本文研究了五种常见寄主对瓜实蝇Dacus cucurbitae Coquillett实验种群增长的影响。用五种寄主(黄瓜、水瓜、南瓜、苦瓜和西瓜)饲养的瓜实蝇,对未成熟期各阶段(卵,幼虫,蛹)的发育历期和成活率的影响差异不显著。但对成虫寿命和繁殖力的影响因寄主不同而差异很大。具体表现在平均世代历期,总卵量和孵化率等方面。本文求出了一系列反映瓜实蝇实验种群生长的重要参数,包括繁殖价,以及瓜实蝇各发育阶段在稳定种群中所占的比例。  相似文献

六株虫生真菌的生物学特性及其对油茶象甲的致病力   总被引:2,自引:0,他引:2  
为了筛选防治油茶象甲的虫生真菌菌株,本研究测定了不同绿僵菌(Ma09、Ma25、Ma05、Mac985)和白僵菌(Bb01、Bb02)菌株的生长速度、产孢量以及孢子萌发率,采用孢悬液浸渍法测定了上述菌株对油茶象甲成虫和幼虫的致病力。结果表明,Ma09、Ma05、Bb01、Bb02菌株的菌落直径分别为63.13、62.55、61.99、61.90 mm(25℃、15 d),生长速度显著快于其余菌株(P0.05);Ma25、Bb01、Ma09菌株的产孢量分别为3.04×10~8、2.88×10~8、2.85×10~8孢子/cm~2,显著高于其余菌株(P0.05);Bb01和Ma09菌株的孢子萌发率较高,分别达到97.80%和96.40%。不同菌株对油茶象甲成虫和幼虫致病力差异显著,其中Bb01和Ma09在1×10~7孢子/mL时,对油茶象甲成虫的LT_(50)分别为4.64 d和4.93 d,处理后15 d累计校正死亡率均达到100.00%,僵虫率分别为76.00%和78.67%;对幼虫的LT_(50)分别为2.15 d和3.48 d,10 d累计校正死亡率均达到100.00%,僵虫率分别为81.00%和86.31%。本研究筛选的Bb01和Ma09菌株具有生长快、产孢量大、孢子萌发率高、对油茶象甲致病力强等特性,在油茶象甲的生物防治中具有潜在的应用价值。  相似文献

通过比较5株白僵菌菌株的产孢量及对刚竹毒蛾的毒力,结果发现:Bgz06菌株的产孢量为(5.4±0.3)×108孢子·cm-2,室内测定对刚竹毒蛾幼虫的LT50为5.6 d;用该菌株生产的粉剂进行林间防治试验,用量为22.5kg·hm-2时,幼虫死亡率超过90%,僵虫率超过85%,说明Bgz06菌株是防治刚竹毒蛾的优良菌株,可以进一步进行推广应用.  相似文献

为明确瓜实蝇对短时高温胁迫的耐受性。利用人工气候箱模拟短时高温胁迫,测定了不同高温处理(34、36、38、40、42、44、46、48℃)12 h,对不同发育阶段瓜实蝇的存活率和生长发育的影响。结果表明短时高温显著影响瓜实蝇的存活,随温度升高,瓜实蝇各虫态的存活率逐渐降低;高温处理12 h后瓜实蝇卵、幼虫、蛹、雌成虫、雄成虫的致死中温度LT 50分别为35.48、37.55、41.85、43.62、43.32℃;34~42℃短时高温胁迫对瓜实蝇各虫态的发育历期无明显影响,44℃时其发育历期均显著增长;46℃、48℃处理下各虫态死亡率较高,不能正常发育;随着处理温度的升高,雌成虫产卵前期不断增长,单雌产卵量呈下降趋势,成虫寿命不断缩短,后代雌性比增大。44℃及以上的短时高温胁迫不利于瓜实蝇的生长发育,40℃及以上的短时高温胁迫不利于瓜实蝇的繁殖,雌性瓜实蝇对短时高温的胁迫的适应性强于雄性,随着处理温度的升高,后代雌性比例增大。  相似文献

【目的】了解瓜实蝇Bactrocera cucurbitae(Coquillett)对低温的耐受性,分析其不同发育阶段耐寒力差异。【方法】以不同发育阶段瓜实蝇为实验材料,分别测定并比较了过冷却点、水分、脂肪、甘油和糖类含量的变化情况。【结果】其不同龄期的过冷却点差异显著,5日龄蛹的过冷却点最低,为﹣17.04℃,1日龄幼虫的过冷却点最高,为﹣11.82℃,同一日龄的雌、雄成虫之间的过冷却点无显著差异;其幼虫期和蛹期虫体的含水量随着龄期的增长而下降,从1日龄幼虫79.64%下降到5日龄蛹65.31%,5日龄蛹的含水量显著低于其他龄期瓜实蝇,成虫期各龄期雌成虫含水量均显著高于雄成虫;5日龄幼虫脂肪含量最高,为32.90%,1日龄雄成虫和14日龄雄成虫脂肪含量最低,均为15%;幼虫期甘油含量随着龄期的增长而下降,从3.32μg/mg下降到1.12μg/mg,而蛹期逐渐上升,14日龄雌成虫甘油含量最高,为5.90μg/mg;1日龄幼虫总糖、海藻糖和糖原含量均最高,分别为7.51、0.94和1.93μg/mg,显著高于其他龄期瓜实蝇,蛹期糖类含量逐渐下降,而成虫期糖类含量随龄期增长而逐渐升高。【结论】本研究结果可为正确评估瓜实蝇的地理适应性和低温检疫处理提供依据。  相似文献

本研究在测定不同球孢白僵菌Beauveria bassiana和金龟子绿僵菌Metarhizium anisopliae菌株的生长速率与产孢量的基础上, 采用孢悬液浸渍法进行了对油茶新害虫--黑足角胸叶甲Basilepta melanopus成虫的生物测定, 旨在筛选出感染该成虫的高致病力菌株, 为防治该虫提供新的生物资源。结果表明: 不同菌株生长速率和产孢量间存在显著差异, MaYTTR-04, BbFZ-17, MaZPTR-01和BbTK-01生长速率和产孢量均显著高于其他菌株。接种后, 叶甲成虫累积死亡率随时间的增加而逐渐增高, 接种白僵菌7 d后, 成虫校正死亡率全部达到100%; 接种MaZPTR-01和MaYTTR-04两个绿僵菌菌株14 d后, 成虫死亡率分别为80.3%和78.8%。而且接种白僵菌后, 叶甲成虫的僵虫率显著较绿僵菌高, 尤其以BbTK-01和BbFZ-17两个菌株较好, 分别达到85.7%和75.8%。BbXJ-01, BbFZ-17和BbTK-01 3个白僵菌菌株的LT50最小, 分别为3.0, 3.3和3.4 d; MaYTTR-04和MaZPTR-01两个绿僵菌的LT50分别为6.0和6.2 d。结果说明, 白僵菌对叶甲成虫的致病力较强, 尤其是BbTK-01和BbFZ-17两个菌株, 不仅致死率高, 且致死速度快, 僵虫率高, 同时这2个菌株生长速度快、 产孢量大, 具有优良的生产特性, 在黑足角胸叶甲的生物防治中将有重要的应用价值。  相似文献

从田间褐飞虱Nilaparvata lugens (St?l)罹病虫体上新分离鉴定一株黄绿绿僵菌Mf82,将其与实验室保存的8株绿僵菌Metarhizium spp.一起,分别测定其对褐飞虱成虫毒力和几丁质酶活力,并用扫描电镜观察侵入昆虫表皮的过程和体表的形态变化,进而分析侵入能力与几丁质酶的相关性。结果表明:菌株Mf82累计校正死亡率和几丁质酶活力均最高,为82.1%和9.78U/mg,与其他菌株差异显著。同时,扫描电镜照片显示Mf82分生孢子既可以由褐飞虱体壁节间膜和凹陷处侵入,还可以从含几丁质较多的胸部背板侵入。比较9株绿僵菌菌株菌落生长速度、产孢初始时间、产孢量和萌发率显示,Mf82菌株具有生长速度快、产孢初始时间短、萌发率高和产孢量大等优点,可见筛选的Mf82菌株致病力强,在褐飞虱的微生物防治方面将有良好的应用前景。  相似文献

栎旋木柄天牛高毒力球孢白僵菌菌株的筛选   总被引:9,自引:0,他引:9  
栎旋木柄天牛Aphrodisium sauteri是近年来在安徽黄山风景区大面积暴发的危险性林业有害生物之一,其发生危害已经严重影响了风景区自然与生态景观。本研究选择分离自不同天牛成虫的8个球孢白僵菌Beauveria bassiana菌株,依据菌落形态、菌株生长速度、产孢量、萌发率、抗旱力和产胞外蛋白酶水平等生物学性状进行初步筛选。在此基础上,筛选出生物学性状优良菌株Bb202、Bb646和Bb1898,并进一步对栎旋木柄天牛幼虫进行生物测定。基于此种筛选模式的菌株筛选方法筛选出了具有菌落生长速度快、萌发率高、产孢和抗旱能力强、胞外蛋白酶产酶水平高及毒力强的球孢白僵菌Bb202菌株。该菌株对栎旋木柄天牛的校正死亡率达92.8%,侵染率达93.3%,在浓度为1×107/mL时的半致死中时仅为5.86天,显示出对目标天牛的极强毒力,在栎旋木柄天牛的生物防治中将有重要的应用价值。  相似文献

在武昌,蚕豆象Bruchus rufimanus Boheman每年发生一代,以成虫越冬。成虫必须取食蚕豆花后才能正常交配和产卵。4月为交配盛期。4月中、下旬为产卵盛期,最喜欢把卵产到生长已有1l—20天的嫩蚕豆荚上,但不在豌豆荚上产卵。产卵历期约为9天。4月下旬至5月上旬为孵化盛期。幼虫共四龄,在豆粒内的死亡率随着单个豆粒上侵入孔的增多而增高。8月为化蛹盛期,8月中旬到9月上旬为羽化盛期。羽化出的成虫绝大部分藏在豆粒内越冬。从卵发育到成虫羽化约需120天。成虫寿命一般为212天,最长达295天,但不能度过两个冬季。  相似文献

【背景】夜蛾科害虫易对化学杀虫剂产生高抗性,但一些化学农药可以对部分虫生真菌的毒力作用效果起增幅作用,目前缺乏对莱氏野村菌(Nomuraea rileyi)的该方面研究。【目的】探究对常用有机磷杀虫剂敌敌畏具有较强耐药性的紫外线诱变莱氏野村菌突变菌株的生理特性,包括菌丝生长、产孢情况和产几丁质酶活性。【方法】在紫外线诱变莱氏野村菌结合药剂驯化筛选出的突变株Nr-UVY1和Nr-UVY6基础上,以添加不同含量敌敌畏的平板培养基对突变株进行菌丝生长抑制率、继代产孢量、几丁质酶活性测定。【结果】在含1291mg/L敌敌畏的培养基上,敌敌畏对出发菌株菌丝生长抑制率达到100%,突变株Nr-UVY1(42.38%)和Nr-UVY6(37.01%)均远低于出发菌株。处理浓度大于1291 mg/L时,2株突变株的菌丝生长抑制率曲线平稳上升且远低于出发菌株,说明突变株对敌敌畏的抗药性显著且稳定。在各浓度敌敌畏含量的培养基上,突变株Nr-UVY6得到的菌丝抑制率均小于Nr-UVY1,说明突变株Nr-UVY6对敌敌畏的抗药性更强。在同样的培养条件下,2株突变株的开始产孢时间略迟于出发菌株。突变株各代的产孢量均明显高于出发菌株,尤其是突变株Nr-UVY6各代的产孢量始终高于出发菌株2倍以上且更稳定。透明圈法测定几丁质酶活性,突变株Nr-UVY6高于Nr-UVY1和出发菌株。【结论】筛选出的经过紫外线诱变的突变菌株对敌敌畏的抗性远高于出发菌株,在生长和繁殖方面表现出更优秀的性状,并且由于其几丁质酶的活性高,可进一步探究其致病性。  相似文献

曲霉Aspergillus spp.是自然界中分布极为广泛的一大类真菌,在工业上如黑曲霉等已经被人们广泛利用于食品添加剂等生产。因此,曲霉在工业、农业、医药领域均发挥着重要的作用。然而,有些曲霉如黄曲霉能够分泌致癌物黄曲霉毒素从而污染农产品;在临床上还有一类以烟曲霉为主的引起侵袭性真菌感染的条件致病曲霉菌。因此,曲霉就像一把双刃剑影响着人们的生活。曲霉菌丝具有发达的隔膜形成多细胞菌丝体并能在分生孢子梗上产生大量的分生孢子进行无性繁殖。研究曲霉的基因编辑技术对于控制医学和农业上有害曲霉的增殖和促进工业上有益曲霉的生长和代谢都具有非常重要的意义。长期以来,同源重组和随机整合一直是被用于研究曲霉基因功能的传统基因编辑方法,然而其操作费时费力且效率低、难以达到预期的要求。随着第三代基因编辑技术CRISPR/Cas9即成簇的规律间隔的短回文重复序列及其相关系统CRISPR/Cas9(Clustered regulatory interspaced short palindromic repeats-associated protein 9)建立以来,CRISPR/Cas9以编辑高效、操作简便等优势被广泛应用到不同物种中。目前,世界上多个研究团队已经建立了有效地用于不同曲霉物种的CRISPR-Cas9 基因编辑体系。本文概述了有关曲霉基因编辑的历史和进展,以期为尚未建立完整遗传编辑体系的其他曲霉物种或者丝状真菌引入高效CRISPR-Cas9体系提供帮助。  相似文献

The efficacy of Bacillus sphaericus and B. thuringiensis israelensis produced on a new potato-based culture medium as well as the conventional culture medium of Luria Bertani was compared against Culex quinquefasciatus in the field. After sporulation, the spores/crystals were harvested and used. The bacterial samples controlled the larvae and pupae for three weeks. Mortality due to the bacterial toxins produced from the new culture medium was very high and comparable to that produced using conventional medium. But the cost for cultivating these bacteria using potato extract was much lower as compared with that of the conventional medium.  相似文献

The potential of different Aspergillus strains in carrying out the biotransformation of cyclic ketones was investigated. All the strains employed showed alcohol dehydrogenase and Baeyer–Villiger monooxygenase activities. trans-2-Methylcyclohexanol and trans-4-methylcyclohexanol were prepared in a single isomeric form by the use of Aspergillus terreus SSP 1498 and the corresponding ketones. Baeyer–Villiger oxidation of cyclic ketones by all the fungi Aspergillus led to chiral lactones in good enantioselectivity.  相似文献

In laboratory and field cage experiments, the horizontal transmission of the entomopathogenic fungus Beauveria bassiana (Boverol®) between adults of the spruce bark beetle Ips typographus was examined. In a laboratory experiment, conidia transfer between treated and untreated beetles at different ratios resulted in 96% mortality (MST 4.3 days) at a ratio of 1:1 and 90% mortality (MST 4.5 days) at a ratio of 1:2. At ratios of 1:5, 1:10 and 1:20, the mortalities at 7 days were 83, 77 and 75%, respectively. In a second laboratory experiment, the efficacy of B. bassiana was investigated after transmission from contaminated to healthy beetles over a period of 5 days. Two weeks later, the mortality of treated and untreated beetles was 99% (control mortality was 44%), while the rate of mycosis was 79% in the treatment compared to 10% in the control beetles. A single contact between one treated male (2.0×105 conidia/beetle) and an untreated female was found to be sufficient to transmit a lethal dose of B. bassiana of about 1.2×104 conidia. Two experiments in field cages in a spruce stand demonstrated horizontal transmission of B. bassiana (Boverol®) between beetles. In the first experiment the transmission was investigated following the introduction of B. bassiana-inoculated beetles into a population of untreated beetles. Significant reductions were observed in the length of maternal galleries, and the number of larvae and pupae. In the second field cage experiment, natural populations of beetles were lured into a flight cage using a pheromone trap, and treated with B. bassiana or left untreated on alternate days. In this experiment, significant reductions were observed in the number of bore holes and the length of maternal galleries, while no larvae, pupae or juveniles were found under the bark of spruce trunks in the Boverol® transmission treatment. It is concluded that B. bassiana has potential for control of I. typographus, and further field experiments are warranted to investigate its efficacy under more practical conditions.  相似文献

The effect of temperature regimes on conidia germination, radial growth and virulence of Beauveriabassiana and Metarhizium anisopliae against the spotted stalk borer Chilo partellus was investigated with Ethiopian isolates. Conidia germination, radial growth and sporulation of all isolates were retarded at 15 and 35°C. A suitable temperature range for the isolates was between 20 to 30°C. Conidia germination was less tolerant of low temperature (15°C) than radial growth. Radial growth and sporulation reacted differently to temperature. At both 25 and 30°C, all isolates induced 100% mortality to C. partellus larvae in six days. The LT50 decreased for the isolates with increasing temperature.  相似文献

The susceptibility of third instar Helicoverpa armigera to seven strains of three entomopathogenic fungal species, i.e. Metarhizium anisopliae, Beauveria bassiana and Paecilomyces fumosoroseus, was tested under laboratory conditions using the larval immersion method. High efficacies ranging from 68 to 100% corrected mortality were recorded with more profound effects in treatments with B. bassiana and P. fumosoroseus strains. The median lethal concentration (LC50) for L3 was 6.0×105 in M. anisopliae 79, 1.5×105 in B. bassiana 124 and 4.2×104 in P. fumosoroseus 14. These three strains were further used to characterize the age-dependent mortality of different larval stages (L2-L5) and the effect against pupae of H. armigera. Larval stages did not differ in their mortality but differed i in median lethal time, with shorter values recorded in the second instar. Tested fungi also caused a high reduction between 74.4 and 100% in the emergence of pupae using the soil inoculation method and the pupal immersion technique. All three fungal species, especially P. fumosoroseus, have a high potential for biocontrol of H. armigera larvae and also as a soil treatment targeting the pupae.  相似文献

为获取对草地贪夜蛾Spodoptera frugiperda具有高致病力的生防真菌,从福建省不同地区分离得到8株寄主为鳞翅目和半翅目幼虫僵虫的绿僵菌Metarhizium,采用浸渍法测定了其对草地贪夜蛾2龄幼虫和蛹的致病力,并根据形态学和分子生物学方法对高致病力菌株进行种类鉴定。结果表明,8株绿僵菌菌株对草地贪夜蛾2龄幼虫和蛹均表现出不同程度的致病力,其中菌株FJMR2和FJXY7表现出较强的致病力。在5×107个/mL孢子浓度下,FJMR2和FJXY7对草地贪夜蛾2龄幼虫的致死率分别为88.76%和82.13%,对蛹的致死率分别为86.57%和84.00%;对草地贪夜蛾2龄幼虫的LT50分别为4.81 d和4.93 d,对蛹的LT50分别为4.94 d和4.83 d。经鉴定菌株FJMR2和FJXY7均为莱氏绿僵菌Metarhizium rileyi。本研究获得2株对草地贪夜蛾2龄幼虫和蛹具有高致病力的莱氏绿僵菌菌株,在草地贪夜蛾的生物防治中具有较大应用潜力。  相似文献

我国果胶酶制剂使用广泛但专一性不高,高效、专一的果胶酶制剂在市场上仍然匮乏。利用基因工程技术改造果胶酶生产菌株——黑曲霉来生产单一成分的果胶酶成为解决果胶酶应用需求的一种有效方案。构建一种高效的CRISPR-Cas9基因编辑技术,可为构建高产单一性果胶酶的黑曲霉底盘菌株提供有效的基因编辑工具。首先敲除产果胶酶黑曲霉基因组上的pyrG基因构建尿嘧啶营养缺陷型菌株AnΔpyrG,并在AnΔpyrG菌株的pyrG基因位点定点整合Cas9基因表达盒和pyrG基因表达盒,构建组成型表达Cas9基因的黑曲霉菌株AnCas9,再构建含有gpdA启动子、锤头结构核酶、HDV核酶的稳定性表达sgRNA的pLM2-sgRNA质粒,建立CRISPR-Cas9基因编辑体系。利用该技术失活AnCas9菌株中的2个聚半乳糖醛酸酶基因4978020和4983861来检测构建的CRISPR-Cas9基因编辑效率并检测4978020基因功能缺失菌株的表型变化和产酶变化,结果表明果胶酶基因编辑效率大于50%,AnΔ4978020的表型和果胶酶酶活性与出发菌株均无明显变化。在黑曲霉中成功构建了高效的Cas9基因编辑技术,4978020基因功能缺失也不影响菌株表型,为构建高产单一性果胶酶黑曲霉底盘菌株奠定基础。  相似文献

【目的】研究昆虫病原真菌蜡蚧霉Lecanicilliurn lecanii(Zimmermann.)菌株No.V3.4504在不同培养基上继代培养,对菌种的菌落生长特性、胞外酶活力和对蚧虫致病力的影响。【方法】试验菌种蜡蚧霉菌株No.V3.4504是从染病蚧虫上分离的。试验蚧虫是沙里院褐球蚧Rhodococcus sariuoni Borchsenius和日本龟蜡蚧Ceroplastes japonicus Green。采用7种培养基继代培养多代。观察菌落形态特征、测定生长速率、产孢量、胞外蛋白酶和几丁质酶活性及对蚧虫的致死率。【结果】在PDA培养基上,菌落生长速率最快,但产孢量最低,胞外蛋白酶和几丁质酶的活性均呈逐代下降趋势,对两种蚧虫致死率也最低;增加蛋白胨对改善菌种致病力没有明显效果;在增加蚧虫尸体的D、E、F培养基上,菌落生长速率虽然较慢,但产孢量上升为8.83×106-9.13×106孢子/cm2。蛋白酶和几丁质酶的活性平均达到2.16-2.13 U/g和1.01-1.03 U/g,对两种蚧虫的致死率分别在55%-58%和39%-42%;在活蚧虫上连续培养3代,蛋白酶和几丁质酶的活性最高,为3.08-2.92 U/g和1.45-1.42 U/g,是PDA培养基上的1.6倍。对两种蚧虫的致死率也最高,分别达到71.30%和58.89%。蛋白酶和几丁质酶的活性与蚧虫死亡率呈正直线相关关系。【结论】采用PDA培养基连续多代培养会引起菌株No.V3.4504明显退化;在培养基中加入蚧虫尸体,对于保持菌种活力有明显效果;在活蚧虫体上继代培养对复壮菌种,提高菌种毒力的效果最佳。  相似文献

To investigate the role of aiiA gene in Bacillus thuringiensis, B. thuringiensis strain BMB171 was selected and three derivatives were constructed, such that (1) aiiA was knocked out (aiiA-KO), (2) aiiA-KO was reversed (aiiA-Rev) and (3) the expression of aiiA was elevated (aiiA-Elevated). First, we found that mutation of aiiA increased the ethanol tolerance in B. thuringiensis. Omitting ethanol from the medium resulted in a decreased generation time for aiiA-Elevated. In contrast, aiiA-KO had the highest ethanol tolerance when the medium contained 2 or 4% ethanol. Second, we found that more melanin was produced at 37 °C when aiiA expression was elevated. Third, we found inhibition of swarming motility in the aiiA-Elevated group. Finally, there was delayed sporulation in Luria–Bertani, ICPM, casein and ammonium sulfate medium in the aiiA-Elevated group. The conclusion was that ethanol tolerance, yield of melanin, swarming motility and growth are correlated with the expression levels of aiiA gene in B. thuringiensis.  相似文献
