The aim of this work was to evaluate the ability of a line selected for reproductive longevity (LP) to confront productive challenges compared to a line selected during 31 generations for litter size at weaning (V). A total of 133 reproductive rabbit does were used (72 and 61 from LP and V lines, respectively). Within each line, three groups with different levels of productive effort were planned: PP9, inseminated at day 4 after the first partum and with nine kits during the second partum, and inseminated after first weaning (30 days) and with nine (PW9) or five kits (PW5) during the second partum. The reproductive performance, body condition (perirrenal fat thickness (PFT)) and lipolytic response were controlled. LP does showed greater mean live weight (LW; +128 g; P < 0.05), PFT (+0.47 mm; P < 0.05) and estimated body energy (EBE; +0.29 MJ/kg; P < 0.01) than V does at second partum. However, LP does that mated at first post partum did not significantly differ in EBE relative to V does at second partum. During the first week of lactation, dry matter (DM) intake was similar for both lines (94 and 95 g DM/kg LW0.75 day for V and LP does, respectively). There was a significant difference in milk yield between both lines during the first week when litter size was nine (60 v. 54 g of milk/kg LW0.75 day for LP and V does, respectively; P < 0.01), but no difference when litter size was five. Consequently, when litter size was nine, LP does showed a lower recovery of PFT (0.6 mm less; P < 0.05) than V does during the first 10 days of lactation. However, when litter size was five, LP does showed a higher LW (+210 g; P < 0.05) than V does at 10 days of lactation and a similar recovery of PFT. During the last 3 weeks of the lactation, LP does showed a higher feed intake (+6 g DM/kg LW0.75 day; P < 0.05) and milk yield (+27 g/day; P < 0.001) than V does when litter size was nine, resulting in no significant differences in LW at 30 days of lactation. However, when litter size was five, both lines showed similar feed intake and milk yield, maintaining their differences in LW at 30 days of lactation (+206 g for LP does; P < 0.05). These results show that the rabbit line selected for reproductive longevity is more robust with respect to coping with productive challenges, than a line selected for reproductive intensity.  相似文献

Anthropogenic disturbance may lead to the spread of vector-borne diseases through effects on pathogens, vectors, and hosts. Identifying the type and extent of vector response to habitat change will enable better and more accurate management strategies for anthropogenic disease spread. We compiled and analyzed data from published empirical studies to test for patterns among flea and small mammal diversity, abundance, several measures of flea infestation, and host specificity in 70 small mammal communities of five biomes and three levels of human disturbance: remote/wild areas, agricultural areas, and urban areas. Ten of 12 mammal and flea characteristics showed a significant effect of disturbance category (six), biome (four), or both (two). Six variables had a significant interaction effect. For mammal-flea communities in forest habitats (39 of the 70 communities), disturbance affected all 12 characteristics. Overall, flea and mammal richness were higher in remote versus urban sites. Most measures of flea infestation, including percent of infested mammals and fleas/mammal and fleas/mammal species increased with increasing disturbance or peaked at intermediate levels of disturbance. In addition, host use increased, and the number of specialist fleas decreased, as human disturbance increased. Of the three most common biomes (forest, grassland/savanna, desert), deserts were most sensitive to disturbance. Finally, sites of intermediate disturbance were most diverse and exhibited characteristics associated with increased disease spread. Anthropogenic disturbance was associated with conditions conducive to increased transmission of flea-borne diseases.  相似文献

BACKGROUND: 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) exposure prior to chick embryo incubation (GD 0) induces dilated cardiomyopathy, and reduces myocardial hypoxia, vascular endothelial growth factor A (VEGF-A) expression, and coronary vascularization. We investigated whether reduced coronary vascularization 1) occurs in the absence of changes in cardiac morphology and 2) is associated with altered secretion of VEGF-A and/or an antivasculogenic factor. METHODS: Chicken eggs were treated with control (corn oil) or TCDD (0.075-0.3 pmol of TCDD/gm) on GD 5. In vivo cardiac morphology and artery number were determined on GD 10, while in vitro vascular outgrowth and VEGF-A secretion were determined from cardiac explants on GD 6. Effects of recombinant VEGF-A (rcVEGF-A), soluble flt-1 (sFlt-1) receptor plus rcVEGF-A, and control conditioned media were assessed in TCDD explants, while effects of TCDD-conditioned media was assessed in control explants. RESULTS: TCDD reduced coronary artery number in vivo by 53 +/- 8% and induced a dose-related reduction in tube outgrowth in vitro, but had no effect on cardiac morphology. All TCDD doses reduced explant VEGF-A secretion equally (43 +/- 3%), compared to control. sFlt-1 blocked outgrowth in control cultures and blocked rcVEGF-A-mediated rescue of outgrowth in TCDD explants. Control conditioned media partially rescued outgrowth from TCDD explants, while conditioned media from TCDD explants had no effect on controls. CONCLUSIONS: TCDD inhibition of coronary vascularization can occur in the absence of changes in cardiac morphology and is associated with reduced VEGF-A secretion but not an antivasculogenic factor. Since control media only partly rescues TCDD's inhibitory effect, we suggest that TCDD-exposed endothelial cells are less responsive to vasculogenic stimuli.  相似文献

For several years progesterone in milk or blood has been recognized as an indicator of different cow states related to reproduction. For this study, an existing simulation model was modified in order to analyze the technical and economic effects of including information on progesterone status in an automatic inline monitoring system. Implementation of an inline progesterone indicator was assumed to directly affect the estrus detection rate, the period until treatment for post-partum anestrus and the number of mistimed AIs. Different implementations of an inline progesterone indicator were simulated in a typical Danish dairy herd with 120 cows and in other herd situations represented by: a herd with poor reproduction efficiency, a herd with a high estrus detection rate and a herd using a 9 week postponed AI period for primiparous cows. It was concluded that implementation of an inline progesterone indicator in a dairy herd previously using visual estrus detection has a break-even price of 3-81 euros per cow-year depending on differences in implementation type and herd reproduction management. The highest break-even price was found using the assumptions that simulated a herd with initially poor reproductive efficiency. With the assumptions that simulated a typical Danish herd the break-even price was 46 euros per cow-year. Attributable effects of using the indicator, including effects of labor time, are discussed.  相似文献

Reproductive management, in particular timely oestrus detection, is important for profitable dairy production. The aim of this study was to develop a biological model to predict reproductive state on the basis of milk progesterone measures. A number of additional inputs were incorporated to make use of other known effectors of reproductive performance that are not reflected in progesterone levels. These are: days from calving, breed, parity, signs of behavioural oestrus, insemination dates, pregnancy determinations, energy status, body fat status, milk urea content and reproductive disorders associated with calving. A dynamic, deterministic model was developed. It is designed to run each time a new trigger input (progesterone, behavioural oestrus, inseminations, pregnancy determinations) occurs using the current and previous values and can run in the absence of the additional inputs. The milk progesterone values are smoothed using an extended Kalman filter before being processed in the biological component of the model. The model predicts the reproductive status of the cow, which can be one of three mutually exclusive states: postpartum anoestrus, oestrus cycling, and potentially pregnant. The other model outputs are all reproductive status specific with the exception of days to next sample (DNS), which is calculated in each model run regardless of reproductive status. DNS is designed to feedback to the sampling system so that the frequency of milk sampling (i.e. progesterone measurement) can be varied according to the predicted likelihood of a future reproductive event, such as onset of oestrus cycling. The other model outputs are: risk of prolonged postpartum anoestrus, risk and type of ovarian cyst, onset of oestrus, likelihood of a potential insemination succeeding, and likelihood of being pregnant (following oestrus). The model was evaluated using three simulated datasets consisting of a timeseries of progesterone values centred on each of the three reproductive statuses and including relevant additional information. Test runs were carried out on the full datasets and then on reduced data. The data reductions were made by using only those values that would have been available if the model days to next sample function was used to control sampling frequency. The sensitivity of the model to noise in the raw progesterone data was examined by adding 1, 2, or 3 residual standard deviations (1.85 ng/ml) random variation to the original data and evaluating model performance. The model was found to be able to readily identify and distinguish reproductive states. A reduction in sampling frequency to 36% of original sample resulted in an average increase in days to detection of oestrus of 0.36. The addition of 1 S.D. noise did not cause additional oestruses to be detected and all oestruses were correctly identified. However, when 2 or 3 S.D. noise were added, the model found on average 1.4 and 3 extra oestruses. It was concluded that reproductive status can be predicted from milk progesterone values using a biological model and that such a model is robust to reductions in sampling frequency number and to a doubling in the random variation in the raw progesterone values. It therefore has the potential to provide the basis for a useful reproductive management tool.  相似文献

Involvement of rabphilin-3A-like (RPH3AL), or Noc2, the potential effector of Ras-associated binding proteins Rab3A and Rab27A in the regulation of exocytotic processes in the endocrine pancreas has been demonstrated in experimental models. Noc2 expression together with other regulatory molecules of the exocytotic machinery in human tissues, however, has not been studied. We evaluated immunohistochemical expression of the key molecules of the exocytotic machinery, Noc2, Rab3A, Rab27A, and RIM2, together with the characteristic islet cell hormones, insulin and glucagon in normal and endocrine tumor tissues of human pancreas. Normal pancreatic islets were stained for all of these proteins and showed strong cytoplasmic localization. A similar pattern of strong cytoplasmic expression of these proteins was observed in the majority of endocrine tumors. By contrast, the exocrine portions of normal appearing pancreas completely lacked Rab27A staining and showed decreased expression of the proteins, Noc2, Rab3A, and RIM2. The staining pattern of Noc2 and Rab27A was similar to the staining pattern of glucagon-producing cells within the islets. The concomitant expression of Noc2 with these molecules suggests that Noc2 may serve as an effector for Rab3A and Rab27A and that it is involved in the regulation of exocytosis of the endocrine pancreas in humans.  相似文献

Aims

In the Swedish sub-Arctic, mountain birch (Betula pubescens ssp. czerepanovii) forests mediate rapid soil C cycling relative to adjacent tundra heaths, but little is known about the role of individual trees within forests. Here we investigate the spatial extent over which trees influence soil processes.

Methods

We measured respiration, soil C stocks, root and mycorrhizal productivity and fungi:bacteria ratios at fine spatial scales along 3 m transects extending radially from mountain birch trees in a sub-Arctic ecotone forest. Root and mycorrhizal productivity was quantified using in-growth techniques and fungi:bacteria ratios were determined by qPCR.

Results

Neither respiration, nor root and mycorrhizal production, varied along transects. Fungi:bacteria ratios, soil organic C stocks and standing litter declined with increasing distance from trees.

Conclusions

As 3 m is half the average size of forest gaps, these findings suggest that forest soil environments are efficiently explored by roots and associated mycorrhizal networks of B. pubescens. Individual trees exert influence substantially away from their base, creating more uniform distributions of root, mycorrhizal and bacterial activity than expected. However, overall rates of soil C accumulation do vary with distance from trees, with potential implications for spatio-temporal soil organic matter dynamics and net ecosystem C sequestration.

In this study, features of progesterone profiles were examined in relation to the outcome of insemination. Three groups of estrous cycles were analyzed: resulting in pregnancy, not resulting in pregnancy and resulting in lost pregnancy. The aim of the study was to identify a complex of progesterone profile features associated with successful insemination. The features used were (1) from the estrous cycle preceding the artificial insemination: estrus progesterone concentration, post-estrus maximum rate of increase in progesterone, luteal phase peak, pre-estrus maximum rate of decline in progesterone and the length of follicular and luteal phase and (2) from the estrous cycle following insemination: estrus progesterone concentration, post-estrus maximum rate of increase in progesterone and days from estrus to post-estrus maximum rate of increase in progesterone. A discriminant analysis did not reveal clear differences between the groups. However, the analysis correctly classified 75% of true pregnant cows. Conversely, only 60% of not pregnant animals were classified as such by the discriminate analysis. Individual analysis of progesterone profile features in pregnant and not pregnant groups of estrous cycles showed that a shorter follicular phase preceding insemination is associated with proper timing of post-ovulatory luteinisation and therefore is more likely to result in pregnancy.  相似文献

Climate models suggest that extreme rainfall events will become more common with increased atmospheric warming. Consequently, changes in the size and frequency of rainfall will influence biophysical drivers that regulate the strength and timing of soil CO2 efflux – a major source of terrestrial carbon flux. We used a rainfall manipulation experiment during the summer monsoon season (July–September) to vary both the size and frequency of precipitation in an arid grassland 2 years before and 2 years after a lightning‐caused wildfire. Soil CO2 efflux rates were always higher under increased rainfall event size than under increased rainfall event frequency, or ambient precipitation. Although fire reduced soil CO2 efflux rates by nearly 70%, the overall responses to rainfall variability were consistent before and after the fire. The overall sensitivity of soil CO2 efflux to temperature (Q10) converged to 1.4, but this value differed somewhat among treatments especially before the fire. Changes in rainfall patterns resulted in differences in the periodicity of soil CO2 efflux with strong signals at 1, 8, and 30 days. Increased rainfall event size enhanced the synchrony between photosynthetically active radiation and soil CO2 efflux over the growing season before and after fire, suggesting a change in the temporal availability of substrate pools that regulate the temporal dynamics and magnitude of soil CO2 efflux. We conclude that arid grasslands are capable of rapidly increasing and maintaining high soil CO2 efflux rates in response to increased rainfall event size more than increased rainfall event frequency both before and after a fire. Therefore, the amount and pattern of multiple rain pulses over the growing season are crucial for understanding CO2 dynamics in burned and unburned water‐limited ecosystems.  相似文献

Tree planting is increasingly being proposed as a strategy to combat climate change through carbon (C) sequestration in tree biomass. However, total ecosystem C storage that includes soil organic C (SOC) must be considered to determine whether planting trees for climate change mitigation results in increased C storage. We show that planting two native tree species (Betula pubescens and Pinus sylvestris), of widespread Eurasian distribution, onto heather (Calluna vulgaris) moorland with podzolic and peaty podzolic soils in Scotland, did not lead to an increase in net ecosystem C stock 12 or 39 years after planting. Plots with trees had greater soil respiration and lower SOC in organic soil horizons than heather control plots. The decline in SOC cancelled out the increment in C stocks in tree biomass on decadal timescales. At all four experimental sites sampled, there was no net gain in ecosystem C stocks 12–39 years after afforestation—indeed we found a net ecosystem C loss in one of four sites with deciduous B. pubescens stands; no net gain in ecosystem C at three sites planted with B. pubescens; and no net gain at additional stands of P. sylvestris. We hypothesize that altered mycorrhizal communities and autotrophic C inputs have led to positive ‘priming’ of soil organic matter, resulting in SOC loss, constraining the benefits of tree planting for ecosystem C sequestration. The results are of direct relevance to current policies, which promote tree planting on the assumption that this will increase net ecosystem C storage and contribute to climate change mitigation. Ecosystem‐level biogeochemistry and C fluxes must be better quantified and understood before we can be assured that large‐scale tree planting in regions with considerable pre‐existing SOC stocks will have the intended policy and climate change mitigation outcomes.  相似文献
