We studied the development and feeding behaviour of the pea aphid, Acyrthosiphon pisum (Harris) (Homoptera: Aphididae), on the Radius and Sapko alfalfa ( Medicago sativa L.) (Fabaceae) cultivars. Three saponins and flavones were identified in the alfalfa cultivars after thin layer chromatography separation. Cultivar Radius differed from Sapko in that it had a higher level of saponins, including zanhic acid tridesmoside and 3-GlcA,28-AraRhaXyl medicagenic acid glycoside. The flavones identified, including 7- O -β-D-glucuronopyranosyl-4'- O- [2'- O- E-feruloyl- O -β-D-glucuronopyranosyl(1→2)- O -β-D-glucuronopyranoside] apigenin, 7- O -{2- O- E-feruloyl-[β-D-glucuronopyranosyl(1→3)]- O -β-D-glucuronopyranosyl(1→2)- O -β-D-glucuronopyranoside} apigenin, and 4'- O- [2'- O -E-feruloyl- O -β-D-glucuronopyranosyl(1→2)- O -β-D-glucuronopyranoside] apigenin, occurred in tissues of both alfalfa cultivars. However, cv. Radius had very low mean flavonoid concentrations in comparison to cv. Sapko. Pea aphids that fed on cv. Radius plants showed a reduction in reproduction and survival. The aphid pre-reproductive period on cv. Radius was prolonged and the reproductive and post-reproductive periods on cv. Radius were reduced, compared to those on cv. Sapko. Cultivar Radius also negatively influenced aphid probing behaviour. This was especially the case during the initial period of the pathway phase. The results suggested that alfalfa cv. Radius, which had a higher level of saponins and a lower level of flavonoids, was less accepted by the pea aphid.  相似文献

Electrical penetration graphs (DC EPG) were used to monitor the feeding behavior of the pea aphid Acyrthosiphon pisum Harris (Hemiptera: Aphididae) exposed to mixtures of saponins: 3GlcA, 28AraRhaXyl medicagenic acid glycoside (MAG) and zanhic acid tridesmoside (ZAD) with flavonoid apigenin, on agarose-sucrose gels. In general, mixtures of saponins with apigenin incorporated into agarose-sucrose gels resulted in a reduction in both the number of aphid probes and delayed their duration. Aphid feeding on gels impregnated with mixtures of the tested chemicals also showed a reduction in salivation into the gels and elongation of passive ingestion from the gels (EPG patterns g-E1 and g-E2, respectively). There were significant differences among the feeding behaviors of the pea aphids on gels containing mixtures of either MAG or ZAD with apigenin. These differences were also depended on the concentration and proportions of these compounds.  相似文献

Triterpene saponins are a group of bioactive compounds abundant in the genus Medicago , and have been studied extensively for their biological and pharmacological properties. In this article, we evaluated the effects of the ectopic expression of AsOXA1 cDNA from Aster sedifolius on the production of triterpene saponins in barrel medic ( Medicago truncatula Gaertn.). AsOXA1 cDNA encodes β-amyrin synthase, a key enzyme involved in triterpene saponin biosynthesis. One of the four transgenic lines expressing AsOXA1 accumulated significantly larger amounts of some triterpenic compounds in leaf and root than did control plants. In particular, the leaf exhibited significantly higher levels of bayogenin, medicagenic acid and zanhic acid. The amounts of medicagenic acid and zanhic acid, which represent the core of the M. truncatula leaf saponins, were 1.7 and 2.1 times higher, respectively, than the amounts extracted from the control line. In root, the production of bayogenin, hederagenin, soyasapogenol E and 2β-hydroxyoleanolic acid was increased significantly. The increase in the total amounts of triterpenic compounds observed in the leaves of transgenic lines correlated with the AsOXA1 expression level. Interestingly, the plants expressing AsOXA1 showed, under different growth conditions, improved nodulation when compared with the control line. Nodulation enhancement was also accompanied by a significant change in the soyasapogenol B content. Our results indicate that the ectopic expression of AsOXA1 in barrel medic leads to a greater accumulation of triterpene saponins and enhanced root nodulation.  相似文献

Summary The allelopathic effect of alfalfa (Medicago media Pers.) and red clover (Trifolium pratense L.) root saponins on winter wheat seedling growth and the fate of these chemicals in soil environments were studied. Seed germination, seedling and test fungus growth were suppressed by water and by alcohol extracts of alfalfa roots, and by crude saponins of alfalfa roots, indicating that medicagenic acid glycosides are the inhibitor. Powdered alfalfa roots inhibited wheat seedling growth when added to sand. At concentrations as low as 0.25% (w/w) the root system was completely destroyed whereas seedling shoots suffered little damage. Red clover roots caused some wheat growth inhibition when incorporated to sand, but their effect was much lower than in the alfalfa root treatment. Soil textures had a significant influence on the inhibitory effect of alfalfa roots. The inhibition of seedling growth was more pronounced on light than on heavy soils. This was attribted to the higher sorption of inhibitors by heavy soils. Incubation of alfalfa roots mixed into loose sand, coarse sand, loamy sand and clay loam for a period of 0–8 days resulted in decreased toxicity to bothT. viride and wheat seedlings. This decrease occurred more quickly in heavier soils than in loose sand, due to the hydrolysis of glycosides by soil microorganisms. Soil microbes were capable of detoxifying medicagenic acid glycosides by partial hydrolysis of sugar chain to aglycone. These findings illustrate the importance of medicagenic acid glycosides as an inhibitor of wheat seedling growth, and of their fate in different soil environments.  相似文献

This research aims to examine the effect of phenolics on pea aphid (Acyrthosiphon pisum) (Homoptera: Aphididae) development and feeding behaviour, on leaves of selected low-saponin lines of Radius alfalfa (Medicago sativa). There was a slight, negative correlation (Spearman rank correlation r s = −0.80) between concentrations of saponins and phenols. Lines with higher concentrations of saponins had less phenolics. Levels of phenolics in low-saponin lines of alfalfa cv. Radius were related to their acceptance by the pea aphid. Our data revealed an inverse relationship between level of phenolics and the aphid abundance and its biology on studied alfalfa lines. Larval development of the pea aphid was longer, reproduction period was shorter, and the fecundity was lower on low-saponin lines with higher level of phenolics. There were observed some tendencies in the pea aphid feeding behaviour on these lines: prolonging the probing of the peripheral tissues (epidermis and mesophyll) and shortening the period of phloem sap ingestion. The better hosts for the pea aphid were low-saponin lines with low levels of phenolic compounds.  相似文献

The bean aphid, Megoura crassicauda Mordvilko, feeds selectively on plants belonging to the genus Vicia (Fabaceae). However, it never infests the tiny vetch, V. hirsuta (L.) Gray. The aphid appeared to discriminate between host and non-host plants by tasting specific chemicals during penetration of its stylet into the plant tissues. The aphid, after being stimulated by specific probing stimulants, deposited characteristic proteinous stylet sheaths through a parafilm membrane, which has one side in contact with an extract solution of Vicia angustifolia. However, an addition of a V. hirsuta extract to the medium strongly inhibited the salivary sheath formation. A specific probing deterrent was isolated from a V. hirsuta extract by monitoring the inhibitory effect, and identified as (E)-2-methyl-2-butene-1,4-diol 4-O-beta-d-glucopyranoside. A mixture of the glycoside and the stimulatory V. angustifolia fraction in the same equivalency found in plants significantly decreased the probing activity in M. crassicauda. Since the stylet insertion process is a crucial step for the aphid's settlement on a plant, the glycoside seems to act as an effective chemical barrier for V. hirsuta.  相似文献

Among alfalfa pests in Iran three aphid species, green alfalfa aphid Acyrthosiphon pisum Harris spotted alfalfa aphid Therioaphis trifolii forma maculata Buckton and blue alfalfa aphid, Acyrthosiphon kondoi Shinji are important pests. The green alfalfa aphid can be observed all along the growing season particularly from late May to mid June at Karaj climate conditions. During this period, the mean monthly maximum temperature and relative humidity were about 28 degrees C and 60-65% respectively. This aphid overwinters as nymph and viviparous female. Sexual forms and eggs could not be seen under field conditions. Spotted alfalfa aphid, Therioaphis trifolii fonna maculata is the most prevalent aphid in summer time, when the mean monthly maximum temperature and relative humidity are about 33-34 degrees C and 44-58% respectively. Sexual individuals have been observed in the laboratory but not in the field. Among predators (Coccinella septempunctata, Adonia variegata, Syrphus cinctus, S. corolae, S. grassulariae, Chrysoperla carnea and Nabis capsiformis) one coccinellid species, C. septempunctata, had greatest impact on fluctuations of population. Among hymenopterous parasitoids two species have been collected from alfalfa field they were Aphidius ervi and Praon palitans. These parasitoids destroyed a good percent of aphids and statistically proved to lower aphid populations significantly.  相似文献

本文对新疆博州豇豆豆蚜Aphis craccivora (Koch)田间消长规律进行调查,研究了0.3%印楝素与脂肪酸甲酯喷雾助剂联合对豆蚜的防治效果,并对豆蚜天敌消退率进行了调查。结果表明,在博州,豆蚜以卵在苦豆子和苜蓿上越冬,3月下旬至4月初开始孵化,5月上中旬出现有翅蚜在杂草上繁殖蔓延,5月中旬开始迁入棉花上危害,6月中旬陆续迁入豇豆上危害,7月上中旬在豇豆上达到危害高峰时期,7月中下旬慢慢迁入苜蓿地,9月中旬出现性蚜,9月下旬-10月中旬在苜蓿和杂草上产卵越冬。施药后1 d,20%啶虫脒可湿性粉剂1 000倍液对豆蚜的防效最高,达到82.91%; 施药后3 d,20%啶虫脒可湿性粉剂1 000倍液防效仍最高为97.92%;施药后7 d,0.3%印楝素乳油60 mL/667m2+脂肪酸甲酯喷雾助剂1 000倍液防效达97.38%,显著高于其他处理。总体来说,在豇豆豆蚜发生期施用1次0.3%印楝素+脂肪酸喷雾助剂防治豇豆豆蚜,防效好、持效期长,对豇豆及蚜虫天敌安全性较好。  相似文献

Host evaluation behaviour was examined in three species of aphid parasitoids,Aphidius ervi haliday,A. pisivorus Smith, andA. smithi Sharma & Subba Rao (Hymenoptera: Aphidiidae). Parasitoids were provided under laboratory conditions with three kinds of hosts representing two aphid species: (green) pea aphid,Acyrthosiphon pisum (Harris), and green and pink colour morphs of the alfalfa aphid,Macrosiphum creelii Davis. Females of all threeAphidius species distinguished between aphids on the basis of colour, movement, and host species. Patterns of host acceptance by parasitoids were species-specific. InA. ervi, host preference was the same in light and dark conditions: pea aphid>green alfalfa aphid≫pink alfalfa aphid. In contrast,A. pisivorus attacked and accepted pea aphid and green alfalfa aphid equally in the light and preferred both of these over pink alfalfa aphid; however, it made no distinction between pea aphid and pink alfalfa aphid in the dark. Females ofA. smithi attacked all three kinds of hosts (pea aphid>green alfalfa aphid≫pink alfalfa aphid) but apparently laid eggs only in pea aphid. The frequencies of attack and oviposition by all wasps were higher on ‘normal’ pea aphids than on those anaesthetized with CO2. Host recognition is confirmed by chemical cues in the aphid cuticle that are detected during antennation, and host acceptance is dependent on an assessment of host quality during ovipositor probing.  相似文献

The antimycotic activity of medicagenic acid and of some synthetic derivatives thereof was tested against plant pathogenic fungi. In general they all possess antimycotic activity. Furthermore, in the case of Sclerotium rolfsii, compounds where the hydroxyl functions of the aglycon remained unchanged (medicagenic acid and its dimethyl ester) or could be enzymically released (3-0-β-D-glucoside of medicagenic acid dimethyl ester) were significantly more active than compounds where these functions were modified by acetylation or methylation. Selective 2-0-methylation of medicagenic acid and comparison of the antimycotic activity of the resulting derivative against S. rolfsii to that of other derivatives suggests that a potential free hydroxyl at position 3 is essential to antimycotic activity.  相似文献

The effects of five burning treatments of alfalfa (Medicago sativa L.) stubble combined with two insecticide treatments on populations of alfalfa weevil, Hypera postica (Gyllenhal), and the pea aphid, Acyrthosiphon pisum (Harris) were evaluated over an eight year period. The effects of the burn treatments were dependent on insect species, time of burn and year. Immature alfalfa weevil population levels were significantly reduced by the burn-every-autumn treatment in 3 of the 8 years, in 2 out of 8 years by the burn-e very-spring treatment, and by the alternate-year-burn treatment in 1 out of 4 years. Burning had little or no effect on pea aphid populations.  相似文献

Aphids are pests of chrysanthemum that employ plant volatiles to select host plants and ingest cell contents to probe host quality before engaging in prolonged feeding and reproduction. Changes in volatile and nonvolatile metabolite profiles can disrupt aphid–plant interactions and provide new methods of pest control. Chrysanthemol synthase (CHS) from Tanacetum cinerariifolium represents the first committed step in the biosynthesis of pyrethrin ester insecticides, but no biological role for the chrysanthemol product alone has yet been documented. In this study, the TcCHS gene was over‐expressed in Chrysanthemum morifolium and resulted in both the emission of volatile chrysanthemol (ca. 47 pmol/h/gFW) and accumulation of a chrysanthemol glycoside derivative, identified by NMR as chrysanthemyl‐6‐O‐malonyl‐β‐D‐glucopyranoside (ca. 1.1 mM), with no detrimental phenotypic effects. Dual‐choice assays separately assaying these compounds in pure form and as part of the headspace and extract demonstrated independent bioactivity of both components against the cotton aphid (Aphis gossypii). Performance assays showed that the TcCHS plants significantly reduced aphid reproduction, consistent with disturbance of aphid probing activities on these plants as revealed by electropenetrogram (EPG) studies. In open‐field trials, aphid population development was very strongly impaired demonstrating the robustness and high impact of the trait. The results suggest that expression of the TcCHS gene induces a dual defence system, with both repellence by chrysanthemol odour and deterrence by its nonvolatile glycoside, introducing a promising new option for engineering aphid control into plants.  相似文献

Three new oleanane-type saponins, leptocarposide B-D (13), were isolated from the whole plant of Ludwigia leptocarpa (Nutt.) Hara, together with ten known compounds 4–13.The structures of these compounds were determined by interpretation of their spectral data, mainly HR-TOFESIMS, 1D-NMR (1H, 13C) and 2D-NMR (1H–1H COSY, HSQC, HMBC, and NOESY), and by comparison with the literature data. The structures of the new compounds were established as 28-O-β-d-xylopyranosyl-(1  4)-α-l-rhamnopyranosyl-(1  2)-[α-l-arabinopyranosyl-(1  3)]-4-O-(3′-hydroxybutanoyloxy-3-hydroxybutanoyloxy)-β-d-fucopyranosyl zanhic acid (1); 3-O-β-d-glucopyranosyl-28-O-β-d-xylopyranosyl-(1  4)-α-l-rhamnopyranosyl-(1  2)-4-O-(3′-hydroxybutanoyloxy-3-hydroxybutanoyloxy)-β-d-fucopyranosyl medicagenic acid (2); 3-O-β-d-glucopyranosyl-(1  4)-β-d-glucopyranosyl-28-O-β-d-xylopyranosyl-(1  4)-α-l-rhamnopyranosyl-(1  2)-[α-l- arabinopyranosyl-(1  3)]-4-O-(3′-hydroxybutanoyloxy-3-hydroxybutanoyloxy)-β-d-fucopyranosyl zanhic acid (3).  相似文献

Eight saponins were isolated from alfalfa roots (Medicago sativa). The sensitivity of Trichoderma viride to the saponin varied with the individual saponin isolate. Seven isolates appeared to contain the aglycone, medicagenic acid, and while the other did not, it inhibited the growth of the fungus at higher concentrations than the other isolates. One pair and a triplet of saponins with divergent Rfs evoked near identical biological responses suggesting structural similarity toxic to T. viride.  相似文献

Bathyplectes curculionis (Thomson) is an introduced natural enemy of the alfalfa weevil in North America. The wasp requires carbohydrate foods as an adult. Adult wasps have increased longevity and fecundity when provided access to pea aphid, Acyrthosiphon pisum (Harris), honeydew in the laboratory, and adults respond positively to the presence of pea aphids in alfalfa fields. However, it is unknown how these wasps find aphid honeydew in the field. In a series of Y-tube olfactometer experiments, we evaluated the response of naïve and experienced adult female B. curculionis to odors from pea aphids, alfalfa, and pea aphids on alfalfa. Naïve adult females did not respond positively to pea aphid odor even when hungry. But adult females were able to learn aphid odor, and the mechanism of learning appears to be associative rather than by sensitization. Naïve females also showed no preference for alfalfa odor but learned alfalfa odor through sensitization. The wasps did not distinguish between alfalfa with aphids and alfalfa without aphids, even after exposure to aphids or alfalfa with aphids. However, they preferred pea aphid odor to alfalfa odor after a feeding experience in the presence of pea aphid odors. But after exposure to mixed odors of aphids and alfalfa while feeding, B. curculionis females preferred the odor of alfalfa to the odor of pea aphids. These results suggest that alfalfa odors mask or override aphid odors when aphids are associated with alfalfa (as happens naturally), thus interfering with the wasp's ability to respond to learned aphid odors. Therefore, although the wasps are capable of learning to find pea aphids and their honeydew in a simplified laboratory setting, it appears unlikely that they do so in the field.  相似文献

The spotted alfalfa aphid (SAA), Therioaphis trifolii maculata (Buckton), causes a characteristic veinal chlorosis and necrosis in the growing tips of susceptible cultivars of alfalfa. The pea aphid, Acyrthosiphon pisum (Harris), causes general degenerative changes in alfalfa but no specific, local symptoms. Biochemical and electrophoretic analyses detected similar enzymes in the ejected saliva of either species: pectin methylesterase, endopolygalacturonase and at least three isozymes of a copper dependent oxido-reductase that showed both catechol oxidase and peroxidase activity. Pectinase and catechol oxidase activities per unit of soluble protein were much greater in the saliva of pea aphid compared with that of SAA. The isozymes of the oxidase from SAA were roughly half the molecular weights of the corresponding isozymes from pea aphid, however, and radiotracer studies showed that soluble secretions injected into alfalfa by SAA travelled to growing tips considerably faster than the secretions of pea aphid. It is suggested that differences in the lesions caused by these aphids may be due to reaction kinetics rather than specific salivary toxins; that the rate of arrival of salivary components, possibly the oxidases, at phloem unloading sites may determine whether the plant's local defensive system is able to repress the immediate challenge or undergoes a run-away reaction leading to necrosis.  相似文献

CUF 101, a resistant cultivar of alfalfa, was exposed to 15 clones of Acyrthosiphon pisum Harris collected from alfalfa fields in three regions of France (east, south, central west) to determine whether the level of resistance varied across the different clones. The survival of alfalfa seedlings infested at the cotyledon stage was assessed using a standardized method. Although no difference in seedling mortality was detected between clones grouped by region, there was a significant variation among the 15 pea aphid clones. In particular, two clones of southern origin were more aggressive. In addition, the different pea aphid clones were characterized using allozyme and RAPD-PCR markers. Among the 15 clones, seven allozyme genotypes (plus one when adding colour polymorphism) and 12 RAPD-PCR genotypes were distinguished. The two southern clones differing by their aggressiveness on the resistant alfalfa belonged to the same allozyme and RAPD genotype which was distinct from the other pea aphid clones. Our results reinforce the need to take into account aphid genetic diversity in breeding programmes for resistance in cultivated plants.  相似文献

  • 1 We tested switching behaviour in four species of aphidiid parasitoids, using a two-aphid experimental system consisting of second-instar nymphs of pea aphid (Acyrthosiphon pisum (Harris)) and alfalfa aphid (Macrosiphum creelii Davis) feeding on broad beans in the laboratory.
  • 2 Aphidius ervi Haliday, A.pisivorus Smith, A.smithi Sharma & Subba Rao, and Pram pequodorum Viereck showed an innate preference for pea aphid when both host species were provided in equal numbers.
  • 3 Wasps encountered both aphid species equally but differed in their acceptance of alfalfa aphid. Females of A.pisivorus and P.pequodorum accepted alfalfa aphids when few pea aphids were available, but A. smithi always concentrated attacks on pea aphid. Aphidius ervi super-parasitized an increasing proportion of pea aphids as their availability declined.
  • 4 Switching to the alfalfa aphid occurred in A.ervi and P.pequodorum (but not in A.pisivorus and A.smithi) under the condition of a 1:3 ratio of pea aphids:alfalfa aphids. Wasps did not switch when more pea aphids than alfalfa aphids were provided (3:1 ratio).
  • 5 Alfalfa aphids were more likely than pea aphids to escape from parasitoid attack.
  • 6 Switching to the most abundant host may not be adaptive in these four species of aphid parasitoids. A foraging wasp incurs a potentially higher cost in lost opportunity time when attacking (and failing to oviposit in) alfalfa aphids. In addition, alfalfa aphids may have lower host quality than pea aphids, a difference that could influence offspring fitness.

The occurrence of a secondary bacterial symbiont (PASS) of pea aphid, Acyrthosiphon pisum (Harris), was detected by polymerase chain reaction (PCR) with specific nucleotide primers based on PASS 16S rDNA nucleotide sequences from over 80% (50/57) of clones of pea aphid collected from widely separated locations in California. PASS was also detected by PCR in both red and green phenotypes of rose aphid, Macrosiphum rosae (L.), but not in six other species of aphids examined, including blue alfalfa aphid (A. kondoi Shinji). The nucleotide sequences of the PCR-amplified, partial 16S rDNAs (1060 bp) from pea aphid and rose aphid were identical and 99.9% similar to the published 16S rDNA of PASS. PASS and a recently described new rickettsia of pea aphid (PAR) were transmitted by needle injection of hemolymph from positive pea aphid clones into negative clones and into blue alfalfa aphids. 研究了蚜虫危害胁迫后不同抗蚜性苜蓿品种叶片内丙二醛含量及防御性酶活性的动态变化。结果表明:在蚜虫刺吸诱导的过程中,高感品种的MDA含量始终高于高抗品种,并且高感和高抗品种均保持上升的趋势;高感品种的SOD、POD和PAL活性始终低于高抗品种,其中高感和高抗品种的SOD和POD活性均表现先上升后下降的趋势,而PAL活性上升到高峰后均趋于稳定;CAT活性在高感和高抗品种间表现为交替的上升下降;高抗品种的PPO活性前期低于高感品种,而后期高于高感品种。由此可见,在蚜虫危害胁迫下,高感和高抗品种间MDA、SOD、POD、PAL和PPO活性的变化与苜蓿的抗蚜性密切相关,均可作为苜蓿抗蚜性鉴定的生理指标,而CAT活性变化与苜蓿抗蚜性的联系有待进一步研究。  相似文献
