Egg yolk lipoprotein,a new supplement for the growth of mammalian cells in serum-free medium

Egg yolk lipoprotein promoted growth of a wide variety of mammalian cell lines, including plasma-cytomas and epithelial cell lines, in serum-free medium. The lipoprotein was active for cell growth when used with insulin, transferrin, ethanolamine and selenite. The most active lipoprotein fraction (YLP-pI7.5) was purified to give a single peak by chromatofocusing and gel filtration, and was homogeneous on a 0.35% agarose gel electrophoretogram. The lipoprotein was characterised as a very low density lipoprotein with a protein content of only 1.3%. This lipoprotein had an optimal concentration of 300 g/ml (4 g protein/ml). It was easily separable from proteinous molecules secreted into the serum-free medium by the cells, since it floated on the surface of the medium after addition of ammonium sulfate, to precipitate protein, and centrifugation. An associated structure of lipid and protein seemed to be still necessary for the lipoprotein to exhibit a growth promoting activity.     

The usefulness of CHAPS as a non-cytotoxic stabilizing agent in purification of growth factors

Among several detergents, a zwitterionic detergent, 3-[(3-cholamidopropyl)dimethylammonio]-1-propane sulfonate (CHAPS), was found to be least cytotoxic for cultured mammalian cells. CHAPS improved the activity recovery and elution profile of crude and purified fibroblast growth factors (FGFs) during chromatographies. Diluted preparations of FGFs were stabilized by CHAPS against the loss during storage. Amino acid sequence analysis was not disturbed by CHAPS. CHAPS was removable by reversed-phase high-performance liquid chromatography. 图论在中国慈姑属数量分类研究中的应用

武汉地区硅质磷片金藻的初步研究

1986年11月,在武汉植物所的3个池塘及东湖附近的池塘内采集的标本中,含有大量的具硅质磷片的金藻,共11种,分别隶属于Spiniferomonas, Paraphysomonas, Chrysosphaevella, Mallomonas和Synura 5个属。本文提供了这些藻的硅质磷片、刺及毛的电镜照片。这些藻在中国研究得很少,许多种都是第一次报道,它们中的大多数在世界上其它一些地区也是普遍出现的,有些则毫无疑问地属于世界性分布。     

植物激素对草莓叶片不定芽形成的影响

用试管内生长的草莓幼嫩叶片作外植体,培养在MS基本培养基上附加1.5—2.5毫克/升6—BA和0.1毫克/升NAA,可直接诱导成不定芽,诱导率可达20%。如果不定芽继代培养在同样浓度的培养基上,继而可形成大量的丛生芽。能使叶外植体形成不定芽的植物激素组合而不能使其愈伤组织分化成芽。IAA与6—BA的不同浓度组合对不定芽形成效果不明显。     

湖北贝母单鳞片砂培繁殖研究

本文针对湖北贝母生产中存在繁殖系数低的问题,研究了单鳞片砂培繁殖对提高鳞茎繁殖率的效果和原理。试验结果表明:1.单鳞片繁殖率为对照种鳞茎的5—9倍,2.低温(2—10℃)预处理4—8周和暗条件培养,能有效地提高子球形成率,促使子球迅速长大,3.植物激素(6-BA、KT、2,4-D)处理,有利于促进鳞片不定芽原基的分化,繁殖率为种茎繁殖的9—11倍;4.单鳞片繁殖的小鳞茎主要发生在鳞片基部的茎盘上,还可发生在鳞片的远轴面上,但不发生在近轴面。     

钠离子在念珠菌素修饰的双层脂膜中的运送

本文研究了念珠菌素所修饰的双层脂膜的离子渗透性、离子选择性和pH—膜电流的关系。发现该膜在一定的钠离子浓度范围内,可以作为Na⁺离子传感器,当膜两侧溶液存在着质子梯度时,念珠菌素起着一个钠离子载体的作用。     

应用蛋白印迹法检测鼻咽癌病人血清中抗Epstein—Barr病毒IgA/EA抗体
