Traditionally regarded as a genetic disease of the cardiac sarcomere, hypertrophic cardiomyopathy (HCM) is the most common inherited cardiovascular disease and a significant cause of sudden cardiac death. While the most common etiologies of this phenotypically diverse disease lie in a handful of genes encoding critical contractile myofilament proteins, approximately 50% of patients diagnosed with HCM worldwide do not host sarcomeric gene mutations. Recently, mutations in genes encoding calcium-sensitive and calcium-handling proteins have been implicated in the pathogenesis of HCM. Among these are mutations in TNNC1- encoded cardiac troponin C, PLN-encoded phospholamban, and JPH2-encoded junctophilin 2 which have each been associated with HCM in multiple studies. In addition, mutations in RYR2-encoded ryanodine receptor 2, CASQ2-encoded calsequestrin 2, CALR3-encoded calreticulin 3, and SRI-encoded sorcin have been associated with HCM, although more studies are required to validate initial findings. While a relatively uncommon cause of HCM, mutations in genes that encode calcium-handling proteins represent an emerging genetic subset of HCM. Furthermore, these naturally occurring disease-associated mutations have provided useful molecular tools for uncovering novel mechanisms of disease pathogenesis, increasing our understanding of basic cardiac physiology, and dissecting important structure-function relationships within these proteins.  相似文献

Background  

Immune response pathways have been relatively well-conserved across animal species, with similar systems in both mammals and invertebrates. Interestingly, honey bees have substantially reduced numbers of genes associated with immune function compared with solitary insect species. However, social species such as honey bees provide an excellent environment for pathogen or parasite transmission with controlled environmental conditions in the hive, high population densities, and frequent interactions. This suggests that honey bees may have developed complementary mechanisms, such as behavioral modifications, to deal with disease.  相似文献

Interleukin-8 (IL8) is believed to play a role in the pathogenesis of bronchiolitis, a common viral disease of infancy, and a recent U.K. family study identified an association between this disease and the IL8-251A allele. In the present study we report data, from a different set of families, which replicate this finding; combined analysis of 194 nuclear families through use of the transmission/disequilibrium test gives P = .001. To explore the underlying genetic cause, we identified nine single-nucleotide polymorphisms (SNPs) in a 7.6-kb segment spanning the IL8 gene and its promoter region and used six of these SNPs to define the haplotypic structure of the IL8 locus. The IL8-251A allele resides on two haplotypes, only one of which is associated with disease, suggesting that this may not be the functional allele. Europeans show an unusual haplotype genealogy that is dominated by two common haplotypes differing at multiple sites, whereas Africans have much greater haplotypic diversity. These marked haplotype-frequency differences give an F(ST) of.25, and, in the European sample, both Tajima's D statistic (D = 2.58, P = .007) and the Hudson/Kreitman/Aguade test (chi(2) = 4.9, P = .03) reject neutral equilibrium, suggesting that selective pressure may have acted on this locus.  相似文献

A quantitative examination of the reef fish assemblage at Orpheus Island, Great Barrier Reef, contrasting clove oil and rotenone, sampled 365 individuals of 47 species with clove oil v. 536 individuals of 53 species with rotenone. Number of species and individuals were not found to differ significantly between the two techniques, largely due to variation among samples. Neopomacentrus bankieri (Pomacentridae) and Eviota queenslandica (Gobiidae) were the most dominant in the samples using either technique. Although the samples appeared to be comparable, only 31 species (45%) in eight families were common to both techniques. Fishes often recovered before collection when using clove oil and were not driven out of the reef during induction to anaesthesia. Although the samples collected with clove oil approximate the results obtained using rotenone, enclosed rotenone stations are the preferred method for providing relatively complete quantitative samples.  相似文献

Phosphorus, an essential plant nutrient, may become toxic when accumulated by plants to high concentrations. Certain plant species such as Verticordia plumosa L. suffer from P toxicity at solution concentrations far lower than most other plant species. In this study, exposure of V. plumosa plants to a solution containing as low as 3 mg l–1 P resulted in significant growth inhibition and typical symptoms of P toxicity. In a wide range of P levels studied, micronutrient concentrations in V. plumosa leaves were within the range considered adequate for optimal growth. Notably, tomato plants with high hexokinase activity due to overexpression of Arabidopsis hexokinase (AtHXK1) exhibited senescence symptoms similar to those of P toxic V. plumosa. The resemblance in senescence symptoms between P-toxic tomato plants and those with high hexokinase activity suggested that increased sugar metabolism could play a role in P toxicity in plants. To test this hypothesis, we determined the amount of hexose phosphate, the product of hexokinase, in V. plumosa leaves grown at various P levels in the nutrient solution. Positive correlations were found between concentration in the medium, P concentration in the plant, hexose phosphate concentration in leaves and P toxicity symptoms. Foliar Zn application suppressed P toxicity symptoms and reduced the level of hexose phosphate in leaves. Furthermore, Zn also inhibited hexokinase activity in vitro. Based on these results we suggest that P toxicity involves sugar metabolism via increased activity of hexokinase that accelerates senescence  相似文献

The traditional view regarding the pathogenesis of cervical lymphadenitis in guinea pigs is that Lancefield Group C Streptococcus gains access to cervical lymph nodes via an abraded oral mucosa. In this study, it is established that inoculation of intact nasal and conjunctival mucous membranes with Streptococcus zooepidemicus (Lancefield Group C) also can produce the disease. Weanling (SPF) guinea pigs (Cavia porcellus) were divided into two experimental groups of 10 and two control groups of four each. Guinea pigs from each group were individually housed in separate cubicles. Group I was inoculated with 0.05 ml of culture containing 2.8 x 10(7) CFU/ml of S. zooepidemicus into the conjunctiva of the left eye. Group II received a similar inoculum into the left nares. Control groups received 0.05 ml of TSB broth in the same sites. Five of ten guinea pigs in Group II died four to nine days postinoculation. Surviving guinea pigs were euthanatized at intervals between days 4-13 postinoculation. All guinea pigs were necropsied, cultured and examined for evidence of infection. S. zooepidemicus was recovered from 30/50 and 39/46 sites cultured from Groups I and II, respectively. Lymphadenitis was found in cervical lymph nodes from 8/10 guinea pigs in Group I and 10/10 in Group II. The conjunctival and nasal mucosa, therefore, represent potential sites of entry resulting in cervical lymphadenitis in guinea pigs.  相似文献
