Oleoylethanolamide (OEA) is a lipid mediator that inhibits food intake by activating the nuclear receptor peroxisome proliferator-activated receptor-alpha. In the rodent small intestine OEA levels decrease during food deprivation and increase upon refeeding, suggesting that endogenous OEA may participate in the regulation of satiety. Here we show that feeding stimulates OEA mobilization in the mucosal layer of rat duodenum and jejunum but not in the serosal layer from the same intestinal segments in other sections of the gastrointestinal tract (stomach, ileum, colon) or in a broad series of internal organs and tissues (e.g. liver, brain, heart, plasma). Feeding also increases the levels of other unsaturated fatty acid ethanolamides (FAEs) (e.g. linoleoylethanolamide) without affecting those of saturated FAEs (e.g. palmitoylethanolamide). Feeding-induced OEA mobilization is accompanied by enhanced accumulation of OEA-generating N-acylphosphatidylethanolamines (NAPEs) increased activity and expression of the OEA-synthesizing enzyme NAPE-phospholipase D, and decreased activity and expression of the OEAdegrading enzyme fatty acid amide hydrolase. Immunostaining studies revealed that NAPE-phospholipase D and fatty acid amide hydrolase are expressed in intestinal enterocytes and lamina propria cells. Collectively, these results indicate that nutrient availability controls OEA mobilization in the mucosa of the proximal intestine through a concerted regulation of OEA biosynthesis and degradation.  相似文献

Mammalian Dicer interacts with double-stranded RNA-binding protein TRBP or PACT to mediate RNA interference and micro-RNA processing. TRBP and PACT are structurally related but exert opposite regulatory activities on PKR. It is not understood whether TRBP and PACT are simultaneously required for Dicer. Here we show that TRBP directly interacts with PACT in vitro and in mammalian cells. TRBP and PACT form a triple complex with Dicer and facilitate the production of small interfering RNA (siRNA) by Dicer. Knockdown of both TRBP and PACT in cultured cells leads to significant inhibition of gene silencing mediated by short hairpin RNA but not by siRNA, suggesting that TRBP and PACT function primarily at the step of siRNA production. Taken together, these findings indicate that human TRBP and PACT directly interact with each other and associate with Dicer to stimulate the cleavage of double-stranded or short hairpin RNA to siRNA. Our work significantly alters the current model for the assembly and function of the Dicer-containing complex that generates siRNA and micro-RNA in human.  相似文献

Human apolipoprotein D (ApoD) occurs in plasma associated with high density lipoprotein. Apart from the involvement in lipid metabolism, its binding activity for progesterone and arachidonic acid plays a role in cancer development and neurological diseases. The crystal structures of free ApoD and its complex with progesterone were determined at 1.8A resolution and reveal a lipocalin fold. The narrow, mainly uncharged pocket within the typical beta-barrel accommodates progesterone with its acetyl side chain oriented toward the bottom. The cavity adopts essentially the same shape in the absence of progesterone and allows complexation of arachidonic acid as another cognate ligand. Three of the four extended loops at the open end of the beta-barrel expose hydrophobic side chains, which is an unusual feature for lipocalins and probably effects association with the high density lipoprotein particle by mediating insertion into the lipid phase. This mechanism is in line with an unpaired Cys residue in the same surface region that can form a disulfide cross-link with apolipoprotein A-II.  相似文献

The objective of the present study was to investigate the effects of fermented oyster mushroom (Pleurotus ostreats) by-production (FOMP) supplementation on the growth performance, blood parameters, carcass traits and meat quality in finishing Berkshire pigs. FOMP was made by mixing oyster mushroom by-production with rice bran and barley bran and this mixture was fermented for 60 days. The experimental diets were 0, 3, 5 and 7% of FOMP added to C, T1, T2 and T3 in the basis diet for 7 weeks. Average daily gain (kg/day) was higher in C and T1 than in T2 and T3 ( P < 0.05). Average daily feed intake (kg/day) and feed conversion increased by the addition of FOMP ( P < 0.05). Total cholesterol and high-density lipoprotein cholesterol were higher in T3 than other treatments ( P < 0.05). Carcass weight (kg) was higher in C and T1 than in T2 and T3 ( P < 0.05). Dressing (%) was higher in C than in T3 ( P < 0.05). Crude protein was lower in T3 than in other treatments ( P < 0.05). Crude fat was higher in T2 and T3 than in C ( P < 0.05). pH24 was higher in C than in other treatments ( P < 0.05). Cooking loss (%) was higher in T1 than T2 ( P < 0.05). Water-holding capacity (%) was higher in C than in T1 ( P < 0.05). In meat colour, CIE a* was lower by the addition of FOMP ( P < 0.05). CIE b* was higher in C than in other treatments ( P < 0.05). In backfat colour, CIE L* was lower in T3 than other treatments ( P < 0.05). CIE b* was lower by addition of FOMP ( P < 0.05). Palmitoleic and oleic acid were higher in T3 than in other treatments ( P < 0.05). Linoleic and arachidonic acids were higher in T2 than in other treatments ( P < 0.05). The results indicate that 3% of FOMP affected the growth performance, carcass traits, meat quality and fatty acid in contrast to addition of 5% of FOMP for Berkshire pigs during the finishing period.  相似文献

Chinese hamster ovary cells show endogenous high-affinity Na^+ -dependent glutamate transport activity. This transport activity is kinetically similar to a glutamate transporter family strategically expressed in the central nervous system and is pharmacologically unlike glutamate transporter- 1 or excitatory amino acid carrier 1. The cDNA of a glutamate/aspartate transporter (GLAST)-like transporter was obtained and analyzed. The deduced amino acid sequence showed high similarity to human, mouse, and rat GLAST. We concluded that a GLAST-like glutamate transporter exists in Chinese hamster ovary cells that might confer the endogenous high-affinity Na^+ -dependent glutamate transport activity evident in these cells.  相似文献

There is a crucial need in the study of global change to understand how terrestrial ecosystems respond to the climate system. It has been demonstrated by many researches that Normalized Different Vegetation Index (NDVI) time series from remotely sensed data, which provide effective information of vegetation conditions on a large scale with highly temporal resolution, have a good relation with meteorological factors. However, few of these studies have taken the cumulative property of NDVI time series into account. In this study, NDVI difference series were proposed to replace the original NDVI time series with NDVI difference series to reappraise the relationship between NDVI and meteorological factors. As a proxy of the vegetation growing process, NDVI difference represents net primary productivity of vegetation at a certain time interval under an environment controlled by certain climatic conditions and other factors. This data replacement is helpful to eliminate the cumulative effect that exist in original NDVI time series, and thus is more appropriate to understand how climate system affects vegetation growth in a short time scale. By using the correlation analysis method, we studied the relationship between NOAA/AVHRR ten-day NDVI difference series and corresponding meteorological data from 1983 to 1999 from 11 meteorological stations located in the Xilingole steppe in Inner Mongolia. The results show that: (1) meteorological factors are found to be more significantly correlation with NDVI difference at the biomass-rising phase than that at the falling phase; (2) the relationship between NDVI difference and climate variables varies with vegetation types and vegetation communities. In a typical steppe dominated by Leymus chinensis, temperature has higher correlation with NDVI difference than precipitation does, and in a typical steppe dominated by Stipa krylovii, the correlation between temperature and NDVI difference is lower than that between precipitation and NDVI difference. In a typical steppe dominated by Stipa grandis, there is no significant difference between the two correlations. Precipitation is the key factor influencing vegetation growth in a desert steppe, and temperature has poor correlation with NDVI difference; (3) the response of NDVI difference to precipitation is fast and almost simultaneous both in a typical steppe and desert steppe, however, mean temperature exhibits a time-lag effect especially in the desert steppe and some typical steppe dominated by Stipa krylovii; (4) the relationship between NDVI difference and temperature is becoming stronger with global warming. __________ Translated from Acta Phytoecologica Sinica, 2005, 29(5): 753–765 [译自: 植物生态学报]  相似文献

本文旨在探讨分子佐剂C3d3与hCGβ融合在基因免疫中增强抗hCGβ体液免疫效应的机制。分别用质粒pCMV4-hCGB-C3d3、pCMV4-hCGβ和pCMV4免疫BALB/c小鼠,间接ELISA法检测免疫小鼠外周血IgG/IgA类抗hCGβ抗体水平;ELISPOT分析免疫鼠脾脏组织IgG/IgA类抗体分泌细胞水平(ASC);RT-PCR分析免疫鼠脾脏B细胞趋化因子受体表达,RT-PCR和FCM分析CXCR4表达水平;RT-PCR和ELISA检测脾脏组织CXCL12表达水平。结果显示,pCMV4- hCGβ-C3d3免疫组外周血IgG类抗hCGβ抗体水平明显高于pCMV4-hCGβ免疫组;而IgA类抗hCGβ抗体水平在两组间无明显差异。pCMV4-hCGβ-C3d3免疫组脾脏组织IgG类ASCs水平明显高于pCMV4-hCGβ组;两组间IgA类ASCs水平无明显差异。经pCMV4-hCGB、pCMV4-hCGβ- C3d3免疫鼠脾脏B细胞CXCR4表达明显高于对照组;且pCMV4-hCGβ-C3d3组明显高于pCMV4-hCGβ免疫组。CXCR4~ 细胞与ASCs呈正相关,r=0.966,(P<0.05)。pCMV4-hCGβ-C3d3和pCMV4-hCGβ组脾脏组织CXC L12表达均显著高于对照组。结果表明,分子佐剂C3d3与hCGβ基因融合,在基因免疫小鼠后能够显著升调节脾脏ASCs CXCR4表达,从而可能增强抗hcGβ基因疫苗的体液免疫效应。  相似文献

目的:构建小鼠CDC6基因的RNAi真核表达载体PGCsilencer TM u6/Neo/GFP/RNAi,观察其转染小鼠肝细胞前后CDC6的表达变化。方法:根据GenBank中CDC6的序列,设计特异性siRNA序列,将模板序列克隆至PGCsilencer U6/Neo/GFP质粒中,通过测序鉴定后,用脂质体将重组子转染至正常小鼠肝细胞中,用RT-PCR检测CDC6的mRNA的表达及用Western blot方法检测CDC6蛋白水平的表达,并比较转染前后其表达水平的变化。结果:经测序,模板序列与设计序列完全正确,经过RT-PCR及Western blot方法检测,转染干扰质粒后,小鼠肝细胞中CDC6表达在mRNA及蛋白水平都有明显的下降。结论:成功构建了CDC6基因的RNAi真核表达载体并转染至小鼠肝细胞中,为下一步探讨CDC6在肝再生的作用奠定了基础。  相似文献
