An aging solution of ammoniacal silver hydroxide was used to impregnate paraffin sections of formalin-fixed tissue by a procedure based on Gordon and Sweet's method (Amer. J. Path., 12: 545-51, 1936). The solution was first used when it was 1 day old, and subsequently used at 2 day intervals for a total period of 30 days. On the final using it was found that although the staining was slightly reduced in intensity, the stain was still adequate for diagnostic purposes.  相似文献

Procedures having enhanced reliability over older methods for both Bielschowsky and Cajal types of stain are described.

Fixation of embryos in a solution containing 4% formaldehyde and 0.5% trichloracetic acid greatly improved the staining of neural elements by Bielschowsky's method.

Among the variations of Cajal's type of staining tried, a modification of Ranson's pyridin-silver method gave the most complete staining of neurofibrillar elements. Washing for 0.5 to 1 hour after silver impregnation and shortening of the reduction time from 24 to 4 hours corrected the tendency of the method to overstain.  相似文献

Several investigators have challenged the widely held view that the hydroxyl radical is the primary oxidant formed in the reaction between the ferrous ion and hydrogen peroxide. In recent studies, using the ESR spin trapping technique, Yamazaki and Piette found that the stoichiometry of oxidant formation in the reaction between Fe2+ and H2O2 often shows a marked deviation from the expected value of 1:1 (I. Yamazaki and L. H. Piette (1990) J. Am. Chem. Soc. 113, 7588-7593). In order to account for these observations, it was suggested that additional oxidizing species are formed, such as the ferryl ion (FeO2+), particularly when iron is present at high concentration and chelated to EDTA.

In this paper it is shown that secondary reactions, involving the redox cycling of iron and the oxidation of the hydroxyl radical adduct of the spin trap 5,5-dimethyl-1-pyrroline-N-oxide(DMPO) by iron, operate under the reaction conditions employed by Yamazaki and Piette. Consequently, the stoichiometry of oxidant formation can be rationalized without the need to envisage the formation of oxidizing species other than the hydroxyl radical. It is also demonstrated that the iron(III) complex of DETAPAC can react directly with DMPO to form the DMPO hydroxyl radical adduct (DMPO/OH) in the absence of hydrogen peroxide. Therefore, to avoid the formation of (DMPO/OH) as an artefact, it is suggested that DETAPAC should not be used as a reagent to inactivate containating adventitious iron in experiments using DMPO.  相似文献

本研究检视了采自中国沿海的银口天竺鲷属标本314尾, 形态学鉴定为8种: 斑鳍银口天竺鲷(Jaydia carinata (Cuvier, 1828))、细条银口天竺鲷(J. lineata (Temminck & Schlege, 1842))、新几内亚银口天竺鲷(J. novaeguineae (Valenciennes, 1832))、黑鳃银口天竺鲷(J. poeciloptera (Cuvier, 1828))、史密斯氏银口天竺鲷(J. smithi Kotthaus 1970)、横带银口天竺鲷(J. striata (Smith & Radcliffe, 1912))、印度洋银口天竺鲷(J. striatodes (Gon, 1997))和黑边银口天竺鲷(J. truncata (Bleeker, 1854))。结合GenBank中的同种序列, 对史密斯氏银口天竺鲷进行DNA条形码分析, 发现中国群体和地中海群体分为两个组群, 两者平均组间遗传距离为0.044, 表明其中存在隐存种。因该种模式产地为亚丁湾, 推测中国种群为隐存种Jaydia sp.。结合标本和文献考证, 我们认为中国已知有银口天竺鲷属鱼类9种, 未采集到的烟台银口天竺鲷J. tchefouensis (Fang, 1942)可能为J. lineata次定同种异名。我们整理了各种的同种异名、形态特征和地理分布, 编制了检索表, 探讨了分类问题, 修订了错误。中国已记录物种J. elliotiJ. arafuraeJ. albomarginata实际为J. truncataJ. poecilopteraJ. novaeguineae。  相似文献

Spinal cord of cat and rabbit was stained, after experimental lesions, by variations of Marchi's method. The following conclusions were drawn:

1. The presence of an oxidizing agent (K2Cr2O7, NaIO3, or KCIO3) in the osmic acid solution is of primary importance and a preliminary oxidation in Mueller's fluid is unnecessary or even detrimental.

2. Acetic acid added to Marchi's fluid, accentuates the action of the oxidizing agent in restraining the staining of normal myelin.

3. Too high concentration of oxidizing agent or of acid may inhibit staining of degenerating myelin.

4. Marchi's and Busch's methods have been modified as follows: Fix one day in 10% formalin and transfer without washing to the staining mixture, either A or B. Staining mixture A: Marchi's fluid plus 1 to 3% glacial acetic acid. B: An aqueous solution containing KCIO3 0.25%, osmic acid 0.33%, and acetic acid 1%. Stain about one week. These methods worked on spinal cord and medulla, but cannot be recommended for brain.

5. The detrimental effects of long post mortem autolysis or of prolonged fixation in formalin may be counteracted to some degree by increasing the concentration of the acid in Marchi's fluid up to 5% or of the KCIO3 up to 0.4% in the modified Busch's fluid.  相似文献

In previous papers (Colotta, V. et al. Arch. Pharm. Pharm. Med. Chem. 1999, 332, 39. Colotta, V. et al. J. Med. Chem. 2000, 43, 1158) we reported the synthesis and binding affinity at bovine (b) A1 and A2A and human (h) A3 adenosine receptors (ARs) of the 4-amino-6-benzylamino-2-phenyl-1,2,4-triazolo[4,3-a]quinoxalin-1-one (compound A) which resulted in a potent and selective A2A AR antagonist. Compound A provided the lead compound of a series of 6- or 8-(hetero)arylalkylamino-4-amino-2-phenyl-1,2,4-triazolo[4,3-a]quinoxalin-1-one derivatives (compounds 1–20) which are the object of this paper. Most of the newly synthesized compounds are inactive at hA3 ARs while they possess both nanomolar bA2A affinities and different degrees of bA2A versus bA1 selectivity. The binding data show that hydrophilic substituents on the benzyl moiety are the most profitable for bA2A receptor affinity. Furthermore, their steric hindrance seems to play an important role for the bA2A AR interaction, thus suggesting that the 6-aralkylamino moiety of these ligands interacts with a size-limited binding pocket of this AR subtype. Thus, the SAR studies provided us some new insights about the structural requirements of the bA2A AR recognition site.  相似文献

Several samples of J.S.B. stain (Jaswant Singh and Bhattacharjee, 1944) solution 1 (polychrome methylene blue) were prepared with 3-8 hr for dichromate-acid oxidation and addition of varying quantities of Na2HPO4 buffer for pH adjustment. Storage under severe tropical conditions and periodical checks by staining Plasmodium cynomolgi smears revealed that staining solutions oxidized 6-7 hr with a final pH of 7.8 gave optimum results. Some precipitation of azures, due to heat after 5 mo, adversely affected the quality of staining solutions, while cooler storage conditions were most favorable. Spectrophotometric and chromatographic studies indicated that the J.S.B. solution 1 was composed of blue and purple components, corresponding to higher methylene azures with methylene blue and thionin with allied products respectively.  相似文献
