Inert particles (iron saccharate or latex beads) injected in the haemocoel of Locusta migratoria, are taken up by pericardial cells (iron saccharate only), reticular cells of the haemopoietic tissue and certain haemocytes: plasmatocytes and coagulocytes; these two haemocyte types are also the main phagocytic blood cells in Galleria mellonella.Necrosis of phagocytic haemocytes, following injection of an overdose of iron saccharate, explains the profound modifications of the haemogram observed during the first 24 hr following injection; the macrophagic evolution of reticular cells slows down the haemopoietic differentiation of these cells and explains the long term disturbances of the blood picture.Clearance of latex beads injected in larvae of Locusta complies to an exponential function of time; we can determine a granulopectic index which will permit comparisons to be made between clearance of inert and of ‘antigenic-like’ particles. 相似文献

Effect of wind-borne pollens on the population dynamics ofAmblyseius hibisci [Acarina: Phytoseiidae]
Populations ofAmblyseius hibisci (Chant), an important predator of citrus red mite,Panonychus citri (McGregor) exhibited varying responses to field applications of cat-tail,Typha latifolia L., pollen in San Joaquin Valley citrus groves. Predator densities increased to significantly higher levels after pollen treatments during the fall months, and to a limited extent after late winter treatments, but not after early spring treatments. The amount of response appeared to be inversely related to the levels of natural foods present during the various seasons. Results of the wind-borne pollen monitoring studies indicated that population growth inA. hibisci was correlated with the seasonal pattern of pollen fallout. Population growth was slowest in years when pollen catches were low during the late winter and early spring, and fastest in years when peak pollen catches occurred in late winter or early spring. Population growth inA. hibisci did not appear to be markedly responsive toP. citri populations although suppression of prey densities was indicated in ca. 50% of the cases. In total hip arthroplasty and particularly in revision surgery, computer assisted pre-operative prediction of the best possible anchorage strategy for implant fixation would be a great help to the surgeon. Computer simulation relies on validated numerical models. In the current study, three density–elasticity relationships (No. 1–3) from the literature for inhomogeneous material parameter assignment from CT data in automated finite element (FE) modeling of long bones were evaluated for their suitability for FE modeling of human pelvic bone. Numerical modal analysis was conducted on 10 FE models of hemipelvic bone specimens and compared to the gold standard provided by experimental modal analysis results from a previous in-vitro study on the same specimens. Overall, calculated resonance frequencies came out lower than measured values. Magnitude of mean relative deviation of numerical resonance frequencies with regard to measured values is lowest for the density–elasticity relationship No. 3 (−15.9%) and considerably higher for both density–elasticity relationships No. 1 (−41.1%) and No. 2 (−45.0%). Mean MAC values over all specimens amount to 77.8% (No. 1), 78.5% (No. 2), and 83.0% (No. 3). MAC results show, that mode shapes are only slightly influenced by material distribution. Calculated resonance frequencies are generally lower than measured values, which indicates, that numerical models lack stiffness. Even when using the best suited (No. 3) out of three investigated density–elasticity relationships, in FE modeling of pelvic bone a considerable underestimation of model stiffness has to be taken into account. 相似文献

For mammalian TRPM8, the amino acid residues asparagine-799 and aspartate-802 are essential for the stimulation of the channel by the synthetic agonist icilin. Both residues belong to the short sequence motif N-x-x-D within the transmembrane segment S3 highly conserved in the entire superfamily of voltage-dependent cation channels, among them TRPM8. Moreover, they are also conserved in the closely related TRPM2 channel, which is essentially voltage-independent. To analyze the differential roles of the motif for the voltage-dependent and voltage-independent gating, we performed reciprocal replacements of the asparagine and aspartate within the S3 motif in both channels, following the proposed idea that specific electrostatic interactions with other domains take place during gating. Wild-type and mutant channels were heterologeously expressed in HEK-293 cells and channel function was analyzed by whole-cell patch-clamp analysis as well as by Ca2+-imaging. Additionally, the expression of the channels in the plasma membrane was tested by Western blot analysis, in part after biotinylation. For the mutations of TRPM8, responses to menthol were only compromised if also the expression of the glycosylated channel isoform was prevented. In contrast, responses to cold were consistently and significantly attenuated but not completely abolished. For TRPM2, surface expression was not significantly affected by any of the mutations but channel function was only retained in one variant. Remarkably, this was the variant of which the corresponding mutation in TRPM8 exerted the most negative effects both on channel function and expression. Furthermore, we performed an exchange of the inner pair of residues of the N-x-x-D motif between the two channels, which proved deleterious for the functional expression of TRPM8 but ineffective on TRPM2. In conclusion, the N-x-x-D motif plays specific roles in TRPM8 and TRPM2, reflecting different requirements for voltage-dependent and voltage-independent channel gating. 相似文献

Sour rot is a disease complex that causes serious damage in viticulture. The common vinegar fly Drosophila melanogaster (Diptera: Drosophilidae) is associated with sour rot in overripe or otherwise damaged grapes. Drosophila suzukii (Diptera: Drosophilidae) is an invasive species, which is suspected to induce sour rot in previously undamaged grapes due to the flies' ability to infest healthy, undamaged soft fruits with its serrated ovipositor. As a consequence, infection of healthy grapes by D. suzukii may facilitate the colonization by D. melanogaster. We investigated the single and combined effects of D. suzukii and D. melanogaster on sour rot development by measuring volatile acidity under near-natural conditions in the vineyard, along with laboratory experiments under controlled climate. In 2017, the combined field and laboratory experiments suggested that the presence of D. suzukii and D. melanogaster increased the volatile acidity levels at a similar rate. In 2018, the field experiments showed an only marginal increase in sour rot development in treatments with both Drosophila species. Under more favourable laboratory conditions, the presence of D. suzukii, but not D. melanogaster triggered sour rot emergence. A facilitating effect of D. suzukii infestation for D. melanogaster was not detectable. These findings suggest that D. suzukii does in fact have the potential to trigger sour rot, but will probably rarely do so under field conditions in the vineyard, at least in the studied region. Instead, our study showed that D. melanogaster can have a similar impact on sour rot development as D. suzukii, emphasizing the need of comparative studies. 相似文献

Nations throughout the Indo‐Pacific region use pyrethroid insecticides to control Aedes aegypti, the mosquito vector of dengue, often without knowledge of pyrethroid resistance status of the pest or origin of resistance. Two mutations (V1016G + F1534C) in the sodium channel gene (Vssc) of Ae. aegypti modify ion channel function and cause target‐site resistance to pyrethroid insecticides, with a third mutation (S989P) having a potential additive effect. Of 27 possible genotypes involving these mutations, some allelic combinations are never seen whereas others predominate. Here, five allelic combinations common in Ae. aegypti from the Indo‐Pacific region are described and their geographical distributions investigated using genome‐wide SNP markers. We tested the hypothesis that resistance allele combinations evolved de novo in populations versus the alternative that dispersal of Ae. aegypti between populations facilitated genetic invasions of allele combinations. We used latent factor mixed‐models to detect SNPs throughout the genome that showed structuring in line with resistance allele combinations and compared variation at SNPs within the Vssc gene with genome‐wide variation. Mixed‐models detected an array of SNPs linked to resistance allele combinations, all located within or in close proximity to the Vssc gene. Variation at SNPs within the Vssc gene was structured by resistance profile, whereas genome‐wide SNPs were structured by population. These results demonstrate that alleles near to resistance mutations have been transferred between populations via linked selection. This indicates that genetic invasions have contributed to the widespread occurrence of Vssc allele combinations in Ae. aegypti in the Indo‐Pacific region, pointing to undocumented mosquito invasions between countries. 相似文献

Wolbachia are maternally inherited endosymbiotic bacteria found within many insect species. Aedes mosquitoes experimentally infected with Wolbachia are being released into the field for Aedes‐borne disease control. These Wolbachia infections induce cytoplasmic incompatibility which is used to suppress populations through incompatible matings or replace populations through the reproductive advantage provided by this mechanism. However, the presence of naturally occurring Wolbachia in target populations could interfere with both population replacement and suppression programs depending on the compatibility patterns between strains. Aedes aegypti were thought to not harbor Wolbachia naturally but several recent studies have detected Wolbachia in natural populations of this mosquito. We therefore review the evidence for natural Wolbachia infections in A. aegypti to date and discuss limitations of these studies. We draw on research from other mosquito species to outline the potential implications of natural Wolbachia infections in A. aegypti for disease control. To validate previous reports, we obtained a laboratory population of A. aegypti from New Mexico, USA, that harbors a natural Wolbachia infection, and we conducted field surveys in Kuala Lumpur, Malaysia, where a natural Wolbachia infection has also been reported. However, we were unable to detect Wolbachia in both the laboratory and field populations. Because the presence of naturally occurring Wolbachia in A. aegypti could have profound implications for Wolbachia‐based disease control programs, it is important to continue to accurately assess the Wolbachia status of target Aedes populations. 相似文献
