Watermelon ( Citrullus vulgaris Schrad. cv. Fairfax) cotyledons were excised from the embryo and grown in the dark for 4 days. They were then transferred to 10 μm benzyladenine (BA) solution or illuminated with white light. We have compared changes in ultrastructure of the plastids and of their polypeptide pattern induced by the two treatments.
At the end of the 4-day-period in the dark the plastids differentiated to amyloplasts and had few polypeptides: only the two subunits of ribulose bisphosphate carboxylase (RuBP) were clearly observed. Both light and BA induced starch depletion and gratia formation after 12–24 h. BA was less efficient than light in inducing thylakoid formation and more efficient in inducing starch depletion. After 6 h both factors induced the appearance of the same new polypeptides in the 28–53 kDa range. Most prominent among them is a 32 kDa band. Light is much more effective in inducing the formation of a 29 kDa band than is BA. In mature chloroplasts this band stains very strongly, while the 32 kDa band disappears. We suggest that the 29 kDa polypeptide is the light harvesting complex (LHC), since a purified LHC preparation from cotyledons grown either on water in light or on BA in the dark migrates on the polyacrylamide gel as a single 29 kDa band.  相似文献

The placement of Pinus ponderosa embryos on a benzyladenine (BA) supplemented medium induces the differentiation of two distinct morphological types of cotyledons. Cotyledons in contact with the medium (lower cotyledons) do not elongate, and rapidly respond to BA by forming multiple buds along their entire length. Cotyledons not in contact with the medium (upper cotyledons) elongate, do not form buds, and are similar both morphologically and in protein profiles to seedling cotyledons that do not respond to BA. Using sodium dodecyl sulfate polyacrylamide electrophoresis, protein differences between these two cotyledon types can be detected as early as one week. These protein differences include the reduced level of a 52,000 Kilodalton (52 Kd) peptide and the appearance of a 4 Kd peptide in the lower cotyledons. These peptide differences may prove useful as markers for tissue that will respond to BA.  相似文献

The leaf apoplast is a dynamic compartment in contact with plant pathogenic bacteria after infection. Among the very first interaction events is the receptor-mediated perception of bacterial surface molecules such as flagellin or other conserved microbe-associated molecular patterns (MAMPs). Apoplast proteins likely play a role in basal resistance (BR) or pattern-triggered immunity (PTI). Here, a proteomic approach was carried out on water soluble — potentially the most mobile — apoplast proteins from flagellin-treated tobacco (Nicotiana tabacum) leaves. As the quickness of BR/PTI seems crucial for its efficacy, samples were taken as early as 2.5 and 7 h post inoculation. Proteins were separated by two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and identified by liquid chromatography tandem mass spectrometry (LC-MS/MS). Forty-nine different proteins from 28 protein spots changed in their density compared to the water-inoculated control. Eleven protein spots appeared de novo in response to EBR induction. There are glycohydrolases and redox-active proteins besides pathogenesis-related proteins among them, predicting plant cell wall structural modifications and more direct antimicrobial effectors as earliest changes related to BR/PTI.  相似文献

Here we describe a proteomic approach to detect fibrinolytic enzymes from the culture supernatant of Bacillus subtilis 168. Following isoelectric focusing without dithiothreitol, two gels, one for sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and the other for zymography, were run in parallel. After silver staining of SDS-PAGE and activity staining of zymography gel, the two gels were superimposed to detect protein spots that coincided with clear zones on the zymography gel. We identified four protein spots and characterized them with matrix-assisted laser desorption/ionization mass spectrometry. Database search revealed that four spots contained at least one of the extracellular serine proteases such as WprA and Vpr. This combined method of two-dimensional gel and zymography can be used as a powerful tool to detect proteases from various organisms.  相似文献

Cotyledons of 3- to 4-week-old seedlings of Douglas-fir [ Pseudotsuga menziesii (Mirb). Franco] were treated with shoot induction medium (SIM) containing 5 μ M 6N-benzylaminopurine (BAP) and 5 n M naphthaleneacetic acid (NAA). Fresh weight, dry weight and soluble protein levels were not altered within the first 48 h of SIM treatment. SIM-treated cotyledons were labelled in vivo with 35SO42-, and TCA-insoluble proteins were analyzed electrophoretically by a 2-dimensional system consisting of non-equilibrium electrophoresis (NEPHGE) followed by sodium dode-cylsulfate polyacrylamide gel electrophoresis (SDS-PAGE). A basic polypeptide with a relative molecular weight of 14.5 kDa was detected 32 h after induction and after 48 h a number of polypeptides in the 14 to 35 kDa range were induced. Translation products of poly-A+ RNA isolated from cotyledons treated with SIM for 4, 16, 32 and 48 h were analyzed by using 2-dimensional NEPHGE-SDS-PAGE. Both qualitative and quantitative differences in the translation products were observed at all time points investigated. Expression of a specific RNA coding for a 30 kDa polypeptide was demonstrated as early as 4 h after culture on SIM. This RNA was also present at 16 h, but decreased with longer SIM treatments. Thus, culture of excised cotyledons on a medium inducing the formation of adventitious shoots results in rapid quantitative and qualitative changes in the polypeptide composition and translatable RNA population prior to morphological evidence of shoot induction.  相似文献

Two major endopeptidases were present in cotyledons of germinating Vigna mungo seeds, as detected by the zymogram after polyacrylamide gel electrophoresis. They were not detectable in cotyledons of dry seeds, but their intensities on the zymogram increased during germination. During incubation of detached cotyledons, however, the activities showed only a slight increase for 5 days. These two endopeptidases could be separated by Sephacryl S-200 column chromatography. One of them was found to be a serine-endopeptidase as judged by phenylmethylsulfonylfluoride and diisopropyl fluorophosphate inhibition. The other was a sulfhydryl-endopeptidase because of its dependency on 2-mercaptoethanol and inhibition by leupeptin, chymostatin, and antipain. Analysis by sodium dodecyl sulfate polyacrylamide gel electrophoresis indicatd that the two endopeptidases digested the Vigna mungo seed globulin subunits at different rates. The serine enzyme digested the 56 kilodalton subunit at first, but the sulfhydryl enzyme digested the 54 kilodalton peptide more efficiently than the 56 kilodalton peptide. The pattern of digestion of globulin by the combination of the serine- and sulfhydryl-endopeptidases was similar to that using crude enzyme extracts.  相似文献

The vegetative mycelium of Sphaerostilbe repens Berkeley and Broome (strain CBS 275-60) gives rise, within 48 h, to aggregated organs composed of coremia and rhi-zomorphs. Developmental changes in polypeptide patterns were studied by one- and two-dimensional polyacrylamide gel electrophoresis after cells had been induced to undergo synchronized differentiation. One-dimensional gel electrophoresis revealed only minor changes during the morphogenesis. Of the 300 polypeptides resolved by two-dimensional gel electrophoresis, nearly 12% either increased or decreased during coremium and rhizomorph differentiation. Some polypeptides appeared to be unique to one or the other of the cell preparations and represented apparent qualitative differences. During the first 24 h of differentiation, about 20 polypeptide spots appeared, 6 were enhanced, 4 were reduced and 32 disappeared. Over the next 24 h changes in the population of proteins were less marked: 14 new proteins were revealed and 9 increased in intensity while 15 declined and 9 were no longer detectable. Five proteins which were present at a significant level only during the first stages of differentiation, may therefore, putatively be designated as aggregation-specific polypeptides.  相似文献

Excised zygotic embryos,cotyledons and hypocotyls of juvenile seedlings of masson pine were grown on DCR medium supplemented with several concentrations of various plant phytohormones.BA(1.0mg/L) in combination with NAA(0.05mg/L) in DCR medium was found to increase the formation of adventitious buds from mature zygotic embryos,but most of them were formed at the tips of embryonic cotyledons.Adventitious buds were obtained from cotyledons and hypocotyls from juvenile seedlings when they were cultured on DCR medium containing BA 3-5 mg/L and NAA 0.1-0.2 mg/L.Elongation of buds were observed on hormone-free DCR medium with or without activated charcoal(0.5%).Root initiation was achieved with full or half strength DCR medium supplemented with IBA 1.0 mg/L and NAA 0.25-0.5 mg/L.Approximately 11-20 axillary buds formed on each explant when juvenile seedling explants were treated(3-20h) with BA 50-100 mg/L,followed by transfer to hormone-free DCR medium.The maximum number of shoots obtained per explant within six months was 33.  相似文献

This paper describes a two-dimensional gel electrophoresis method for separating large RNA molecules such as messenger RNAs. RNAs were at first separated on a polyacrylamide plus agarose composite gel and subjected to a second dimension electrophoresis on a polyacrylamide gel containing urea. This method is illustrated by analyses of poly(A)+ yeast RNAs. About 80 discrete spots were detected on the gel, when RNAs from 1000 to 3500 nucleotides in size were examined.  相似文献

Proteinases were excreted by strains of Bacillus subtilis during fermentation of African locust bean cotyledons. Those excreted by one strain were purified and characterized by ammonium sulphate precipitation, ion-exchange chromatography (IEC), gel filtration, inhibition tests and polyacrylamide gel electrophoresis (PAGE). Three proteinases and an esterase without proteolytic activity were identified. A serine proteinase which showed a high degree of hydrophobicity and a neutral proteinase were present. The third proteinase showed both proteolytic and esterolytic activities, and had multiple electrophoretic mobilities on polyacrylamide gel.  相似文献

Embryogenic and non-embryogenic calli from leaf sections of Coffea arabica cv. Catimor, were analyzed under denaturing conditions in one- and two-dimensions by polyacrylamide gel electrophoresis. The protein patterns revealed qualitative and quantitative differences in size and charge. In non-embryogenic calli, two dimensional analysis reveals seven distinctive polypeptides in the range of 15 to 70 kDa. Four of the polypeptides are acidic, three of 70 kDa and one of 15 kDa. Similarly, in embryogenic calli there are seven characteristic polypeptides with molecular weight from 23 to 35 kDa in a broad pI from acid to basic. Five of them are found in the neutral to acid pI, and are probably related to storage protein-like polypeptides detected in zygotic embryos and seeds of Coffea arabica cv. Catimor. ADP-glucose pyrophosphorylase (AGP) was purified from tomato (Lycopersicon esculentum Mill.) fruit to apparent homogeneity. By sodium dodecyl sulfate-polyacrylamide gel electrophoresis the enzyme migrated as two close bands with molecular weights of 50,000 and 51,000. Two-dimensional polyacrylamide gel electrophoresis analysis of the purified enzyme, however, revealed at least five major protein spots that could be distinguished by their slight differences in net charge and molecular weight. Whereas all of the spots were recognized by the antiserum raised against tomato fruit AGP holoenzyme, only three of them reacted strongly with antiserum raised against the potato tuber AGP large subunit, and the other two spots (with lower molecular weights) reacted specifically with antisera raised against spinach leaf AGP holoenzyme and the potato tuber AGP small subunit. The results suggest the existence of at least three isoforms of the AGP large subunit and two isoforms of the small subunit in tomato fruit in vivo. The native molecular mass of the enzyme determined by gel filtration was 220 +/- 10 kD, indicating a tetrameric structure for AGP from tomato fruit. The purified enzyme is very sensitive to 3-phosphoglycerate/inorganic phosphate regulation.  相似文献

Somatic embryos were induced in lettuce cotyledons culturedon Murashige and Skoog's (MS) medium containing either 2 mgl–1 6-benzylaminopurine (BA) and 0.2 mg l–1 naphthaleneaceticacid (NAA) or 0.2 mg l–1 BA and 2 mg l–1 NAA. Bothcombinations induced a frequency of over 70%. The explants culturedonly in the presence of 2,4-dichlorphenoxyacetic acid (2,4-D)did not produce somatic embryos. The development of the embryoidswas studied histologically and by scanning electron microscopy.Peroxidase activity was assayed and the isoenzyme pattern ofcalluses was determined by polyacrylamide gel electrophoresis.Callus from an embryogenic line showed a much higher peroxidaseactivity than that from a non-embryogenic line, one extra peroxidaseisozyme band being present and typical of the embryogenic callus.No qualitative differences were detectable between the embryogeniccalluses. We constructed a novel database of the proteome of DLD-1 colon cancer cells by two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) of fluorescence-labeled proteins followed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF) analysis. The database consists of 258 functionally categorized proteins corresponding to 314 protein spots. The majority of the proteins are oxidoreductases, cytoskeletal proteins and nucleic acid binding proteins. Phosphatase treatment showed that 28% of the protein spots on the gel are phosphorylated, and mass spectrometric analysis identified 21 of them. Proteins of DLD-1 cells and of laser-microdissected colon cancer tissues showed similar distribution on 2D gels, suggesting the utility of our database for clinical proteomics.  相似文献

Jasmonates play a critical role in plant defense against pathogens through regulation of the expression of defense-related genes. To study the role of jasmonic acid (JA) in the rice self-defense mechanism, a proteomic approach was applied. When 3-week-old rice cv. Java 14 was treated with 100 microM JA for 3 days, numerous necrotic brown spots were observed on the leaf blade. Three-week-old rice was treated with JA and proteins from cytosolic and membrane fractions of leaf blade were separated by two-dimensional polyacrylamide gel electrophoresis. A total of 305 proteins were detected in both cytosolic and membrane fractions. When rice plant was treated with 100 microM JA for 2 days, 12 proteins were up-regulated and 2 proteins were down-regulated. Out of them, 8 proteins were changed in dose dependence manner, while 4 proteins were changed in a time course manner. Among them, pathogenesis-related protein 5 (PR5) and probenazole inducible protein 1 (PBZ1) were significantly induced by 100 microM JA for 2 days. These results suggest that PR5 and PBZ1 are important proteins expressed down-stream of JA signals in rice cv. Java 14.  相似文献

Floral induction in seedlings of Pharbitis nil Choisy cv. Violet, with one cotyledon removed, was manipulated by applying various photoperiodic treatments to the remaining cotyledon. Populations of polyadenylated RNA from treated cotyledons were examined to identify messages specifically involved in floral induction. The RNA was translated in vitro using a wheat-germ system, and the resulting translation products were analysed by two-dimensional polyacrylamide gel electrophoresis. Substantial qualitative and quantitative differences were found between mRNA from cotyledons of seedlings kept in continuous light (non-induced) and of seedlings given a 16-h dark period (induced). 通过二维聚丙烯酰胺凝胶电泳以及计算机辅助的图像分析技术,对荔枝开花后40d的正常与败育胚蛋白质图谱进行初步分析。结果表明,100个蛋白质点在表达丰度上有明显差异;选择仅在正常发育胚胎胶上表达的蛋白点15个和仅在败育胚胎胶上表达的蛋白点50个,进行基质辅助激光解吸附电离飞行时间质谱(MALDI—TOFMASS)分析,鉴定出9个与胚发育相关的蛋白,这些蛋白可能参与了胚败育的调节和控制。  相似文献

Two-dimensional polyacrylamide gel electrophoresis comparisons were made for the non-histone “Chromatin fraction II” proteins of normal, phytohemagglutinin-stimulated and acute leukemic lymphocytes. The “Chromatin fraction II” proteins were extracted from the nuclear residue fraction after initial treatment with (a) 0.075 M NaCl containing 0.025 M EDTA, pH 8; (b) 0.01 M Tris-HCl, pH 8; and (c) 0.4 N H2SO4. Most of the proteins found earlier in the “Chromatin fraction II” of rodent liver and hepatomas were also found in the human cells. Some changes such as the decrease in amount of protein BA of normal rodent cells were found in the comparison of normal and stimulated human cells. By comparison with normal lymphocytes, the phytohemagglutinin-treated cells had decreased spot densities and sizes for proteins BA and Bv and an increase in densities and sizes of proteins CB, C25, CS and CT. In the acute lymphocytic leukemic cells there was a decrease in spots A24, BA, Bv, CD and CD′ by comparison with the normal lymphocytes. Protein CG′ which was found earlier in the hepatomas was found in acute lymphocytic leukemic cells but not in the control or phytohemagglutinin-treated cells. These studies show that there is a loss in specific Chromatin proteins BA and Bv from the Chromatin of rapidly turning over cells. Concomitantly, increases are observed for the amounts of protein spots CB, C25, CS and CT in the actively growing cell samples.  相似文献

Benzyladenine (BA) stimulated 5-aminolevulinic acid (ALA) accumulation in the presence of levulinic acid during illumination with 43 μmol m−2 s−1 light in excised etiolated cotyledons of cucumber ( Cucumis sativus L. cv. Aonagajibai). A short dark-pretreatment (6 h) with BA eliminated the lag phase of ALA accumulation. The rate of ALA accumulation during the steady-state phase in cotyledons pretreated with BA for a long period (14 h) was considerably accelerated compared to that in cotyledons pretreated with BA for 6 h. The rate of ALA accumulation during the lag phase was saturated at a very low light fluence (<1.4 μmol m−2 s−1) in both BA-pretreated and water-control cotyledons. The steady-state rate of ALA accumulation increased with increasing light fluence up to 43 μmol m−2 s−1 (parallel to that of Chl formation) in water-control cotyledons. In contrast, in cotyledons pretreated with BA for either 6 or 14 h, the steady-state rate reached a plateau at a very low light fluence. Based on the above results together with our finding that there are two components of Chl formation (M. Dei, 1984. Physiol. Plant. 62: 521–526) possible intermediate steps of Chl biosynthesis pathway affected by BA and light intensity are discussed.  相似文献
