ABSTRACT. The sesquiterpenoid euplotin C is a secondary metabolite produced by the ciliated protist Euplotes crassus and provides a mechanism for damping populations of potential competitors. Indeed, E. crassus is virtually resistant to its own product while different non-producer species representing an unbiased sample of the marine, interstitial, ciliate diversity are sensitive. For instance, euplotin C exerts a marked disruption of different homeostatic mechanisms in Euplotes vannus . We demonstrate by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay that euplotin C quickly decreases viability and mitochondrial function of E. vannus with a very high efficacy and at micromolar potency. In addition, euplotin C induces apoptosis in E. vannus as 4,6-diamino-2-phenylindole and terminal transferase dUTP nick end labeling staining show the rapid condensation and fragmentation of nuclear material in cells treated with euplotin C. These effects occur without detectable permeabilisation or rupture of cell membranes and with no major changes in the overall morphology, although some traits, such as vacuolisation and disorganized microtubules, can be observed by transmission electron microscopy. In particular, E. vannus show profound changes of the mitochondrial ultrastructure. Finally, we also show that caspase activity in E. vannus is increased by euplotin C. These data elucidate the pro-apoptotic role of euplotin C and suggest a mechanism for its impact on natural selection.  相似文献

The recognition of broad biogeographic provinces provides an important framework for ecological and conservation biological research. Marine biologists have long recognized distinct biogeographic provinces in southern Australia, primarily on the basis of qualitative differences in intertidal species assemblages. Here we provide an a priori test for these traditional eastern (Peronian), western (Flindersian) and south‐eastern (Maugean) provinces. Specifically, we analyse distributional data for approximately 1500 algal species using the newly available Australian Virtual Herbarium, an online database of herbarium specimens. Our quantitative algal analyses across southern Australia identify three distinct biogeographic assemblages, consistent with traditional qualitative provinces. We argue that these broad provinces provide a highly effective framework for understanding and managing Australia's marine biodiversity. In particular, biogeographic provinces provide a regional framework for integrating the ongoing discovery of biological variation at finer scales. More broadly therefore we recommend that biologists undertake quantitative analyses to test provincial biogeographic boundaries around the globe.  相似文献

The Corsica–Sardinia archipelago is a hotspot of Mediterranean biodiversity. Although tempo and mode of arrival of species to this archipelago and phylogenetic relationships with continental species have been investigated in many taxa, very little is known about the current genetic structure and evolutionary history subsequent to arrival. In the present study, we investigated genetic variation within and among populations of the Tyrrhenian treefrog Hyla sarda, a species endemic to the Corsica–Sardinia microplate and the surrounding islands, by means of allozyme electrophoresis. Low genetic divergence (mean Dnei = 0.01) and no appreciable differences in the levels and distribution of genetic variability (HE: 0.06–0.09) were observed among all but one populations (Elba). Historical demographic and isolation‐by‐distance analyses indicated that this diffused genetic homogeneity could be the result of recent demographic expansion. Along with paleoenvironmental data, such expansion could have occurred during the last glacial phase, when wide and persistent land bridges connected the main islands and a widening of lowland areas occurred. This scenario is unprecedented among Corsica–Sardinia species. Together with the lack of concordance even among the few previously studied species, this suggests either that species had largely independent responses to paleoenvironmental changes, or that most of the history of assembly of the Corsica–Sardinia biota is yet to be written. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 103 , 159–167.  相似文献

A morphometric study on five populations of the Onosma echioides complex was carried out through univariate analysis, principal coordinate analysis and cluster analysis (the unweighted pair‐group method with arithmetic mean; UPGMA). In consideration of the quantitative morphological variability, of the known karyological homogeneity and of the allopatric distribution of the taxa, we propose here to treat this complex as a single species subdivided in four subspecies. The names O. angustifolia Lehm., O. canescens J. et C. Presl, O. dalmatica Scheele, O. echioides var. columnae Lacaita, O. echioides var. veronensis Lacaita and O. javorkae Simonkai are typified. The new combinations O. echioides subsp. angustifolia (Lehm.) Peruzzi & N. G. Passal., O. echioides subsp. canescens (J. et C. Presl) Peruzzi & N. G. Most geophilomorph centipedes have segmental clusters of exocrine glands whose opening pores are arranged in more or less well-defined sternal pore areas. We describe here the cuticular structures forming and/or accompanying the gland openings on the sternites and the shape of the pore areas along the body axis in representatives of most geophilomorph families. The cuticular ring around the pore may exhibit either of two forms. In Himantariidae ( Himantarium ) and in Dignathodontidae ( Henia ) the ring looks like a continuous ribbon with a visible suture, whereas in the representatives of the remaining families no suture is seen. As to the distribution of the pores on ventral surface of the body, we record the presence of pores on the last leg-bearing segment of Clinopodes flavidus , whereas that segment was described as poreless in all geophilomorphs. We also provide a taxonomic survey of shape and distribution of pore areas in the individual families, where the pore areas may take very different shapes that we regard as transformational homologues. As for the segmental distribution of sternal pore areas, there is a considerable amount of complexity along the trunk of geophilomorph centipedes, in contrast to the apparently uniform trunk structure.  相似文献

High pressure liquid chromatography with electrochemical detection (HPLC-ED) was employed in conjugation with a sensitive and specific salicylate hydroxylation assay to evaluate the immediate effects of hydroxyl radical (·OH) attack on four catechol intermediates of eumelanin, dopamine (3,4-dihydroxyphenylethylamine), its precursor dopa (3,4-dihydroxyphenylalanine), and their respective neurotoxic trihydroxyphenyl derivatives, 6-hydroxydopamine (2,4,5-trihydroxyphenylethylamine,6-OHDA) and 6-hydroxydopa(2,4,5-trihydroxyphenylalanine, TOPA). Semiquinone and quinone species were identified as the initial products of the oxidation of these four catechol substrates. The enhanced oxidations of the catechols when exposed to ·OH attack was accompanied by marked decreases in the level of each semiquinone species. Quinone levels were elevated in reactions involving ·OH attack on dopamine and 6-OHDA, but absent in reactions involving radical attack on dopa or TOPA, suggesting that dopaquinone (DOQ) and TOPA p-quinone (TOPA p-Q) are oxidized more rapidly by‘OH than are the quinones of dopamine and 6-OHDA. The formation of 6-OHDA p-quinone (6-OHDA p-Q) in incubations involving DA and ·OH suggest that the ·OH-mediated hydroxylation of DA may be a mechanism for generating this potentially cytotoxic trihydroxyphenyl. The results of this study demonstrate for the first time that semiquinone and quinone intermediates of eumelanin are the initial products derived from the ·OH-mediated oxidations of dopa, DA, TOPA, and 6-OHDA. These observations suggest that if ·OH is generated beyond the capabilities of cytoprotective mechanisms, the radical can rapidly oxidize catechol precursors, augment melanogenesis, and generate additional cytotoxic quinoid intermediates of eumelanin.  相似文献

ABSTRACT. The systematics of the green algal class Ulvophyceae have been difficult to resolve with ultrastructural and molecular phylogenetic analyses. Therefore, we investigated relationships among ulvophycean orders by determining the distribution of two discrete genetic characters previously identified only in the order Dasycladales. First, Acetabularia acetabulum uses the core translation GTPase Elongation Factor 1α (EF-1α) while most Chlorophyta instead possess the related GTPase Elongation Factor-Like (EFL). Second, the nuclear genomes of dasycladaleans A. acetabulum and Batophora oerstedii use a rare non-canonical genetic code in which the canonical termination codons TAA and TAG instead encode glutamine. Representatives of Ulvales and Ulotrichales were found to encode EFL, while Caulerpales, Dasycladales, Siphonocladales, and Ignatius tetrasporus were found to encode EF-1α, in congruence with the two major lineages previously proposed for the Ulvophyceae. The EF-1α of I. tetrasporus supports its relationship with Caulerpales/Dasycladales/Siphonocladales, in agreement with ultrastructural evidence, but contrary to certain small subunit rRNA analyses that place it with Ulvales/Ulotrichales. The same non-canonical genetic code previously described in A. acetabulum was observed in EF-1α sequences from Parvocaulis pusillus (Dasycladales), Chaetomorpha coliformis , and Cladophora cf. crinalis (Siphonocladales), whereas Caulerpales use the universal code. This supports a sister relationship between Siphonocladales and Dasycladales and further refines our understanding of ulvophycean phylogeny.  相似文献

Two new Italian species of the genus Islamia (Prosobranchia:Hydrobiidae), one living in eastern Sicily (I. cianensis), andone on Elba Island (Tuscan Archipelago, Italy) (I. gaiteri)are described. The two species are distinguished on the basisof shell and anatomical characters, mainly those of the malegenitalia. I. cianesis n. sp. is characterized by a valvatoidshell and a penial lobe with internal band of glandular tissuenot distinct in its lower portion from the penis body but bulgingapically as a small knob. Measurements were made of respiration in the roots of tomato cultivars susceptible and resistant to Meloidogyne incognita. Nematode infestation of the susceptible cv. Roma VF caused an initial stimulation of the total respiration with an increase in the CN-sensitivity; the effect decreased and then disappeared as the seedlings aged. In the resistant cv. Rossol nematode infestation initially caused an inhibition of total respiration and a decrease in the CN-sensitivity; respiration and CN-sensitivity increased with seedling age. The activity of the alternative respiration pathway (m-chlorobenzhydroxamic acid-sensitive) was unchanged by nematode infestation in the resistant cultivar and increased markedly in the susceptible cultivar during the early stages of infestation.  相似文献
