Glucose reacts with proteins nonenzymatically under physiological conditions. Such glycation is exacerbated in diabetic patients with high levels of blood sugar and induces various complications. Human albumin serum (HSA) is the most abundant protein in plasma and is glycated by glucose. The glycation sites on HSA remain controversial among different studies. Here, we report two protein crystal structures of HSA in complex with either glucose or fructose. These crystal structures reveal the presence of linear forms of sugar for both monosaccharides. The linear form of glucose forms a covalent bond to Lys-195 of HSA, but this is not the case for fructose. Based on these structures, we propose a mechanism for glucose ring opening involving both residues Lys-195 and Lys-199. These results provide mechanistic insights to understand the glucose ring-opening reaction and the glycation of proteins by monosaccharides.  相似文献

The requirement for complex therapeutic proteins has resulted in mammalian cells, especially CHO cells, being the dominant host for recombinant protein manufacturing. In creating recombinant CHO cell lines, the expression vectors integrate into various parts of the genome leading to variable levels of expression and stability of protein production. This makes mammalian cell line development a long and laborious process. Therefore, with the intention to accelerate process development of recombinant protein production in CHO systems, UCOEs are utilized to diminish instability of production by maintaining an open chromatin surrounding in combination with MTX amplification. Chromosome painting and FISH analysis were performed to provide detailed molecular evaluation on the location of amplified genes and its relationship to the productivity and stability of the amplified cell lines. In summary, cell lines generated with vectors containing UCOEs retained stable GFP expression with MTX present (but instability was observed in the absence of MTX). UCOE cell lines displayed a higher frequency of integration into >1 chromosome than non‐UCOE group. Cell populations were more homogenous in terms of transgene location at the end of Long‐term culture (LTC). Overall our findings suggest variation in eGFP fluorescence may be attributed to changes in transgene integration profile over LTC.  相似文献

Question: What is the effect of gap size on the seedling emergence, growth and survival of four common tree species in wooded pastures? Location: A pasture in the Jura mountains, Switzerland. Methods: Seeds were sown in a complete three‐way factorial design with eight blocks in May 2003. Each block consisted of a competition treatment (four gap sizes including zero) and a mowing treatment (mown and unmown). Emergence, survival and total biomass of tree seedlings of three species (Picea abies, Acer pseudoplatanus and Fagus sylvatica) were measured. A fourth species (Abies alba) failed to germinate. Results: Gaps had a positive influence on the early stages of tree development for all species. Larger gaps favoured growth and survival more than small gaps. Seedling growth was higher when vegetation around the openings was mown. Mowing the vegetation at gap size zero enhanced both growth and survival compared to unmown vegetation. Mown gaps larger than zero had increased seedling desiccation but decreased seedling predation. Species showed similar trends in their emergence and growth responses to gap size and mowing treatments but for Picea emergence rate was higher and survivorship was lower than for Acer and Fagus. Conclusions: Gap size does matter for tree seedling success but even in more favourable large gaps only a small percentage of seedlings emerged and survived. The effects of gaps on tree seedling establishment are complex as a result of interactions between biotic and abiotic changes caused by gaps.  相似文献

1994年4~9月调查结果表明,东山湾海域霞水母的密度高峰期出现在6月25日~7月10日,生物量高峰期在7月10~8月5日;群体的伞径分布范围15~580mm,平均247.48mm,体重分布范围0.4~10250g,平均1274g;高次方程模拟的伞径生长方程式为Φt=15.1726+16.7204t+0.1926t2-0.0014t3—0.0004t4,指数高次方程模拟体重生长方程式为logeWt=-0.6885+1.3397t-0.0959t2+0.0034t3—0.00005t4;以生物经济学原理确定合理开捕为7月15日,合理开捕伞径为360mm.还讨论了表层水温和盐度与霞水母生物量和密度的关系及其资源开发等问题.  相似文献

In order to understand how MutS recognizes mismatched DNA and induces the reaction of DNA repair using ATP, the dynamics of the complexes of MutS (bound to the ADP and ATP nucleotides, or not) and DNA (with mismatched and matched base‐pairs) were investigated using molecular dynamics simulations. As for DNA, the structure of the base‐pairs of the homoduplex DNA which interacted with the DNA recognition site of MutS was intermittently disturbed, indicating that the homoduplex DNA was unstable. As for MutS, the disordered loops in the ATPase domains, which are considered to be necessary for the induction of DNA repair, were close to (away from) the nucleotide‐binding sites in the ATPase domains when the nucleotides were (not) bound to MutS. This indicates that the ATPase domains changed their structural stability upon ATP binding using the disordered loop. Conformational analysis by principal component analysis showed that the nucleotide binding changed modes which have structurally solid ATPase domains and the large bending motion of the DNA from higher to lower frequencies. In the MutS–mismatched DNA complex bound to two nucleotides, the bending motion of the DNA at low frequency modes may play a role in triggering the formation of the sliding clamp for the following DNA‐repair reaction step. Moreover, MM‐PBSA/GBSA showed that the MutS–homoduplex DNA complex bound to two nucleotides was unstable because of the unfavorable interactions between MutS and DNA. Floret opening of excised spikelets in Sorghum bicolor (L.) Moench and Sorghum Sudanesis (Piper) Stapf was significantly stimulated by immersing into 2 mM methyl jasmonate (MeJA) solution. In male sterile (MS) lines of Sorghum bicolor (L.) Moench, floret opening was more sensitive to MeJA than that in maintainer (MT) lines. Salicylic acid (SA) could abolish the effect of MeJA on the opening of spikelets. These data indicate that MeJA plays a role in floret opening in sorghum, and that SA interacts with MeJA in regulating of this process.  相似文献
