梅花鹿的微卫星多态性及种群的遗传结构   总被引:8,自引:1,他引:7  
梅花鹿是我国极度濒危的鹿科动物,其野生种群已濒临灭绝。为了探讨我国野生梅花鹿的保护和管理对策,我们选用了16 个微卫星位点检测来自东北、四川、江西和浙江种群的122 份样品,以此分析我国野生梅花鹿种群的遗传多样性和遗传结构。费希尔确切性检验表明,四个种群中均存在偏离哈迪- 温伯格平衡的现象。与其它的濒危动物相比,中国梅花鹿有着相对较高的遗传多样性:每个位点的平均等位基因数为4 个,平均期望杂合度为0. 559。四个种群中,东北种群拥有最高的平均等位基因数(n = 3. 688),东北种群、四川种群、江西种群以及浙江种群的平均期望杂合度分别为0. 584,0. 477,0. 585 和0.589,它们之间不存在显著的差异。同时,利用逐步突变模型、双相突变模型和无限等位基因突变模型检测了种群的瓶颈效应,结果表明:除四川种群外,其他种群在近期内都经历过遗传瓶颈。费希尔确切性检验及配对样品F ST 的结果均表明:四个梅花鹿种群间存在显著的遗传分化(P < 0. 001)。因此,我们建议将我国梅花鹿的野生种群划分为4 个管理单元进行保护和

该研究利用 ISSR 分子标记,对不同生境下的6个河口莲座蕨种群进行遗传多样性分析。结果表明:从44条ISSR引物中筛选出8条能够扩增出清晰、稳定条带的引物,对6个河口莲座蕨种群进行基因组 DNA扩增,共扩增出144条带,其中多态性条带119个,多态性条带比率为93.7%,Nei’s遗传多样性指数(H)为0.296,Shannon多样性指数(I)为0.457,遗传分化指数(Gst)为0.1520,种群遗传一致度和遗传距离分别为0.9138~0.9548和0.0584~0.0901;UPGMA聚类分析表明,种群间遗传距离与空间距离和生境类型有关。河口莲座蕨在不同干扰程度的生境中,种群具有高水平的遗传多样性,一定强度或频率的干扰生境中,种群遗传多样性较高,与海拔、坡度和坡向无相关性。  相似文献

东北梅花鹿(Cervus nippon hortulorum)的野生种群已濒临灭绝,但其圈养种群遍布全国各地,是我国圈养梅花鹿的主要品种(亚种)。为了探讨圈养梅花鹿种群作为东北梅花鹿野外放归项目资源种群的可行性,测定了来自9个圈养种群45只梅花鹿个体的线粒体DNA控制区的部分序列,以此分析我国圈养梅花鹿种群的遗传多样性和遗传结构。结果表明,我国圈养梅花鹿种群的遗传多样性并不贫乏,种群之间并没有发生显著的遗传分化。因此,东北地区的圈养梅花鹿种群可以作为野外放归项目的资源种群,而野外放归项目的建群者应来源于东北地区的多个圈养种群。  相似文献

保护自然生态系统的完整性是我国建立国家公园的主要目标之一, 《国家公园设立规范》(GB/T 39737-2021)已将生态系统完整性作为国家公园的准入条件生态重要性的认定指标之一, 但目前对国家公园生态系统完整性的内涵还没有统一定义, 并缺乏具体的评价体系。本文在国内外相关研究的基础上, 结合我国国家公园体制试点成果及第一批国家公园设立实践, 提出了国家公园生态系统完整性的基本内涵及评价指标体系, 以期为今后设立国家公园时划定边界范围或评估国家公园管理成效提供理论依据。作者提出, 应从生态系统结构和过程完整性、功能完整性、空间格局完整性3个维度对我国国家公园生态系统完整性进行综合评价。以东北虎豹国家公园为例, 体制试点以来, 东北虎(Panthera tigris altaica)和东北豹(P. pardus orientalis)等顶级食肉动物的食物链相对完整, 虎、豹野生种群从俄罗斯进入、定居、繁衍、扩散的路径清晰, 种群稳定增长、栖息地质量持续改善, 生物多样性日益丰富, 国家公园正式设立范围划定时充分考虑了生态系统完整性保护, 生态系统完整性评级为优秀, 为国家公园可持续发展奠定了良好基础。  相似文献

青藏高原垂穗披碱草遗传变异的地理因素分析   总被引:14,自引:2,他引:12  
采用等位酶标记对我国青藏高原地区垂穗披碱草的9个种群进行遗传变异和系统关系分析.结果表明,不同种群间遗传距离范围在0.010 6-0.377 8,种群间遗传距离和地理环境关系密切.采用聚类分析和二维、三维Mantel检验,对青藏高原垂穗披碱草(Elymusnutans Griseb.)不同种群间遗传距离与环境条件的关系(纬度、经度和海拔)进一步分析发现,海拔和地理位置(纬度和经度)均明显影响垂穗披碱草种群遗传差异.种群间遗传距离和海拔(R=0.476 8)、地理位置(R=0.447 1)均呈现显著正相关,种群间遗传距离随着海拔、地理位置差距的增大而增加,但海拔是影响其遗传结构的最重要因素,其次是经纬度.  相似文献

梅花鹿东北亚种(Cervus nippon hortulorum)曾被认为已野外灭绝,近年来在黑龙江东南部和吉林东部临近边境地区发现少量分布,其生境隔离、面积狭小,破碎化严重。亟需对其种群的遗传变化,特别是遗传多样性和近交衰退等种群遗传信息开展进一步评价,增强保护与管理的针对性。本研究在大、小兴安岭和长白山设计9个重点研究区域,共收集673份疑似梅花鹿粪样样本。首先基于线粒体DNA Cyt b基因测序技术开展物种鉴定,并对鉴定为梅花鹿的阳性样本利用微卫星技术进行个体识别。结果证实,东北梅花鹿仅在老爷岭东部山脉有分布,106份梅花鹿粪便DNA中识别出33只个体(穆棱保护区20只,老爷岭保护区13只)。33个Cyt b基因序列共检测出6个变异位点和5个单倍型,单倍型多样性指数(Hd)为0.621,核苷酸多样性指数(Pi)为0.006 7;微卫星检出种群平均等位基因数(Na)7.1个,观测杂合度(Ho)0.604,期望杂合度(He)0.712,固定系数(Fis)0.152。结果表明,东北梅花鹿种群遗传多样性丰富,但也存在一定程度的杂合度不足和近亲繁殖;种群近期经历了瓶颈效应,未发生种群扩张;群体间无遗传分化,可作为一个管理单元加以保护。建议,对东北梅花鹿稀有单倍型个体重点监测和保护,恰当时期考虑圈养种群野外放归,以提高野外个体间基因交流和快速种群恢复。  相似文献

2009年3~6月,采用样方法对四川省洪雅县人工林赤腹松鼠危害相关的生境因子进行了调查。共设置了240个样方。样方调查中记录了2个反应危害等级的变量(危害株数和乔木总数)和11个生境变量(森林类型、海拔、坡向、坡度、坡位、距最近道路距离、距最近水源距离、乔木均高、乔木平均胸围、灌木盖度和草本盖度)。通过广义线性模型研究这些因子与人工林赤腹松鼠危害程度的关系。结果显示,赤腹松鼠危害程度与森林类型、坡度、坡向、乔木平均胸围呈显著性相关,与其他因子无显著相关性,在柳杉-杉木林中的危害高于纯柳杉林,在阴坡的危害低于其他坡向,危害程度随坡度和乔木平均胸围的增加而增加。  相似文献

采用筛选的8条ISSR引物对濒危植物观光木13个野生种群的遗传多样性及遗传结构进行分析,结果显示:物种和种群水平上多态性位点百分率分别为79.67%和46.84%,Shannon表型多样性指数分别为0.3880和0.2192,与木兰科其他近缘植物相比,遗传多样性水平较高;分子方差分析表明,观光木野生种群间遗传分化系数为0.3752,种群间存在显著的遗传距离;STRUCTURE分析和UPGMA聚类表明,参试的13个野生种群可分为3大类群;相关性分析表明,种群间遗传距离与其地理距离呈显著正相关。南昆山、笔架山、弄相山种群的遗传多样性最丰富,建议优先加以保护。  相似文献

川金丝猴湖北种群是川金丝猴分布的最东缘,现主要分布于神农架国家公园和巴东自然保护区,其作为单独的管理单元,相对较低的遗传多样性、孤立的遗传状态和较小的种群数量,使得该种群面对环境变化脆弱性高。以神农架为研究区域,首先基于物种分布模型获得川金丝猴移动的阻力图层,利用电路理论分析适宜生境连接度;其次,设置道路和旅游两个人为干扰情景,运用最小费用距离模型与电路理论模型,分析道路和旅游活动对川金丝猴移动路径以及对不同亚群内和亚群间连通性的影响;最后,利用CDPOP软件模拟道路对川金丝猴遗传多样性的影响。结果表明:(1)神农架川金丝猴适宜生境分布较集中,生境斑块间存在多个潜在扩散区域;(2)道路改变了川金丝猴往来于南北生境间的最优路径,旅游干扰对川金丝猴移动路径无明显影响,其干扰主要在于川金丝猴对其具有回避性,回避距离为100 m;(3)道路显著降低金猴岭亚群内的连通性,并显著增加了所有亚群间的移动阻力,旅游活动对亚群内和亚群间的连通性无显著影响;(4)川金丝猴种群在150年内的遗传多样性呈下降趋势,道路的影响将加剧遗传多样性下降的程度。基于以上结果本文提出了针对性的保护建议,对于该物种的保护和其他濒危物种的类似研究具有较强的参考价值和借鉴意义。  相似文献

选择坡度、海拔、坡向、到道路和河流及居民点的最近距离等作为植被变化的空间驱动因子,分别采用传统Logistic和AutoLogistic回归模型,对比研究湖南张家界女儿寨小流域植被变化的驱动力,并用ROC方法检验这两个模型的解释能力。结果表明:(1)在女儿寨小流域,各类植被变化和空间驱动因子均表现出一定的空间正自相关,且这种空间自相关性随着距离的增加而逐渐减弱。(2)考虑空间自相关性的AutoLogistic模型比传统Logistic模型的精度更高,这说明在分析小流域植被变化的空间驱动力时不能忽略空间自相关性的影响。考虑空间自相关后,显著的空间驱动因子的个数比未考虑空间自相关性的个数少。(3)自然环境和人类活动因子对小流域植被变化的影响力不同:坡度对耕地变化、果园变化、常绿阔叶林变化和针叶林变化等均影响较大;坡向对耕地变化、果园变化、常绿阔叶林变化和灌木林地变化的影响显著;海拔对该小流域各种植被变化类型的影响相对较小;人类活动的影响对落叶阔叶林的变化影响显著。  相似文献

Sika Cervus nippon are native to Japan and East Asia but are now naturalized in many parks of the world, including Britain. In contrast to the substantial body of research on the extent of hybridization between sika and red deer Cervus elaphus in Scottish populations, there has been little genetic analysis of the English populations of sika. Sika in England still have a patchy and discontinuous distribution; populations are thus still genetically isolated and may be expected to show higher variability in genetic type. The current paper uses DNA-based techniques to explore the genetic composition of sika in the New Forest (Hampshire) and that of sika from the largest population in England, in the Purbeck region (Dorset). The study aims were to determine whether New Forest sika show signs of recent interbreeding with New Forest red deer populations, whether New Forest sika are genetically distinct from Purbeck sika, and whether New Forest and Purbeck sika show evidence of past hybridization or whether they are pure (non-hybrid) strains. Microsatellite analysis was used to compare the genetic profiles of individual deer. Results showed that sika and red deer in the New Forest were genetically distinct, indicating that there is no large-scale in situ hybridization occurring between these feral populations. In terms of overall genetic composition, there was no significant difference between the sika in the New Forest and Purbeck. However, a more detailed analysis found that New Forest sika showed a lower level of introgression with red deer compared with the Purbeck sika. We conclude that, overall, the New Forest sika deer do appear to be more genetically pure bred than the Purbeck sika.  相似文献

Sika deer (Cervus nippon), native to Asia, formed two well-established free-living populations in the Czech Republic over the last century and continue to spread. Sika are also maintained in a large number of enclosures; these continue to introduce new individuals from the places of its origin as well as from other European countries. Despite extensive research into the morphology and ethology of the Czech sika deer, conducted over the last three decades, no study using genetic methods has been done. This study aimed to determine the genetic variability and the geographic origin of the Czech sika deer population. Two mitochondrial markers, the cytochrome b and the control region were analyzed in this study. Analysis of the two markers confirmed that the founder individuals of the Czech population originated from both native island (Japanese Islands) and native mainland (Far East Russia) populations. Results showed that the genetic variability of the Czech sika deer population is lower than the variability of the native Japanese population, but higher than that of the sampled part of the native Russian population. Also, the genetic variability was found to be higher within the samples from enclosures.  相似文献

We assessed genetic differentiation and diversity in 14 populations of sika deer (Cervus nippon) from Japan and four populations of sika deer introduced to the UK, using nine microsatellite loci. We observed extreme levels of differentiation and significant differences in diversity between populations. Our results do not support morphological subspecies designations, but are consistent with previous mitochondrial DNA analyses which suggest the existence of two genetically distinct lineages of sika deer in Japan. The source of sika introduced to the UK was identified as Kyushu. The underlying structure of Japanese populations probably derives from drift in separate glacial refugia and male dispersal limited by distance. This structure has been perturbed by bottlenecks and habitat fragmentation, resulting from human activity from the mid-nineteenth century. Most current genetic differentiation and differences in diversity among populations probably result from recent drift. Coalescent model analysis suggests sika on each of the main Japanese islands have experienced different recent population histories. Hokkaido, which has large areas of continuous habitat, has maintained high levels of gene flow. In Honshu the population is highly fragmented and is likely to have been evolving by drift alone. In Kyushu there has been a balance between gene flow and drift but all the populations have experienced high levels of drift. Habitat fragment size was not significantly associated with genetic diversity in populations but there was a significant correlation between habitat fragment size and effective population size.  相似文献

The introduction of non‐native species can have long‐term effects on native plant and animal communities. Introduced populations are occasionally not well understood and offer opportunities to evaluate changes in genetic structure through time and major population changes such as bottleneck and or founder events. Invasive species can often evolve rapidly in new and novel environments, which could be essential to their long‐term success. Sika deer are native to East Asia, and their introduction and establishment to the Delmarva Peninsula, USA, is poorly documented, but probably involved ≥1 founder and/or bottleneck events. We quantified neutral genetic diversity in the introduced population and compared genetic differentiation and diversity to the presumed source population from Yakushima Island, Japan, and a captive population of sika deer in Harrington, Delaware, USA. Based on the data from 10 microsatellite DNA loci, we observed reduced genetic variation attributable to founder events, support for historic hybridization events, and evidence that the population did originate from Yakushima Island stocks. Estimates of population structure through Bayesian clustering and demographic history derived from approximate Bayesian computation (ABC), were consistent with the hypothesized founder history of the introduced population in both timing and effective population size (approximately five effective breeding individuals, an estimated 36 generations ago). Our ABC results further supported a single introduction into the wild happening before sika deer spread throughout the Delmarva. We conclude that free‐ranging sika deer on Delmarva are descended from ca. five individuals introduced about 100 years ago from captive stocks of deer maintained in the United Kingdom. Free‐ranging sika deer on Delmarva have lost neutral diversity due to founder and bottleneck events, yet populations have expanded in recent decades and show no evidence of abnormalities associated with inbreeding. We suggest management practices including increasing harvest areas and specifically managing sika deer outside of Maryland.  相似文献

The distribution of suitable habitat influences natal and breeding dispersal at small spatial scales, resulting in strong microgeographic genetic structure. Although environmental variation can promote interpopulation differences in dispersal behavior and local spatial patterns, the effects of distinct ecological conditions on within‐species variation in dispersal strategies and in fine‐scale genetic structure remain poorly understood. We studied local dispersal and fine‐scale genetic structure in the thorn‐tailed rayadito (Aphrastura spinicauda), a South American bird that breeds along a wide latitudinal gradient. We combine capture‐mark‐recapture data from eight breeding seasons and molecular genetics to compare two peripheral populations with contrasting environments in Chile: Navarino Island, a continuous and low density habitat, and Fray Jorge National Park, a fragmented, densely populated and more stressful environment. Natal dispersal showed no sex bias in Navarino but was female‐biased in the more dense population in Fray Jorge. In the latter, male movements were restricted, and some birds seemed to skip breeding in their first year, suggesting habitat saturation. Breeding dispersal was limited in both populations, with males being more philopatric than females. Spatial genetic autocorrelation analyzes using 13 polymorphic microsatellite loci confirmed the observed dispersal patterns: a fine‐scale genetic structure was only detectable for males in Fray Jorge for distances up to 450 m. Furthermore, two‐dimensional autocorrelation analyzes and estimates of genetic relatedness indicated that related males tended to be spatially clustered in this population. Our study shows evidence for context‐dependent variation in natal dispersal and corresponding local genetic structure in peripheral populations of this bird. It seems likely that the costs of dispersal are higher in the fragmented and higher density environment in Fray Jorge, particularly for males. The observed differences in microgeographic genetic structure for rayaditos might reflect the genetic consequences of population‐specific responses to contrasting environmental pressures near the range limits of its distribution.  相似文献

Spatial autocorrelation statistics have been studied in theoretical population genetic models and widely used in experimental studies of spatial structure in many plant and animal populations. However, the statistical properties of spatial autocorrelation statistics have remained uncharacterized. Little is known about how values of spatial autocorrelation statistics in population samples depend on the level of dispersal and scheme of sampling. In this paper, we characterize the statistical properties of join-count spatial autocorrelation statistics for population genetic surveys under various conditions of dispersal and sampling. The results indicate generally high statistical power. These results can provide a method to estimate gene dispersal based on standing spatial patterns of genetic variation observed within populations.  相似文献

The effect of sex-biased dispersal in mammalian ecology and evolution can be elucidated by focusing on maternally or paternally inherited DNA polymorphisms. In sika deer, the genetic structure of the maternal lineage has been clarified by studies based on mitochondrial DNA variation. However, the genetic structure of the paternal lineage has not been well analyzed due to the limited number of point mutations in Y chromosome sequences. In this study, we focused on mutations of highly polymorphic simple sequence repeats (SSRs) in the Y chromosome and developed 16 Y chromosome SSR markers to evaluate male-biased dispersal in sika deer. In total, 55 alleles and 31 multi-locus haplotypes were detected from these 16 loci, revealing clear genetic differentiation among populations (FST = 0.783). In particular, there were unique alleles for the native individuals on Tanegashima and Yakushima Islands and introduced exotic individuals from Taiwan. These markers are highly useful for evaluating not only historical male-mediated dispersal, genetic structure, and demography of the native populations in Japan, but also the impact of artificial introductions on hybridization, especially the introgression of alleles from escaped farmed individuals to native populations.  相似文献

In southern Kantoh, Japanese sika deer (Cervus nippon) are distributed discontinuously due to large urban areas and developed road networks. To assess the impact of habitat fragmentation on sika deer subpopulations, we examined mitochondrial D-loop sequences from 435 individuals throughout southern Kantoh. About 13 haplotypes were detected, and their distributions revealed spatial genetic structure. Significant genetic differentiation was observed among seven of eight subpopulations. We found no significant correlation between pairwise F ST and geographical distance among subpopulations. Genetic diversity indices suggested that seven of eight subpopulations had probably experienced population bottlenecks in the recent past. Therefore, and in the light of the results of a nested clade analysis of these haplotypes, we conclude that recent fluctuations in population size and the interruption of gene flow due to past and present habitat fragmentation have played major roles influencing the spatial genetic structure of the sika deer population. This is the first evidence of spatial genetic population structure in the highly fragmented sika deer population in Honshu, Japan.  相似文献

BACKGROUND AND AIMS: In plant populations the magnitude of spatial genetic structure of apparent individuals (including clonal ramets) can be different from that of sexual individuals (genets). Thus, distinguishing the effects of clonal versus sexual individuals in population genetic analyses could provide important insights for evolutionary biology and conservation. To investigate the effects of clonal spread on the fine-scale spatial genetic structure within plant populations, Hosta jonesii (Liliaceae), an endemic species to Korea, was chosen as a study species. METHODS: Using allozymes as genetic markers, spatial autocorrelation analysis of ramets and of genets was conducted to quantify the spatial scale of clonal spread and genotype distribution in two populations of H. jonesii. KEY RESULTS: Join-count statistics revealed that most clones are significantly aggregated at < 3-m interplant distance. Spatial autocorrelation analysis of all individuals resulted in significantly higher Moran's I values at 0-3-m interplant distance than analyses of population samples in which clones were excluded. However, significant fine-scale genetic structure was still observed when clones were excluded. CONCLUSIONS: These results suggest that clones enhance the magnitude of spatial autocorrelation due to localized clonal spread. The significant fine-scale genetic structure detected in samples excluding clones is consistent with the biological and ecological traits exhibited by H. jonesii including bee pollination and limited seed dispersal. For conservation purposes, genetic diversity would be maximized in local populations of H. jonesii by collecting or preserving individuals that are spaced at least 5 m apart.  相似文献
