Autosomal dominant polycystic kidney disease (ADPKD), the most common inherited cause of kidney failure, is caused by mutations in either PKD1 (85%) or PKD2 (15%). The PKD2 protein, polycystin-2 (PC2 or TRPP2), is a member of the transient receptor potential (TRP) superfamily and functions as a non-selective calcium channel. PC2 has been found to form oligomers in native tissues suggesting that it may form functional homo- or heterotetramers with other subunits, similar to other TRP channels. Our experiments unexpectedly revealed that PC2 mutant proteins lacking the known C-terminal dimerization domain were still able to form oligomers and co-immunoprecipitate full-length PC2, implying the possible existence of a proximal dimerization domain. Using yeast two-hybrid and biochemical assays, we have mapped an alternative dimerization domain to the N terminus of PC2 (NT2-1-223, L224X). Functional characterization of this domain demonstrated that it was sufficient to induce cyst formation in zebrafish embryos and inhibit PC2 surface currents in mIMCD3 cells probably by a dominant-negative mechanism. In summary, we propose a model for PC2 assembly as a functional tetramer which depends on both C- and N-terminal dimerization domains. These results have significant implications for our understanding of PC2 function and disease pathogenesis in ADPKD and provide a new strategy for studying PC2 function.  相似文献

Several blue copper proteins are known to change the active site structure at alkaline pH (alkaline transition). Spectroscopic studies of Met16Phe, Met16Tyr, Met16Trp, and Met16Val pseudoazurin variants were performed to investigate the second sphere role through alkaline transition. The visible electronic absorption and resonance Raman spectra of Met16Phe, Met16Tyr, and Met16Trp variants showed the increasing of axial component at pH 11 like wild-type PAz. The visible electronic absorption and far-UV CD spectra of Met16Val demonstrated that the destabilization of the protein structure was triggered at pH > 11. Resonance Raman (RR) spectra of PAz showed that the intensity-weighted averaged Cu–S(Cys) stretching frequency was shifted to higher frequency region at pH 11. The higher frequency shift of Cu–S(Cys) bond is implied the stronger Cu–S(Cys) bond at alkaline transition pH 11. The visible electronic absorption and far-UV CD spectra of Met16X PAz revealed that the Met16Val variant is denatured at pH > 11, but Met16Phe, Met16Tyr, and Met16Trp mutant proteins are not denatured even at pH > 11. These observations suggest that Met16 is important to maintain the protein structure through the possible weak interaction between methionine –SCH3 part and coordinated histidine imidazole moiety. The introduction of π–π interaction in the second coordination sphere may be contributed to the enhancement of protein structure stability.  相似文献

Cdc7 is a serine-threonine kinase that regulates initiation and progression of DNA replication. The activity of purified Cdc7 kinase is significantly stimulated by polyamines such as spermine or spermidine. Positively charged polymers of lysine or arginine also stimulate its kinase activity, whereas the negatively charged substances such as polyglutamate or nucleic acids significantly inhibit the kinase activity. Spermine affects both the K(m) and V(max) of Cdc7 kinase for a minichromosome maintenance (MCM) substrate. We also found that histones, lysine- and arginine-rich basic proteins, can stimulate Cdc7 kinase activity, and a MCM complex in association with histone is a more efficient substrate of Cdc7 than the free MCM complex. These results identify potential cellular inhibitors and stimulators of Cdc7 kinase and suggest that Cdc7 may be another target of cellular polyamines and that histones may stimulate Cdc7-mediated phosphorylation of chromatin-bound substrates. Ectopic expression of an antizyme, known to reduce the cellular polyamine levels, resulted in reduction of Cdc7-mediated phosphorylation of MCM4 protein, suggesting physiological roles of polyamines in regulation of Cdc7 kinase activity in the cells.  相似文献

The flux of emerging aquatic insects from streams can provide a significant energy subsidy to riparian web-building spiders. However, despite the high temporality of aquatic insect emergence, the effects of such aquatic insect dynamics on spider distribution are poorly understood. To examine the relationship, the aquatic insect flux from a headwater stream in a northern Japanese deciduous forest was experimentally manipulated by using a greenhouse-type covering, during May to July. Under natural conditions, the aquatic and terrestrial insect abundances dramatically decreased and increased from May through July, respectively. The experimental reduction of aquatic insect flux depressed the density of horizontal orb weavers (Tetragnathidae) in both May and June, but not in July when aquatic insects were scarce, indicating a temporal limitation on spider distribution by aquatic insect flux. In contrast, the densities of both vertical orb weavers (Araneidae) and sheet weavers (Linyphiidae) were unaffected by the manipulation throughout the study period. These various responses, differing among months or spider guilds, may be attributed to the degree of specialization for aquatic prey in the spiders and their mobility in response to aquatic insect flux. The experimental results provided direct evidence that the temporal dynamics of aquatic insect flux, as well as spider characteristics, were primary factors determining the distributional patterns of riparian web-building spiders.  相似文献

Cholesterol side-chain cleavage activity in mitochondria isolated from the outer and inner zones of the guinea pig adrenal cortex was evaluated in order to clarify the role of the zona reticularis in steroidogenesis. It was found that side-chain cleavage activity was three times higher in the outer zone. In addition, ether stress increased side-chain cleavage activity in the outer zone but not the inner zone. The concentration of total and free cholesterol was also found to be higher in the outer zone. However, when exogenous cholesterol was added to mitochondria, there was no enhancement in side-chain cleavage activity in either zone.  相似文献

The proportion of stromal fat cells to parenchymal cells in 100 normal parathyroid glands was determined by the image analyzing computer technique. The parathyroid glands were resected at the time of thyroidectomy in 86 patients with thyroid tumors. None of the patients had any evidence of parathyroid dysfunction preoperatively. In the histologic sections of the parathyroid glands, the average percentage of stromal fat cell content was 38%. The percentage of stromal fat cells was correlated with the age and the body constitution of the patients, but the percentages of fat cells varied widely among glands in the given age and body constitution ranges. It was therefore not possible to discriminate a normal parathyroid gland from an abnormal gland solely on the basis of microscopic determination of stromal fat cell content.  相似文献
