肉牛的性成熟受下丘脑-垂体-卵巢(HPO)轴的调节,但其具体作用机制尚不清楚.为了探讨HPO轴繁殖相关基因和调控机制,采用高通量转录组RNA-seq技术,研究了成年安格斯牛(30个月大)的下丘脑、垂体和卵巢.结果表明,HPO共有18 179个基因在GO(基因本体论)中注释,37 158个基因在KEGG(京都基因百科全书和基因组)中注释,36 645个基因在KOG(KEGG同源数据库)中注释.GO分析表明,HPO富集了8种生殖相关功能,主要是受精、单个组织生殖过程、子宫胚胎发育和雌激素反应.KEGG分析发现,HPO富集300条信号通路(SP),主要是癌症SP和磷脂酰肌醇3激酶蛋白激酶(PI3K-Akt)SP.与生殖有关的SPs共有16条,主要是Notch SP、神经营养素SP、哺乳动物雷帕霉素(mTOR)SP和血管内皮生长因子(VEG F)SP.为今后肉牛繁殖性能的提高、分子育种和同行参考提供了科学依据.  相似文献

以1、2、3年生的楸树实生苗和嫁接苗(梓树砧木)根系为研究对象,通过对ITS rDNA区域标记扩增子的Illumina MiSeq测序,分析不同苗龄楸树实生苗和嫁接苗根相关真菌的结构组成和多样性。获得根相关真菌OTU共842个,分属4门、24纲、70目、134科、233属、347种;丛枝菌根真菌(arbuscular mycorrhizal fungi)AMF-OTU共42个,分属1门、1纲、3目、3科、3属、13种。根相关真菌和AMF的OTU数量、丰度和多样性在实生苗中均随苗龄的增加而降低,而在嫁接苗中则随着苗龄的增加而增加。门水平上,实生苗与嫁接苗根相关真菌的优势菌都是子囊菌门Ascomycota、担子菌门Basidiomycota和接合菌门Zygomycota,但它们的相对丰度有所差异;属水平上,实生苗和嫁接苗根相关真菌的优势菌种在组成和数量上都具有一定的差异性。楸树根相关真菌拥有3种营养模式和12个生态功能群,其中实生苗根系中病理营养型真菌的比例大于嫁接苗,腐生营养型则差异不大,而共生营养型则小于嫁接苗。生态功能群分析显示大多数楸树根系真菌表现出多种生存策略,部分真菌可以在植物-真菌-动物中跨界侵染。该研究可为楸树根相关真菌的利用提供一定的理论依据和基础。  相似文献

目的:探讨优化两步输注美罗培南联合参麦注射液对重症感染患者血清感染指标、细菌清除率和T淋巴细胞亚群的影响。方法:选取2017年1月-2018年10月在我院重症医学科(ICU)住院的153例重症感染患者,按随机数字表法将患者分为传统输注美罗培南组(A组,54例)、优化两步输注美罗培南组(B组,49例)、优化两步输注美罗培南联合参麦注射液组(C组,50例)。比较治疗后三组患者的总体疗效、血清感染指标和T淋巴细胞亚群水平及不良反应发生情况。结果:B组、C组的临床有效率、细菌清除率、28天生存率高于A组(P<0.05);B组、C组机械通气时间、ICU住院时间、总住院时间少于A组,且C组少于B组(P<0.05);治疗后B组、C组CD3+、CD4+、CD4+/CD8+高于A组,且C组高于B组(P<0.05);B组、C组CD8+、白细胞计数(WBC)、降钙素原(PCT)、C-反应蛋白(CRP)水平低于A组,且C组低于B组(P<0.05)。三组患者不良反应发生率比较无差异(P>0.05)。结论:优化两步输注美罗培南联合参麦注射液治疗重症感染患者疗效确切,可提高细菌清除率,改善患者免疫状态,促进患者康复。  相似文献

以花脸香蘑Lepista sordida为材料,研究其分别在9种碳源和11种氮源液体培养条件下不同阶段pH值和葡萄糖浓度的变化,以及不同碳氮源对其所分泌的木质素过氧化物酶、羧甲基纤维素酶、锰过氧化物酶和漆酶活性的影响。结果表明,pH值在不同碳源培养后期变化显著(P<0.05),而在不同氮源培养阶段无明显变化(P>0.05),葡萄糖浓度和菌丝量在不同碳氮源中也无显著差异(P<0.05)。不同碳源和氮源培养基对花脸香蘑木质素过氧化物酶、羧甲基纤维素酶、锰过氧化物酶和漆酶活性均具有影响(P<0.05)。木糖和米糠有利于花脸香蘑分泌羧甲基纤维素酶(P<0.05),红糖和牛肉浸膏有利于其分泌漆酶(P<0.05),白砂糖和豆粉有利于其分泌锰过氧化物酶(P<0.05),木糖和尿素有利于其分泌木质素过氧化物酶(P<0.05)。本研究为选择合适培养基以提高花脸香蘑生物转化效率提供了理论基础。  相似文献

开展米槁群落物种生态位研究,可了解米槁群落物种对资源的利用特征,为米槁的保护与管理提供科学依据。基于贵州省米槁天然林群落的实际调查数据,通过生态位宽度、生态位重叠指数,分析该群落主要物种的资源利用特征。群落中米槁(Cinnamomum migao)、杜茎山(Maesa japonica)、菝葜(Smilax china)的生态位宽度最大,在群落中处于明显优势地位,具有较强竞争力和资源利用能力;生态位宽度越大的物种间生态位重叠越大,但在相同或相似环境条件下,生态位宽度值小的种对也会拥有较高的生态位重叠;米槁群落主要物种间的生态位重叠指数偏低,说明物种间对资源利用的相似性较低,物种的生态位分化明显,竞争不激烈,群落结构较为稳定,物种间能够较好地相互共存。  相似文献

A new species of rugose coral, Sanidophyllum dubium n. sp., and the typical Emsian (Early Devonian) rugose coral Xystriphylloides nobilis are described from the Mia Le Formation in northern Vietnam. The lower Emsian index conodonts ranging from the Polygnathus excavatus zone to the P. nothoperbonus zone are illustrated. The biostratigraphic correlation between northern Vietnam and South China shows that the Mia Le Formation in northern Vietnam is early Emsian in age, and its upper part can be correlated with the lower part of the Shizhou Member of the Yukiang Formation in Liujing, Guangxi and its equivalents in South China. Based on the study of the lower Emsian biostratigraphic sequence, the disappearance of Xystriphylloides nobilis fauna in the overlying bed of the uppermost Mia Le Formation and the extinction of the “tonkinensis fauna” (sensu lato) in the interval between the basal Si Phai Formation and the uppermost Mia Le Formation demonstrate the influence of the Yujiang Event in northern Vietnam.  相似文献

The intestinal barrier dysfunction is crucial for the development of liver fibrosis but can be disturbed by intestinal chronic inflammation characterized with cyclooxygenase-2 (COX-2) expression. This study focused on the unknown mechanism by which COX-2 regulates intestinal epithelial homeostasis in liver fibrosis. The animal models of liver fibrosis induced with TAA were established in rats and in intestinal epithelial–specific COX-2 knockout mice. The impacts of COX-2 on intestinal epithelial homeostasis via suppressing β-catenin signalling pathway were verified pharmacologically and genetically in vivo. A similar assumption was tested in Ls174T cells with goblet cell phenotype in vitro. Firstly, disruption of intestinal epithelial homeostasis in cirrhotic rats was ameliorated by celecoxib, a selective COX-2 inhibitor. Then, β-catenin signalling pathway in cirrhotic rats was associated with the activation of COX-2. Furthermore, intestinal epithelial–specific COX-2 knockout could suppress β-catenin signalling pathway and restore the disruption of ileal epithelial homeostasis in cirrhotic mice. Moreover, the effect of COX-2/PGE2 was dependent on the β-catenin signalling pathway in Ls174T cells. Therefore, inhibition of COX-2 may enhance intestinal epithelial homeostasis via suppression of the β-catenin signalling pathway in liver fibrosis.  相似文献

Alzheimer''s disease (AD) is a leading cause of dementia in elderly individuals and therapeutic options for AD are very limited. Over‐activation of N‐methyl‐D‐aspartate (NMDA) receptors, amyloid β (Aβ) aggregation, a decrease in cerebral blood flow (CBF), and downstream pathological events play important roles in the disease progression of AD. In the present study, MN‐08, a novel memantine nitrate, was found to inhibit Aβ accumulation, prevent neuronal and dendritic spine loss, and consequently attenuate cognitive deficits in 2‐month‐old APP/PS1 transgenic mice (for a 6‐month preventative course) and in the 8‐month‐old triple‐transgenic (3×Tg‐AD) mice (for a 4‐month therapeutic course). In vitro, MN‐08 could bind to and antagonize NMDA receptors, inhibit the calcium influx, and reverse the dysregulations of ERK and PI3K/Akt/GSK3β pathway, subsequently preventing glutamate‐induced neuronal loss. In addition, MN‐08 had favorable pharmacokinetics, blood‐brain barrier penetration, and safety profiles in rats and beagle dogs. These findings suggest that the novel memantine nitrate MN‐08 may be a useful therapeutic agent for AD.  相似文献

Amyloid‐beta (Aβ) oligomer is known to contribute to the pathophysiology of age‐related macular degeneration. Herein, we aimed to elucidate the in vivo and in vitro effects of Aβ1‐42 application on retinal morphology in rats. Our in vivo studies revealed that intracerebroventricular administration of Aβ1‐42 oligomer caused dysmorphological changes in both retinal ganglion cells and retinal pigment epithelium. In addition, in vitro studies revealed that ARPE‐19 cells following Aβ1‐42 oligomer application had decreased viability along with apoptosis and decreased expression of the tight junction proteins, increased expression of both phosphor‐AKT and phosphor‐GSK3β and decreased expression of both SIRT1 and β‐catenin. Application of conditioned medium (CM) obtained from mesenchymal stem cells (MSC) protected against Aβ1‐42 oligomer‐induced retinal pathology in both rats and ARPE‐19 cells. In order to explore the potential role of peptides secreted from the MSCs, we applied mass spectrometry to compare the peptidomics profiles of the MSC‐CM. Gene ontology enrichment analysis and String analysis were performed to explore the differentially expressed peptides by predicting the functions of their precursor proteins. Bioinformatics analysis showed that 3‐8 out of 155–163 proteins in the MSC‐CM maybe associated with SIRT1/pAKT/pGSK3β/β‐catenin, tight junction proteins, and apoptosis pathway. In particular, the secretomes information on the MSC‐CM may be helpful for the prevention and treatment of retinal pathology in age‐related macular degeneration.  相似文献
