The interaction of two symbionts,Rhizobium (a bacterium) andGigaspora calospora (a VAM fungus) was studied inPisum sativum (a nodulating legume).Pisum sativum plants inoculated with VA mycorrhizal fungi andRhizobium singly and in combination responded favourably as compared to uninoculated control. Characteristically dual inoculation exerted a higher beneficial effect on shoot and root dry mass, phosphorus and nitrogen uptake, nodule formation and degree of mycorrhizal infection than either inoculum alone.

The growth response of Hevea brasiliensis to vesicular-arbuscular mycorrhizal (VAM) fungi inoculation was assessed in two field nursery sites containing indigenous mycorrhizal fungi (IMF). Seedling rootstocks were inoculated with mixed VAM-fungal species in a factorial combination with phosphorus (P) fertilizer application, and planted in randomised blocks on sandy (site 1) and clayey (site 2) soils. Plants were harvested after 26 weeks for measurements of shoot dry weight (DW), stem diameter, height, mycorrhizal root colonization and leaf nutrient contents. At site 1, VAM increased shoot DW, stem diameter and plant height only in treatments without P applied. Increases in shoot DW due to VAM were 70% greater than the uninoculated controls although this was reduced to 5% when P was applied. At site 2, VAM inoculation also increased shoot DW and stem diameter but the magnitude of the increases was smaller. Shoot DW response due to VAM was only 29%. At this second site, applying phosphate to uninoculated plants did not increase shoot yields further. Leaf concentrations of all nutrients were unaffected by VAM at both sites, except for copper (Cu) which was increased by VAM in treatments where P was not applied. However, leaf contents of P, potassium (K), magnesium (Mg) and Cu were increased by VAM at site 1, and of leaf nitrogen (N) and K at site 2. These experiments demonstrate that VAM-fungi could be introduced into field nursery sites to improve growth and P uptake by H. brasiliensis. The relevance of VAM-fungi to H. brasiliensis seedling rootstock development and the influence of IMF in determining field responses is discussed.

Inoculation of finger millet (Eleusine coracana Gaertn.) plants with one of six different vesicular, arbuscular, mycorrhizal (VAM) fungi increased plant biomass, height, leaf area and absolute growth rate; however, effectiveness of the various VAM fungi varied significantly. Maximum root colonization and mycorrhizal efficacy was observed with plants inoculated with Glomus caledonicum. Among five host genotypes tested for mycorrhizal dependency against G. caledonicum, genotype HR-374 gave the highest plant biomass, mycorrhizal efficacy and root colonization, the inoculation resulting in increased mineral (phosphate, nitrogen, Zn2+ and Cu2+) content and uptake in shoots.

The influence of three inoculum densities of Glomus caledonius and G. epigaeus and two temperature/light intensity conditions was investigated on Trifolium repens. The significance of inoculation was compared to the significance of naturally occurring vesicular–arbuscular mycorrhizal (VAM) fungi and to application of soluble phosphate fertilizer. Increasing density of inoculum and the highest temperature/light intensity condition tested increased VAM infection, whereas only small differences were found between efficiency of the two introduced VAM fungi. The presence of naturally occurring VAM fungi proved as efficient in establishing infection as the most successful inoculations. Some interactions among the investigated parameters were found for several recordings. The increase in VAM infection was followed by an increase in number of nodules; in uptake of phosphorus, nitrogen, zinc, and copper; and in growth of roots and shoots. The calculated inflow of phosphate, zinc, and copper into roots was not associated with inoculum density, VAM species or temperature/light conditions. Compared to an uninoculated control without application of phosphate, inoculation with the highest spore density increased (after 18 weeks growth) the dry weight of shoot 52 fold and 7 fold for G. caledonius , and 121 fold and 9 fold for G. epigaeus at low and high temperature/light conditions, respectively. It was also found that VAM increased weight per nodule 52% when roots with no or sparse VAM infection were compared to roots with low to maximal VAM infection and 98% when roots with low VAM infection were excluded. Application of phosphate fertilizer enhanced nodulation and growth of non–mycorrhizal plants to a level similar to that of the most heavily VAM infected plants.

The response ofCicer arietinum to inoculation withGlomus versiforme under field conditions was investigated in a phosphorus deficient sandy loam soil. Inoculation with the mycorrhizal fungusGlomus versiforme increased the rate of VAM development in chickpea. The weight of nodules and the number of nodules per plant were higher in inoculated than in uninoculated plants. The phosphorus content of the shoots and its total uptake, were increased by either the application of single super-phosphate, or by inoculation withG. versiforme. Inoculation increased shoot dry weights and grain yields by 12% and 25% respectively, as compared with the 33% and 60% increases respectively produced by P-treated plants.

The growth and mineral nutrition responses were evaluated of three tropical legumes, cowpea (Vigna unguiculata L. cv Kuromame), pigeonpea [Cajanus cajan L. (Millsp.) cv ICPL 86009] and groundnut (Arachis hypogaea cv Nakateyutaka) inoculated with two different species of VAM fungi, Glomus sp. (Glomus etunicatum-like species) and Gigaspora margarita, and grown in Andosols with different fertilities [Bray II-P: topsoil (72 ppm), subsoil (<0.1 ppm)]. Percent fungal root colonization was high in cowpea and groundnut but relatively low in pigeonpea in both soil types. Despite the low rate of root infection, significant growth responses were produced, especially in the inoculated pigeonpea plant. In all legumes, shoot dry matter production was favoured by the inoculations. Increases in shoot biomass due to mycorrhizae were greater in the subsoil than in the topsoil. Mycorrhization raised shoot concentrations of P and Ca (in cowpea and groundnut) and P and K (in pigeonpea) in the topsoil. Whereas the P concentration in shoots in the subsoil was not positively affected by VAM fungi, particularly in cowpea and pigeonpea, the concentration of K in such plants was significantly increased by VAM treatment. The results also showed that mycorrhizal enhancement of shoot micronutrient concentrations was very rare in all plants, with negative effects observed in certain cases. Cu concentration, in particular, was not affected by VAM formation in any of the plants, and Mn and Fe in pigeonpea and groundnut, respectively, remained the same whether plants were mycorrhizal or not. In both soils the three legumes responded to Glomus sp. better than to Gigaspora margarita, and the effects of the VAM fungi on each of the crops relative to the controls were greater in the subsoil than in the topsoil. However, shoot growth of groundnut was not affected as much as cowpea and pigeonpea by the type of soil used. In spite of the relatively low infection of its root, pigeonpea was generally the most responsive of the three legume species in terms of mycorrhizal/nonmycorrhizal ratios.

The growth response ofCalopogonium caeruleum, a leguminous covercrop in plantation agriculture, to inoculation with two vesicular-arbuscular mycorrhizal (VAM) fungi was investigated in five phosphorus (P)-deficient soils supplied with various levels of rock phosphate. Significant shoot yield increases over the uninoculated controls were obtained in most sterilised or unsterilised soils at all applied P levels, although the inoculant VAM fungi differed in their effectiveness in the soils used. Responses in mycorrhizal root infections, P and nitrogen (N) concentrations in tops and plant nodulation varied. The results are discussed in relation to the edaphic environment of the mycorrhizal association.

Prior field studies have shown that populations of forest herbs on relatively nutrient poor soils have higher vesicular-arbuscular mycorrhizal (VAM) infection intensity than plants on rich soils. However, the growth responses and ability to take up P against the soil nutrient gradient are often not linearly related to infection intensity. To determine if intraspecific differences among populations of the common VAM fungus Glomus occultum could differentially affect growth and nutrient uptake, Geranium robertianum seedlings were inoculated with Glomus occultum isolated from four forest types along a gradient of soil fertility, and grown in a greenhouse at P levels typical of the extremes of that gradient. Plants given inoculum from relatively infertile forest sites generally produced greater root, shoot, and total mass than plants given inoculum from fertile sites or uninoculated plants, especially at the low P supply rate. Total P uptake and both P and N uptake efficiency were also highest in plants given inocula from low fertility sites. These results indicate that local adaptation and intraspecific variations in the ability of VAM fungi to induce growth and nutrient uptake effects on host plants may be as important as interspecific differences among VAM fungus species.

Seven banana cultivars (Musa acuminata, AAA group) were inoculated with two species of vesicular arbuscular mycorrhizal (VAM) fungi (Glomus mosseae and Glomus macrocarpum) in a greenhouse experiment. Inoculated plants had generally greater shoot dry weight and shoot phosphorus concentrations compared to the noninoculated plants. A great variation in dependency on mycorrhizal colonization was observed among the banana cultivars. Cv. Williams showed the highest relative mycorrhizal dependency (RMD) and cv. Poyo the lowest. For all the cultivars studied, inoculation with G. macrocarpum resulted in the highest RMD values. Both root dry weight and root hair length or density of the noninoculated plants were inverserly correlated with the RMD values of cultivars.

The effect of arbuscular mycorrhizal (AM) fungi on the accumulation and transport of lead was studied in a pot experiment on maize plants grown in anthropogenically-polluted substrate. The plants remained uninoculated or were inoculated with different Glomus intraradices isolates, either indigenous to the polluted substrate used or reference from non-polluted soil. A considerably lower tolerance to the conditions of polluted substrate was observed for the reference isolate that showed significantly lower frequency of root colonisation as well as arbuscule and vesicule abundance. Plants inoculated with the reference isolate also had significantly lower shoot P concentrations than plants inoculated with the isolate from polluted substrate. Nevertheless, inoculation with either indigenous or reference G. intraradices isolate resulted in higher shoot and root biomass and inoculated plants showed lower Pb concentrations in their shoots than uninoculated plants, regardless of differences in root colonisation. Root biomass of maize plants was divided according to AM-induced colouration into brightly yellow segments intensively colonised by AM fungus and non-colonised or only slightly colonised whitish ones. Intensively colonised segments of the isolate from polluted substrate contained significantly higher concentrations of phosphorus and lead than non-colonised ones, which suggest significant participation of fungal structures in element accumulation. Summary Tomato plants were inoculated withGlomus fasciculatus mycorrhizal fungi while growing in sand through which recycled nutrient solution was automatically passed several times daily. Concentration of P and N in the solution were maintained at relatively low levels. Roots of inoculated plants became highly infected with mycorrhizal fungi, and yield parameters were significantly increased with inoculation over uninoculated control plants.

Stored topsoil from stripmining operations in western North Dakota was inoculated with mycorrhizal fungi contained in native prairie soil. The effects on plant mycorrhizal infection percentage, growth as shoot dry weight, and phosphorus uptake were determined. The studied topsoil piles were found to contain little or no vesicular arbuscular mycorrhizal (VAM) fungal inoculum at a depth of 120 cm. The inoculum soil was mixed into the stored soil at rates of 10% and 1%, or surface-applied at 1%. In control pots, sterilized inoculum soil was used. Corn plant (Zea mays) bioassays were used. After 30 days growth the percent VAM fungal infection of the test plants increased with both the 10% and 1% soil inocula. Phosphorus concentrations were generally increased by inoculation with 10% soil mixtures but not 1%. Shoot dry weights of the plants were not measurably different between 10% and 1% inoculation. However, when the plant growth period was increased to 60 days, all three parameters were increased over the check plants. When the inoculum was not mixed into the soil, but layered on the surface, there were no differences in any of the parameters.

Summary The effects on cowpea of inoculation with vesicular-arbuscular (VA) mycorrhizal fungi and rock phosphate (RP) fertilization were studied in pots using Alagba and Araromi series soils and in the field on Alagba, Apomu and Egbeda series soils. Inoculation of the plants with VA-mycorrhizal fungi caused very rapid infection of the roots. A higher per cent mycorrhizal infection was maintained during subsequent plant growth in the field. RP application reduced the degree of infection without affecting plant growth in the field and in pot experiments. Nodulation, nitrogen fixation and utilization of RP were increased by inoculation with mycorrhizal fungi in the pot experiments but not in the field experiments. In the pot experiments, inoculated plants supplied with RP flowered earlier, and took up more phosphorus than either inoculated plants without RP or uninoculated plants. The largest response to inoculation in terms of shoot dry matter, nodule yield and nitrogen content of shoots was obtained in Alagba soil under both pot and field conditions.IITA Journal Series Paper No. 136.

Summary A greenhouse study was conducted to determine the effects of stockpiling prairie grassland topsoil for 3 years on mycorrhizal development and root and shoot production of slender wheatgrass. The vesicular-arbuscular mycorrhizal (VAM) fungi involved in the symbiosis were also assessed as was the decomposition potential of the soil. During the first week of growth, VAM development in grasses grown in the stockpiled soil lagged behind that observed for grasses in the undisturbed soil. However, by 3 weeks, the mycorrhizal infection in plants in the stockpiled soil had reached levels similar to that in plants in the undisturbed soil. The dominant species of VAM fungi involved in the symbiosis at 8 weeks after planting shifted fromGlomus fasciculatum in the undisturbed soil toG. mosseae in the stockpiled soil. The delay in initial VAM infection and shift in VAM fungal species did not significantly affect plant productivity which was greatest in the stockpiled soil. The greater shoot production exhibited by grasses in the stockpiled soil was attributed to higher levels of NO3-N in the stockpiled than undisturbed soil. The potential of the soil to decay dead slender wheatgrass roots was not altered by stockpiling.

Eucalyptus tereticornis was grown in a green house in a low phosphorus (0.67 ppm Olsen's P) soil (Typic Haplustalf) inoculated with mixed indigenous arbuscular mycorrhizal (AM) fungi. Soil was amended to achieve P levels of 10, 20, 25, 30 and 40 ppm to evaluate the growth response and dependence of E. tereticornis to inoculation with AM fungi. A positive response to mycorrhizal inoculation was evident at the first two levels of soil P, i.e., at 0.67 and 10 ppm but not at the higher levels of soil P. Dry matter yield of inoculated plants beyond 20 ppm soil P was similar or even less compared to their uninoculated counterparts. Inoculated plants produced maximum dry matter (root and shoot) at 10 ppm soil P, whereas uninoculated plants did not produce until the level reached 20 ppm. The percentage root length colonized by AM fungi decreased from 31% to 3% as the concentration of P increased beyond 10 ppm soil P. Higher levels of soil P depressed the AM colonization significantly. Inoculated plants had higher shoot P and N contents compared to their uninoculated counterparts at all levels of soil P. However, at the first two lower levels of soil P, inoculated plants showed significantly higher shoot P and N contents over their respective uninoculated counterparts. The increasing shoot P accumulation beyond 10 ppm did not enhance dry matter yields. Inoculated plants had lower values of phosphorus utilization efficiency (PUE) and nitrogen utilization efficiency (NUE) at all levels of soil P except at the unamended level (0.67 ppm) where the inoculated plants showed higher values of NUE compared to uninoculated control plants. Taking dry matter yield into consideration, Eucalyptus plants were found to be highly dependent on 10 ppm of soil P for maximum dry matter production. Any further amendment of P to soil was not beneficial neither for AM symbiosis nor plant growth.

Summary Six mycorrhizal fungi were tested as inoculants for pearl millet (Pennisetum americanum Leeke) grown in pots maintained in a greenhouse. VAM fungi varied in their ability to stimulate plant growth and phosphorus uptake. Inoculation withGigaspora margarita, G. calospora andGlomus fasciculatum increased shoot drymatter 1.3 fold over uninoculated control. In another pot trial, inoculation withGigaspora calospora andGlomus fasciculatum resulted in dry matter and phosphorus uptake equivalent to that produced by adding phosphorus at 8 kg/ha.The influence of inoculatingGigaspora calospora on pearl millet at different levels of phosphorus fertilizer (0 to 60 kg P/ha) as triple superphosphate in sterile and unsterile alfisol soil was also studied. In sterile soil, mycorrhizal inoculation increased dry matter and phosphorus uptake at levels less than 20 kg/ha. At higher P levels the mycorrhizal effect was decreased. These studies performed in sterilized soil suggest that inoculation of pearl millet with efficient VAM fungi could be extremely useful in P deficient soils. However, its practical utility depends on screening and isolation of fungal strains which perform efficiently in natural (unsterilized) field conditions.

To test the hypothesis that high levels of soluble phosphate applied in combination with VAM fungi, to citrus plants, can cause growth depression even in the absence of other limiting factors, and also to test if rock phosphate, under these conditions, may be a satisfactory P source, a greenhouse experiment was conducted using sterilized soil with four levels of phosphate (0, 50, 100 and 200 ppm P) supplied either as soluble P or as rock phosphate. Citrus seedlings were either inoculated with the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus etunicatum or left uninoculated. Six months after the start of the experiment, the plants were harvested and shoot dry weight, P and K uptake, root colonization and the number of spores in 50 cm3 of soil were determined. Significant increases were found in dry matter yields and in P and K contents, due to VAM fungus inoculation, at the zero and 50 ppm soluble P levels and at all rock phosphate levels. At 100 ppm soluble P, the development of VAM plants was equilvalent to that of non-VAM plants, and at 200 ppm, growth was significantly less than that of non-VAM plants. Root colonization and sporulation were reduced at higher P levels. The absolute growth depression of VAM plants at the higher P level was likely due to P toxicity. In addition, high leaf P and K concentrations may have interfered with carbohydrate distribution and utilization in these symbioses. Rock phosphate may be used with VAM citrus to substitute for medium amounts of soluble phosphate.

A field trial was conducted to study the response of sunflower (Helianthus annuus L.) to different phosphorus levels (16, 24 or 32 kg P ha–1) and inoculation with vesicular-arbuscular mycorrhizal fungus, Glomus fasciculatum on vertisol during summer 1993. At the vegetative stage of sunflower, percent mycorrhizal root colonization, spore count, dry biomass and P uptake did not differ significantly between inoculated and uninoculated control plants. However, at later stages (flowering and maturity) percent root colonization, spore count, total dry biomass and total P uptake were significantly higher in inoculated plants than in uninoculated control plants. The total dry biomass, P content and seed yield increased with increasing P level in uninoculated plants, whereas no significant difference was observed between 16 and 32 kg P ha–1 in inoculated plants. The positive effect of mycorrhizal inoculation decreased with increasing P level above 16 kg P ha–1, due to decreased percent root colonization and spore count at higher P levels.
