The carbon isotope composition of an animals breath reveals the composition of the nutrients that it catabolizes for energy. Here we describe the use of Keeling plots, a method widely applied in ecosystem ecology, to measure the 13C of respired CO2 of small vertebrates. We measured the 13C of Rufous Hummingbirds (Selasphorus rufus) in the laboratory and of Mourning (Zenaida macroura) and White-winged (Z. asiatica) Doves in the field. In the laboratory, when hummingbirds were fed a sucrose based C3 diet, the 13C of respired CO2 was not significantly different from that of their diet (13CC3 diet). The 13C of respired CO2 for C3 fasted birds was slightly, albeit significantly, depleted in 13C relative to 13CC3 diet. Six hours after birds were shifted to a sucrose based C4 diet, the isotopic composition of their breath revealed that birds were catabolizing a mixture of nutrients derived from both the C3 and the C4 diet. In the field, the 13C of respired CO2 from Mourning and White-winged Doves reflected that of their diets: the CAM saguaro cactus (Carnegeia gigantea) and C3 seeds, respectively. Keeling plots are an easy, effective and inexpensive method to measure 13C of respired CO2 in the lab and the field.  相似文献

The effect of interspecific competition and element additions (N and P) on four grassland species (Poa pratensis, Lolium perenne, Festuca valida, Taraxacum officinale) grown under field conditions was studied. Two grasses (L. perenne, F. valida) grown in monoculture (absence of competition) showed lower carbon isotope discrimination (13C) and enriched 15N values. Nitrogen addition (as urea) had inconsistent effects on species 13C while caused enrichment of 15N of P. pratensis and F. valida but strong depletion of 15N of T. officinale. Phosphorous had no significant effect on 13C but depleted 15N of all species.  相似文献

Temporal variability in the 13C of foliage (13CF), soil (13CS) and ecosystem (13CR) respired CO2 was contrasted between a 17.2-m tall evenly aged loblolly pine forest and a 35-m tall unevenly aged mature second growth mixed broadleaf deciduous forest in North Carolina, USA, over a 2-year period. The two forests are located at the Duke Forest within a kilometer of each other and are subject to identical climate and have similar soil types. The 13CF, collected just prior to dawn, was primarily controlled by the time-lagged vapor pressure deficit (VPD) in both stands; it was used for calculating the ratio of intercellular to ambient CO2 (Ci/Ca). A remarkable similarity was observed in the relationship between Ci/Ca and time-lagged VPD in these two forests despite large differences in hydraulic characteristics. This similarity emerged as a result of physiological adjustments that compensated for differences in plant hydraulic characteristics, as predicted by a recently proposed equilibrium hypothesis, and has implications to ecophysiological models. We found that in the broadleaf forest, the 13C of forest floor CO2 efflux dominated the 13CR, while in the younger pine forest, the 13C of foliage respired CO2 dominated 13CR. This dependence resulted in a more variable 13CR in the pine forest when compared to the broadleaf forest due to the larger photosynthetic contribution. Given the sensitivity of the atmospheric inversion models to 13CR, the results demonstrate that these models could be improved by accounting for stand characteristics, in addition to previously recognized effects of moisture availability, when estimating 13CR.  相似文献

Estimates of nitrogen fixation by trees on an aridity gradient in Namibia   总被引:5,自引:0,他引:5  
Summary Nitrogen (N2) fixation was estimated along an aridity gradient in Namibia from the natural abundance of 15N (15N value) in 11 woody species of the Mimosacease which were compared with the 15N values in 11 woody non-Mimosaceae. Averaging all species and habitats the calculated contribution of N2 fixation (N f ) to leaf nitrogen (N) concentration of Mimosaceae averaged about 30%, with large variation between and within species. While in Acacia albida N f was only 2%, it was 49% in Acacia hereroensis and Dichrostachys cinerea, and reached 71% in Acacia melifera. In the majority of species N f was 10–30%. There was a marked variation in background 15N values along the aridity gradient, with the highest 15N values in the lowland savanna. The difference between 15N values of Mimosaceae and non-Mimosaceae, which is assumed to result mainly from N2 fixation, was also largest in the lowland savanna. Variations in 15N of Mimosaceae did not affect N concentrations, but higher 15N-values of Mimosaeae are associated with lower carbon isotope ratios (13C value). N2 fixation was associated with reduced intrinsic water use efficiency. The opposite trends were found in non-Mimosaceae, in which N-concentration increased with 15N, but 13C was unaffected. We measured leaf dry matter 18O and 13C in parasitic plants and their hosts growing in southwestern Australia. Parasite/host pairs included two mistletoe species, three species of holoparasites, and five species of root hemiparasites. Among these parasite functional types, significant variation was observed in parasite/host isotopic differences for both 18O (P<0.0001, n=65) and 13C (P<0.0001, n=64). Mistletoes were depleted in both 18O and 13C compared to their hosts; parasite/host differences were –4.0 for 18O (P<0.0001) and –1.9 for 13C (P<0.0001). The lower 18O in mistletoe leaf dry matter compared to their hosts is consistent with the frequently observed high transpiration rates of these parasites. Root hemiparasites were also depleted in 18O and 13C compared to their hosts, but not to the same extent as mistletoes; parasite/host differences were –1.0 for 18O (P=0.04) and –1.2 for 13C (P=0.0006). In contrast to mistletoes and root hemiparasites, holoparasites were enriched in both 18O and 13C compared to their hosts; parasite/host differences were +3.0 for 18O (P<0.0001) and +1.5 for 13C (P=0.02). The enrichment in 18O for holoparasite dry matter did not result from more enriched tissue water; holoparasite tissue water 18O was less than host leaf water 18O by a difference of –3.8 when sampled at midday (P=0.0003). Enrichment of holoparasites in 13C compared to their hosts is consistent with a generally observed pattern of enrichment in heterotrophic plant tissues. Results provide insights into the ecology of parasitic plants in southwestern Australia; additionally, they provide a context for the formulation of specific hypotheses aimed at elucidating mechanisms underlying isotopic variations among plants.  相似文献

In order to assess the ability of Porites corals to accurately record environmental variations, high-resolution (weekly/biweekly) coral 18O records were obtained from four coral colonies from the northern Gulf of Aqaba, which grew at depths of 7, 19, 29, and 42 m along one transect. Adjacent to each colony, hourly temperatures, biweekly salinities, and monthly 18O of seawater were continuously recorded over a period of 14 months (April 1999 to June 2000). Contrary to water temperature, which shows a regular and strong seasonal variation and change with depth, seawater 18O exhibits a weak seasonality and little change with depth. Positive correlations between seawater 18O and salinity were observed. The two parameters were related to each other by the equation 18O Seawater (, VSMOW) = 0.281 × Salinity – 9.14. The high-resolution coral 18O records from this study show a regular pattern of seasonality and are able to capture fine details of the weekly average temperature records. They resolve more than 95% of the weekly average temperature range. On the other hand, attenuation and amplification of coral seasonal amplitudes were recorded in deep, slow-growing corals, which were not related to environmental effects (temperature and/or seawater 18O) or sampling resolution. We propose that these result from a combined effect of subannual variations in extension rate and variable rates of spine thickening of skeletal structures within the tissue layer. However, no smoothing or distortion of the isotopic signals was observed due to calcification within the tissue layer in shallow-water, fast-growing corals. The calculations from coral 18O calibrations against the in situ measurements show that temperature (T) is related to coral 18O ( c ) and seawater 18O ( w ) by the equation T (°C) = –5.38 ( c w ) –1.08. Our results demonstrate that coral 18O from the northern Gulf of Aqaba is a reliable recorder of temperature variations, and that there is a minor contribution of seawater 18O to this proxy, which could be ignored.  相似文献

Summary Plants of Drosera species, neighbouring noncarnivorous plants, and arthropods on or near each Drosera sp. were collected at 11 contrasting habitat locations in SW Australia. At three of the sites clones of the rare glandless mutant form of D. erythrorhiza were collected alongside fully glandular counterparts. The 15N value (15N/14N natural isotope composition) of insect-free leaf and stem fractions was measured, and the data then used to estimate proportional dependence on insect N (%NdI) for the respective species and growth forms of Drosera. The data indicated lower %NdI values for rosette than for self-supporting erect or for climbing vine species. The latter two groups showed an average %NdI value close to 50%. The %NdI increased with length and biomass of climbing but not erect forms of Drosera. 15N values of stems were positively correlated with corresponding values for leaves of Drosera. Leaf material was on average significantly more 15N enriched than stems, possibly due to delayed transport of recent insect-derived N, or to discrimination against 15N in transfer from leaf to the rest of the plant. The comparison of 15N values of insects and arthropod prey, glandless and glandular plants of D. erythrorhiza indicated %NdI values of 14.3, 12.2 and 32.2 at the respective sites, while matching comparisons based on 15N of insect, reference plants and glandular plants proved less definitive, with only one site recording a positive %NdI (value of 10.4%) despite evidence at all sites of feeding on insects by the glandular plants. The use of the 15N technique for studying nutrition of carnivorous species and the ecological significance of insect feeding of different growth forms of Drosera growing in a large range of habitats is discussed.  相似文献

Otoliths of ocean-pen reared cod, Gadus morhua, provide a unique opportunity to examine the lifetime history of the fish. Here we report 18 analyses of such otoliths on seasonal (winter and summer) stable oxygen and carbon isotope ratios. The calculated isotopic temperatures from otoliths of reared cod were essentially in agreement with the temperature record during rearing, suggesting that temperature is a dominant factor in the precipitation of otolith aragonite. Carbon isotope ratios increased with age and leveled off at 4 years old, presumed to correlate with sexual maturity of cod. As compared with otoliths of wild-caught cod, significant differences were found in isotope variation and the correlation between 13C and 18O. These differences were probably attributable to the different environmental constraints and food supply.  相似文献

Understanding the links between breeding and wintering areas of migratory species has important ecological and conservation implications. Recently, stable isotope technology has been used to further our understanding. Stable isotope ratios vary geographically with a range of biogeochemical factors and isotope profiles in organisms reflect those in their food and environment. For inert tissues like feathers, isotope profiles reflect the environment in which they were formed. Following large-scale habitat destruction, the globally threatened aquatic warbler Acrocephalus paludicola has a fragmented breeding population across central Europe, largely in Belarus, Poland and Ukraine. The species sub-Saharan African wintering grounds have not yet been discovered, and this significantly hampers conservation efforts. Aquatic warblers grow their flight feathers on their wintering grounds, and we analysed stable isotope ratios (15N, 13C, D) in rectrices of adults from six main breeding sites (subpopulations) across Europe to determine whether different breeding subpopulations formed a single mixed population on the wintering grounds. 15N varies considerably with dietary trophic level and environmental factors, and D with the D in rainfall; neither varied between aquatic warbler subpopulations. Uniform feather 15N signatures suggest no major variation in dietary trophic level during feather formation. High variance and inter-annual differences in mean D values hinder interpretation of these data. Significant differences in mean 13C ratios existed between subpopulations. We discuss possible interpretations of this result, and consider differences in moulting latitude of different subpopulations to be the most parsimonious. 13C in plants and animals decreases with latitude, along a steep gradient in sub-Saharan Africa. Birds from the most north-westerly breeding subpopulation (Karsibor, Poland) had significantly lower variance in 13C and 15N than birds from all other sites, suggesting either that birds from Karsibor are less geographically dispersed during moult, or moult in an area with less isotopic heterogeneity. Mean 13C signatures from winter-grown feathers of different subpopulations were positively correlated with the latitude and longitude of breeding sites, suggesting a strong relationship between European breeding and African winter moulting latitudes. The use of stable isotopes provides novel insights into migratory connectivity and migration patterns in this little-known threatened species.  相似文献

In situ photosynthetic nitrogen-use efficiency (PNUE, photosynthetic capacity per unit leaf nitrogen) was investigated in species that commonly distributed at different altitudes (600–3700m above sea level) on Mount Kinabalu. Photosynthetic nitrogen-use efficiency was lower in species at higher altitudes, with a mean PNUE at 3700m being one-third as large as that at 600m. This difference in PNUE was larger than that explained by the biochemical response to lower air pressures only. Across altitudes a negative correlation between 13C abundance (13C) and PNUE was found. Species at higher altitudes tended to have higher 13C, suggesting that they had a lower conductance for CO2 diffusion from the air to chloroplasts. The lower conductance might be responsible for the lower PNUE in species at higher altitudes. Although leaf nitrogen content per unit area tended to be higher at higher altitudes, it did not seem to contribute to increasing photosynthetic rates. Thus, the idea that a higher nitrogen content at higher altitudes is a compensation for a lower PNUE was not supported. In contrast to the large difference in PNUE among altitudes, PNUE tended to converge within a narrow range among species growing at the same altitude.  相似文献

The 13C signature of organic carbon in estuarine bottom sediment in Louisiana Barataria Basin was used for estimating carbon flux from adjacent marsh. The stable carbon isotope composition of plants, soils and sediments from the basin were determined. The 13C content of marsh vegetation ranged from -26.3 to -27.8% for C3 freshwater vegetation in the upper basin to -13.0 to -13.3% for C4 vegetation in the lower basin. The 13C content of the highly organic marsh soils were similar to 13C content of vegetation present. The 13C content of organic carbon from bottom sediment of open water bodies ranged from 27.3 in the upper basin (freshwater) to 16.4 in bottom sediment of salt marsh ponds. The13C signature of organic carbon in bottom sediment from saline regions corresponded to the size of the body of water. The smaller salt marsh ponds contain sediment with 13C values close to that of the C4 plantSpartina alterniflora. Results suggest that phytoplankton rather thanSpartina alterniffora is the likely organic source in bottom sediment of the larger bay near the coast (e.g. Caminada Bay).  相似文献

Winter and spring precipitation that saturates to deep soil layers precedes summer droughts in the Intermountain West. Occasional summer convection storms relieve summer drought, but are infrequent and unreliable from year to year, leading to the hypothesis that dominant tree species might not invest limited carbon reserves to surface roots to take up summer precipitation in these regions. We compared the hydrogen (D) and oxygen (18O) isotope ratios of winter, spring and summer precipitation to that of xylem sap water in Acer grandidentatum and Quercus gambelii, two dominant trees of this region. By this method we could identify water sources utilized throughout the growing season. Xylem D and 18O values changed significantly when each species leafed-out; this change was not associated with changes in either soil or plant water status (as measured by predawn and midday water potentials). This shift is apparently related increased transpirational flux, which may flush out residual stem water from the previous growing season. D values of xylem sap of both species matched winter precipitation input values throughout most of the summer, indicating a reliance on deep-soil moisture sources throughout the growing season. Mature Q. gambelii did not take up summer precipitation, whereas A. grandidentatum responded slightly to the largest summer rain event. Small trees of both species, particularly A. grandidentatum, showed a limited uptake of summer rains.  相似文献

Mangrove sediments from three different mangrove ecosystems (Coringa Wildlife Sanctuary in the Godavari Delta, Andhra Pradesh, India, and Galle and Pambala, south-west Sri Lanka) were analysed for their organic carbon content, elemental ratios (C:N) and carbon stable isotope composition. Organic carbon content (0.6 – 31.7% dry weight), C/N ratios (7.0 – 27.3) and 13C (between –29.4 and –20.6) showed a wide range of values. Lower stocks of organic carbon coincided with low C/N (atom) ratios and less negative 13C values, indicating import of marine or estuarine particulate suspended matter. High organic carbon stocks coincided with high C/N ratios and 13C values close, but not equal, to those of the mangrove vegetation. The variations observed in this study and published literature data could be adequately described by a simple two-end mixing model, whereby marine/estuarine suspended matter and mangrove litter were taken as end members. Thus, while in some mangrove ecosystems or vegetation zones, organic carbon stocks can be very high and are almost entirely of mangrove origin, there also appear to be cases in which deposited estuarine or marine suspended matter is the dominant source of organic carbon and nitrogen in mangrove sediments. This situation is remarkably similar to that observed in temperate salt marsh ecosystems where the importance of local vascular plant production to the sediment organic carbon pool is equally variable. The observed high variability in organic matter origin is thought to have a major impact on the overall carbon dynamics in intertidal mangrove ecosystems.  相似文献

Samples of the Clusiaceae generaClusia, Oedematopus andDystovomita were collected at various sites and different altitudes in northern and south-western Venezuela. Analyses of stable isotopes of carbon and hydrogen and of leaf-nitrogen levels were performed on the dried samples. Correlations among these variables, i.e. carbon isotope discrimination (), hydrogen isotope ratio (D) and N-levels, and with altitude were assessed. In the samples, where values of above 15 indicate predominant performance of C3 photosynthesis, there were slight tendencies of increasing , D and N-levels with increasing altitude and of increasing with increasing N. Although these correlations taken separately were not statistically significant, they support each other and indicate increasing transpiration and increased leaf-nutrient supply at increasing altitude. Performance of crassulacean acid metabolism (CAM) in species ofClusia appears to be restricted to altitudes below 1500 m a.s.l. There was a significant negative correlation of with altitude in the samples, where values of below 10 indicated predominant performance of CAM. This suggests that phases II and IV of CAM are progressively suppressed towards the upper altitudinal limit of CAM inClusia in northern Venezuela. It is concluded that among the large number of environmental factors and combinations thereof, which determine the expression of CAM inClusia and trigger C3-CAM transitions in C3/CAM intermediate species, low availability of water is the most important.  相似文献

Variation in stable nitrogen isotope ratios (15N) was assessed for plants comprising two wetland communities, a bog-fen system and a flood plain, in central Japan. 15N of 12 species from the bog-fen system and six species from the flood plain were remarkably variable, ranging from –5.9 to +1.1 and from +3.1 to +8.7, respectively. Phragmites australis exhibited the highest 15N value at both sites. Rooting depth also differed greatly with plant species, ranging from 5cm to over 200cm in the bog-fen system. There was a tendency for plants having deeper root systems to exhibit higher 15N values; plant 15N was positively associated with rooting depth. Moreover, an increasing gradient of peat 15N was found along with depth. This evidence, together with the fact that inorganic nitrogen was depleted under a deep-rooted Phragmites australis stand, strongly suggests that deep-rooted plants actually absorb nitrogen from the deep peat layer. Thus, we successfully demonstrated the diverse traits of nitrogen nutrition among mire plants using stable isotope analysis. The ecological significance of deep rooting in mire plants is that it enables those plants to monopolize nutrients in deep substratum layers. This advantage should compensate for any consequential structural and/or physiological costs. Good evidence of the benefits of deep rooting is provided by the fact that Phragmites australis dominates as a tall mire grass.  相似文献

Here we report on the use of iodination of the membrane-bound nicotinic acetylcholine receptor (nAChR) from Torpedo californica electric tissue in order to define surface-exposed portions of the receptor molecule. Membrane-bound nAChR was 125I-iodinated using the oxidation agent Iodo-Gen. The iodinated subunits were separated by preparative gel electrophoresis, desalted, and cleaved with trypsin. The resulting peptides were separated by reverse-phase HPLC and the radioactive peptides were identified by mass spectrometry and protein sequencing. For the -subunit, we identified five iodinated peptides containing the tyrosine residues Tyr17, Tyr74, Tyr365, Tyr372, and Tyr428. The surface exposition of these amino acids is in agreement with the four-transmembrane-segment model (4TM model) of the nAChR, but the assignment to the intra- or extracellular surface is doubtful. According to this model, the N-terminal portion of the receptor subunits including the iodinated residues Tyr17 and Tyr74 is extracellular and Tyr372 as a site of tyrosine phosphorylation is located on the cytoplasmic side. But since this latter residue is among the first to be iodinated using an immobilized iodination agent, its true position with respect to the membrane bilayer is not clear.  相似文献

In Ceropegia dichotoma, Crassula argentea, Esheveria colorata, Kalanchoë beharensis, Opuntia ficus-indica, Sansveria stuckyi and Opuntia inermis the carbon-isotope ratio ( 13C) of tissues close to the epidermis is 2–4.3 more negative than those in the centre of the leaf or cladode. The greatest change in 13C value occurs between the epidermal layer and the layer of mesophyll tissue immediately underneath. Analysis of major metabolic and structural components in successive layers of Crassula argentea grown under controlled environmental conditions conducive to Crassulacean acid metabolism confirmed that much of the variation in 13C values of bulk carbon is caused by differences in chemical composition. Thus the steep gradient in 13C value at the epidermis reflects, in part, the contribution of more-negative 13C values of lipids in these tissues. Moreover, during nocturnal CO2 fixation the amount of malic acid synthesised decreases with depth and the 13C value of the methanol-soluble fraction is less negative with distance away from the upper epidermis. At five European sites, differing in atmospheric Sinputs by a factor of 6, and differing in S isotope signatures ofthese inputs by up to 14 (CDT), we investigated thedirection and magnitude of an assimilation-related 34S shiftand the relationship between atmospheric deposition and Sretention in selected ecosystem compartments. Bulk precipitationand spruce throughfall were collected between 1994 and 1996 inthe Isle of Mull (Scotland), Connemara (Ireland), Thorne Moors(England), Rybárenská slat' and Oceán (both Czech Republic) andanalyzed for sulfate concentrations and 34S ratios. Eighteenreplicate samples per site of living Sphagnum collected inunforested peatlands and 18 samples of spruce forest floorcollected near each of the peatlands were also analyzed for Sconcentrations and 34S ratios. Assimilation of S was associatedwith a negative 34S shift. Plant tissues systematicallypreferred the light isotope 32S, on average by 2. There wasa strong positive correlation between the non-marine portion ofthe atmospheric S input and total S concentration in forest floorand Sphagnum, respectively (R = 0.97 and R = 0.85). Elevated Sinputs lead to higher S retention in these two organic-richcompartments of the ecosystem. It follows that equal emphasismust be placed on organic S as on adsorption/desorption ofinorganic sulfate when studying acidification reversal inecosystems. The sea-shore sites had rainfall enriched in theheavy isotope 34S due to an admixture of sea-spray. The inlandsites had low 34S reflecting 34S of sulfur emitted from localcoal-burning power stations. Sphagnum had always lower S contentsand higher 34S ratios compared to forest floor. The within-siterange of 34S ratios of Sphagnum and forest floor was wide (upto 12) suggesting that at least six replicate samples shouldbe taken when using 34S as a tracer.  相似文献

Temporal and spatial changes in 13C and 15N of seston (mainly phytoplankton) and isotopic relationship between seston and the lake anchovy (Coilia ectenes) were studied in the large eutrophic freshwater Lake Chaohu in China. Much of the spatial and temporal variation in 13C of lake anchovies was explained by variation in seston, indicating a strong link between pelagic primary production and higher order consumers. Because the lake is shallow, there were no significant differences in 13C and 15N of seston between surface and overlying waters. Spatially, the relatively high 13C and 15N of seston in the western part of the lake might be due to high levels of anthropogenically derived N and C introduced from the surrounding cities through sewage drainage systems. The trophic position of the lake anchovy in the food web of Lake Chaohu was estimated to be 2.9–4.1 (3.5 ± 0.4), which agrees well with the previous stomach content analysis suggesting that the lake anchovy fed both on zooplankton and small planktivorous fishes.  相似文献

Measurement of nitrogen isotopic composition (15N) of plants and soil nitrogen might allow the characteristics of N transformation in an ecosystem to be detected. We tested the measurement of 15N for its ability to provide a picture of N dynamics at the ecosystem level by doing a simple comparison of 15N between soil N pools and plants, and by using an existing model. 15N of plants and soil N was measured together with foliar nitrate reductase activity (NRA) and the foliar NO3 pool at two sites with different nitrification rates in a temperature forest in Japan. 15N of plants was similar to that of soil NO3 in the high-nitrification site. Because of high foliar NRA and the large foliar NO3 pool at this site, we concluded that plant 15N indicated a great reliance of plants on soil NO3 there. However, many 15N of soil N overlapped each other at the other site, and 15N could not provide definitive evidence of the N source. The existing model was verified by measured 15N of soil inorganic N and it explained the variations of plant 15N between the two sites in the context of relative importance of nitrification, but more information about isotopic fractionations during plant N uptake is required for quantitative discussions about the plant N source. The model applied here can provide a basis to compare 15N signatures from different ecosystems and to understand N dynamics.  相似文献
