Arabidopsis thaliana histidine phosphotransfer proteins (AHPs) are similar to bacterial and yeast histidine phosphotransfer proteins (HPts), which act in multistep phosphorelay signaling pathways. A phosphorelay pathway is the current model for cytokinin signaling. To assess the role of AHPs in cytokinin signaling, we isolated T-DNA insertions in the five AHP genes that are predicted to encode functional HPts and constructed multiple insertion mutants, including an ahp1,2,3,4,5 quintuple mutant. Single ahp mutants were indistinguishable from wild-type seedlings in cytokinin response assays. However, various higher-order mutants displayed reduced sensitivity to cytokinin in diverse cytokinin assays, indicating both a positive role for AHPs in cytokinin signaling and functional overlap among the AHPs. In contrast with the other four AHPs, AHP4 may play a negative role in some cytokinin responses. The quintuple ahp mutant showed various abnormalities in growth and development, including reduced fertility, increased seed size, reduced vascular development, and a shortened primary root. These data indicate that most of the AHPs are redundant, positive regulators of cytokinin signaling and affect multiple aspects of plant development.  相似文献

Summary Raft membrane domains are envisioned as lateral assemblies of cholesterol and sphingolipids which adopt a liquid-ordered membrane phase. Our understanding of the raft architecture in cell membranes is developing rapidly. The current view describes raft domains as small and highly dynamic subdomains of cell membranes. The size and stability of raft domains are essential parameters which determine the function of raft domains in cells. Astroglia are targets for estrogen and testosterone and are apparently involved in the action of sex steroids on the brain. Sex hormones induce changes in the expression of glial fibrillary acidic protein, the growth of astrocytic processes, and the degree of apposition of astroglial processes to neuronal membranes in the rat hypothalamus. These changes are linked to modifications in the number of synaptic inputs to hypothalamic neurons. These findings suggest that astrocytes may participate in the genesis of androgen-induced sex differences in synaptic connectivity and in estrogen-induced synaptic plasticity in the adult brain. Astrocytes and tanycytes may also participate in the cellular effects of sex steroids by releasing neuroactive substances and by regulating the local accumulation of specific growth factors, such as insulin-like growth factor-I, that are involved in estrogen-induced synaptic plasticity and estrogen-mediated neuroendocrine control. Astroglia may also be involved in regenerative and neuroprotective effects of sex steroids, since astroglia formation after brain injury or after peripheral nerve axotomy is regulated by sex hormones. Furthermore, the expression of aromatase, the enzyme that produces estrogen, is induced de novo in astrocytes in lesioned brain areas of adult male and female rodents. Since astroglia do not express aromatase under normal circumstances, the induction of this enzyme may be part of the program of glial activation to cope with the new conditions of the neural tissue after injury. Given the neuroprotective and growth-promoting effects of estrogen after injury, the local production of this steroid may be a relevant component of the reparative process.  相似文献

We previously reported the cloning of the thousand and one-amino acid protein kinase 1 (TAO1), a rat homolog of the Saccharomyces cerevisiae protein kinase sterile 20 protein. Here we report the complete sequence and properties of a related rat protein kinase TAO2. Like TAO1, recombinant TAO2 selectively activated mitogen-activated protein/extracellular signal-regulated kinase kinases (MEKs) 3, 4, and 6 of the stress-responsive mitogen-activated protein kinase pathways in vitro and copurified with MEK3 endogenous to Sf9 cells. To examine TAO2 interactions with MEKs, the MEK binding domain of TAO2 was localized to an approximately 135-residue sequence just C-terminal to the TAO2 catalytic domain. In vitro this MEK binding domain associated with MEKs 3 and 6 but not MEKs 1, 2, or 4. Using chimeric MEK proteins, we found that the MEK N terminus was sufficient for binding to TAO2. Catalytic activity of full-length TAO2 enhanced its binding to MEKs. However, neither the autophosphorylation of the MEK binding domain of TAO2 nor the activity of MEK itself was required for MEK binding. These results suggest that TAO proteins lie in stress-sensitive kinase cascades and define a mechanism by which these kinases may organize downstream targets.  相似文献

Dietary information was reviewed for 22 bird species typically found in farmland areas across Europe using 100 references from the UK and other European countries for which quantitative data were presented. The number of studies and degree of taxonomic resolution available varied considerably between the species and, for some, dietary information was only available prior to agricultural intensification and may be unrepresentative of current feeding habits. Consequently, some caution is required in interpreting the results because of the different methods of data collection between the studies and the biases in analytical techniques for prey detectability. Food availability was rarely quantified in the study areas, and diet may have reflected availability or preference. Plant families and invertebrate orders were ranked in their importance separately for adult diet in the breeding and nonbreeding seasons and for chicks. The most important plant families overall were Poaceae, Polygonaceae, Caryophyllaceae, Cruciferae, Compositae, Chenopodiaceae and Labiatae, although the last two were unimportant for chicks. A number of key species were also identified. Similarities in dietary preferences were observed among Fringillidae (finches) and members of the Columbidae (doves and pigeons). Fringillidae and Carduelis cannabina (linnet) were associated with plants that occur in seminatural habitats, while Emberiza citrinella (yellowhammer), Phasianidae (partridges) and Columbidae were shown to be more closely associated with plants of agricultural environments. The most important invertebrate orders overall were Coleoptera adults, Hemiptera adults, Arachnida, Lepidoptera larvae/pupae, Diptera adults, Lepidoptera adults, Diptera larvae/pupae and Hymenoptera adults. During the nonbreeding season, a smaller number of invertebrate taxa were important. For adults in the breeding season and for chicks, there were 10 families of invertebrate that were important for four or more bird species (Aphididae, Carabidae, Chrysomelidae, Curculionidae, Elateridae, Formicidae, Scarabeidae, Staphylinidae, Tenthredinidae and Tipulidae). In the nonbreeding season, Curculionidae and Lumbricidae were important in the diet of three species. A few species exhibited a narrow diet range, selecting invertebrates from only one or two families. Similarities in dietary composition were observed among closely related bird species, while that of chicks and adults sometimes differed.  相似文献

Smoking behavior is a complex, which includes multiple stages in the progression from experimentation to continued use and dependence. The experience of subjective effects, such as dizziness, euphoria, heart pounding, nausea and high, have been associated with varying degrees of persistence and subsequent abuse/dependence of marijuana, cocaine, tobacco and alcohol ( Grant et al. 2005 , Wagner & Anthony 2002 ). Previous studies have reported associations between neuronal nicotinic receptor (CHRN) genes and subjective effects to nicotine. We sought to replicate and expand this work by examining eight single nucleotide polymorphisms (SNPs) in a sample of adult smokers (n = 316) who reported subjective effects following cigarette smoking in a controlled laboratory environment. Two SNPs each in the CHRNB2, CHRNB3, CHRNA6 and CHRNA4 genes were examined. A significant association was found between two SNPs and physical effects reported after smoking the first experimental cigarette. SNP rs2072658 is upstream of CHRNB2 (P‐value = 0.0046) and rs2229959 is a synonymous change in exon 5 of CHRNA4 (P value = 0.0051). We also examined possible functional relevance of SNP rs2072658 using an in vitro gene expression assay. These studies provided evidence that the minor allele of rs2072658 may lead to decreased gene expression, using two separate cell lines, P19 and SH‐SY5Y (18% P < 0.001 and 26% P < 0.001 respectively). The human genetic study and functional assays suggest that variation in the promoter region of CHRNB2 gene may be important in mediating levels of expression of the β2 nicotinic receptor subunit, which may be associated with variation in subjective response to nicotine.  相似文献
