Overcoming challenges on using native seeds for restoration of megadiverse resource‐poor environments: a reply to Madsen et al.

Madsen et al. (2016) reviewed several major limiting factors to establishment of seedlings in nonforest ecosystems (NFE), and proposed seed enhancement technologies to overcome these restoration barriers. However, biodiverse nutrient‐poor NFE present additional hurdles that preclude landscape‐scale seed‐based restoration and were not mentioned in their review. Here, we discuss issues related to native seed availability and provenance, and shortfalls in knowledge on seed quality testing and dormancy release that severely hamper restoration of degraded nutrient‐impoverished NFE. We present alternatives for overcoming these challenges and highlight the need for investments to find more practical and cost‐effective options for broad‐scale restoration.

Chironomidae (Insecta: Diptera) from four protected areas in the state of Pernambuco,Brazil

New records and distributional notes of Chironomidae (Insecta, Diptera) are provided for four protected areas in the state of Pernambuco, northeastern Brazil. Additionally, we also present new records and update of distributional ranges from Brazil and the Neotropical Region. In total, 810 specimens belonging to 35 genera within the subfamilies Chironominae (22 taxa), Tanypodinae (11 taxa) and Orthocladiinae (2 taxa) were found. The subfamilies Chironominae and Tanypodinae predominated. Axarus Roback, 1980 Roback, S.S. (1980), ‘New name for Anceus Roback nec Anceus Risso’, Entomological News, 91, 32.[Web of Science ®] , [Google Scholar] and the Tanytarsus ortoni-group were recorded for the first time in the state of Pernambuco, while Nanocladius Kieffer, 1913a Kieffer, J.J. (1913b), ‘Nouvelle étude sur les Chironomides de l'Indian Museum de Calcutta’, Records of the Indian Museum, 9, 119–197. [Google Scholar] was recorded for the first time in the Northeast Region of Brazil. Our results make evident how much and where current knowledge of the northeastern Brazil chironomids remains fragmentary.     

Functional reconstitution of Arabidopsis thaliana plant uncoupling mitochondrial protein (AtPUMP1) expressed in Escherichia coli

The Arabidopsis thaliana uncoupling protein (UCP) gene was expressed in Escherichia coli and isolated protein reconstituted into liposomes. Linoleic acid-induced H+ fluxes were sensitive to purine nucleotide inhibition with an apparent K(i) (in mM) of 0.8 (GDP), 0.85 (ATP), 0.98 (GTP), and 1.41 (ADP); the inhibition was pH-dependent. Kinetics of AtPUMP1-mediated H+ fluxes were determined for lauric, myristic, palmitic, oleic, linoleic, and linolenic acids. Properties of recombinant AtPUMP1 indicate that it represents a plant counterpart of animal UCP2 or UCP3. This work brings the functional and genetic approaches together for the first time, providing strong support that AtPUMP1 is truly an UCP.     

Synthesis and biological evaluation of 6-aryl-6H-pyrrolo[3,4-d]pyridazine derivatives: high-affinity ligands to the alpha 2 delta subunit of voltage gated calcium channels

A novel class of 6-aryl-6H-pyrrolo[3,4-d]pyridazine ligands for the alpha2delta subunit of voltage-gated calcium channels has been described. Substitutions in the aryl ring of the molecule were generally not tolerated, and resulted in diminished binding to the alpha2delta subunit. Modifications to the pyridazine ring revealed numerous permissive substitutions, and detailed SAR studies were carried out in this portion of the molecule. Replacement of the pyridazine ring methyl group with an aminomethyl functionality provided greatly improved potency over the initial lead. The initial lead compound displayed good rat pharmacokinetic properties, and was shown to be efficacious in the Chung model for neuropathic pain in rats.     

Synthesis and pharmacological evaluation of prenylated and benzylated pterocarpans against snake venom

Edunol (3), a pterocarpan isolated from Harpalyce brasiliana, a plant used in the northeast of Brazil against snakebites, was obtained by synthesis and showed antimyotoxic, antiproteolytic and PLA2 inhibitor properties. These proprieties could be improved through the synthesis of a bioisoster (5), where the prenyl group was replaced by the benzyl group.     

Matrilineal genetic structure within and among populations of the cooperatively breeding common marmoset,Callithrix jacchus

Common marmosets are members of the family Callitrichidae, South American primates characterized by highly social group living and cooperative breeding. In this study we analysed 1112 base pairs (bp) of the mitochondrial control region in 59 Callithrix jacchus individuals, sampled mainly from two geographically distinct field sites in N.E. Brazil. Analysis of molecular variation revealed a highly significant genetic structuring of haplotypes between social groups and between populations. Examination of matrilineal genetic structure within social groups revealed that seven of nine recorded breeding pairs were from different maternal lineages, indicating assortative mating and outbreeding. In addition to the breeders, at least six of 10 groups contained adult individuals from different matrilines, with five haplotypes present in one social group of nine animals. Groups of mixed lineages raise questions about potential reproductive conflicts of interest, and the extent of kin-selected altruism in the evolution and maintenance of cooperative breeding in this species.     

In vivo lymphocyte activation and apoptosis by lectins of the Diocleinae subtribe

This paper reports the overall effects of three lectins, extracted from Canavalia brasiliensis, Dioclea violacea, and D. grandiflora, on BALB/c mice popliteal draining lymph nodes. These lectins have presented high stimulatory capacity on lymph node T cells. Additionally, they were able to induce apoptosis and inflammation (frequently associated with high endothelial venule necrosis). The data presented here suggest that the Diocleinae lectins studied can stimulate in vivo T cell activation and apoptosis, as well as present important side effects.
