The freezing tolerance of many plants, such as pea (Pisum sativum),is increased by exposure to low temperature or abscisic acidtreatment, although the physiological basis of this phenomenonis poorly understood. The freezing tolerance of pea shoot tips,root tips, and epicotyl tissue was tested after cold acclimationat 2C, dehydration/rehydration, applications of 10–4M abscisic acid (ABA), and deacclimation at 25C. Tests wereconducted using the cultivar ‘Alaska’, an ABA-deficientmutant ‘wil’, and its ‘wildtype’. Freezinginjury was determined graphically as the temperature that caused50% injury (T50) from electrical conductivity. Endogenous ABAwas measured using an indirect enzyme-linked immunosorbant assay,and novel proteins were detected using 2-dimensional polyacrylamidegel electrophoresis. The maximum decrease in T50 for root tissuewas 1C for all genotypes, regardless of treatment. For ‘Alaska’shoot tips and epicotyl tissue, exogenous ABA increased thefreezing tolerance by –1.5 to –4.0C, while coldtreatment increased the freezing tolerance by –7.5 to–14.8C. Cold treatment increased the freezing toleranceof shoot tips by –9 and –15C for ‘wil’and ‘wild-type’, respectively. Cold acclimationincreased endogenous ABA concentrations in ‘Alaska’shoot tips and epicotyls 3- to 4-fold. Immunogold labeling increasednoticeably in the nucleus and cytoplasm of the epicotyl after7 d at 2C and was greatest after 30 d at the time of maximumfreezing tolerance and soluble ABA concentration. Simulated swards of each of two selection lines of Lolium perennecv. S23 with ‘fast’ and ‘slow’ ratesof ‘mature tissue’ respiration were establishedin growth rooms at 20/15 °C day/night temperatures and studiedover four successive regrowth periods of 46, 30, 26 and 53 daysduration. The ‘slow’ line outyielded the ‘fast’,both in harvestable shoot (above a 5 cm cut) and in root andstubble. Its advantage increased over successive regrowth periodsto 23 per cent (total biomass). Gas analysis measurements onthe entire communities (including roots), during the final regrowthperiod, showed that the ‘slow’ line had a 22–34per cent lower rate of dark respiration per unit dry weight.This enabled it to maintain its greater mass of tissue for thesame cost in terms of CO2 efflux per unit ground area. Halfthe extra dry weight produced by the ‘slow’ line,relative to the ‘fast’, could be attributed to itsmore economic use of carbon. The rest could be traced to a 25per cent greater tiller number which enabled the ‘slow’line to expand leaf area faster (though not at a greater rateper tiller), intercept more light and fix more carbon, earlyin the regrowth period. Honeylocust (Gleditsia triacanthos L.) seedlings were grownin solution culture at pH 4.0) with 50, 150, 600 and 1500 mmolm–3 aluminium. All levels of aiuminium reduced the sizeand weight of roots, shoots and leaves with the exception ofroot elongation at 50 mmol m–3 Al. Aluminium content ofroots was 50 to 100 times that of shoots. With increasing concentrationof aluminium, aluminium content of leaves and roots increasedexponentially while a linear increase was observed for stems.The nutrient content of seedlings was improved in 50 mmol m–3where increases in shoot calcium, magnesium, and phosphorusconcentrations were observed. Aluminium concentrations greaterthan 50 mmol m–3 reduced shoot nutrient content. Presenceof aluminium increased the root phosphorus and calcium levelsbut had no effect on potassium and magnesium concentrations.Results show that honeylocust is an aluminium sensitive treespecies whose growth may be reduced by high soil Al levels. Fiskeby V soya bean was grown from seed germination to seedmaturation with two contrasting patterns of nitrogen metabolism:either wholly dependent on dinitrogen fixation, or with an abundantsupply of nitrate nitrogen, but lacking root nodules. The carbonand nitrogen economies of the plants were assessed at frequentintervals by measurements of photosynthesis, shoot and rootrespiration, and organic and inorganic nitrogen contents. Plantsfixing atmospheric nitrogen assimilated only 25–30 percent as much nitrogen as equivalent plants given nitrate nitrogen:c. 40 per cent of the nitrogen of ‘nitrate’ plantswas assimilated after dinitrogen fixation had ceased in ‘nodulated’plants. The rates of photosynthesis and respiration of the shootsof soya bean were not markedly affected by source of nitrogen;in contrast, the roots of ‘nodulated’ plants respiredtwice as rapidly during intense dinitrogen fixation as thoseof ‘nitrate’ plants. The magnitude of this respiratoryburden was calculated to increase the daily whole-plant respiratory loss of assimilate by 10–15 per cent over thatof plants receiving abundant nitrate. It is concluded that ‘nodulated’plants grew more slowly than ‘nitrate’ plants inthese experiments for at least two reasons: firstly, the symbioticassociation fixed insufficient nitrogen for optimum growth and,secondly, the assimila tion of the nitrogen which was fixedin the root nodules was more energy-demanding in terms of assimilatethan that of plants which assimilated nitrogen by reducing nitratein their leaves.  相似文献

Regulation of in vitro shoot and root formation and the histologicalorigin of newly formed shoots was studied in the apple cultivarÅkerö. Both composition of mineral elements and benzylaminopurine(BAP) concentration affected shoot multiplication. Similar numbersof shoots were obtained with Lepoivre and MS medium after twosucceeding subcultures, but Murashige and Skoog medium was preferabledue to production of longer shoots. The optimum BAP concentrationwax around 8.8 µM. Higher concentrations caused vitrifiedshoots. The rooting ability increased with numbers of subcultures.Also the concentration of indol-3-yl butyric acid (IBA) affectedrooting. A strong interaction between numbers of subculturesand IBA concentration was obtained. After insufficient numbersof subcultures, when shoots were still difficult to root, increasingIBA concentration exerted little effect on rooting. When shootshad reached an ‘easy-to-root condition’ root initiationdepended on IBA concentration, showing an optimum at 2.5 µM.Supraoptimal IBA concentrations delayed root initiation. Dark treatment of shoots during the root-initiation phase increasedrooting ability. The most positive effect was obtained at suboptimalIBA concentrations. Anatomical studies revealed both axillaryand adventitious shoots. Two kinds of adventitious structureswere demonstrated. In two experiments, the functioning and metabolism of nodulesof white clover, following a defoliation which removed abouthalf the shoot tissue, were compared with those of undefoliatedplants. In one experiment, the specific respiration rates of nodulesfrom undefoliated plants varied between 1160 and 1830 µmolCO2 g–1h–1, of which nodule ‘growth and maintenance’accounted for 22 ± 2 per cent, or 27 ± 3.6 percent, according to method of calculation. Defoliation reducedspecific nodule respiration and nodule ‘growth and maintenance’respiration by 60–70 per cent, and rate of N2 fixationby a similar proportion. The original rate of nodule metabolismwas re-established after about 5 d of regrowth; during regrowthnodule respiration was quantitatively related to rate of N2,fixation: 9.1 µmol CO2 µmol–1N2. With the possible exception of nodules examined 24 h after defoliation,the efficiency of energy utilization in nitrogenase functioningin both experiments was the same in defoliated and undefoliatedplants: 2.0±0.1 µmol CO2 µmol–1 C2H4;similarly, there was no change in the efficiency of nitrogenasefunctioning as rate of N2 fixation increased with plant growthfrom 1 to 22 µmol N2 per plant h–1. Exposure of nodulated white clover root systems to a 10 percent acetylene gas mixture resulted in a sharp peak in rateof ethylene production after 1.5–2.5 min; subsequently,rate of ethylene production declined rapidly before stabilisingafter 0.5–1 h at a rate about 50 per cent of that initiallyobserved. Regression of ‘peak’ rate of ethyleneproduction on rate of N2 fixation indicated a value of 2.9 µmolC2H4 µmol–1 N2, for rates of N2 fixation between1 and 22 µmol N2 per plant h–1. The partition of 14C labelled current assimilates to root insimulated swards of Lolium perenne cv. S24 was measured duringthe transition from vegetative growth in autumn to reproductivegrowth in spring under close to natural conditions of lightand temperature. Assimilate partitioning was also measured in‘established’ swards cut three times during thegrowing season and in vegetative ‘seedling’ swardsgrowing in autumn and in spring. All measurements were madewhen the swards had achieved more than 90 per cent light interception,and all swards were abundantly supplied with water and mineralnutrients. During autumn there was a gradual decrease in the proportionof assimilates partitioned to the roots in both the ‘established’and the ‘seedling’ swards. In the established swards,partition to roots was low over winter, increased during earlyspring, but decreased dramatically, later in the spring, whenstem elongation began. In contrast, in the unvernalized vegatativeseedling swards in spring, partition to roots remained high. The seasonal pattern of assimilate partitioning is consideredin relation to changes in the natural environment and the rateat which the crop fixed carbon in photosynthesis. A decreasein the proportion of assimilates partitioned to roots duringlate spring was significant in increasing the production ofshoot at that time but seasonal differences in partition contributedvery little to the marked differences in shoot growth betweenthe spring and autumn crop. Kanninen, M. 1985. Shoot elongation in Scots pine: Diurnal variationsand response to temperature.—J. exp. Bot. 36: 1760–1770. Time series analysis is used to study the diurnal variationsin the rate of shoot elongation of Scots pine (Pinus sylvestrisL.) seedlings, and its response to temperature. The shoot elongation of three 5-year-old Scots pine seedlingswas measured at 2 h intervals over a 12 d period in June 1979.Ambient temperature, measured with a thermocouple, was continuouslyrecorded during the same period. The measurements were carriedout in the field. Time series analysis of the data was carried out by estimatedauto-correlation and partial auto-correlation functions. A multiplicative‘seasonal’ model was applied to both the input andoutput series to ‘prewhiten’ the data set. Serialcross-correlation analysis of the prewhitened series was usedto identify the structure of the transfer function model betweentemperature and growth rate. There was a phase shift between the air temperature and shootextension. The cross-correlation function peaked at 2 h lagvalue. The influence of temperature on the growth rate was adequatelydescribed by a first-order transfer function model. The characteristicfeature of the model was a time constant of 3.3 h and a basetemperature of 6.3 °C for the response of shoot elongationto temperature. Young plants of two selection lines of Lolium perenne cv. S23with ‘fast’ and ‘slow’ rates of ‘maturetissue’ respiration were individually grown from seed,together with plants of S23, their common parent, in 9.2 cmpots in a controlled environment at 20/15 °C day/night temperatures. No significant differences were found between the genotypesin leaf extension and tiller production during this early stageof their growth. They did differ however, by an average of 26%,in the rate of dark respiration of fully expanded leaf laminae.The use of a simple model demonstrated that such a differencein respiration could alone account for the different rates ofdry matter production shown by the selection lines when grownas young crops from seed. Possible penalties of ‘slow’respiration are also considered. A series of experiments was conducted to assess net CO2assimilationand growth responses to waterlogging of grafted and seedlingtrees in the genus Annona. Seedlings of A. glabra, A. muricataandA. squamosa L., and scions of ‘Gefner’ atemoya(A. squamosaxA. cherimola Mill.), ‘49-11’ (‘Gefner’atemoyaxA. reticulata L.), ‘4-5’ (‘Priestley’atemoyaxA. reticulata), A. reticulata grafted onto either A.glabra, A. reticulata orA. squamosa rootstocks were floodedfor up to 60 d. Soil anaerobiosis occurred on the third dayof flooding. Seedlings ofA. glabra and A. muricata, and thescions ‘49-11’, ‘Gefner’ atemoya, andA. reticulata grafted onto A. glabra rootstock were consideredflood tolerant based on their ability to survive and grow inflooded conditions. Scions of the normally flood-sensitive A.reticulata, ‘Gefner’ atemoya, and ‘49-11’tolerated root waterlogging when grafted onto the flood-tolerantspecies, A. glabra. In contrast, flooding of A. squamosa seedlingsand rootstocks, and A. reticulata rootstocks greatly reducedgrowth and net CO2assimilation rates, and resulted in 20–80%tree mortality. Stem anatomical responses to long-term flooding(12 continuous months) were assessed in seedlings of A. glabraand A. muricata, and trees of ‘49-11’ grafted ontoA. glabra. Flooded trees developed hypertrophied stem lenticels,particularly in A. glabra, and enlarged xylem cells resultingin thicker stems with reduced xylem density. Flooding did notincrease air spaces in pre-existing xylem near the pith or inxylem tissue that was formed during flooding. Thus, flood tolerancedid not involve aerenchyma formation in the stem. Extracts of small and mature-size lupin pods yielded four substancesaffecting the growth of wheat-coleoptile sections: one acidpromotor (A), two acid inhibitors(B and X), and one neutralinhibitor(Y). Inhibitor B was extremely active, however, coleoptile sectionsshowed no signs of toxic effects; they resumed growth at a rapidrate after rinsing them and adding ß-indolylaceticand (IAA) to the medium. 1 µg of IAA was required to counteractthe effect of ‘B’ extracted from 230 mg. Of tissue.On an equal fresh weight basis the inhibiting action of ‘B’in lupin pods was 500–1,500 times more potent than thatof ‘inhibitor ß’ in etiolated pea seedlings. Small pods of plants infected with pea-mosaic virus yielded3 times the amount of ‘A’ of healthy plants (equivalentto 1 µg. IAA 0.3 µg. IAA per 25 g. of tissue respectively),and approximately the amount of ‘B’. Mature podsof virus-infected plants again yielded more‘A’,but also 2? times more ‘B’ than pods of healthyplants. Healthy pods yielded more ‘A’ than virus-infectedpods, and there was no difference in ‘X’. A lupin abscission test was developed and the effects of proximaland distal application of -naphthyl acetic acid (NAA) are presented,and discussed with respect to results of other abscission tests. ‘A’ accelerated abscission when applied proximally,and delayed or prevented it when applied distally. ‘B’strongly accelerated abscission when applied in either way.A possible mechanism explaining the abscission-inducing effectof developing pods on later flowers is discussed in terms ofthe substances ‘A’ and ‘B’. Root border cells are living cells that surround root apices of most plant species and are involved in production of root exudates. We tested predictions of the hypothesis that they participate in detection and avoidance of aluminum (Al) toxicity by comparing responses of two snapbean (Phaseolus vulgaris) cultivars (cv Dade and cv Romano) known to differ in Al resistance at the whole-root level. Root border cells of these cultivars were killed by excess Al in agarose gels or in simple salt solutions. Percent viability of Al-sensitive cv Romano border cells exposed in situ for 96 h to 200 microM total Al in an agarose gel was significantly less than that of cv Dade border cells; similarly, relative viability of harvested cv Romano border cells was significantly less than that of cv Dade cells after 24 h in 25 microM total Al in a simple salt solution. These results indicate that Al-resistance mechanisms that operate at the level of whole roots also operate at the cellular level in border cells. Al induced a thicker mucilage layer around detached border cells of both cultivars. Cultivar Dade border cells produced a thicker mucilage layer in response to 25 microM Al compared with that of cv Romano cells after 8 h of treatment and this phenomenon preceded that of observed cultivar differences in relative cell viability. Release of an Al-binding mucilage by border cells could play a role in protecting root tips from Al-induced cellular damage.  相似文献

Photosynthesis, respiration and growth of two cultivars of carrotwith contrasting ratios of shoot: storage root weight at maturity,were compared during initiation of the storage root at 20 °C.Partition of assimilate between shoot, roots and respirationshowed no varietal differences but distribution between storageand fibrous roots was different from the time that the storageroot could be morphologically identified. For both cultivarsover the period investigated, approximately 64% of net photosynthesiswas partitioned to the shoot with 5% lost as respiration duringthe dark and 59% used in growth. Of that exported to the rootsystem (36%), 19% (of net photosynthesis) was used in growthand 17% was lost in respiration. In the cultivar with greatershoot: storage root ratio at maturity, 4.6% was allocated tothe storage root in contrast to 7.5% in the cultivar with alesser shoot: storage ratio at maturity. It is concluded thatgreater dry matter accumulation in the storage root of the lattercultivar does not result from transient differences in respiratoryloss and is not evident in shoot to total root dry matter distributionover this period. This review compares new developmental models on flowering andother vascular plants with evolutionary hypotheses formulatedby Agnes Arber (1879–1960) and like-minded botanists.Special emphasis is laid on philosophical basics such as perspectivism,pluralism about evolutionary modelling, continuum way of thinking,and fuzzy logic. Arber's perspective is best labelled as F uzzyA rberian M orphology (FAM Approach). Its proponents (‘FAMmers’)treat structural categories (e.g. ‘roots’, ‘shoots’,‘stems’, ‘leaves’, ‘stipules’)in vascular plants as concepts with fuzzy borderlines allowingintermediates (including transitional forms, developmental mosaics).The FAM Approach complements Cla ssical Plant M orphology (ClaMApproach), which is the traditional approach in botany. ClaMproponents (‘ClaMmers’) postulate that the structuralcategories of vascular plants are regarded as concepts withclear-cut borderlines and without intermediates. However, duringthe evolution of vascular plants, the root-shoot distinctionand the stem-leaf distinction have become blurred several timesdue to developmental changes, resulting in organs with uniquecombinations of features. This happened, for example, in thebladderworts (Utricularia, Lentibulariaceae). When focusingon the ‘leaf’, the FAM Approach is identical toArber's ‘partial-shoot theory of the leaf’ and Sinha's‘leaf shoot continuum model’. A compound leaf canrepeat the developmental pathway of the whole shoot, at leastto some degree. For example, compound leaves of Chisocheton(Meliaceae)with indeterminate apical growth and three-dimensional branchingmay be seen as developmental mosaics sharing some growth processeswith whole shoots! We focus here on the FAM Approach becausethis perspective is especially promising for developmental geneticistsstudying flowering and other vascular plants. Background and Aims: Plant growth responses to the rare earth elements lanthanum(La) and cerium (Ce) have been reported, but little is knownabout the effects of these two elements on plant mineral nutrition. Methods: Corn (Zea mays ‘Hycorn 82’) and mungbean (Vignaradiata ‘Berken’) were grown in continuous flowingnutrient solutions containing 0, 0·2, 1·0 and5·0 µM La or Ce. At harvest plants were dividedinto roots and shoots, dried, weighed and analysed for macro-and micronutrients, as well as for La and Ce. Key Results: La and Ce did not increase the growth of corn or mungbean. Thedry weight of corn shoots was decreased by 32 % in the presenceof 5·0 µM Ce; the other La and Ce concentrationshad no effect. La and Ce concentrations of 0·9 and 5·0µM decreased the shoot dry weight of mungbean by 75 or95 %, the two elements having closely similar effects. Decreasesin the uptake of Ca, Na, Zn and Mn by corn were observed withincreases in solution La and Ce. For mungbean, the uptake ratesof all measured elements decreased with increases in solutionLa and Ce. The concentrations of La and Ce in the roots of bothspecies were higher than in the shoots and increased stronglywith increasing concentrations of La or Ce in solution. TheLa and Ce concentrations in mungbean shoots were always higherthan in corn shoots. Conclusions: La and Ce did not enhance the growth of corn or mungbean, butdecreased the growth, root function and consequently the nutritionalstatus of mungbean at concentrations >0·2 µMin solution. The purpose of this study was to determine how a plant's responseto colonization by mycorrhizal fungi relates to its abilityto acquire and utilize phosphorus for growth and reproduction.Two tomato genotypes previously found to be either responsive(‘LA1709’) or unresponsive (‘large cherry’)to mycorrhizal colonization during early vegetative growth wereexamined in detail. Plants were grown at four levels of addedphosphate or with mycorrhizal inoculum. Vegetative and reproductivegrowth, phosphorus uptake and root length density were measuredduring the course of plant development. Mycorrhizal symbiosissignificantly increased above-ground dry mass, root length,phosphorus content and yield under low phosphorus conditionsin ‘LA1709’, while it had less effect on these characteristicsin ‘large cherry’. When uninfected, however, ‘LA1709’grew and reproduced poorly unless high amounts of phosphoruswere added to the soil, while ‘large cherry’ grewwell under very low phosphorus conditions. This was because‘large cherry’ had significantly higher root lengthdensities than ‘LA1709’, enabling plants from thisgenotype to explore more soil volume and acquire greater amountsof phosphorus when grown without mycorrhizal fungi in low phosphorussoil. ‘Large cherry’ also had higher phosphorususe efficiency and allocated a greater proportion of phosphorusto reproduction when uninfected than ‘LA1709’. Seeds of Sorghum bicolor (L.) Moench. were germinated on moistfilter paper for 6 d, before the seedlings were transferredto pots containing 500 µmol l-1 Ca(NO3)2 for 2 d. Theseedlings were then treated with 0 or 100 µmol l-1 Alin factorial combination with 0, 1400 or 2800 µmol l-1Si for 8 d. The background solution used throughout was 500µmol l-1 Ca(NO3)2. Aluminium treatment reduced root growthand caused a significant increase in shoot/root ratio. The presenceof silica in the solution significantly ameliorated the effectsof aluminium on root growth. Three treatment were selected for a microanalytical investigationof the basal region of the root: 2800 µmol l-1 Si only;100 µmol l-1 Al only; and a combination of the two. Inthe 2800 µmol l-1 treatment silica was deposited in theendodermis, with the greatest accumulation being in the innertangential wall (ITW). When plants were treated with 100 µmoll-1 Al only, aluminium concentration was highest in the outertangential wall (OTW) of the epidermis. The element was presentin the hypodermal walls and OTW of the endodermis and was notdetectable in the stele. With both 2800 µmol l-1 Si and100 µmol l-1 Al in the nutrient solution the two biomineralizationsites were the ITW of the endodermis, where silicon was themajor element deposited, and atypically in the OTW of the epidermiswhere both aluminium and silicon were present. The effects of nitrogen and plant growth regulators (stem shorteners)on root and shoot characteristics associated with lodging resistancewere investigated in two winter wheat (Triticum aestivum L.)cultivars of contrasting lodging resistance: the susceptibleGalahad and the resistant Hereward. The morphology and mechanicalstrength of the stems and anchorage systems grown at two levelsof nitrogen and with or without growth regulators were measuredand related to the incidence of lodging recorded in a fieldtrial. In both cultivars high levels of nitrogen increased theheight of the stem, thereby increasing the ‘self-weight’moment transmitted into the ground and weakened both the stemsand the anchorage coronal roots. As a result, the anchoragestrength was also reduced, plants failing in the root systemin simulated lodging tests. Growth regulators, in contrast,had little effect on the bending strength of the shoots androot systems, but reduced plant height so that the over turningmoments generated by the weight of the shoot were less. Therewere also differences between cultivars: Galahad plants hadweaker anchorage due to the smaller number and lower strengthof the coronal roots. The morphological and mechanical measureswere used to calculate a safety factor against both stem androot lodging. Five factors were found to influence the safetyfactors, these were: cultivar type, the type of lodging, therate of nitrogen and growth regulator application, and time,being lowest in Galahad plants at high levels of nitrogen andwithout growth regulators and at grain filling when the earswere heaviest. This was consistent with the observed patternof lodging: root lodging occurred at grain filling and onlyin Galahad which had been treated with high nitrogen rates,most strongly in plants without growth regulators.
