为了丰富花生遗传资源,开拓新的育种方法,本研究对离体诱变创造花生新种质、培育花生新品种进行了研究。利用花生品种花育20号胚小叶作为外植体,平阳霉素(PYM)作为诱变剂进行离体诱变培养,然后在含有羟脯氨酸(HYP)的培养基上进行定向筛选,最终获得了15个再生小苗。再生小苗经嫁接移栽田间,从后代中获得了23个高油株系,3个产量显著提高的品系,其中一个高产高油品系2015年通过了安徽省新品种登记鉴定,定名为宇花4号,在参试的品种中名列第一,比对照白沙1016增产16.63%。宇花4号为早熟、小粒、高油花生品种,经农业部油料及制品质量监督检验测试中心(武汉)化验,籽仁含油率达56.10%,达到高油标准,比诱变亲本花育20号(含油率49.50%)高6.6个百分点,荚果产量比花育20号增产15%以上。本研究结果表明,离体诱变结合离体定向筛选是创造花生新种质、培育新品种的有效途径。  相似文献

为开拓新的花生育种方法,对辐照诱变结合组织培养创造花生新种质、培育新品种进行了研究。以我国北方地区主栽花生品种鲁花11号成熟种子为试材,经快中子辐照处理后取种子胚小叶进行组织培养,通过胚胎发生途径获得再生苗。再生苗经嫁接驯化后移栽田间,83个单株获得种子。后代按系谱法进行选育,从83个再生植株后代中获得了107份突变体,分别在主茎高、分枝数、荚果形状和大小、种皮颜色、内种皮颜色、含油率、蛋白含量等性状上发生了明显变异。从突变体后代中选育出了低油早熟耐涝大花生新品种宇花7号,其产量比亲本鲁花11号增产14%以上;其含油率(47.0%)比鲁花11号低5.1个百分点。宇花7号2016年参加辽宁省新品种登记试验,比对照品种白沙1017平均增产13.8%。2018年通过了国家非主要农作物品种登记,登记号为"GPD花生(2018) 370105"。研究结果说明,辐照结合组织培养是创造花生新种质、培育新品种的有效方法。  相似文献

花生胚小叶离体再生体系的优化研究   总被引:3,自引:0,他引:3  
目的:研究培养基中植物激素的种类、浓度和基因型差异对花生胚小叶离体培养再生效率的影响。方法:以花生胚小叶为外值体,比较各处理间的芽丛诱导率、芽丛出苗率和植株再生率,确定每个品种对应的适宜培养基。结果:小花生品种丰花2号的适宜芽丛诱导培养基为MSB+3.0mg/LBA+0.7mg/LNAA,芽丛诱导率为81.49%,再生率为94.39%;适宜芽丛成苗培养基为MSB+2.5mg/LBA,芽丛出苗率为172.00%,再生率为122.10%。大花生品种丰花3号的适宜芽丛诱导培养基为MSB+4.5mg/LBA+0.2mg/LNAA,芽丛诱导率为72.33%,再生率为10.70%;适宜芽丛成苗培养基为MSB+5.0mg/LKIN,芽丛出苗率为33.33%,再生率为4.03%。结论:不同基因型之间的成苗芽丛百分率、芽丛出苗率和植株再生率差异显著。  相似文献

ACC氧化酶(ACC oxidase,ACO)是催化乙烯合成的关键酶之一,乙烯参与植物的盐胁迫反应过程,而盐胁迫严重影响花生产量。本研究通过对AhACOs基因的克隆及功能验证,探究AhACOs在花生盐胁迫响应中的生物学功能,为花生耐盐品种的选育提供基因资源。以花生耐盐突变体M29的cDNA为模板扩增得到基因AhACO1和AhACO2,与植物表达载体pCAMBIA super1300重组后,通过农杆菌介导的花粉管注射法将重组质粒转化到花育22号中。收获后切取籽仁远胚端部分子叶,利用PCR检测筛选阳性籽仁。利用qRT-PCR分析AhACOs基因表达量,通过毛细管柱气相色谱法检测植株的乙烯释放量。阳性籽仁和对照籽仁种植21 d后浇盐水,观察其表型变化。结果发现,盐胁迫后,转基因植株生长状况好于对照组花育22号,并且其叶绿素相对含量SPAD(soil and plant analyzer development)值和净光合速率(net photosynthesis rate,Pn)均高于对照组花生。另外,AhACO1和AhACO_(2)转基因植株的乙烯释放量分别为对照组花生的2.79倍和1.87倍。这些结果表明AhACO1和AhACO2可显著提高花生的耐盐能力。  相似文献

选用中抗赤霉病的春小麦品种和品系的花药进行离体培养,以小麦赤霉病29号菌株产生的致病培养滤液为选择剂,结合物理诱变处理,进行抗病鉴定,用赤霉病菌分生孢子直接接种在愈伤组织和再生植株筛选抗病突变体。在83块愈伤中有53块抗病。在11株的再生植株中有9株均比未经培养滤液处理的对照提高了抗病性,从中选出4株抗病性接近或超过“苏麦3号”品种。  相似文献

以枇杷属Eriobotrya植物11个野生种、1个远缘杂交后代材料为试材,采用胚为外植体,在不同植物生长调节剂配比培养条件下,进行胚离体培养和植株再生研究。结果表明,胚培养的最佳培养基配方为MS + 6-BA 1.0 mg·L-1 + KT 1.0 mg·L-1 + NAA 0.1 mg·L-1。在此培养基中,所有试验材料均获得成功,其中栎叶枇杷E. prinoides的萌发率和丛芽率均达100%,有效地建立了枇杷属野生种的胚离体培养及其植株再生体系。  相似文献

近年来我们在甘蔗、芦笋及羊草等多种植物上开展脯氨酸超常积累的细胞突变体的筛选研究,现简要报道如下。在离体组织培养技术基础上获得的胚性愈伤组织上进行本实验。培养物经过或不经过细胞诱变措施(诱变时用0.1%EMS 进行24小时诱变处理)。选用羟脯氨酸(HYP)作为压力选择剂,经过加选择压——去选择压——加选择压反复筛选培养,获得抗性细胞系,这些抗性细胞系在经过多次无选择压的继代培养后,回到含有 HYP 的压力培养基上多数仍表现有  相似文献

以花生上胚轴为外植体,研究不同Picloram(毒莠定)浓度处理和不同基因型对体细胞胚胎发生及植株再生的影响,并利用基因枪将含GUS基因的pCAMBIA2301质粒载体轰击体胚,对基因枪转化条件进行了初步探索.结果表明:外植体在添加5 mg/ L Picloram+1 mg/L Glutamine(谷氨酰胺)的MB培养基上诱导的体胚发生率、产胚数及植株再生率最高.不同基因型以‘中花8号'体胚再生率最高(体胚诱导率为55.36%,植株再生率为56.79%).在氦气压力为1 100 psi,轰击距离为9 cm时,体细胞胚GUS瞬时表达率可达到22.95%.  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

传染性法氏囊病(Infection bursal disease, IBD)是由鸡传染性法氏囊病毒(Infectious bursal disease virus, IBDV)引起的鸡和火鸡的一种高度接触性传染病,给世界各国的禽养殖业带来了巨大损失.自IBDV发现至今新的变异株不断出现,分子结构的改变导致病毒致病力的改变及宿主对疫苗应答的改变,使得传统的疫苗已不能控制其流行,因此各国学者对其基因组结构和功能进行了广泛深入的研究,并积极研制新型有效的疫苗以达到防治的目的.  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献
