三品系小型猪35个微卫星基因座的遗传学研究   总被引:35,自引:5,他引:30  
利用35个微卫星基因座对中国三个品系的小型猪(贵州小型香猪、广西巴马小型猪、版纳小耳猪近交系)进行了遗传检测。计算出三个小型猪品系个体样本在35个微卫星基因座的纯合率,并对其进行t检验。计算出各品系的平均杂合度,多态信息含量(PIC)及品系间的遗传距离,并进行了系统聚类。结果表明三个品系的小型猪其基因纯合率均较高,其中版纳小耳猪近交系的基因纯合率最高;PIC和平均杂合度均较低;贵州小型香猪和广西巴马小型猪亲缘关系较近,并均与版纳小耳猪近交系的亲缘关系略远。 Abstract:The polymorphism of 35 microsatellites in the three miniature pig breeds in China(Guizhou miniature pig, Guangxi Bama miniature pig, Banna miniature pig inbred) was analysed. Rates of homozygote for 35 microsatellite loci in three miniature pig breeds were calculated,and t-test for them were performed. Mean heterozygosity and polymorphism information content(PIC) were calculated for all breeds, and genetic distances between these breeds were estimated. The dendrograms were obtained based on genetic distances. The results suggest that rates of homozygote in the three breeds are all high, and that is the highest in Banna miniature pig inbred. The results also suggest that polymorphism information content and mean heterozygosity in all the three breeds are low, and the genetic relationship between Guizhou miniature pig and Guangxi Bama miniature pig is closer than their relationship with Banna miniature pig inbred.  相似文献

分析中国三种实验用小型猪线粒体DNA(mtDNA)D-loop的多态性,建立各品种品系猪的遗传标记,为各品种品系猪的鉴别提供依据.应用PCR对西双版纳近交系小耳猪、广西巴马小型猪、贵州小型香猪和长白猪血液总DNA样品中mtDNA D-loop进行扩增,用23种限制性内切酶消化,观察其酶切多态.PCR扩增其mtDNA D-lcop 5′端227 bp高变区域,应用PCR-SSCP和PCR直接测序分析,观察其单链构象多态和序列多态.结果显示:3种小型猪之间未见酶切长度多态、单链构象多态和序列多态;与长白猪之间表现出单链构象多态和序列多态.本研究认为:3种实验用小型猪之间mtDNA多态性贫乏,证明其在母系起源和进化上的一致性,亲源关系很近,应用PCR-RFLP、PCR-SSCP和PCR直接测序分析,尚不能作为3种实验用小型猪品种品系鉴定的依据,但与长白猪等欧系猪比较有一定差异.  相似文献

应用190个RAPD引物对贵州小型猪,巴马小型猪、西双版纳近交系小耳猪JB和JS亚系、荣昌猪Ⅰ系、长白猪的亲缘关系进行了初步分析,其中39个引物的扩增结果具有非常明显的品系间多态性,将其扩增结果以RAPDistance package Version 1?04程序进行分析,计算相对遗传距离指数1-F,再用NJ法进行聚类分析,构建系统树。结果表明,三种小型猪相互间亲缘关系较近,尤以贵州小型猪与巴马小型猪更近。荣昌猪Ⅰ系和长白猪关系较近,但与小型猪关系较远。 关键词:随机扩增多态DNA;小型猪;亲缘关系 The phylogenetic relationship of five species of pigs including Guizhou miniature pig,Bama pig,Xisuangbanna inbred pig,Rongchang Pig and Landrance was studied by RAPD analysis.Thirty-nine single polymorphic primers were selected out of 190 primers.The amplified fragments of thirty-nine primers were analysed by the RAPDistance package version 1.04 and the phylogenetic tree was constructed using NJ method.The results indicated as the following: three strains of miniature pigs have close phylogenetic relationship and the phylogenetic relationship between Guizhou miniature pig and Bama miniature pig was much closer.Landrance and Rongchang pig have close phylogenetic relationship too,but their phylogenetic relationship is far from the three strains of miniature pigs.  相似文献

The competitive equilibrium of fatty acid biosynthesis and oxidation in vivo determines porcine sub-cutaneous fat thickness(SFT) and intramuscular fat(IMF) content.Obese and lean-type pig breeds show obvious differences in adipose deposition;however, the molecular mechanism underlying this phenotypic variation remains unclear.We used pathway-focused oligo microarray studies to examine the expression changes of 140 genes associated with meat quality and carcass traits in backfat at five growth stages(1―5 months) of Landrace(a leaner, Western breed) and Taihu pigs(a fatty, indigenous, Chinese breed).Variance analysis(ANOVA) revealed that differences in the expression of 25 genes in Landrace pigs were significant(FDR adjusted permutation, P<0.05) among 5 growth stages.Gene class test(GCT) indicated that a gene-group was very significant between 2 pig breeds across 5 growth stages(PErmineJ<0.01), which consisted of 23 genes encoding enzymes and regulatory proteins associ-ated with lipid and steroid metabolism.These findings suggest that the distinct differences in fat deposition ability between Landrace and Taihu pigs may closely correlate with the expression changes of these genes.Clustering analysis revealed a very high level of significance(FDR adjusted, P<0.01) for 2 gene expression patterns in Landrace pigs and a high level of significance(FDR adjusted, P<0.05) for 2 gene expression patterns in Taihu pigs.Also, expression patterns of genes were more diversified in Taihu pigs than those in Landrace pigs, which suggests that the regulatory mechanism of micro-effect polygenes in adipocytes may be more complex in Taihu pigs than in Landrace pigs.Based on a dy-namic Bayesian network(DBN) model, gene regulatory networks(GRNs) were reconstructed from time-series data for each pig breed.These two GRNs initially revealed the distinct differences in physiological and biochemical aspects of adipose metabolism between the two pig breeds;from these results, some potential key genes could be identified.Quantitative, real-time RT-PCR(QRT-PCR) was used to verify the microarray data for five modulated genes, and a good correlation between the two measures of expression was observed for both 2 pig breeds at different growth stages(R=0.874±0.071).These results highlight some possible candidate genes for porcine fat characteristics and provide some data on which to base further study of the molecular basis of adipose metabolism.  相似文献

采用蛋白电泳技术研究了云南地方猪种血液蛋白多态性。共分析了3个云南地方猪种32-36个遗传位点,其中AKP、CAT、DIA、ES、G6PD、PA、6PGD、PHI、TF等9个 位点检测到多态性,多态位点百分比为0.1875-0.2121,平均杂合度为0.0712-0.1027。结果表明,云南地方猪种血液蛋白多态程度较高,反映在蛋白水平上的遗传多样性较为丰富。 Abstract:In this paper,protein electrophoresis was used to analyze the blood protein polymorphism in Yunnan local pig breeds and 32~36 genetic loci in the Yunnan local pig breeds were surveyed,Nine of them,such as AKP、CAT、DIA、ES、G6PD、PA、6PGD、PHI and TF,were found to be polymorphic,the mean heterozygosit(H)was 0.0712~0.1027.The results indicated that the blood protein polymorphism in the Yunnan local pig breeds is high,the Yunnan local pig breeds are wealthy in genetic diversity in point of their protein level.  相似文献

以一个鸡的小卫星cMS18为探针,对五指山小型猪、长白猪及枫泾猪进行了DNA指纹分析。结果表明,五指山小型猪的相似系数(0.721)显著高于长白猪(0.503)和枫泾猪(0.484),在遗传背景上也与后两者相距甚远。家系分析表明, 鸡的小卫星探针产生的猪的DNA指纹图的遗传符合孟德尔规律。 Abstract:A chicken minisatellite probe cMS18 was used to generate DNA fingerprints of Wuzhisshan microping,Landrace pig and Fengjing pig.It was revealed that similarity coefficient of Wuzhishan microping(0.721)was much higher than both that of Landrace pig(0.503)and of Fengjing pig(0.484).It was shown that Wuzhishan micropig was genetically distant to Landrace pig and Fengjing pig.Analysis of a porcine pedigree using cMS18 probe manifested that the segregation of DNA fingerprint bands was consistent with the Mendelian fashion.  相似文献

应用100条RAPD引物对三江白猪及其杂交亲本长白猪和东北民猪,以及大约克夏、圣特西和法系大白猪进行亲缘关系的DNA多态分析。其中15条引物的扩增结果具有明显的多态,将其扩增结果进行聚类分析,发现三江白猪与长白猪(遗传距离d = 0.090)、与东北民猪(d = 0.175)的亲缘关系较近,与大约克夏(d = 0.263)、法系大白(d = 0.223)、圣特西猪(d = 0.580)的亲缘关系较远。Abstract: One hundred primers were used to analyze the polymorphism of kinship of Sanjiang White pig and its parents, Landrace and Dongbei Min pig, Yorkshire, French Hybrids(Fh 019 strain),French Yorkshire by RAPD. Amplification results of 15 primers showed obvious polymorphisms, which were analyzed by clustering. The results indicate that the genetic relationship of Sanjiang White Pig, and Landrace, and Dongbei Min pig are respectively closer(d=0.090,,0.175), and that of Sanjiang White pig and Yorkshive(d=0.263), French Yorkshire(d = 0.223), Fh 019strain (d = 0.580) are respectively further.  相似文献

报道了AFLP标记在研究贵州小型香猪遗传多态性方面的应用和该品系猪个体基因组DNA的AFLP扩增结果,分析了贵州小型香猪的群体遗传结构.实验应用10条AFLP引物,用PstⅠ酶切,对17头猪基因组DNA进行AFLP反应,共获得116个AFLP标记,单引物获得的标记数在2~22间,贵州小型香猪群体相似系数AFLP研究结果为0.866(0.760~0.967),该研究为评价贵州小型香猪的遗传稳定性提供了相关的参数,准确评价尚待和其它品种猪对比研究后确定。 Abstract:We reported the application of AFIP markers to detect genetic polymorphic loci in Guizhou miniaturc pig and their amplified AFLP results.Their genetic construction Of the population was also analysed.TenAFLP primers were used,genomic DNAs from 17 pigs were restrictive by pst I,116 AFLP markers were obtained,the obtaining marker numbers of individual primer were between 2~22.The results indicate as the following:(1)AFLP marker is suitable for analysing genetic polymorphism in pig;(2)The similarty index of population in Guizhou miniaturc pigs was 0.866(0.760~0.967).The study provides a uscful parameter with appraise genetic stability of Guizhou miniature pigs.  相似文献

An insertion fragment in porcine FSHβ subunit gene was cloned by PCR. Sequencing data show that the insertion is a retroposon of 292 bp siting in intronⅠ at the site between +809 and +810 base. Based on these results, a PCR programme was created to genotype animal individuals in different pig breeds at FSHβ locus and polymorphism of FSHβ gene was analyzed. With the combination of genotype and litter size of sows, it was demonstrated that FSHβ locus is closely associated with major gene controlling litter size in commercial pig breeds, such as Yorkshire, Landrace, Durco. Averagely the AA sows give more 1.5 piglets than BB sows do per litter.  相似文献

SSCP analysis of pig mitochondrial DNA D-loop region polymorphism   总被引:10,自引:0,他引:10  
The sequence polymorphism that occurs in the mitochondrial DNA (mtDNA) displacement (D)-loop region is useful as a cytoplasmic DNA marker. We cloned the mtDNA D-loop regions of five breeds of pig by polymerase chain reaction (PCR) and determined their sequences. The sequence diversities in D-loop regions among five breeds of pig were located in the starting area of heavy-strand replication. From these sequences, we designed primers for PCR-mediated single-strand conformation polymorphism (PCR-SSCP) analysis that amplified the most polymorphic 227 bp fragment of the D-loop region. The results of PCR-SSCP analysis clearly showed that four types of polymorphism (A to D) are found in Landrace (A), Large White (A, B), Duroc (A), Göttingen miniature pig (B) and Meishan (C, D). The same polymorphisms were also detected from each porcine embryo by this method. Our results show that PCR-SSCP analysis is useful in detecting polymorphisms in the D-loop region of pigs and pig embryos.  相似文献

目的分析五指山小型猪、巴马小型猪和贵州香猪线粒体DNA控制区碱基序列,比较研究不同猪种的遗传标志。方法应用PCR技术分别对这三种小型猪的血液总DNA样品中线粒体DNA D-loop区进行扩增,测序比对。结果猪的线粒体DNA D-loop区分三个区域。I区(靠近5’端区域)704bp,五指山小型猪在此区共有6个变异位点,通过6个变异位点中归纳出3个单倍体,而巴马小型猪在此区有9个变异位点,通过9个变异位点归纳出4个单倍体,贵州香猪在此区共有6个变异位点,通过6个变异位点归纳出3个单倍体。Ⅱ区(串联重复序列区),五指山小型猪、巴马小型猪和贵州香猪序列相同。Ⅲ区(靠近3’端区域)三种小型猪的序列几乎相同。结论五指山小型猪、巴马小型猪和贵州香猪三种小型猪之间线粒体DNA碱基序列变异位点较少,五指山小型猪和巴马小型猪亲缘关系较近。  相似文献

西藏小型猪的线粒体DNA控制区分析   总被引:1,自引:1,他引:0  
目的研究西藏小型猪的遗传标记以及与其他国内地方猪的亲缘关系。方法扩增102头西藏小型猪以及16头巴马小型猪、17头贵州香猪的线粒体DNA控制区,测序并与国内其他猪进行比较。结果西藏小型猪线粒体DNAD-loop区分三个区域。串联重复序列区处于中间位置,包含有15~29个10 bp的重复片段,分为A、B两种类型。D-loop 3′端340 bp,与国内其他猪的序列相同比较保守;5′端704 bp,共有22个变异位点。由22个变异位点中归纳出25个单倍型,其中有两种主要的单倍型,分别占34.4%和36.6%。根据三个转换位点:305、500、691,将西藏小型猪分成了两组,几乎与串联重复序列所分的A、B两组类型相对应。与西藏小型猪相比,巴马小型猪和贵州香猪D-loop 5′端变异位点较少,分别只有4种和2种单倍型,串联重复区也只有一个类型。结论西藏小型猪可能有两个母系祖先并且与我国西南地区的品种猪有较近的亲缘关系;不同的串联重复片段类型和5′端的变异位点可以联合组建西藏小型猪的遗传标记。  相似文献

Phylogenetic relationships among Asian and European pig breeds were assessed using 1036 bp of mitochondrial DNA (mtDNA) D-loop sequences. An unweighted pair-group method with arithmetic mean (UPGMA) tree was constructed on the basis of maximum likelihood distances using sequences determined for three Cheju (Korea), 11 Chinese, one Westran (Australian feral origin) and two European pigs (Berkshire and Welsh), and also published sequences for four Japanese (including two Wild Boars), one Yucatan miniature, five European (including Large White, Landrace, Duroc, Swedish and Wild Boar) and two Meishan pigs. The Colombian collared peccary (Tayassu tajacu) sequence was also determined and used as an outgroup. The maximum parsimony with heuristic search method was used to determine bootstrap support values. Asian-type pigs clustered together (bootstrap support 33%), but were separate from European-type pigs that also clustered together (93%). The Westran pig, derived from the feral descendants of pigs inhabiting Kangaroo Island of South Australia, clustered with Asian pigs, demonstrating Asian origin of their mitochondria. Berkshire and Large White clustered with Asian pigs, indicating that Asian pigs were involved in the development of these breeds. Our findings clearly demonstrate that pigs indigenous to China, Korea and Japan are only recently diverged from each other and distinctly different from European-type pigs. European pig breeds consist of pigs with mitochondria of Asian and non-Asian type, some of which were formed from closely related maternal ancestors, if not from a single ancestor.  相似文献

Somatic cell nuclear transfer (SCNT) has been established for the transmission of specific nuclear DNA. However, the fate of donor mitochondrial DNA (mtDNA) remains unclear. Here, we examined the fate of donor mtDNA in recloned pigs through third generations. Fibroblasts of recloned pigs were obtained from offspring of each generation produced by fusion of cultured fibroblasts from a Minnesota miniature pig (MMP) into enucleated oocytes of a Landrace pig. The D-loop regions from the mtDNA of donor and recipient differ at nucleotide sequence positions 16050 (A→T), 16062 (T→C), and 16135 (G→A). In order to determine the fate of donor mtDNA in recloned pigs, we analyzed the D-loop region of the donor's mtDNA by allele-specific PCR (AS-PCR) and real-time PCR. Donor mtDNA was successfully detected in all recloned offspring (F1, F2, and F3). These results indicate that heteroplasmy that originate from donor and recipient mtDNA is maintained in recloned pigs, resulting from SCNT, unlike natural reproduction.  相似文献

The evaluation of the genetic structure of autochthonous pig breeds is very important for conservation of local pig breeds and preservation of diversity. In this study, 18 microsatellite loci were used to detect genetic relationship between autochthonous pig breeds [Black Slavonian (BS), Turopolje pig (TP), and Croatian wild boar] and to determine phylogenetic relationship among Croatian autochthonous pig breeds and certain Asian and European pigs using the mitochondrial DNA (mtDNA) D-loop sequence polymorphism. Relatively high degree of genetic variation was found between the observed populations. The analysis of mtDNA showed that haplotypes of the studied pig populations are different from the other European and Chinese haplotypes. BS pigs showed some similarities with Mangalitsa and Duroc breeds. The genetic distances of TP can be explained by high degree of inbreeding during the past century. Despite the European origin of Croatian pig breeds with some impact of Chinese breeds in the past, the results of present study show that genetic diversity is still pronounced within investigated breeds. Furthermore, the genetic diversity is even more pronounced between Croatian breeds and other European and Chinese pig breeds. Thus, conservation of Croatian pig breeds will contribute to overall genetic diversity preservation of pig breeds.  相似文献

Nucleotide sequences of mitochondrial DNA (mtDNA) cytochrome B gene (1140 bp) and control region (707 bp) were used to determine the phylogenetic relationships among 51 pig samples representing ancient and current varieties of Iberian pigs (26), Spanish wild boars (seven) and other domestic pigs (18) of cosmopolitan (Duroc, Large White, Landrace, Pietrain and Meishan) and local (Spotted Black Jabugo, Basque and Mangalitza) breeds. A neighbour-joining tree constructed from pairwise distances provide evidence of the European origin of both Iberian pigs and Spanish wild boars. The introgression of Asian mtDNA haplotypes in the genetic pool of the Iberian breed seems unlikely. Four estimates of sequence divergence between European and Asian clades were calculated from the two main domains of the D-loop region and the synonymous and nonsynonymous nucleotide substitutions in the cytochrome B gene. The time since the divergence of pig ancestors was estimated at about 600,000 years before present.  相似文献

采用世界粮农组织(FAO)和国际动物遗传学会(ISAG)推荐的27个微卫星,对久仰香猪、剑白香猪、从江香猪、环江香猪、黑香猪(贵州省种猪场)、五指山猪和滇南小耳猪等我国7小型猪种(类群)及杜洛克、长白和大白等3个外来猪种的群内遗传变异性和群间遗传差异进行了分析。结果表明,7个小型猪品种(类群)均有较高的群内遗传变异,但久仰、剑白、从江和环江4个香猪类群的群内遗传变异(平均多态信息含量(PIC)为0.61~0.64)显著低于其他3个小型猪种(平均PIC为0.80~0.84)。久仰、剑白、从江和环江4个香猪类群彼此间的遗传差异较小(奈氏标准遗传距离为0.12~0.22),但它们与其他3个小型猪种有较大的遗传差异(奈氏标准遗传距离为1.61~1.96),与3个外来猪种的遗传差异更大(奈氏标准遗传距离为1.99~3.30)。通过聚类分析,可将这些猪种清晰地分为3大类,久仰、剑白、从江和环江4个香猪类群紧密地聚为一类,其他3个小型猪种聚为一类,3个外来猪种聚为另一类。这一结果基本符合这些猪种的地理分布和品种来源。

Vietnam is one of the most important countries for pig domestication, and a total of 26 local breeds have been reported. In the present study, genetic relationships among the various pig breeds were investigated using 90 samples collected from local pigs (15 breeds) in 15 distantly separated, distinct areas of the country and six samples from Landrace pigs in Hanoi as an out‐group of a common Western breed. All samples were genotyped using the Illumina Porcine SNP60 v2 Genotyping BeadChip. We used 15 160–15 217 SNPs that showed a high degree of polymorphism in the Vietnamese breeds for identifying genetic relationships among the Vietnamese breeds. Principal components analysis showed that most pigs indigenous to Vietnam formed clusters correlated with their original geographic locations. Some Vietnamese breeds formed a cluster that was genetically related to the Western breed Landrace, suggesting the possibility of crossbreeding. These findings will be useful for the conservation and management of Vietnamese local pig breeds.  相似文献
