Human glioblastomas may be hierarchically organized. Within this hierarchy, glioblastoma‐initiating cells have been proposed to be more resistant to radiochemotherapy and responsible for recurrence. Here, established stem cell markers and stem cell attributed characteristics such as self‐renewal capacity and tumorigenicity have been profiled in primary glioblastoma cultures to predict radiosensitivity. Furthermore, the sensitivity to radiotherapy of different subpopulations within a single primary glioblastoma culture was analyzed by a flow cytometric approach using Nestin, SRY (sex‐determining region Y)‐box 2 (SOX2) and glial fibrillary acidic protein. The protein expression of Nestin and SOX2 as well as the mRNA levels of Musashi1, L1 cell adhesion molecule, CD133, Nestin, and pleiomorphic adenoma gene‐like 2 inversely correlated with radioresistance in regard to the clonogenic potential. Only CD44 protein expression correlated positively with radioresistance. In terms of proliferation, Nestin protein expression and Musashi1, pleiomorphic adenoma gene‐like 2, and CD133 mRNA levels are inversely correlated with radioresistance. Higher expression of stem cell markers does not correlate with resistance to radiochemotherapy in the cancer genome atlas glioblastoma collective. SOX2 expressing subpopulations exist within single primary glioblastoma cultures. These subpopulations predominantly form the proliferative pool of the primary cultures and are sensitive to irradiation. Thus, profiling of established stem cell markers revealed a surprising result. Except CD44, the tested stem cell markers showed an inverse correlation between expression and radioresistance.

Adenoid cystic carcinoma (ACC) is an aggressive malignant neoplasm of the salivary glands in which c-Kit is overexpressed and activated, although the mechanism for this is as yet unclear. We analyzed 27 sporadic ACC tumor specimens to examine the biologic and clinical significance of c-Kit activation. Mutational analysis revealed expression of wild-type c-Kit in all, eliminating gene mutation as a cause of activation. Because stem cell factor (SCF) is c-Kit's sole ligand, we analyzed its expression in the tumor cells and their environment. Immunohistochemistry revealed its presence in c-Kit–positive tumor cells, suggesting an activation of autocrine signaling. We observed a significant induction of ERK1/2 in the cells. SCF staining was also found in other types of non-cancerous cells adjacent to tumors within salivary glands, including stromal fibroblasts, neutrophils, peripheral nerve, skeletal muscle, vascular endothelial cells, mucous acinar cells, and intercalated ducts. Quantitative PCR showed that the top quartile of c-Kit mRNA expression distinguished ACCs from normal salivary tissues and was cross-correlated with short-term poor prognosis. Expression levels of SCF and c-Kit were highly correlated in the cases with perineural invasion. These observations suggest that c-Kit is potentially activated by receptor dimerization upon stimulation by SCF in ACC, and that the highest quartile of c-Kit mRNA expression could be a predictor of poor prognosis. Our findings may support an avenue for c-Kit-targeted therapy to improve disease control in ACC patients harboring the top quartile of c-Kit mRNA expression.  相似文献

A convenient approach for the synthesis of narrowly dispersed polypeptide copolymers of defined compositions is presented. The controlled denaturation of the proteins serum albumin and lysozyme followed by an in situ stabilization with polyethylene(oxide) chains yields polypeptide side chain copolymers of precisely defined backbone lengths as well as the presence of secondary structure elements. Supramolecular architectures are formed in solution because of the presence of hydrophobic and hydrophilic amino acids along the polypeptide main chain. Polypeptide copolymers reported herein reveal excellent solubility and stability in aqueous media and no significant cytotoxicity at relevant concentrations, and they can be degraded via proteolysis, which is very attractive for biomedical applications. This "semi-synthetic chemistry" approach is based on a novel and convenient concept for producing synthetic polypeptides from native protein resources, which complements traditional polypeptide synthesis and expression approaches and offers great opportunities for the preparation of diverse polypeptides with unique architectures.  相似文献

As some malaria control programs shift focus from disease control to transmission reduction, there is a need for transmission data to monitor progress. At lower levels of transmission, it becomes increasingly more difficult to measure precisely, for example through entomological studies. Many programs conduct regular cross sectional parasite prevalence surveys, and have access to malaria treatment data routinely collected by ministries of health, often in health management information systems. However, by themselves, these data are poor measures of transmission. In this paper, we propose an approach for combining annual parasite incidence and treatment data with cross-sectional parasite prevalence and treatment seeking survey data to estimate the incidence of new infections in the human population, also known as the force of infection. The approach is based on extension of a reversible catalytic model. The accuracy of the estimates from this model appears to be highly dependent on levels of detectability and treatment in the community, indicating the importance of information on private sector treatment seeking and access to effective and appropriate treatment.  相似文献

Cytokine-inducible SRC homology 2 domain protein (CISH) is a suppressor of cytokine signaling that controls interleukin-2 signaling pathway. We investigated the single nucleotide polymorphism (SNP) -292A>T in 473 Vietnamese hepatitis B virus (HBV) carriers and 416 healthy controls. CISH variants at -292A>T were associated to HBV infection (Allelic: OR, 1.22 95% CI, 1–1.49; P = 0.04; Recessive: OR, 1.69 95% CI 1.23–2.54; P = 0.007). A gene dose effect for the risk allele -292T was observed (P = 0.04). The level of interleukin 2 and liver enzymes such as alanine transaminase, aspartate transaminase, total bilirubin, and direct bilirubin were not associated to CISH polymorphism at position -292A>T This study associated the vital role of CISH SNP -292A>T variant to hepatitis B virus infection in a Vietnamese population.  相似文献

A syndrome presenting as gross abdominal distension and diagnosed as acquired megacolon was observed in five adult female long-tailed macaques. Gastrointestinal signs included diarrhea, mucus in the stool, anorexia, and failure to pass stool, with repeated episodes of extreme abdominal distension and accumulation of gas and feces in greatly enlarged colons. Medical management was unsuccessful. A partial colectomy with a standard end-to-end colonic anastomosis was performed to remove the section of distended colon in each animal. Histologically, affected colons had degeneration and fibrosis, primarily in the longitudinal layer of the lamina muscularis. Hemograms, serum chemistries, and histopathologic features were not diagnostic of a specific etiology for megacolon. Four of five animals had undergone at least one obstetrical surgery. Two of these had the first episode of colonic distension within 3 days postoperatively. Intra-abdominal adhesions were noted during exploratory surgery in all animals. Three of five had colonic volvulus observed during colectomy. Recovery post-colectomy was uneventful and animals remained free of clinical signs of megacolon.  相似文献

The retinoblastoma gene product (Rb), a cellular growth suppressor, complexes with viral and cellular proteins that contain a specific binding domain incorporating three invariant residues: Leu-X-Cys-X-Glu, where X denotes a nonconserved residue. Hydrophobic and electrostatic properties are strongly conserved in this segment even though the nonconserved amino acids vary considerably from one Rb-binding protein to another. In this report, we present a diagnostic computer pattern for a high-affinity Rb-binding domain featuring the three conserved residues as well as the conserved physico-chemical properties. Although the pattern encompasses only 10 residues (with only 4 of these explicitly defined), it exhibits 100% sensitivity and 99.95% specificity in database searches. This implies that a certain pattern of structural and physico-chemical properties encoded by this short sequence is sufficient to govern specific Rb binding. We also present evidence that the secondary structural conformation through this region is important for effective Rb binding.  相似文献

As an alternative for radioimmunoassays a new enzyme immunoassay (EIA) for the determination of 13,14-dihydro-15-keto PGF2 alpha (PGFM) has been developed. Biocytin was linked to PGFM by the N-hydroxysuccinimide method and the product (biocytinyl-PGFM) purified by reversed phase column chromatography. Biocytinyl-PGFM was used in the EIA as a bridge between the immobilized PGFM-antibody and streptavidin-peroxidase. The absolute sensitivity of the assay was about 160 amol (92% rel. binding) and required only 2 microliters plasma for PGFM estimation within the whole physiological range (0.08-20 pmol/ml). All variabilities were less than 14%. The described assay procedure may be of general applicability for other prostaglandins.  相似文献
