Re-examination, using molecular tools, of the diversity of haemosporidian parasites (among which the agents of human malaria are the best known) has generally led to rearrangements of traditional classifications. In this study, we explored the diversity of haemosporidian parasites infecting vertebrate species (particularly mammals, birds and reptiles) living in the forests of Gabon (Central Africa), by analyzing a collection of 492 bushmeat samples. We found that samples from five mammalian species (four duiker and one pangolin species), one bird and one turtle species were infected by haemosporidian parasites. In duikers (from which most of the infected specimens were obtained), we demonstrated the existence of at least two distinct parasite lineages related to Polychromophilus species (i.e., bat haemosporidian parasites) and to sauropsid Plasmodium (from birds and lizards). Molecular screening of sylvatic mosquitoes captured during a longitudinal survey revealed the presence of these haemosporidian parasite lineages also in several Anopheles species, suggesting a potential role in their transmission. Our results show that, differently from what was previously thought, several independent clades of haemosporidian parasites (family Plasmodiidae) infect mammals and are transmitted by anopheline mosquitoes.  相似文献

Voltage‐gated sodium channels are essential for electrical signalling across cell membranes. They exhibit strong selectivities for sodium ions over other cations, enabling the finely tuned cascade of events associated with action potentials. This paper describes the ion permeability characteristics and the crystal structure of a prokaryotic sodium channel, showing for the first time the detailed locations of sodium ions in the selectivity filter of a sodium channel. Electrostatic calculations based on the structure are consistent with the relative cation permeability ratios (Na+ ≈ Li+ ≫ K+, Ca2+, Mg2+) measured for these channels. In an E178D selectivity filter mutant constructed to have altered ion selectivities, the sodium ion binding site nearest the extracellular side is missing. Unlike potassium ions in potassium channels, the sodium ions in these channels appear to be hydrated and are associated with side chains of the selectivity filter residues, rather than polypeptide backbones.  相似文献

The biosynthesis of the majority of biologically active peptides ends with an obligatory alpha-amidation step that is catalyzed only by peptidylglycine alpha-hydroxylating monooxygenase (PHM). The utility of two mechanisms proposed for this copper- and ascorbate-dependent monooxygenase was examined using site-directed mutagenesis and intrinsic tryptophan fluorescence. Retention of full activity by PHMccGln(170)Ala and -Asn eliminates a critical role for Gln(170) in a substrate-mediated electron transfer pathway. The 20-fold reduction in V(max) observed for PHMccGln(170)Glu and -Leu is consistent with a key role for conformational changes in this region. Mutation of Tyr(79), situated near Cu(A), to Trp reduced V(max) 200-fold. Measurement of changes in intrinsic fluorescence allowed determination of a K(d) for copper (0.06 microM) and for a peptidylglycine substrate, Phe-Gly-Phe-Gly (0.8 microM). Although the peptidylglycine substrate bound more tightly at pH 7.0 than at pH 5.5, V(max) decreased 25-fold at neutral pH. Total quenching of the signal from Trp(79) in apoPHMccTyr(79)Trp along with its greatly reduced V(max) defines a critical role for Cu(A) in the rate-limiting step of the reaction. Taking into account our data and the results of kinetic, spectroscopic, and crystallographic studies, we propose a mechanism in which substrate-mediated activation of molecular oxygen binding at Cu(A) completes a pathway for electron transfer from Cu(B).  相似文献

Spine function requires precise control of the actin cytoskeleton. Kalirin-7, a GDP/GTP exchange factor for Rac1, interacts with PDZ proteins such as PSD-95, colocalizing with PSD-95 at synapses of cultured hippocampal neurons. PSD-95 and Kalirin-7 interact in vivo and in heterologous expression systems. In primary cortical neurons, transfected Kalirin-7 is targeted to spines and increases the number and size of spine-like structures. A Kalirin-7 mutant unable to interact with PDZ proteins remains in the cell soma, inducing local formation of aberrant filopodial neurites. Kalirin-7 with an inactivated GEF domain reduces the number of spines below control levels. These results provide evidence that PDZ proteins target Kalirin-7 to the PSD, where it regulates dendritic morphogenesis through Rac1 signaling to the actin cytoskeleton.  相似文献

This paper critiques the theorisation of culture as difference that dominates Australian cultural studies and cultural anthropology. It argues that a perspective on culture that gets beyond current preoccupations with difference has more to be said for it than practitioners in either discipline have hitherto recognised. Ethnography has long been recognised as crucial to the study of culture—which suggests that we might best appreciate what it is that is being ignored and neglected (not to say demeaned) in contemporary studies of culture if we turn our attention to the way we study it. This paper reconsiders what we mean by the assertion that ethnography constitutes an experiential mode of knowledge which constitutes the most sustained and rigorous application of the principle of verstehen available. In contesting the stress on difference that now dominates the study of culture, a case is made for cultures conceived of as ensembles of mystery and meaning. Whether born into a culture or researching it, as observers and participants we struggle to make sense of what the world is about. As ethnographers, we seek to do this empathically; this paper argues that to do it effectively we must fashion a sensibility adequate to it. An appreciation of the significance of sensibility, it is suggested, provides a much needed alternative to the reduction of culture to difference.  相似文献

辽宁是东北林蛙Rana dybowskii主要分布地之一,种群数量巨大,其群体遗传多样性有待评估.本研究应用ISSR标记技术对东北林蛙4个种群105个样本进行研究,5个引物共获得44条清晰谱带,4个种群的多态位点率均大于75%,Nei's基因多样性为0.2851,Shannon信息指数为0.4476,显示了较高的遗传多样性.对遗传分化系数、Nei's遗传距离、AMOVA分子变异巢式方差分析和F-统计量等遗传参数的统计结果表明,辽宁东北林蛙种群间已经出现一定程度的遗传分化,分析认为,自然屏障(高山和平原等)以及栖息地片段化是其遗传分化形成的主要因素.  相似文献
