Previously we reported that, lactic acid bacteria (LAB) can induce human dermal fibroblast (HDF) cells to form multipotent cell clusters which are able to transdifferentiate into three germ layer derived cell lineages. Later on, we confirmed that ribosome is responsible for the LAB-induced transdifferentiation and ribosomes from diverse organisms can mimic the LAB effect on HDF cells. In our present study we have shown that, upon incorporation of ribosomes, non-small cell lung cancer cell line A549 and gastric tubular adenocarcinoma cell line H-111-TC are transformed into spheroid like morphology those can be transdifferentiated into adipocytes and osteoblast. Our qPCR analysis has revealed that, during the formation of ribosome induced cancer cell spheroids, the expression of the cancer cell associated markers and cell cycle/proliferation markers were altered at different time point. Through our investigation, here we report a novel and a non-invasive approach for cancer cell reprogramming by incorporating ribosomes.  相似文献

This study demonstrates the effects of simulated microgravity on E. coli K 12 MG1655 grown on LB medium supplemented with glycerol. Global gene expression analysis indicated that the expressions of hundred genes were significantly altered in simulated microgravity conditions compared to that of normal gravity conditions. Under these conditions genes coding for adaptation to stress are up regulated (sufE and ssrA) and simultaneously genes coding for membrane transporters (ompC, exbB, actP, mgtA, cysW and nikB) and carbohydrate catabolic processes (ldcC, ptsA, rhaD and rhaS) are down regulated. The enhanced growth in simulated gravity conditions may be because of the adequate supply of energy/reducing equivalents and up regulation of genes involved in DNA replication (srmB) and repression of the genes encoding for nucleoside metabolism (dfp, pyrD and spoT). In addition, E. coli cultured in LB medium supplemented with glycerol (so as to protect the cells from freezing temperatures) do not exhibit multiple stress responses that are normally observed when cells are exposed to microgravity in LB medium without glycerol.  相似文献

Specialized microenvironment, or neurogenic niche, in embryonic and postnatal mouse brain plays critical roles during neurogenesis throughout adulthood. The subventricular zone (SVZ) and the dentate gyrus (DG) of hippocampus in the mouse brain are two major neurogenic niches where neurogenesis is directed by numerous regulatory factors. Now, we report Akhirin (AKH), a stem cell maintenance factor in mouse spinal cord, plays a pivotal regulatory role in the SVZ and in the DG. AKH showed specific distribution during development in embryonic and postnatal neurogenic niches. Loss of AKH led to abnormal development of the ventricular zone and the DG along with reduction of cellular proliferation in both regions. In AKH knockout mice (AKH−/−), quiescent neural stem cells (NSCs) increased, while proliferative NSCs or neural progenitor cells decreased at both neurogenic niches. In vitro NSC culture assay showed increased number of neurospheres and reduced neurogenesis in AKH−/−. These results indicate that AKH, at the neurogenic niche, exerts dynamic regulatory role on NSC self-renewal, proliferation and differentiation during SVZ and hippocampal neurogenesis.  相似文献

Coral sands off Réunion Island and Rodrigues (Mascarene archipelago, Western Indian Ocean) support diverse diatom communities, particularly rich in Monoraphidineae. Recent surveys of reef environments (June 2005 and June 2007) permitted the recognition of several taxa belonging to the diatom genus Cocconeis, among which are two new, relatively small taxa, epipsammic on coral sand. Cocconeis coronata Riaux-Gobin et Romero sp. nov. is similar to C. scutellum but has its own distinctive features. In external view, the concave raphe valve possesses uniseriate striae and a subsident hyaline marginal rim, while the strongly convex sternum valve has a submarginal, generally continuous crest, a hexagonal pattern of areolation, short, rounded spines that are often regularly arranged around valve face areolae, a complex areola hymen and small marginal chambers. Some morphometric differences were noted between the populations of C. coronata at Réunion and Rodrigues. Cocconeis margaritata Riaux-Gobin et Al-Handal, sp. nov. is smaller, possesses a sternum valve like that of C. placentula and C. neothumensis, but has denser striation and small pearl-like concretions around the areola aperture on the sternum valve. Several other taxa that resemble, or may be varieties of, C. margaritata require further study before being formally described. Cocconeis margaritata and C. coronata are small and relatively rare, so their study requires scanning electron microscopy.  相似文献

Interferons are important proteins for the immune system because of their antiviral, anti‐proliferating and immunomodulatory activities. Therapeutic value of these proteins against certain types of tumors caused interest and investigations aimed to obtain highly purified interferons. Molecular imprinting is an efficient method for purification with high selectivity, specificity and good reproducibility. In this study, we utilized advantages of molecular imprinting technique for the purification of interferon from human gingival fibroblast culture. For this purpose, interferon α‐2b imprinted poly(hydroxyethyl methacrylate) cryogel (hIFN‐α‐MIP) was prepared. Optimum adsorption conditions were determined, and maximum adsorption capacity of hIFN‐α‐MIP cryogel was found as 254.8 × 104 IU/g from aqueous solution. All interferon measurements are expressed as International Unit (IU), which is a unit measurement used to quantify biologically active substances like interferon based on their biological activity or effect. Selectivity experiments were performed using competitive proteins and repeated adsorption–desorption studies showed that the adsorption capacity maintained almost at a constant value after ten cycles. For the purification of interferon from human gingival fibroblast culture, fast protein liquid chromatography was used and the specific activity of the purified interferon α‐2b on HeLa cell line was found between the values 3.45 × 108 IU/mg and 3.75 × 108 IU/mg. Caregivers of HIV-positive children were interviewed in the Mbarara and Isingiro districts of Uganda to identify current trends in practices related to HIV testing and the disclosure of HIV status to the child. A total of 28 caregivers of at least one HIV-positive child participated in semi-structured interviews exploring when and why they tested the child for HIV, when the child was informed of their positive status, and what the caregiver did to prepare themselves and the child for status disclosure. For a majority (96%) of respondents, the decision to test the child for HIV was due to existing illness in either the child or a relative. Other common themes identified included the existence of stigma in the caregivers’ communities and doubt that the children truly understood what was being explained to them when their status was disclosed. Most (65%) children were informed of their HIV status between the ages of 5 and 9, with the mean age of disclosure occurring at the age of 7. General provision of HIV information typically began at the same age as disclosure, and as many as two thirds (64%) of the caregivers sought advice from an HIV counsellor prior to disclosure. How a caregiver chose to prepare themselves and the child did not affect the caregiver’s perception of whether the disclosure experience was beneficial or not. These findings suggest that the HIV disclosure experience in Mbarara and Isingiro districts differs from current guidelines, especially with respect to age of disclosure, how caregivers prepare themselves and the child, and approaching disclosure as an ongoing process. The doubts expressed by caregivers regarding the child’s level of HIV understanding following the disclosure experience suggest the children may be insufficiently prepared at the time of the initial disclosure event. The findings also suggest that examining the content of pre-disclosure counselling and HIV education, and how health care professionals are trained to facilitate the disclosure process as important avenues for further research.  相似文献

Background an aims

Nutrient use overlap among sympatric Streptomyces populations is correlated with pathogen inhibitory capacity, yet there is little information on either the factors that influence nutrient use overlap among coexisting populations or the diversity of nutrient use among soil Streptomyces.

Methods

We examined the effects of plant host and plant species richness on nutrient use of Streptomyces isolated from the rhizosphere of Andropogon gerardii (Ag) and Lespedeza capitata (Lc) growing in communities of 1 (monoculture) or 16 (polyculture) plant species. Growth on 95 carbon sources was assessed over 5d.

Results

Cumulative growth was significantly greater for polyculture vs. monoculture isolates, and for Lc vs. Ag isolates. Isolates from monocultures, but not polycultures, exhibited a drop in growth rates between 24 h and 72 h post-inoculation, suggesting resource allocation to non-growth functions. Isolates from high-carbon (polyculture) or high-nitrogen (Lc) soils had larger niche widths than isolates from low-C (monocultures) or low-N (Ag) soils. Sympatric isolates from polycultures were significantly more differentiated from one another in preferred nutrients for growth than sympatric isolates from monocultures.

Conclusions

These results suggest that Streptomyces populations respond to selection imposed by plant host and plant community richness and that populations from polyculture but not from monoculture, mediate resource competition via niche differentiation.

The (Na+ + K+)-stimulated Mg2+-ATPase, but not the Mg2+-ATPase, is irreversibly inhibited when turtle bladder microsomes were incubated with hydroxylamine.

The Mg2+-dependent or the (Mg2+ + Na+)-dependent phosphorylation of ADP by the phospho-protein (the exchange reaction) is reversibly inhibited when the microsomes are incubated with hydroxylamine.

The Na+-induced increment of 32P-labelling of microsomes previously incubated with [λ-32P]ATP is completely eliminated by hydroxylamine, but the Mg2+-dependent 32P-labelling of such microsomes is unaffected by hydroxylamine.

It is concluded that the phospho-enzyme formed during the Mg2+-dependent hydrolysis does not contribute to the Mg2+-dependent exchange reaction. Instead, the phosphoenzyme formed during the (Na+ + K+)-stimulated hydrolysis is apparently the only substance which phosphorylates ADP in the exchange reaction, even in the absence of Na+ and/or K+.

The hydroxylamine-sensitive nature of the sodium form of the phospho-enzyme in the (Na+ + K+)-stimulated ATPase sequence is consistent with the existence of an enzyme-acyl-phosphate bond of high internal energy with respect to that of ADP.

On the other hand, the hydroxylamine-resistant nature of the phospho-enzyme in the Mg2+-ATPase sequence suggests the existence of a non-acyl type of enzyme phosphate bond with low internal energy relative to that of ADP.  相似文献
