玉米种质资源抗南方锈病鉴定   总被引:4,自引:1,他引:3  
玉米南方锈病已成为近几年我国夏玉米生产区间歇性暴发流行的病害,对玉米生产构成严重威胁,病害流行年份可造成10%以上的产量损失。目前,已确认的抗病自交系非常有限,而抗病育种急需不同抗性控制背景的自交系。为发掘和丰富可利用的南方锈病抗源,于2008~2012年,在广西南宁采用田间人工接种方法对1589份玉米种质资源进行抗南方锈病鉴定。通过高病害压力和连续多年的鉴定,从1589份玉米种质中鉴定出高抗(HR)材料26份,占鉴定总数的1.64%;抗病(R)材料137份,占鉴定总数的8.62%;中抗(MR)水平的材料382份,占鉴定总数的24.04%;感病(S)材料489份,占鉴定总数的30.77%;高感(HS)材料555份,占鉴定总数的34.93%。总体上抗南方锈病种质较少,引进种质中抗病类型材料的比例略高。经重复鉴定,筛选出赤556等18份自交系、老来秕等3份地方品种、A69等4份来自津巴布韦的材料、引自CIMMYT的Dr11表现稳定高抗南方锈病,为今后我国玉米抗南方锈病育种提供了新的抗性资源。  相似文献

南方锈病是玉米生产上的重要病害。2013-2015年在广西南宁和北京昌平对903份玉米种质资源进行了抗南方锈病的初步鉴定与评价,并利用SSR标记对筛选出的部分抗性材料进行了遗传多样性分析。结果表明,在903份种质中,8份自交系在广西南宁和北京昌平均对南方锈病表现高抗(HR),占总鉴定种质的0.9%;29份材料表现为抗病(R),占比3.2%,包括27份自交系和2份农家种;中抗种质(MR)100份,占比11.1%;感病(S)和高感(HS)种质分别为181和585份,占鉴定材料的20.0%和64.8%。由此可见,玉米资源中高抗南方锈病的种质较为匮乏,在不同地点均表现高抗的材料是难得的抗源。不同地理来源的玉米种质对南方锈病的抗性水平存在较大差异,其中抗性资源较为丰富的是源自内蒙古和山西的种质。42对多态性SSR引物在50份抗锈病材料中,共扩增出141个条带,多态性条带139个,多态位点百分率(PPB)为98.58%。平均等位基因数(Na)1.98,平均有效等位基因数(Ne)1.59,平均Nei's基因多样性(H)0.34,平均多态性信息含量(PIC)0.78,平均Shannon's信息指数(I)0.51;通过UPGMA聚类分析,50份抗病材料被划分为2个类群,其中,第Ⅰ类群又可划分为5个亚类,表现出较高的遗传多样性,为抗病育种中抗源的选择和利用提供参考信息。  相似文献

安徽梨主栽品种对锈病的抗性差异分析结果显示,最为感病的是砀山酥梨,其次为明月、大青皮.莱阳梨居中.较抗病的是开桦梨和康德梨.感病品种锈病在大田中呈聚集分布.抗病品种开桦梨也属聚集分布,而康德梨和莱阳梨锈病在田间为均匀分布.树上病情分布格局分析结果显示,无论感病或抗病品种均为聚集分布.  相似文献

为了筛选出高抗疮痂病马铃薯品种(系),本研究以36份马铃薯品种(系)资源为材料,分别在南宁市和玉林市种植,通过调查发病率和病情指数,用隶属函数法对36份材料的疮痂病抗性田间自然病圃鉴定结果进行分析。结果表明,发病率(x)和病情指数(y)呈正态分布,y是x的指数函数;明确了其中有2份高抗疮痂病马铃薯品种(系)和1份高感疮痂病马铃薯材料,田间种植法与人工接种鉴定结果基本一致,为马铃薯疮痂病情预测和抗疮痂病育种提供了参考。  相似文献

我国选育的优良玉米自交系齐319高抗南方玉米锈病。通过用病原菌接种齐319分别与5个不同感病自交系的杂交,回交4个世代的P1,P2,F1,F2,BC1F110个分离群体和抗病感病参数的调查结果表明:F1整齐一致,表现抗病;F2和BC1F1抗感分离,经x^2-检验分别符合3:1和1:1的分离比例。据此,齐319携带的南方玉米锈病抗性基因是由显性单基因所控制。  相似文献

采用人工接种黑星病菌的方法,对国家果树种质兴城梨资源圃保存的197份梨种质资源进行了抗病性鉴定,结果表明:不同梨种类发病率差异很大,其中白梨和砂梨最易感病,秋子梨和种间杂交选育品种较易感病,新疆梨较抗病,西洋梨最抗病;对病情指数在各梨种类分布进行了分析;在白梨、砂梨、秋子等各系统分别筛选出黄鸡腿、甩梨、酸梨、锦香等一批抗病资源;对田间自然感病与人工接种感病结果进行了比较。  相似文献

Nonhost resistance is the most common type of resistance in plants. Understanding the factors that make plants susceptible or resistant may help to achieve durably effective resistance in crop plants. Screening of 109 barley (Hordeum vulgare L.) accessions in the seedling stage indicated that barley is a complete nonhost to most of the heterologous rust fungi studied, while it showed an intermediate status with respect to Puccinia triticina, P. hordei-murini, P. hordei-secalini, P. graminis f. sp. lolii and P. coronata ff. spp. avenae and holci. Accessions that were susceptible to a heterologous rust in the seedling stage were much more or completely resistant at adult plant stage. Differential interaction between barley accessions and heterologous rust fungi was found, suggesting the existence of rust-species-specific resistance. In particular, many landrace accessions from Ethiopia and Asia, and naked-seeded accessions, tended to be susceptible to several heterologous rusts, suggesting that some resistance genes in barley are effective against more than one heterologous rust fungal species. Some barley accessions had race-specific resistance against P. hordei-murini. We accumulated genes for susceptibility to P. triticina and P. hordei-murini in two genotypes called SusPtrit and SusPmur, respectively. In the seedling stage, these accessions were as susceptible as the host species to the target rusts. They also showed unusual susceptibility to other heterologous rusts. Abstract The broad bean rust fungus Uromyces viciae-fabae exhibits chitin only on surfaces of those infection structures which in nature are formed on the plant cuticle, but not on those differentiated in the intercellular space of the host leaf. Chitin deacetylase, an enzyme which converts chitin to chitosan, has been studied during in vitro differentiation of rust infection structures. Radiometrie and gel electrophoretic analyses of crude extracts and extracellular washing fluids have shown that chitin deacetylase activity massively increases when the fungus starts to penetrate through the stomata, and that formation of the enzyme is strictly differentiation-specifically controlled. The extracellular portion of chitin deacetylase activity was about 53% in 24-h-old differentiated infection structures. Five isoforms with apparent molecular masses of 48.1, 30.7, 25.2, 15.2 and 12.7 kDa were detectable after substrate SDS-PAGE. The enzyme is temperature-sensitive and has a pH optimum of 5.5-6.0.  相似文献

Six accessions of Triticum turgidum var. dicoccoides L. (4x, AABB) of diverse origin were tested with 10 races of leaf rust (Puccinia recondita f.sp. tritici Rob. ex Desm.) and 10 races of stem rust (P. graminis f.sp. tritici Eriks. &Henn.). Their infection type patterns were all different from those of lines carrying the Lr or Sr genes on the A or B genome chromosomes with the same races. The unique reaction patterns are probably controlled by genes for leaf rust or stem rust resistance that have not been previously identified. The six dicoccoides accessions were crossed with leaf rust susceptible RL6089 durum wheat and stem rust susceptible 'Kubanka' durum wheat to determine the inheritance of resistance. They were also crossed in diallel to see whether they carried common genes. Seedlings of F1, F2, and BC1F2 generations from the crosses of the dicoccoides accessions with RL6089 were tested with leaf rust race 15 and those from the crosses with 'Kubanka' were tested with stem rust race 15B-1. The F2 populations from the diallel crosses were tested with both races. The data from the crosses with the susceptible durum wheats showed that resistance to leaf rust race 15 and stem rust race 15B-1 in each of the six dicoccoides accessions is conferred by a single dominant or partially dominant gene. In the diallel crosses, the dominance of resistance appeared to be affected by different genetic backgrounds. With one exception, the accessions carry different resistance genes: CI7181 and PI 197483 carry a common gene for resistance to leaf rust race 15. Thus, wild emmer wheat has considerable genetic diversity for rust resistance and is a promising source of new rust resistance genes for cultivated wheats.  相似文献

The taxonomic relationship between two microcyclic rust species, Puccinia melampodii and Puccinia xanthii, recorded from a number of Asteraceae hosts, was explored by comparing teliospore morphology, host specificity and sequence data. Teliospore morphology varied between and within individual rust accessions but, in general, teliospores of P. xanthii were larger than those of P. melampodii. Field observations and cross-inoculation greenhouse studies showed that P. melampodii accessions were highly specific to their original host species and unable to establish compatible interactions with related Asteraceae species. Though small amounts of genetic variation were detected between P. melampodii and P. xanthii accessions, the ITS and TEF phylogenetic analyses comprising several other rust species grouped these accessions within a well supported clade. Our data indicate that despite differences between P. xanthii and P. melampodii accessions these rusts cannot readily be separated at the species level, supporting earlier taxonomic conclusions and the recent proposal to group them into a morphospecies. Based on host-specificity data, the name Puccinia xanthii var. parthenii-hysterophorae is proposed to accommodate all Mesoamerican records of P. melampodii associated with the host Parthenium hysterophorus.  相似文献

Summary The epidemiology of rust caused by the fungus Melampsora lini and the effects of infection by this pathogen on its host, the herbaceous perennial Linum marginale, were determined in the field and in garden experiments. There was considerable natural variability in disease levels over the four years (1986–1989) of the study. In two years (1986, 1989) major rust epidemics occurred. In the field, the main effect of disease was to reduce survivorship during the winter following infection. Plants which were heavily infected during the 1986 or 1989 growing seasons had reduced survivorship relative to more lightly infected plants. Melampsora lini infections did not appear to affect survivorship in either 1987 or 1988. Flowering was correlated with environmental factors and the number of stems a plant possessed. A severe drought in 1987 completely inhibited flowering. In the other three years the number of flowers produced by a plant was strongly positively correlated with the number of stems it possessed. Disease levels had no consistent effect on flowering. Controlled garden experiments were also used to examine the response of seedlings and adult plants to infection. These showed that both the timing and severity of disease appears to determine the effect of M. lini infections on L. marginale. Early, severe infection reduced growing season and overwintering survivorship as well as capsule production. However, plants in the field were most often infected only after flowering had begun, and the predominant effect of infection was a reduction in overwintering survivorship. The high variability in disease levels from year to year and the deferred nature of the effect of the rust on its host have significant implications for the design of experiments aimed at assessing the role of diseases in plant communities.  相似文献

从1983~1995年,作者在秦岭地区采集了以树木锈菌为主的森林植物锈菌30属90余种。对这些锈菌的生态和生物地理分析表明:秦岭锈菌以温带型占明显优势(65%),且多分布于海拔1300~2800m的中山针阔叶林带中。该地区锈菌区系的地理成分以东亚成分为主,尤以中国-日本成分为主;欧亚-北美成分和旧世界温带成分在秦岭占有相当比例(约30%)。古热带成分等其它地理成分的锈菌也汇集于此。秦岭地区锈菌区系起源古老。  相似文献

Homoeologous group 1 chromosomes of wheat contain important genes that confer resistance to leaf, stem and stripe rusts, powdery mildew and Russian wheat aphid. A disease resistance gene analog encoding nucleotide binding site-leucine rich repeat (NBS-LRR), designated RgaYr10, was previously identified at the stripe rust resistant locus, Yr10, located on chromosome 1BS distal to the storage protein, Gli-B1 locus. RgaYr10 identified gene members in the homoeologous region of chromosome 1DS cosegregating with the leaf rust resistance gene, Lr21, which originally was transferred from a diploid D genome progenitor. Four RgaYr10 gene members were isolated from chromosome 1DS and compared to two gene members previously isolated from the chromosome 1BS homeologue. NBS-LRR genes tightly linked to stripe rust resistance gene Yr10 on chromosome 1BS were closely related in sequence and structure to NBS-LRR genes tightly linked to leaf rust resistance gene Lr21 located within the homoeologous region on chromosome 1DS. A collection of 917 accessions of Spanish durum and bread wheat was screened for resistance to leaf rust (Puccinia triticina) under field conditions at three locations. Resistance levels ranged from very low to very high, high susceptibility being most frequent. Relative disease severity (referred to the most susceptible accession = 100%) was lower than 20% in about 6% of the accessions in each location. In the collection most of the lines (84%) displayed a susceptible infection type. A final selection of seven accessions (one of them durum) displaying low severity level in the field and high infection type in a growth chamber was chosen for further studies. High levels of partial resistant with longer latency period and high percentage of early aborted colonies without necrosis were found. They might be used in breeding programmes.  相似文献

Brown rust resistance genes were sought in 23 resistant common wheat accessions with alien genetic material of Aegilops speltoides, Ae. triuncialis, and Triticum kiharae from the Arsenal collection. The genes were identified by common phytopathological tests and PCR analysis with STS markers directed to the known Lr genes. None of the methods identified the resistance genes in two accessions. In the other accessions, the combination of the two methods broadened the spectrum of detectable genes and, in some cases, allowed double verification of the presence of a resistance gene. Most accessions proved to contain several brown rust resistance genes, combining juvenile and adult plant ones. The accessions were found to contain gene combinations that ensured field resistance and immunity under the conditions of the Non-Chernozem region (Lr13 + Lr10 and Lr12 + Lr34). Accessions with alien genetic material contained a unique combination of five or six resistance genes. Since the accessions were rich in brown rust resistance genes, including effective ones, and carried rare combinations of these genes, they were proposed as donors to be universally employed in breeding for immunity in all regions of Russia.  相似文献
