Maintenance of bone mass and integrity requires a tight balance between resorption by osteoclasts and formation by osteoblasts. Exocytosis of functional proteins is a prerequisite for the activity of both cells. In the present study, we show that synaptotagmin VII, a calcium sensor protein that regulates exocytosis, is associated with lysosomes in osteoclasts and bone matrix protein-containing vesicles in osteoblasts. Absence of synaptotagmin VII inhibits cathepsin K secretion and formation of the ruffled border in osteoclasts and bone matrix protein deposition in osteoblasts, without affecting the differentiation of either cell. Reflecting these in vitro findings, synaptotagmin VII-deficient mice are osteopenic due to impaired bone resorption and formation. Therefore, synaptotagmin VII plays an important role in bone remodeling and homeostasis by modulating secretory pathways functionally important in osteoclasts and osteoblasts.  相似文献

Physical and functional interactions define the molecular organization of the cell. Genetic interactions, or epistasis, tend to occur between gene products involved in parallel pathways or interlinked biological processes. High-throughput experimental systems to examine genetic interactions on a genome-wide scale have been devised for Saccharomyces cerevisiae, Schizosaccharomyces pombe, Caenorhabditis elegans and Drosophila melanogaster, but have not been reported previously for prokaryotes. Here we describe the development of a quantitative screening procedure for monitoring bacterial genetic interactions based on conjugation of Escherichia coli deletion or hypomorphic strains to create double mutants on a genome-wide scale. The patterns of synthetic sickness and synthetic lethality (aggravating genetic interactions) we observed for certain double mutant combinations provided information about functional relationships and redundancy between pathways and enabled us to group bacterial gene products into functional modules.  相似文献

The primary purpose of this study was to investigate whether the athlete who has high performance in hang power clean, a common weightlifting exercise, has high performances in sprinting, jumping, and changing of direction (COD). As the secondary purpose, relationships between hang power clean performance, maximum strength, power and performance of jumping, sprinting, and COD also were investigated. Twenty-nine semiprofessional Australian Rules football players (age, height, and body mass [mean +/- SD]: 21.3 +/- 2.7 years, 1.8 +/- 0.1 m, and 83.6 +/- 8.2 kg) were tested for one repetition maximum (1RM) hang power clean, 1RM front squat, power output during countermovement jump with 40-kg barbell and without external load (CMJ), height of CMJ, 20-m sprint time, and 5-5 COD time. The subjects were divided into top and bottom half groups (n = 14 for each group) based on their 1RM hang power clean score relative to body mass, then measures from all other tests were compared with one-way analyses of variance. In addition, Pearson's product moment correlations between measurements were calculated among all subjects (n = 29). The top half group possessed higher maximum strength (P < 0.01), power (P < 0.01), performance of jumping (P < 0.05), and sprinting (P < 0.01). However, there was no significant difference between groups in 5-5 COD time, possibly because of important contributing factors other than strength and power. There were significant correlations between most of, but not all, combinations of performances of hang power clean, jumping, sprinting, COD, maximum strength, and power. Therefore, it seems likely there are underlying strength qualities that are common to the hang power clean, jumping, and sprinting.  相似文献

We compared delayed-onset muscle soreness (DOMS) induced by anaerobic resistance exercises with and without aerobic cardioacceleration before each set, under the rationale that elevated heart rate (HR) may increase blood perfusion in muscles to limit eccentric contraction damage and/or speed muscle recovery. In two identical experiments (20 men, 28 women), well-conditioned athletes paired by similar physical condition were assigned randomly to experimental or control groups. HR (independent variable) was recorded with HR monitors. DOMS (dependent variable) was self-reported using Borg's Rating of Perceived Pain scale. After identical pre-training strength testing, mean DOMS in the experimental and control groups was indistinguishable (P > or = 0.19) for musculature employed in eight resistance exercises in both genders, validating the dependent variable. Subjects then trained three times per week for 9 (men) to 11 (women) weeks in a progressive, whole-body, concurrent training protocol. Before each set of resistance exercises, experimental subjects cardioaccelerated briefly (mean HR during resistance training, 63.7% HR reserve), whereas control subjects rested briefly (mean HR, 33.5% HR reserve). Mean DOMS among all muscle groups and workouts was discernibly less in experimental than control groups in men (P = 0.0000019) and women (P = 0.0007); less for each muscle group used in nine resistance exercises in both genders, discernible (P < 0.025) in 15 of 18 comparisons; and less in every workout, discernible (P < 0.05) in 32% (men) and 55% (women) of workouts. Most effect sizes were moderate. In both genders, mean DOMS per workout disappeared by the fourth week of training in experimental but not control groups. Aerobic cardioacceleration immediately before each set of resistance exercises therefore rapidly eliminates DOMS during vigorous progressive resistance training in athletes.  相似文献

Nonhuman primates (NHPs) are imported to the United States for use in research, domestic breeding, and propagation of endangered populations in zoological gardens. During the past 60 years, individuals responsible for NHP importation programs have observed morbidity and mortality typically associated with infectious disease outbreaks. These outbreaks have included infectious agents such as tuberculosis, Herpesvirus sp., simian hemorrhagic fever, and filovirus infections such as the Ebola and Marburg viruses. Some outbreaks have affected both animal and human populations. These epizootics are attributable to a variety of factors, including increased population density, exposure of na?ve populations to new infectious agents, and stress. The practice of quarantining animals arriving in the United States was first applied by individual research programs to improve animal health and ensure the quality of animals entering research programs. The development of government regulations for nonhuman primate quarantine accompanied the recognition that imported NHPs could pose a risk to public health. This article briefly reviews the history of US NHP importation and the factors behind the development of NHP quarantine regulations. The focus is on regulations concerned with infectious disease, public health, and the health of domestic primate colonies. These regulations have had the dual benefit of protecting public health as well as reducing animal morbidity and mortality during importation and quarantine. We review current practices and facilities for nonhuman primate quarantine and identify challenges for the future.  相似文献

Seventy-four hominoid primary teeth have been recovered from the middle Miocene site of Pa?alar, Turkey, constituting the largest sample of deciduous teeth for any species of fossil ape. Morphological features that characterize the permanent teeth of Griphopithecus alpani from the site have also been identified in some of these deciduous teeth, including a lingual pillar on the di(1)s. These features plus the overwhelming preponderance of G. alpani permanent teeth at the site suggest that all of the deciduous teeth belong to this species. Contrary to the situation in the permanent teeth, nothing in the morphology of the primary dentition suggests the representation of a second species. The age profile of the non-adult hominoids was reconstructed based on the degree and type of wear recorded on the dp4s, the most abundant deciduous tooth in the sample, assuming a similar eruption chronology to that of Pan troglodytes. This analysis indicates underrepresentation of very young individuals in the sample and high mortality for individuals belonging to the 3-5-years age cohort, a situation that could be due to the effects of stress related to weaning. The coefficient of variation and range-index values obtained for the majority of tooth types are equal to or greater than the comparable values in a sample of P. troglodytes, in some cases at much smaller sample sizes. One possible explanation for this is that there was greater sexual dimorphism in the G. alpani deciduous dentition than in Pan, which would mirror the condition of the permanent dentition.  相似文献
