Summary Thein vitro culture ofActinidia deliciosa petioles results in a decline of cytokinin content and an increase of auxin levels. The addition of plant growth regulators (PGRs) to the medium lead to recovery of the initial auxin content, and callus induction occurs at the basal end of the explants. Endogenous auxin/cytokinin ratio was higher at this side than in the apical one, due to unequal distribution of endogenous PGRs in the cultured petioles. Some of the induced calluses showed shoot formation when they were transferred to proliferation medium. Most important differences found in hormonal content between organogenic and non-organogenic callus concerned benzyladenine levels. Plant regeneration from embryogenic cells of two Rosa hybrida cultivars, Kardinal and Classy, was increased by dispersing embryogenic callus in liquid medium for 3 h followed by size-fractionation to isolate proembryogenic masses that were smaller than 530 m. Dispersed callus of three cultivars, Kardinal, Classy, and Tineke, produced 61–135 cotyledonary-stage embryos/100 mg fresh weight (FW) as compared to intact callus that had not been dispersed, which produced only zero to three cotyledonary-stage embryos/100 mg FW. Over 500 cotyledonary-stage embryos/100 mg FW callus developed from proembryogenic masses of Kardinal, Classy, and Tineke following 2 months of culture on solidified Murashige and Skoogs basal salts medium supplemented with 0.25% activated charcoal. Cotyledonary-stage embryos of Classy that developed from both dispersed callus and fractionated cells of various sizes showed a significantly higher conversion frequency to plants (28%) than cotyledonary-stage embryos isolated from intact callus (9%). An improved procedure for the induction, proliferation and regeneration of embryogenic callus from coffee leaf explants has been developed. The optimal culture conditions for callus induction and somatic embryogenesis yielded so-called high frequency embryogenic callus ofCoffea canephora P. ex Fr., Arabusta and Congusta, more rapidly and abundantly than other published procedures.Coffea arabica L. genotypes, however, were less responsive to the procedure. The highest multiplication rate of embryogenic callus in liquid culture, which avoided the differentiation of embryos, was obtained by culture at an inoculum density of 10 g callus 1-1 in a modified MS medium containing 4.5 M 2,4-dichlorophenoxyacetic acid, under 3 mol m-2 s-1 illumination, and subcultured every 7–10 days. The best long-term maintenance of embryogenic potential was obtained by culture of aggregates (250–1000 m in diameter) at an inoculum density of 5 g 1-1, with medium renewed every 3–4 weeks. Under these conditions, embryogenic potential ofC. canephora callus was maintained for over 2 years. Analysis of nutrients absorbed by the callus cultures demonstrated that half strength MS macro- and micro-salts were not depleted during at least 3 weeks of sustained culture. The highest regeneration of embryogenic callus required the omission of 2,4-D and a reduced culture density of 1 g 1-1. Under these conditions of culture, 1 g ofC. canephora or Arabusta callus produced 1.2 and 0.9×105 somatic embryos, respectively, after 8–10 weeks in liquid regeneration medium. A protocol for large-scale propagation of Phragmites communis Trin. by somatic embryogenesis has been established. Plants were regenerated through somatic embryogenesis from stem segments of R5002-12, a salt-tolerant variant line of Phragmites communis Trin. Stem segment explants produced hard white callus on the semi-solid Murashige and Skoog (MS) medium supplemented with 9.05 M 2,4-dichlorophenoxy acetic acid (2,4-D) for 4 weeks. The induction frequency was 36.7%. Then, the callus was transferred to MS medium supplemented with 4.52 M 2,4-D. After 4 weeks in culture, yellow embryogenic callus with some nodular structures was formed. When the embryogenic callus was transferred to differentiation medium (MS supplemented with 0.45 M 2,4-D), differentiation was initiated to form small green islands on the surface of the callus after 2 weeks in culture. Within 4 weeks, a large number of somatic embryos were formed with a frequency of 86.7%. Six weeks later, they developed into strong plantlets. When the plantlets (about 1 cm in length) were cultured on propagation medium (MS supplemented with 13.31 M BA+5.37 M NAA), a great number of regenerated plants were obtained. After the plants were cultured on liquid 1/2 MS medium with 2.69 M NAA added 2.46 M IBA roots developed. The rooted plants were transferred to soil with over 85% survival. Using this methodology, more than 20000 regenerated plants of salt tolerant variant line of Phragmites communis Trin. have been produced.  相似文献

Summary Monoterpenoids with 1-p-menthene-structure can be transformed by Corynespora cassiicola DSM 62 474, DSM 62 475, and Diplodia gossypina ATCC 10 936 to chiral 1,2-trans-diols in yields of more than 60% with only minor amounts of side-products. Whereas the substrates (S)-(-)-limonene, -terpinene, -terpinene, and terpinolene are converted to the (1R,2R)-p-menthene-1,2-diols, (R)-(+)-limonene and (R)-(-)-phellandrene yield the (1S,2S)-1,2-diols. Callus of tobacco (Nicotiana tabacum L. cv. Wisconsin 38) was grown in the light on shoot-forming medium in the presence of Na2SO4 for over a year. An increase in Na2SO4 concentration resulted in decreasing callus growth, decreasing percentage of calli producing shoots and number of shoots per callus, and increasing callus percent dry weight. Regeneration of shoots from callus grown in the absence of Na2SO4 began to decline after 14 months in culture, and shoot regeneration capacity was completely lost after 18 months. In contrast, 18-month old callus continuously grown in the presence of Na2SO4 retained the ability to form shoots. The highest percent of callus pieces that formed shoots and the maximum number of shoots per callus occurred at 70 mM (1%) Na2SO4. All plants arising from the 18-month old callus were polyploid. Callus tissues derived from seedlings of roselle (Hibiscus sabdariffa L.) were shown to produce two cyanidin glycosides as major anthocyanin pigments. Both callus growth and anthocyanin synthesis were remarkably stimulated by 2,4-dichlorophenoxyacetic acid. The highest anthocyanin yield was observed when 1 M 2,4-D in combination with 0.1–1 M kinetin was supplemented to the culture medium. Callus and suspension plant cell cultures of Tribulus terrestris L., a valuable medicinal plant producing steroidal glycosides, were obtained. The seeds from an American population of T. terrestris were used as explants. Regulation of the production and growth of cell cultures, as well as the biosynthetic characteristics of the cell lines, were studied. The combination of phytohormones of 2,4-D (2.0 mg/L) and BAP (1.0 mg/L) was found to be optimal for callus induction and cultivation. Suspension cell culture obtained in liquid medium of the same composition showed such high growth characteristics during prolonged cultivation (more than 2 years) as a maximum accumulation of dry biomass of 13 g/L, specific growth rate at exponential phase of 0.24 day–1, and economical coefficient of 0.39. A semicontinuous mode of cultivation was used to grow the plant cell suspension in a lab-scale bioreactor. Screening of the steroidal glycosides in the obtained cell cultures was carried out. Steroidal glycosides were not found in the callus cultures. However, as was demonstrated by TLC and UPLC ESI MS methods, the suspension culture contained furostanol glycosides, and their amount increased during the cultivation process. These results support the hypothesis of the autoselection of cultivated cells containing compounds promoting their proliferation in vitro.  相似文献

Summary Zygotic embryos ofArabidopsis thaliana showed three different types of developmental response, when cultured in vitro: (1) normal development, (2) formation of morphogenetic callus, and (3) somatic embryogenesis. Early zygotic embryos were mechanically isolated and inoculated into different volumes of various culture media. It was possible to isolate embryos to the octant stage. Survival and further development in culture were observed in embryos to the early globular stage. Culture success increased with the initial size of the cultivated embryos. Neither the volume of the culture medium nor its composition were found to significantly influence the proportion of embryos developing in vitro. Whereas normal development from stages beyond 35 m diameter was possible without phytohormones, callus formation was frequently observed in the presence of phytohormones, even if used at very low concentrations. Embryos smaller than 35 m formed callus even without added phytohormones, and the proportion of embryos undergoing callus formation decreased with increasing embryo size at the time of culture initiation. Shoot morphogenesis was easily induced in embryo derived callus. The level of ethylene accumulated in morphogenic callus cultures of Heliconia psittacorum L.f. was only one quarter that of non-morphogenic cultures. The rate of ethylene production in the morphogenic callus cultures during early stages of differentiation of protocorm-like bodies leading to plantlet regeneration was 10-fold higher than that during callus proliferation. In cultures sealed with gastight serum caps, fresh weight gain was reduced 2-to 3-fold compared to those that were closed with Kaputs. Treatment with 1-aminocyclopropane-1-carboxylic acid ( 100 M) caused complete inhibition of plant regeneration from the morphogenic callus on subsequent culture under inductive conditions. Silver nitrate and aminoethoxyvinylglycine also reduced plant regeneration. The effects of ficoll in liquid culture media have been contradictory in previous reports. The objective of this study was to determine the functional properties of ficoll in potato 4 (P4) liquid induction medium and their influence on anther culture responses of wheat. Ficoll addition significantly (p0.01) reduced callus production from the anthers of spring wheat cv. Pavon 76. The reduction was directly related to the concentration of ficoll added within the range of 50 to 200 g l-1 medium. Although the addition of ficoll significantly (p0.01) increased the percentage of regenerable calli and the ratio of green vs. albino plants, the final yield of green plants per 100 anthers was significantly lower. Consistent results also were obtained with four other spring wheat genotypes (Chris, Butte 86, WA 6916, and Edwall). Ficoll concentration affected the density, viscosity, and osmolality of the liquid media. The higher medium density caused by ficoll addition increased the percentage of floating calli, as well as the percentage of regenerable calli and the ratio of green vs. albino plants. However, the increased medium viscosity by ficoll addition significantly (p0.01) reduced callus production. Ficoll addition also increased medium osmolality, which affected callus production by interacting with the sugar concentration of the induction media. Using response functions, the estimated maltose concentration for maximum callus production was 105 g l-1 for the standard P4 media, compared with 68 g l-1 for the ficoll-containing P4 media. These results clearly demonstrate that ficoll addition to the liquid P4 induction medium containing high sucrose concentration (90 g l-1) is deleterious to the maximum production of green plants from wheat anther culture.  相似文献

The chemical defenses in Rhizophora mangle L. are largely carbon based. The family has long been exploited for the high proanthocyanidin (condensed tannin) content of its wood, bark and leaves. In this paper, we quantify the overall pools of plant phenolics in R. mangle leaves, identify the major constituents of these pools and document their changes during leaf maturation and senescence. Overall, polyphenolics account for approximately 23% of the total leaf dry weight. The leaves contain at least seven flavonoid glycosides, five of them based on quercetin. Additional minor constituents are myricetin and kaempferol diglucosides. The aglycone, quercetin, was found only in senescing leaves. Also during senescence, a new compound, 5,4-dimethoxy-7,3,5-trihydroxyflavone, appeared. The flavonoids were accompanied by a complex mixture of condensed tannins based mainly on (+)-catechin and (–)-epicatechin with A-type and B-type linkages; this pool is also distinguished by having previously unreported, high contributions of (+)-catechin and (–)-epicatechin glycosides. During senescence, but prior to leaf abscission, the polyphenolic pools become simplified: flavonol glycosides and low oligomeric tannins largely disappear, leaving only the largest tannin polymers. The ecological and physiological significance of these compounds as they appear in R. mangle is discussed.  相似文献

Methods were developed for the production of somatic embryos of asparagus (Asparagus officinalis L.) in suspension culture and subsequent conversion to plantlets on solidified medium. Stem-derived callus that was subcultured twice on Murashige and Skoog (MS) medium + 0.54 M naphthaleneacetic acid (NAA) and 1.4 M 2-isopentenyladenine (2iP) was used to initiate suspension cultures. Six out of 15 such cell suspensions (MS medium with 54 to 107 M NAA) had a high embryogenic capacity. These cell suspensions consisted primarily of single elongated cells (about 90% of all single cells), embryogenic cell clusters (2571/ml), and globular translucent embryos (32/ml). The latter converted to plantlets within four weeks on embryo development medium (EDM), which was solidified MS medium containing 0.54 M NAA and 0.98 M 2iP. Suspension-derived embryos formed secondary globular embryos at high frequencies (251 to 258/g callus) when placed on EDM with a low carbohydrate (sucrose, glucose or fructose) level (2%). In contrast, EDM with a high carbohydrate level (10%) caused a reduction in the frequency of secondary embryos (30 to 85/g callus), while resulting in the promotion of embryo growth and conversion, 3.6 to 8.5 times higher than 2% carbohydrates. Rye chromosomes of wheat-rye addition lines were successfully identified by means of an RFLP analysis with 30 probes. Our results are in agreement with previous cytological data concerning the identity of lines F (+1R), D (+2R), C (+3R), A (+4R), E (+5R) and B (+7R). Two categories of chromosomal rearrangements have been distinguished, namely: (1) deletions: the current line D possesses a chromosome 2R deleted on its short arm and the line G a chromosome 3R deleted on its long arm; we have also noticed a deletion on the long arm of wheat chromosome 1A in line F61; and (2) evolutionary reciprocal translocations in rye relative to wheat which have been previously mentioned in the literature. The anther culture response of the different lines was studied. A significant difference between FEC 28 and the addition lines was observed for embryo production and plant regeneration. It appears that genes located on S 10 chromosome arm 3RL and on FEC 28 chromosome arm 1AL increase embryo frequency whereas gene(s) located on S 10 chromosome 5R reduce(s) it. Plant regeneration results suggest that genes increasing regeneration ability and green-plant frequency are located on S 10 chromosome 4R. The long arm of chromosome 1A seems to be involved positively in green-plant regeneration whereas chromosomes 1R and 3R limit plant regeneration.  相似文献

Summary InBetula platyphylla var.japonica, colonies were induced efficiently from mesophyll protoplasts cultured in half strength MS (1/2MS) liquid medium containing 0.6 M mannitol, 0.09M sucrose and 1 M 4-PU and 1 M NAA at a cell density of 5 × 104/ml. The colonies grew actively and developed into callus after 3 months of culture.Roots differentiated from the protoplast-derived white calluses cultured on the 1 /2MS solid media supplemented with 0.1–1 M 4-PU and 1 M NAA, and 10 M zeatin with no supplementation of NAA. Furthermore, the protoplast-derived green callus differentiated shoots with 1/2MS solid medium containing 1 M 4-PU or 10 M zeatin with no supplementation of NAA. When shoots obtained were cultured on the cytokinin-free MS solid medium with 2.5 M IBA and 0.1 M NAA, they rooted and developed into plantlets after one month of culture.The phenylurea-type cytokinin, 4-PU, was effective for plantlet regeneration from the mesophyll protoplasts ofB. platyphylla var.japonica. Summary In order to develop techniques for efficient callus production and regeneration in Carica papaya (Var. Honey Dew), lamina, petiole, stem and root explants from in vitro plantlets were cultured in media supplemented with 2.0 mg/1 IBA and 0.5 mg/1 BAP. Use of in vitro-grown plantlets as an explant source helped to avoid contamination common in papaya tissue culture. Callusing was maximum in root explants cultured in a modified MS (half-strength) medium. Shoot reganeration was maxium in root-derived callus grown in full-strength modified MS medium supplemented with 0.5 mg/1 IBA and 1 to 2 mg/1 kinetin. A histological study indicated that shoot buds originated from peripheral cell layers of the callus. Each shoot regenerated from callus was subcultured using a multiplication medium. Summary Plant regeneration in Kentucky bluegrass (Poa pratensis L. cv. Touchdown) via culture of seedling tissues was investigated. When coleoptile, leaf, and stem sections of dark-germinated seedlings were cultured on Murashige and Skoog (MS) medium, different types of callus were produced, depending on the expiant source and growth regulator combinations. Only compact-friable callus (type 3) and moderately compact, friable callus (type 2) produced shoots upon subculture. The nonstructured watery callus (type 4) produced roots without shoots. Shoot differentiation from callus tissues was highest when the culture medium contained 0.2 mgL–1 picloram + 0.01 mgL–1 -naphthaleneacetic acid (NAA). Calli grown from coleoptiles had higher shoot regeneration frequency (32%) than that obtained from either stem sections (12%) or young leaf tissues (2%) of the same seedlings. Some organogenic callus lines produced exclusively green plants, while others produced albino shoots or a mixture of green and albino shoots. The green plants were multiplied in a medium containing 0.1 mgL–1 BAP plus either 0.2 mgL–1 picloram or 0.1 mgL–1 indole-3-acetic acid (IAA). Over 90% of the cultures in the shoot proliferation medium produced roots in 4 weeks. The Rubus species R. parviflorus, R. spectabilis and R. strigosus interfere with conifer seedling establishment on forest regeneration sites in Canada and the United States. As a first step towards microbial metabolite-based control, callus and cell suspension cultures of the Rubus species were developed as a bioassay system to detect phytotoxic compounds that may have relevance in a vegetation control context. Rapidly growing friable callus and suspension cultures were obtained from leaf disks of the three weedy Rubus species using similar culture media conditions (modified Murashige and Skoog) but required different plant growth regulators (R. parviflorus, 4.5 M 2,4-D; R. spectabilis, 26.9 M NAA/0.5 M zeatin; and R. strigosus, 12.4 M picloram). Cell growth and health attributes including callus circumference, degree of browning and suspension culture cell viability as measured by the TTC vital stain assay were developed and were rapid and convenient to use. We have established Rubus tissue culture systems that will make it possible for large scale screening of phytotoxic metabolites.  相似文献

The effect of lowered concentrations of carbon, nitrogen, and phosphorus sources in the medium on the specific growth rate of the R, S, and M dissociants of the hydrocarbon-oxidizing strain Pseudomonas aeruginosaK-2, culture pH, and the population composition was studied. Within the first 16 hours of cultivation in all of the four media tested, the R, S, and M dissociants have virtually identical . The maximal values of were reached by the 20th h of growth in the basal medium (R and S dissociants) and in the carbon-deficient medium containing 0.4% glucose (M dissociant). The R and M dissociants showed the most rapid decrease in in the nitrogen-deficient medium containing 0.55% NaNO3. By the end of cultivation in the basal medium, the pH of the R, S, and M cultures decreased to 6.3, 5.3, and 3.3, respectively. In the case of the carbon-deficient medium, the drop in the culture pH was lower. After a 2.5-day incubation of the S dissociant in the phosphorus- deficient medium containing 0.028% NaH2PO4· 2H2O and of the M dissociant in the basal medium supplemented with chalk powder, these dissociants were completely displaced from the media.  相似文献

Callus cultures of 7 potato cultivars were initiated from tuber tissue and maintained on Gelrite-solidified media with 1–20 M picloram as the only PGR. Ten M picloram was the optimal concentration for callus induction. By 4–6 weeks after explanting, there was sufficient callus produced for subculture to maintenance media which contained 1–20 M picloram as the only PGR. When grown in the dark at 25°C, subcultured callus typically increased 10-fold in wet weight in 4–5 weeks. The callus produced was friable and a light grey to cream color. Callus cultures were used to establish cell suspension cultures.
