscheidendem Maße die biologische Aktivität dieser Tiere beeinflussen. Laboratoriumsanalysen degradierter Wollzellulosefilter lassen eine überraschend starke Vermehrung des AscomycetenLulworthia erkennen und gleichzeitig die Entwicklung einer beachtlichen Fauna assoziierter Nematodenarten, insbesondere vonViscosia macramphida undLeptolaimus plectoides. Im Verlaufe der weiteren Degradation der Wollzellulosematrix kommt es bei der Nematodenfauna zu entsprechenden Sukzessionen.

A glutaredoxin was purified from rabbit bone marrow, and its amino acid sequence was determined by high performance tandem mass spectrometry. The sequences of peptides generated by digestion with trypsin alone or in combination with thermolysin were determined from their collision-induced dissociation (CID) mass spectra. Alignment of these sequences and additional sequence information were obtained from the collision-induced dissociation mass spectra of peptides obtained from digestion of the intact protein with Staphylococcus aureus V8 protease and alpha-chymotrypsin. This glutaredoxin strongly resembles the corresponding calf and pig proteins (known as glutaredoxin and thioltransferase, respectively) with respect to its primary structure and enzymatic activity as a GSH:disulfide thioltransferase, an activity also found for the glutaredoxin from Escherichia coli. However, rabbit glutaredoxin was not active as a hydrogen donor for the reduction of ribonucleotides in the presence of the ribonucleotide reductases from rabbit bone marrow, Lactobacillus leichmannii, and Corynebacterium nephridii. 相似文献

Pseudomonas strain AT3, isolated by elective culture with atropine, hydrolyzed atropine and grew diauxically, first on the tropic acid and then on the tropine. Tropine was also used as a sole carbon and energy source. The methyl group of tropine was eliminated as formaldehyde, and the nortropine thus formed was a precursor of 6-hydroxycyclohepta-1,4-dione. Ammonia was detected as a product of nitrogen elimination. 6-Hydroxycyclohepta-1,4-dione was oxidized to cyclohepta-1,3,5-trione by an induced NAD(sup+)-specific dehydrogenase. Although cyclohepta-1,3,5-trione is a (beta)-diketone with two potential hydrolytic cleavage sites, an induced hydrolase was specific for one of these sites, with 4,6-dioxoheptanoate as the only hydrolysis product. Unlike the alternative cleavage product (3,6-dioxoheptanoate), this compound is also a (beta)-diketone, and a second hydrolytic cleavage formed succinate and acetone. Although Pseudomonas strain AT3 was not capable of growth with acetone, the compound was not detected in the culture medium and may have been lost to the atmosphere. Exhaustive experimentation with a wide range of conditions did not result in detection of the enzymes required for cleavage of the carbon-nitrogen bonds leading to the formation of nortropine and 6-hydroxycyclohepta-1,4-dione. 相似文献

Summary The cellular structures of acid rain-irrigated needles of several provenances of Norway spruce (Picea abies L. Karst) seedlings were studied after winter experimental freezing. Frost injuries and recovery were characterized by visual damage scoring and classification of mesophyll cell alterations, also using histochemical methods for carbohydrate fluorescent staining. The treatment with-30° C during the late dormancy period was sufficient to cause significant injuries and intracellular degradation in the tissues of the green needles. The most affected seedlings in terms of visual injury scoring were found among those treated with clean water or at pH 3, while freezing injury, defined as an occlusion of phenolic substances in the central vacuole of the mesophyll cells, was most abundant in the needles from spruces irrigated either with clean water or at pH 4 or pH 3. Electron microscopy revealed the details of the injury, e. g. thinning out of the cytoplasm and chloroplast stroma, darkening of the chloroplasts and eventually swelling of the chloroplasts and protoplast. PAS and ConA reactions in the needle tissue revealed intense starch accumulation in the mesophyll and transfusion tissues as early as in March, with a tendency to increase, especially in the untreated needles during the recovery period. Plasma membrane disturbances were indicated by histochemical identification of callose deposits in the mesophyll cell walls, these being most abundant in the acid rain-treated needles. All these findings suggest that freezing at –30° C was more deleterious to the seedlings pretreated with acid or clean water than to those not given additional irrigation. 相似文献

Abstract In haplodiploid Hymenoptera, unfertilized eggs produce haploid males while fertilized eggs lead to diploid females under most circumstances. Diploid males can also be produced from fertilization under a system of sex determination known as complementary sex determination (CSD). Under single-locus CSD, sex is determined by multiple alleles at a single sex locus. Individuals heterozygous at the sex locus are female while hemizygous and homozygous individuals develop as haploid and diploid males, respectively. In multiple-locus CSD, two or more loci, each with two or more alleles, determine sex. Diploid individuals are female if one or more sex loci are heterozygous, while a diploid is male only if homozygous at all sex loci. Diploid males are known to occur in 43 hymenopteran species and single-locus CSD has been demonstrated in 22 of these species. Diploid males are either developmentally inviable or sterile, so their production constitutes a genetic load. Because diploid male production is more likely under inbreeding, CSD is a form of inbreeding depression. It is crucial to preserve the diversity of sex alleles and reduce the loss of genetic variation in biological control. In the parasitoid species with single-locus CSD, certain precautionary procedures can prevent negative effects of single-locus CSD on biological control. 相似文献
