The oxidation and reduction of cytochrome f and P-700 is measured spectrophotometrically in leaves of low-light and high-light plants. After illumination with red light, an induction phenomenon for cytochrome f oxidation is observed which indicates a regulation of photosystem I activity through energy distribution between the pigment systems by the energy state of the membrane. After far-red excitation the reduction of cytochrome f in the dark is much slower in low-light leaves. This shows that cyclic electron transport is not improved in low-light plants under these conditions. P-700 is oxidized on excitation with far-red light. However, with high intensities of far-red light, P-700 is partially reduced again which is due to a low extent of photosystem II excitation with the far-red used in the experiments. The low-light leaves show greater sensitivity of photosystem II to this excitation. The initial rate of the cytochrome f oxidation-rate is the same in low-light and high-light leaves. This shows that several P-700 are connected with only one electron transport chain. The consequences of these results concerning the tripartite concept and the photosynthetic unit are discussed. In the high-light plants the experimental data can be well explained by the tripartite organization of the photosynthetic unit. In low-light plants, however, a multipartite organization has to be postulated. Rosa meillandina plants were used to study the effects of water deficit on photosynthesis and chlororespiration. Plants showed high tolerance to heat and high illumination in controlled conditions that ensured that there was no water deficit. However, when heat and high illumination were accompanied by low watering photosynthetic linear electron transport was down regulated, as indicated by the reduced photochemistry efficiency of PS II, which was associated with an increase in the non-photochemical quenching of fluorescence. In addition to the effects on the photosynthetic activity, changes were also observed in the plastidial NDH complex, PTOX and PGR5. In plants exposed to heat and high illumination without water deficit, the activities and amounts of the chlororespiration enzymes, NDH complex and PTOX, remained similar to the control and only increased in response to drought, high light and heat stress, applied together. In contrast, both the PS I activity and the amount of PGR5 polypeptide were higher in plants exposed to heat and high illumination without water deficit than in those with water deficit. The results indicated that in the conditions studied, the contribution of chlororespiration to regulating photosynthetic electron flow is not relevant when there is no water deficit, and another pathway, such as cyclic electron flow involving PGR5 polypeptide, may be more important. However, when PS II activity is inhibited by drought, chlororespiration, together with other routes of electron input to the electron transfer chain, is probably essential.  相似文献

This paper explores the effects of high light stress on Fe-deficient plants. Maize (Zea mays) plants were grown under conditions of Fe deficiency and complete nutrition. Attached, intact leaves of Fe-deficient and control plants were used for gas exchange experiments under suboptimal, optimal and photoinhibitory illumination. Isolated chloroplasts were used to study photosynthetic electron transport system, compromised by the induction of Fe deficiency. The reaction centers of PS II (measured as reduction of Q, the primary electron acceptor of P 680) and PS I (measured as oxidation of P 700) were estimated from the amplitude of light induced absorbance change at 320 and 700 nm, respectively. Plants were subjected to photoinhibitory treatment for different time periods and isolated chloroplasts from these plants were used for electron transport studies. Carbon dioxide fixation in control as well as in Fe-deficient plants decreased in response to high light intensities. Total chlorophyll, P 700 and Q content in Fe-deficient chloroplasts decreased, while Chl a/b ratio and Q/P 700 ratio increased. However, electron transport through PS II suffered more after photoinhibitory treatment as compared to electron transport through PS I or whole chain. Electron transfer through PS I+PS II, excluding the water oxidation complex showed a decrease in Fe-deficient plants. However, electron transport through this part of the chain did not suffer much as a result of photoinhibition, suggesting a defect in the oxidising side of PS II.  相似文献

Transthylakoid proton transport based on Photosystem I-dependent cyclic electron transport has been demonstrated in isolated intact spinach chloroplasts already at very low photon flux densities when the acceptor side of Photosystem I (PS I) was largely closed. It was under strict redox control. In spinach leaves, high intensity flashes given every 50 s on top of far-red, but not on top of red background light decreased the activity of Photosystem II (PS II) in the absence of appreciable linear electron transport even when excitation of PS II by the background light was extremely weak. Downregulation of PS II was a consequence of cyclic electron transport as shown by differences in the redox state of P700 in the absence and the presence of CO2 which drained electrons from the cyclic pathway eliminating control of PS II. In the presence of CO2, cyclic electron transport comes into play only at higher photon flux densities. At H+/e=3 in linear electron transport, it does not appear to contribute much ATP for carbon reduction in C3 plants. Rather, its function is to control the activity of PS II. Control is necessary to prevent excessive reduction of the electron transport chain. This helps to protect the photosynthetic apparatus of leaves against photoinactivation under light stress.  相似文献

以辣椒品种“超辣九号”为试材,采用15%的PEG6000模拟干旱,研究了0.1μmol·L^-1外源24-表油菜素内酯(EBR)处理对干旱胁迫下辣椒叶片快速叶绿素荧光诱导动力学曲线(OJIP)的影响。结果表明:干旱胁迫降低了辣椒叶片的光化学效率和光合性能,导致干旱光抑制的发生。干旱胁迫既损伤了辣椒叶片PSⅡ供体侧放氧复合体(OEC),同时也对PSⅡ反应中心和受体侧造成伤害,阻碍了光合电子传递;干旱胁迫还导致单位叶面积有活性反应中心数目(RC/CS)的下降,并降低了单位叶面积吸收的光能(ABS/CS)、捕获的光能(TRo/CS)和进行电子传递的能量(ETo/CS),同时诱导了单位叶面积热耗散(DIo/CS)的增加。这说明辣椒遭受干旱胁迫后启动了相应的防御机制,一方面通过PSⅡ的可逆失活减少光能吸收与传递,另一方面通过促进热耗散减少过剩激发能的积累。EBR处理改善了干旱胁迫下辣椒叶片PSⅡ受体侧的电子传递,缓解了单位叶面积有活性反应中心数目的减少,优化了光合电子传递的进行,并维持相对较高的热耗散能力,从而减轻了干旱光抑制程度,对干旱胁迫下辣椒叶片光合机构和光合性能起到保护作用。  相似文献

Glycoglycerolipids are dominant lipids of photosynthetic organisms, i.e. higher plants and cyanobacteria. X-ray crystallographic localization of glycerolipids revealed that they are present at functionally and structurally important sites of both the PS I and PS II reaction centres. Phosphatidylglycerol (PG) is an indispensible member of glycerolipids, including the formation of functionally active oligomers of the reaction centres both PS I and PS II. Lipids assist in the assembly of protein subunits of the photosynthetic machinery by pasting the individual protein components together. PG is needed to glue CP43 to the reaction centre core. PG and digalactosyldiacylglycerol (DGDG) interact in photosynthetic processes: PG alone controls electron transport at the acceptor site of PS II, and together with DGDG is involved in electron transport at the donor site of PS II. PG is crucial for the formation of division rings and is implicated in the fission of cyanobacteria.  相似文献

In addition to the linear electron transport, several alternative Photosystem I-driven (PS I) electron pathways recycle the electrons to the intersystem electron carriers mediated by either ferredoxin:NADPH reductase, NAD(P)H dehydrogenase, or putative ferredoxin:plastoquinone reductase. The following functions have been proposed for these pathways: adjustment of ATP/NADPH ratio required for CO(2) fixation, generation of the proton gradient for the down-regulation of Photosystem II (PS II), and ATP supply the active transport of inorganic carbon in algal cells. Unlike ferredoxin-dependent cyclic electron transport, the pathways supported by NAD(P)H can function in the dark and are likely involved in chlororespiratory-dependent energization of the thylakoid membrane. This energization may support carotenoid biosynthesis and/or maintain thylakoid ATPase in active state. Active operation of ferredoxin-dependent cyclic electron transport requires moderate reduction of both the intersystem electron carriers and the acceptor side of PS I, whereas the rate of NAD(P)H-dependent pathways under light depends largely on NAD(P)H accumulation in the stroma. Environmental stresses such as photoinhibition, high temperatures, drought, or high salinity stimulated the activity of alternative PS I-driven electron transport pathways. Thus, the energetic and regulatory functions of PS I-driven pathways must be an integral part of photosynthetic organisms and provides additional flexibility to environmental stress.  相似文献

The effects of drought stress and high irradiance and their combination were studied under laboratory conditions using young plants of a very drought-resistant variety, ICMH 451, of pearl millet (Pennisetum glaucum) and three varieties of sorghum (Sorghum bicolor)—one drought-resistant from India, one drought-tolerant from Texas, and one drought-sensitive variety from France. CO2 assimilation rates and photosystem II fluorescence in leaves were analyzed in parallel with photosynthetic electron transport, photosystem II fluorescence, and chlorophyll-protein composition in chloroplasts isolated from these leaves. High irradiance slightly increased CO2 assimilation rates and electron transport activities of irrigated plants but not fluorescence. Drought stress (less than −1 megapascal) decreased CO2 assimilation rates, fluorescence, and electron transport. Under the combined effects of drought stress and high irradiance, CO2 assimilation rates and fluorescence were severely inhibited in leaves, as were the photosynthetic electron transport activities and fluorescence in chloroplasts (but not photosystem I activity). The synergistic or distinctive effect of drought and high irradiance is discussed. The experiments with pearl millet and three varieties of sorghum showed that different responses of plants to drought and light stresses can be monitored by plant physiological and biochemical techniques. Some of these techniques may have a potential for selection of stress-resistant varieties using seedlings.  相似文献

The photoacoustic technique was used to study in vivo cyclicelectron flow through PS I in intact plant leaves irradiatedwith far-red light. Appreciable PS I-cyclic ATP formation wasshown to occur in both C-3 and C-4 plants. It was also observedthat various environmental/experimental conditions leading toa significant inhibition of the linear photosynthetic electrontransport were associated with a stimulation of PS I-cyclicenergy storage. In contrast, in vivo adaptation of leaves tolight-states 1 and 2 did not induce any photoacoustically measurablechanges in the capacity of PS I for cyclic electron transfer.Consequently, the presented data do not support the recent hypothesisthat the main function of light-induced state transitions isto regulate the balance between linear (PS I+PS II) and cyclic(PS I) electron transport in the chloroplasts. Radix Isatidis (Isatis indigotica Fort.) is one of the most important traditional Chinese medicine plants. However, there is no suitable herbicide used for weed control in Radix Isatidis field during postemergence stage. To explore the safety of sulfonylurea herbicide nicosulfuron on Radix Isatidis (Isatis indigotica Fort.) seedlings and the photosynthetic physiological response of the plant to the herbicide, biological mass, leaf area, photosynthetic pigment content, photosynthetic rate, chlorophyll fluorescence characteristics, and P700 parameters of Radix Isatidis seedlings were analyzed 10 d after nicosulfuron treatment at 5th leaf stage in this greenhouse research. The results showed that biological mass, total chlorophyll, chlorophyll a, and carotenoids content, photosynthetic rate, stomatal conductance, PS II maximum quantum yield, PS II effective quantum yield, PS II electron transport rate, photochemical quenching, maximal P700 change, photochemical quantum yield of PS I, and PS I electron transport rate decreased with increasing herbicide concentrations, whereas initial fluorescence, quantum yield of non-regulated energy dissipation in PS II and quantum yield of non-photochemical energy dissipation due to acceptor side limitation in PS I increased. It suggests that nicosulfuron ≥1 mg L−1 causes the damage of chloroplast, PS II and PS I structure. Electron transport limitations in PS I receptor side, and blocked dark reaction process may be the main cause of the significantly inhibited growth and decreased photosynthetic rate of Radix Isatidis seedlings.  相似文献

以叶绿素快相荧光动力学曲线(OJIP)为探针,研究了外源钙对干旱胁迫下烤烟幼苗光系统Ⅱ(PSⅡ)功能的影响.结果表明:干旱胁迫降低了烤烟幼苗PSⅡ原初光能转换效率(Fv/Fm)和电子传递速率(ETR),抑制了光合作用的原初过程,烤烟幼苗叶片发生了明显的光抑制.叶面喷施10.0 mmol·L-1CaCl2溶液后烤烟叶片的光合电子传递能量比例(ФEo)在干旱胁迫下的降低幅度明显小于对照(喷施清水),电子转运效率(ET0/RC)在干旱胁迫下明显高于对照.叶面喷施CaC12溶液增加了PSⅡ捕获光能用于光合电子传递的比例、剩余有活性反应中心的效率和电子传递链中的能量传递,使烤烟叶片的光系统Ⅱ在干旱胁迫下保持相对较高的活性,从而提高了烤烟幼苗的抗旱能力.  相似文献

Micropropagated rose plants (Rosa hybrida L., cv. New Dawn) were inoculated with the arbuscular mycorrhizal (AM) fungus Glomus intraradices (Schenk and Smith) and subjected to different drought regimens. The dual objectives of these experiments were to investigate the mechanism and the extent to which AM can prevent drought damages and whether physiological analyses reveal enhanced drought tolerance of an economically important plant such as the rose. In a long-term drought experiment with four different water regimens, visual scoring of wilt symptoms affirmed that AM in a selected host–symbiont combination increased plant performance. This effect was mostly expressed if moderate drought stress was constantly applied over a long period. In a short-term experiment in which severe drought stress was implemented and plants were allowed to recover after 4 or 9 days, no visual differences between mycorrhizal and non-mycorrhizal roses were observed. Therefore, the early physiological steps conferring drought tolerance were prone to investigation. Proline content in leaves proved to be an unsuitable marker for AM-induced drought tolerance, whereas analysis of chlorophyll a fluorescence using the JIP test (collecting stress-induced changes of the polyphasic O-J-I-P fluorescence kinetics in a non-destructive tissue screening) was more explanatory. Parameters derived from this test could describe the extent of foliar stress response and help to differentiate physiological mechanisms of stress tolerance. AM led to a more intense electron flow and a higher productive photosynthetic activity at several sites of the photosynthetic electron transport chain. A K step, known as a stress indicator of general character, appeared in the fluorescence transient only in drought-stressed non-mycorrhizal plants; conversely, the data elucidate a stabilising effect of AM on the oxygen-evolving complex at the donor site of photosystem (PS) II and at the electron-transport chain between PS II and PS I. If drought stress intensity was reduced by a prolonged and milder drying phase, these significant tolerance features were less pronounced or missing, indicating a possible threshold level for mycorrhizal tolerance induction.  相似文献

Excessive light conditions repressed the levels of mRNAs accumulation of multiple Lhc genes encoding light-harvesting chlorophyll-a/b (LHC) proteins of photosystem (PS)II in the unicellular green alga, Chlamydomonas reinhardtii. The light intensity required for the repression tended to decrease with lowering temperature or CO(2) concentration. The responses of six LhcII genes encoding the major LHC (LHCII) proteins and two genes (Lhcb4 and Lhcb5) encoding the minor LHC proteins of PSII (CP29 and CP26) were similar. The results indicate that the expression of these Lhc genes is coordinately repressed when the energy input through the antenna systems exceeds the requirement for CO(2) assimilation. The Lhc mRNA level repressed under high-light conditions was partially recovered by adding the electron transport inhibitor 3-(3,4-dichlorophenyl)-1,1-dimethylurea, suggesting that redox signaling via photosynthetic electron carriers is involved in the gene regulation. However, the mRNA level was still considerably lower under high-light than under low-light conditions even in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea. Repression of the Lhc genes by high light was prominent even in the mutants deficient in the reaction center(s) of PSII or both PSI and PSII. The results indicate that two alternative processes are involved in the repression of Lhc genes under high-light conditions, one of which is independent of the photosynthetic reaction centers and electron transport events.  相似文献

Effects of foliar application of 100 mmol/L glycinebetaine (GB) on PS II photochemistry in wheat (Triticum aestivum) flag leaves under drought stress combined with high irradiance were investigated. The results show that GB-treated plants maintained a higher net photosynthetic rate during drought stress than non-GB treated plants. Exogenous GB can preserve the photochemical activity of PSII, for GB-treated plants maintain higher maximal photochemistry efficiency of PSII (F(v)/F(m)) and recover more rapidly from photoinhibition. In addition, GB-treated plants can maintain higher anti-oxidative enzyme activities and suffer less oxidative stress. Our data suggest that GB may protect the PSII complex from damage through accelerating D1 protein turnover and maintaining anti-oxidative enzyme activities at higher level to alleviate photodamage. Diethyldithiocarbamate as well as streptomycin treatment can impair the protective effect of GB on PSII. In summary, GB can enhance the photoinhibition tolerance of PSII.  相似文献

The effects of drought on chlorophyll fluorescence characteristics of PSII, photosynthetic pigments, thylakoid membrane protein (D1), and proline content in different varieties of mung bean plants were studied. Drought stress inhibits PSII activity and induces alterations in D1 protein. We observed a greater decline in the effective quantum yield of PSII, electron transport rate, and saturating photosynthetically active photon flux density (PPFDsat) under drought stress in var. Anand than var. K-851 and var. RMG 268. This may possibly be due to either downregulation of photosynthesis or photoinhibition process. Withholding irrigation resulted in gradual diminution in total Chl content at Day 4 of stress. HPLC analysis revealed that the quantity of β-carotene in stressed plants was always higher reaching maxima at Day 4. Photoinactivation of PSII in var. Anand includes the loss of the D1 protein, probably from greater photosynthetic damage caused by drought stress than the other two varieties.  相似文献

The effects of exposure of a field-grown winter cultivar of barley (Hordeum vulgare L.) to Photosystem I (PS I) photoinhibitory conditions in the form of bright day-light combined with chilling conditions were investigated. PS I photoinhibition was manifested by damage to the Fe-S centers of PS I and to the PS I-A/B polypeptides. Up to 20% of the PS I complexes were photoinactivated. Upon transfer to room temperature, the plants partially recovered from PS I photoinhibition, although damage was still detectable after one week. These results demonstrate that PS I photoinhibition is a physiologically relevant phenomenon in chilling-tolerant plants grown under field conditions. In order to study the induction of cyclic electron transport around PS I by PS I photoinhibitory conditions, antibodies raised against the NDH-I subunit of the NDH complex (a component of cyclic electron transport) were used to measure NDH levels in the exposed plants. A marked increase in the amount of NDH complex and a corresponding increase in NADPH dehydrogenase activity in the thylakoids were observed. The data indicate that the response to PS I-photoinhibitory conditions may involve regulated changes in cyclic electron transport around PS I. The studies were carried out in order to estimate differences in the physiological state between triticale and maize plants subjected to drought stress followed by rehydration. The physiological state of the plants was evaluated by measurements of leaf water potential, net photosynthesis, transpiration and stomatal conductance. Spectrofluorimetric methods for the study of blue, green and red fluorescence were applied. We observed that the soil drought induced a greater water loss in triticale leaves than in maize and consequently caused greater injuries to the photosynthetic apparatus. Moreover, triticale plant recovery was slower than in maize plants during the rehydration phase. The effect was probably connected with the higher functional and structural disorganisation of the photosynthetic apparatus observed during drought stress in triticale. Water stress is responsible for damages to photosystem PS II. The worst light utilisation in photosynthetic light conversion was recorded as an increase in the intensity of red fluorescence. Drought stress induced a strong increase in the intensity of blue and green fluorescence in the studied species and it was still high in maize plants during the first day of rehydration. Increase in the intensity of blue and green fluorescence in maize seems to be the effect of the photoprotection mechanism which prevents damage to PS II through utilisation of excess energy.  相似文献

By the method of hydroponic culture, this paper studied the effects of exogenous gamma-aminobutyric acid (GABA) on the photosynthetic pigment contents, photosynthesis, and chlorophyll fluorescence parameters of muskmelon seedlings under hypoxia stress. Hypoxia stress induced a significant decrease of photosynthetic pigment contents, resulting in the decrease of photosynthesis. Applying GABA could significantly increase the photosynthetic pigment contents, net photosynthetic rate (P(n)), stomatal conductance (G(s)), intercellular CO2 concentration (C(i)), carboxylation efficiency (CE), maximal photochemical efficiency of PS II (F(v)/F(m)), photochemical quenching (q(P)), apparent photosynthetic electron transfer rate (ETR), and quantum yield of PS II electron transport (phi(PS II)), and decrease the stomatal limitation value (L(s)), minimal fluorescence (F(o)), and non-photochemical quenching (NPQ) under both hypoxic and normal conditions. The alleviation effect of GABA on photosynthetic characteristics was more obvious under hypoxia stress. However, simultaneously applying GABA and VGB could significantly decrease the alleviation effect of GABA under hypoxia stress.  相似文献

The contents of pigments and chlorophyll-protein complexes, fluorescence characteristics and electron transport rate were compared for wheat seedlings grown under different light intensities. Leaves of wheat seedlings grown under low-light intensity (2 klx) had lower chlorophyll and carotenoid contents on leaf area or fresh weight basis, a lower ratio of chlorophyll a/b, lower CPIa and CPI contents in photosynthetic membranes than those of wheat seedlings grown under high-light intensity (20 klx). However, the LHCP content in photosynthetic membranes was higher in the former. The kinetic studies of fluorescence induction showed that wheat seedlings grown under low-light intensity possessed a bigger photosynthetic unit, lower PSⅡ activity and lower efficiency of primary energy conversion than those grown under high-light intensity. Moreover. lower electron transport rate was found in the chloroplasts of the former.  相似文献

Photosynthetic electron transport can involve either a linear flow from water to NADP, via Photosystems (PS) II and I or a cyclic flow just involving PSI. Little is known about factors regulating the relative flow through each of these pathways. We have examined photosynthetic electron transport through each system in plants of Arabidopsis thaliana in which either the PSI-D1 or PSI-E1 subunits of PSI have been knocked out. In both cases, this results in an imbalance in the turnover of PSI and PSII, such that PSII electron transport is limited by PSI turnover. Phosphorylation of light-harvesting complex II (LHCII) and its migration to PSI is enhanced but only partially reversible and not sufficient to balance photosystem turnover. In spite of this, cyclic electron flow is able to compete efficiently with PSI across a range of conditions. In dark-adapted leaves, the efficiency of cyclic relative to linear flow induced by far-red light is increased, implying that the limiting step of cyclic flow lies in the re-injection of electrons into the electron transport chain. Illumination of leaves with white light resulted in transient induction of a significant non-photochemical quenching in knockout plants which is probably high energy state quenching induced by cyclic electron flow. At high light and at low CO(2), non-photochemical quenching was greater in the knockout plants than in the wildtype. Comparison of PSI and PSII turnover under such conditions suggested that this is generated by cyclic electron flow around PSI. We conclude that, when the concentration of PSI is limiting, cyclic electron flow is still able to compete effectively with linear flow to maintain a high DeltapH to regulate photosynthesis.  相似文献
