The rupture risk of unruptured intracranial aneurysms is known to be dependent on the size of the aneurysm. However, the association of morphological characteristics with ruptured aneurysms has not been established in a systematic and location specific manner for the most common aneurysm locations. We evaluated posterior communicating artery (PCoA) aneurysms for morphological parameters associated with aneurysm rupture in that location. CT angiograms were evaluated to generate 3-D models of the aneurysms and surrounding vasculature. Univariate and multivariate analyses were performed to evaluate morphological parameters including aneurysm volume, aspect ratio, size ratio, distance to ICA bifurcation, aneurysm angle, vessel angles, flow angles, and vessel-to-vessel angles. From 2005–2012, 148 PCoA aneurysms were treated in a single institution. Preoperative CTAs from 63 patients (40 ruptured, 23 unruptured) were available and analyzed. Multivariate logistic regression revealed that smaller volume (p = 0.011), larger aneurysm neck diameter (0.048), and shorter ICA bifurcation to aneurysm distance (p = 0.005) were the most strongly associated with aneurysm rupture after adjusting for all other clinical and morphological variables. Multivariate subgroup analysis for patients with visualized PCoA demonstrated that larger neck diameter (p = 0.018) and shorter ICA bifurcation to aneurysm distance (p = 0.011) were significantly associated with rupture. Intracerebral hemorrhage was associated with smaller volume, larger maximum height, and smaller aneurysm angle, in addition to lateral projection, male sex, and lack of hypertension. We found that shorter ICA bifurcation to aneurysm distance is significantly associated with PCoA aneurysm rupture. This is a new physically intuitive parameter that can be measured easily and therefore be readily applied in clinical practice to aid in the evaluation of patients with PCoA aneurysms.  相似文献

Therapeutic proteins are utilized in a variety of clinical applications, but side effects and rapid in vivo clearance still present hurdles. An approach that addresses both drawbacks is protein encapsulation within in a polymeric nanoparticle, which is effective but introduces the additional challenge of destabilizing the nanoparticle shell in clinically relevant locations. This study examined the effects of crosslinking self-assembled poly(l -lysine)-grafted-poly(ethylene glycol) nanoparticles with redox-responsive 3,3′-dithiobis(sulfosuccinimidyl propionate) (DTSSP) to achieve nanoparticle destabilization in a reductive environment. The polymer-protein nanoparticles (DTSSP NPs) were formed through electrostatic self-assembly and crosslinked with DTSSP, which contains a glutathione-reducible disulfide. As glutathione is upregulated in various cancers, DTSSP NPs could display destabilization within cancer cells. A library of DTSSP NPs was formed with varying copolymer to protein (C:P) and crosslinker to protein (X:P) mass ratios and characterized by size and encapsulation efficiency. DTSSP NPs with a 7:1 C:P ratio and 2:1 X:P ratio were further characterized by stability in the presence proteases and reducing agents. DTSSP NPs fully encapsulated the model protein and displayed 81% protein release when incubated with 5 mM dithiothreitol for 12 hr. This study contributes to understanding stimulus-responsive crosslinking of polymeric nanoparticles and could be foundational to clinical administration of therapeutic proteins.  相似文献

Cell-survival (of DON and L1210 cells) after treatment with cytotoxic compounds was assessed by measuring cloning efficiency, exclusion of trypan blue and erythrosin B, [51Cr] release, and attachment of DON cells to glass. Cell survival as measured by cloning efficiency did not correlate with survival measured by any of the other methods. We found that the stainability of cells after drug exposure depended on the cell line used. For example, after 3 h exposure to tubercidin although 100% of both DON and L1210 cells were killed (on basis of cloning efficiency), only 11% of DON cells and 68% of L1210 cells were dead as indicated by staining with erythrosin B. The stainability of cells also depended on the particular drug used. For example, after 24 h exposure of L1210 cells to adriamycin and tubercidin (both killed >99% of cells on basis of cloning efficiency) 21% of cells exposed to adriamycin and 99% of cells exposed to tubercidin were stained. The results obtained with several other cytotoxic compounds are discussed.  相似文献

A sequential coring approach was used to measure root biomass and production over 1 year in four different communities within the Great Dismal Swamp. A second method, an implanted bag technique, was also used to measure root production, and values were generally lower using this technique. On all sites, fine roots were the most dynamic root component. Both biomass (1,887 g/m2) and production (354–989 g m 2 yr-1) were highest on the mixed hardwood site, the least flooded site, and second highest on the cedar site, the site with the longest duration of soil saturation (1,033 g/m2 and 274–366 g m-2 yr-1). The maple-gum (696 g/m2 and 59–91 g m-2 yr-1) and cypress (824 g/m2 and 68–308 g m-2 yr-1) sites had similarly low amounts of biomass and rates of production. Environmental parameters that influenced production include frequency and duration of flooding, and soil type. Peaks in belowground production were observed on the most productive sites (mixed hardwood and cedar) in summer and late fall-winter; the other two sites exhibited little seasonal variability. The least flooded stand appears to allocate a greater percentage of net primary production belowground than the more extensively flooded stands. The ratio of above- and belowground allocation appears to change in response to a flooding gradient. This has major implications for ecosystem functions as carbon allocation patterns determine the array of litter types generated (leaves vs. roots) which affect decomposition rates and nutrient availability.  相似文献

Molecular biologists strive to infer evolutionary relationships from quantitative macromolecular comparisons obtained by immunological, DNA hybridization, electrophoretic or amino acid sequencing techniques. The problem is to find unrooted phylogenies that best approximate a given dissimilarity matrix according to a goodness-of-fit measure, for example the least-squares-fit criterion or Farris'sf statistic. Computational costs of known algorithms guaranteeing optimal solutions to these problems increase exponentially with problem size; practical computational considerations limit the algorithms to analyzing small problems. It is established here that problems of phylogenetic inference based on the least-squares-fit criterion and thef statistic are NP-complete and thus are so difficult computationally that efficient optimal algorithms are unlikely to exist for them. Ninety-nine sexually mature, non-pregnant gilts were checked for estrus daily with a mature boar and then allocated at estrus (D O) to receive 2 kg/d of a diet containing 0, 1, 5 or 10 ppm purified zearalenone between D 5 and 20 of the estrous cycle during two seasons of the year (winter and summer). None of the gilts exhibited any visual signs of "hyperestrogenism" and there was no effect of season on interestrous interval (P > 0.05). A significant effect of zearalenone dose on inter-estrous interval was detected (P < 0.001). Gilts receiving 0 or 1 ppm had similar inter-estrous intervals (21.0 +/- 0.3 and 21.5 +/- 0.8 d, respectively) whereas gilts receiving 5 and 10 ppm had extended cycles (29.2 +/- 2.9 and 32.7 +/- 3.3 d, respectively). Plasma progesterone concentrations at D 19 to 21 were higher in gilts with extended cycles (P < 0.001) and corpora lutea (CL) were present at laparotomy. Some 86% of these retained CL underwent spontaneous regression resulting in the onset of estrus within the next 30 d. Fecal zearalenone concentrations rose during ingestion of contaminated diets and declined to pretreatment values within 2 d (1 ppm) to 8 d (10 ppm) of the cessation of treatment. These data show that feeding zearalenone at concentrations of 5 to 10 ppm from D 5 to 20 of the estrous cycle causes luteal maintenance and extended inter-estrous intervals. Spontaneous regression of these CL usually occurs within 30 d after zearalenone is removed from the diet. Fecal zearalenone analysis does not appear to be an effective method for determining prior exposure to zearalenone when carried out more than a few days following the last ingestion of zearalenone.  相似文献

Entire bucks (N = 7) exhibited pronounced liveweight gains over spring and summer months (October-February), to reach a peak mean weight of 59.8 kg, and rapid liveweight losses over the rutting period (April-May) with a minimum mean liveweight of 54.2 kg. Mean neck girth and serum testosterone levels increased during late summer (January-March) and peaked at 387 mm and 12 ng/ml respectively immediately before the onset of the rut (April). Thereafter both measures declined during winter and spring months (June-December). Bucks castrated prepubertally (N = 11) exhibited similar but less pronounced changes in mean liveweight and neck girth, in the absence of any change in testosterone secretion. Peak mean testicular diameter of entire bucks (39 mm) occurred immediately before the rut and was followed by testicular regression over winter and spring months (June-November), such that the testes attained their minimum mean size of 18 mm diameter in early summer (November). Motile spermatozoa were absent from ejaculates collected in summer (November 1983, 1984; January 1984). However, ejaculates collected pre-rut (late March), immediately post-rut (June) and in early spring (September) contained successively increasing numbers of motile spermatozoa. A further 14 polled, entire bucks were given orally 5 mg (N = 7; Group A) or 20 mg (N = 7; Group B) melatonin at 15:30 h daily from 1 December 1983 to 14 January 1984 (45 days). Seven control bucks (Group C) received vehicle ration only. The measurements taken for bucks in Groups A and B were not significantly different (P greater than 0.1) on any sampling date and the data for these 2 groups were pooled. Mean serum testosterone concentrations and mean ejaculate volume were not significantly different between melatonin-treated and control bucks on any sampling date, although other measures exhibited significant differences (P less than 0.05) at various treatment or post-treatment dates: melatonin-treated bucks showed a transiently greater increase in neck muscle development during and immediately after treatment, a slight retardation of liveweight gain between 45 and 75 days after treatment, an earlier peak in maximum mean testicular diameter and an earlier onset of sperm presence in ejaculates.  相似文献
