Cholinesterase activity is known in representatives of all living organisms phyla but the origin of the cholinergic system as known in bilaterian animals is still undeciphered. In particular the implication of cholinesterases in the nervous system of non-bilaterian Metazoa is not well known. We thus chose to investigate this activity in the Clytia hemisphaerica (Cnidaria) medusa. In toto histochemical staining revealed an acetylcholinesterase activity in the tentacle bulbs but not in the nervous system. Sequences homologous to acetylcholinesterase were searched within Clytia ESTs and compared to other sequences found in public databases.  相似文献

Cakile maritima is a halophyte with potential for ecological, economical and medicinal uses. We address here the impact of salinity on its growth, photosynthesis and seed quality. Whole plant growth rate and shoot development were stimulated at moderate salinity (100–200 m M NaCl) and inhibited at higher salt concentrations. Although diminished in the presence of salt, potassium and calcium uptake per unit of root biomass was maintained at relatively high value, while nutrient-use efficiency (NUE) was improved in salt-treated plants. Chl and carotenoid concentrations decreased at extreme salinities, but anthocyanin concentration continuously grew with salinity. Net photosynthetic rate (A), stomatal conductance, maximum quantum efficiency of PSII and quantum yield were stimulated in the 100–200 m M NaCl range. Higher salinity adversely affected gas exchange and changed PSII functional characteristics, resulting in a reduction of A per leaf area unit. This phenomenon was associated with increased non-photochemical quenching. Harvest index, silique number and seeds per fruit valve were maximal at 100 m M NaCl. Despite the decreasing salt accumulation gradient from the vegetative to the reproductive organs, high salinities were detrimental for the seed viability and increased the proportion of empty siliques. Overall, the salt-induced changes in the plant photosynthetic activity resulted into analogous responses at the vegetative and reproductive stages. The enhancement of NUE, the absence of pigment degradation, the reduction of water loss and the concomitant PSII protection from photodamage through thermal dissipation of excess excitation significantly accounted for Cakile survival capacity at high salinity.  相似文献

Zusammenfassung Eier von Sperbern aus Brandenburg und aus Nordhrein-Westfalen wurden auf ihren Gehalt an PCB, DDT, HCH, HCB und die Schwermetalle Blei und Cadmium untersucht. Bei den PCB und beim DDT und seinen Abbauprodukten traten signifikante Unterschiede in der Belastung der beiden Populationen auf, während die anderen untersuchten Stoffe in den Eiern beider Untersuchungsgebiete nur in geringen Mengen nachzuweisen waren. Dabei waren in den Proben aus der ehemaligen DDR die DDT-Konzentrationen deutlich höher als in den Eiern aus dem westfälischen Münsterland. Bei den PCB-Konzentrationen ergab sich ein genau umgekehrtes Ergebnis, im Münsterland waren diese erheblich höher. Die Ergebnisse stehen im klaren Zusammenhang mit der unterschiedlichen Anwendung und dem zeitlich unterschiedlich erfolgtem Verbot der untersuchten Schadstoffe in den beiden ehemaligen deutschen Staaten. Trotz der Belastungsunterschiede bezüglich der PCB ist das PCB-Muster in den Eiern aus den Untersuchungsgebieten aufgrund des artspezifischen Metabolismus nahezu identisch.
Comparison between the burden of xenobiotics in eggs of the Sparrowhawk (Accipiter nisus) from Brandenburg and North-Rhine-Westphalia, Germany
Summary Eggs from Sparrowhawks from Brandenburg (former GDR) and North-Rhine-Westphalia (former FRG) were examined for their burden of PCB, DDT, HCH, HCB and the heavy metals lead and cadmium. Significant differences emerged in the cases of PCB and DDT and its metabolites between the populations of the two regions. The other examined xenobiotics were detectable only in low concentrations. The DDT concentrations were distinctly higher in the samples from the former GDR, while PCB concentrations were higher in the eggs from North-Rhine-Westphalia. The results reflect clearly the different use and prohibition of the examined xenobiotics in the two former german states. In spite of the differences in the PCB burden, the PCB pattern in the eggs of the different regions is nearly identical. Eggshell indices correlate negatively with DDT burden in both sample areas.

Summary Adhesive interactions of trophoblast cells with the endometrium are essential for embryo implantation in the uterus. Choriocarcinoma cells, the malignant counterpart of trophoblast, show pronounced invasiveness and are of interest for model studies. We describe here an in vitro model system for the study of adhesion of human JAR choriocarcinoma multicellular spheroids to different human endometrial epithelial cell lines (RL95-2, HEC-1A, KLE, AN3-CA) grown as monolayers. Cell characterization showed JAR spheroids to secrete the placental hormones human chorionic gonadotropin and progesterone into the culture medium; distinct patterns of keratin, vimentin, and uvomorulin expression were seen in the endometrial cell lines. Spheroid attachment to endometrial monolayers was quantified using a centrifugal force-based adhesion assay, and morphology was examined by light and electron microscopy. Results showed the JAR spheroids to attach to three of the endometrial monolayers (RL95-2, HEC-1A, KLE) progressively over a 24-h period (by which time ≥80% of the spheroids attached). Significant differences in spheroid attachment were most pronounced at 5 h (RL95-2 > HEC-1A > KLE and poly-d-lysine control, i.e. 90:45:17:17% attached). JAR spheroids did not attach to the endometrial cell line AN3-CA. Morphology revealed choriocarcinoma cells to begin to intrude between the uterine RL95-2 epithelial cells at 5 h. At 24 h, this intrusive type of penetration continued to be seen only with the RL95-2 monolayer. The assay system thus identifies differences in attachment properties between choriocarcinoma cells and various endometrial cell lines and forms the basis for further studies on the molecular interactions involved.  相似文献

The 5' exon donor in nematode trans-splicing, the SL RNA, is a small (approximately 100 nt) RNA that resembles cis-spliceosomal U snRNAs. Extensive analyses of the RNA sequence requirements for SL RNA function have revealed four essential elements, the core Sm binding site, three nucleotides immediately downstream of this site, a region of Stem-loop II, and a 5' splice site. Although these elements are necessary and sufficient for SL RNA function in vitro, their respective roles in promoting SL RNA activity have not been elucidated. Furthermore, although it has been shown that assembly of the SL RNA into an Sm RNP is a prerequisite for function, the protein composition of the SL RNP has not been determined. Here, we have used oligoribonucleotide affinity to purify the SL RNP and find that it contains core Sm proteins as well as four specific proteins (175, 40, 30, and 28 kDa). Using in vitro assembly assays; we show that association of the 175- and 30-kDa SL-specific proteins correlates with SL RNP function in trans-splicing. Binding of these proteins depends upon the sequence of the core Sm binding site; SL RNAs containing the U1 snRNA Sm binding site assemble into Sm RNPs that contain core, but not SL-specific proteins. Furthermore, mutational and thiophosphate interference approaches reveal that both the primary nucleotide sequence and a specific phosphate oxygen within a segment of Stemloop II of the SL RNA are required for function. Finally, mutational activation of an unusual cryptic 5' splice site within the SL sequence itself suggests that U5 snRNA may play a primary role in selecting and specifying the 5' splice site in SL addition trans-splicing.  相似文献

Eusynaptomyces benjaminii is described as a new species of the ectoparasiticLaboulbeniales (Ascomycetes). It exists only on two very restricted areas of the body (= position specifity) of its hostEnochrus testaceus (F.) (Coleoptera, Hydrophilidae): on the claws of the right fore-leg and on the lower side of the frontal border of the pronotum. In these two habitatsEu. benjaminii develops two extremely different growth-forms. Male and female hosts are parasitized on somewhat different parts of their body. This can be explained by their mating behaviour. The growth-forms ofEu. benjaminii are so different that one ignorant of the biology of hosts and parasites, might regard them as members of different species or even genera. They are to be interpreted as adaptations of one species to growth positions and mating behaviour of the host. There is no sex-of-host specifity as assumed by certain authors for several species of theLaboulbeniales.

For the federal state of Baden-Wiirttemberg, Germany, the decision tool “Umweltbilanz von Altlastensanierungsverfahren” has been developed and found suitable for the quantification and evaluation of environmental impacts caused by remediation of contaminated sites. The developed tool complements the remediation toolbox of Baden-Wiirttemberg. The tool includes a streamlined life cycle assessment (LCA) and a synopsis of the LCA results with the results of a risk assessment of the contaminated site. The risk assessment tool is not explained here. The data base for the life cycle inventory includes several techniques used in remedial actions. The life cycle impact assessment utilises 14 impact categories. The method allows comparisons between remedial options for specific contaminated sites. A software tool has been developed to be available in 1999.  相似文献

Guanine nucleotide binding proteins (G proteins) mediate a variety of cellular responses to external stimuli. Pure G protein, receptor, and effector are sufficient to reconstitute hormonal activation of an effector in phospholipid vesicles, but other components may be important for specificity or localization in vivo. If another protein associates with GO, the molecular weight of GO solubilized from membranes would be larger than the molecular weight of GO after purification. We find that GO solubilized from bovine brain membranes by Triton X-100 behaves as a single population of molecules on sucrose density gradients and gel filtration columns. Its molecular mass is about 40 kDa larger than pure GO. Association of GO with the other protein is fragile as the proteins dissociate on further purification. There was no difference in ADP-ribosylation or tryptic cleavage of GO in larger and smaller form. These studies provide a basis for future experiments to stabilize the interaction and identify the protein.  相似文献
