Contents of total free [PA(S)] and conjugated polyamines [PA(SH), PA(PH)] were higher in turnip (Brassica rapa L. cv. Rapa) seeds during imbibition (0–36 h) and radicle protrusion (36–48 h) than during the further growth (10 d). Ethylene production was activated with the protrusion, reaching a maximum at the second day of germination and dropping afterwards. The application of ethrel accelerated radicle emergence but the direct intervention of ethylene in the breaking of the seed coat was not clear from the use of ethylene-biosynthesis inhibitors (CoCl2 and AVG). Finally, in this work the gene BrACO2 was characterized. Although its expression was not detected in seeds through zygotic embryogenesis, it increased concomitantly with the germination process.  相似文献

The germination of viable seeds is the basis for new plant growth and development. Seeds lose viability during storage, but the biochemical mechanisms of seed death are not fully understood. This study aimed to investigate degradation patterns of nucleic acids during seed ageing and subsequent water uptake. Seeds of Pisum sativum L. were artificially aged at 50°C and 12% seed water content (WC). Nucleic acids degradation was studied during ageing and during imbibition of four seed lots with differential viability from highly viable to dead. As seeds lost viability during ageing, DNA was gradually degraded into internucleosomal fragments, resulting in ‘DNA laddering’, in conjunction with disintegration of 18S and 28S rRNA bands. During imbibition, non-aged controls had high levels of DNA and RNA integrity through to radicle protrusion. In an aged seed lot with 85% total germination (TG) DNA fragmentation decreased upon imbibition probably due to nucleosome degradation, while rRNA integrity did not improve. In an aged seed lot with 44% TG, neither DNA nor rRNA integrity improved upon imbibition. Dead seeds showed DNA degradation as laddering throughout imbibition along with extensive degradation of rRNA. We present a model in which interlinked programmed and non-programmed events contribute to seed ageing, and suggest that protection of nucleic acids during ageing is key to seed longevity.  相似文献

To study broccoli and radish seed germination under different temperature regimes the germination test has been used to assess final germination percentage, start and rate. This method has been integrated with a computer‐aided image analysis test which is more accurate in monitoring the extent of imbibition phases through the assessment of seed area increase and timing of radicle emergence detected on single seeds. In addition, seed area increase has been used also to establish a close relationship with radicle elongation rate in the time range when ‘visible germination’ is scored by a classical germination test. The results suggest that this image analysis parameter may be considered as a reliable seed imbibition marker to integrate the germination parameters obtained by a germination test.  相似文献

It is not known how embryos of seeds of the Pinaceae protrude from their enclosing tissues to complete germination. Prior to protrusion of the radicle there is an increase in endo-β-1,4-mannanase (EC 3.2.1.78) activity associated with weakening of the micropylar megagametophyte/nucellus from seeds of white spruce ( Picea glauca [Moench.] Voss). Mannanase activity is present as three isoforms (pI values 5.0, 4.8, 4.7) in both the embryo and surrounding structures (megagametophyte and nucellus) prior to and during imbibition. Activity of all the isoforms increases in the chalazal and micropylar megagametophyte during germination. Activity then declines after the testa splits, typically 1 day prior to radicle protrusion, due partially to its leaching from the seed into the surrounding water. Activity increases in the cotyledons and axis as the embryo commences elongation. Seeds from dormant seedlots exhibit a lower germination percentage, relative to seeds from nondormant seedlots, and the force necessary for the embryo to puncture the surrounding structures tends to be greater. Although similar mannanase activities are present in unimbibed seeds of dormant and nondormant seedlots, during germination, enzyme activity in seeds of dormant seedlots is lower. Moist chilling alleviates dormancy in the seeds of the Pinaceae and, during 3 weeks of this treatment, mannanase activity slowly increases. After 3 weeks of moist chilling and regardless of whether the seedlot was dormant or not prior to moist chilling, the force necessary to puncture the micropylar megagametophyte and nucellus is lower, and the speed of germination greater. Seeds from previously dormant seedlots also complete germination to a greater percentage, relative to unchilled seeds from dormant seedlots. Upon transfer to 25°C, mannanase activity in moist-chilled seeds decreases during germination of all seedlots regardless of their previous dormancy status.  相似文献

Lettuce (Lactuca sativa L. cv Minetto) seeds were primed in aerated solutions of 1% K3PO4 or water at 15°C in the dark for various periods of time to determine the manner by which seed priming bypasses thermodormancy. Seeds which were not primed did not germinate at 35°C, whereas those which were primed for 20 h in 1% K3PO4 or distilled H2O had up to 86% germination. The rate of water uptake and respiration during priming were similar regardless of soak solution. Cell elongation occurred in both water and 1% K3PO4, 4 to 6 h prior to cell division. Both processes commenced sooner in water than K3PO4. Radicle protrusion (germination) occurred in the priming solution at 21 h in water and 27 h in 1% K3PO4.

Respiration, radicle protrusion and cell division consistently occurred sooner in primed (redried) seeds compared to nonprimed seeds when they were imbibed at 25°C. Cell division and elongation commenced after 10 h imbibition in primed (redried) seeds imbibed at 35°C. Neither process occurred in nonprimed seeds. Respiratory rates were higher in both primed and nonprimed seeds imbibed at 35°C compared to those imbibed at 25°C, although radicle protrusion did not occur in nonprimed seeds which were imbibed at 35°C. It is apparent that cell elongation and division are inhibited during high temperature imbibition in nonprimed lettuce seeds. Seed priming appears to lead to the irreversible initiation of cell elongation, thus overcoming thermodormancy.

We tested the effects of cold stratification, temperature, light and NaCl on seed germination and germination recovery and of NaCl on radicle growth and radicle elongation recovery of Kalidium caspicum, a small leafy succulent shrub dominant in saline deserts in northwest China. In all conditions of temperature and light/darkness, germination percentages and rates of cold-stratified seeds were significantly higher than those of nonstratified seeds. Germination of a high percentage of both nonstratified and stratified seeds was inhibited by 0.2 M NaCl, and 0.6 M NaCl completely inhibited germination. Nongerminated seeds germinated after they were transferred from NaCl solutions to distilled water. Radicle elongation significantly decreased with increase in salinity, and it was completely inhibited by ≥1.0 M NaCl; radicle elongation recovered in young seedlings pretreated by 10 days of incubation in ≤0.4 M NaCl. Results show that seed germination and early seedling growth of K. caspicum are salt tolerant, and these characteristics help explain why this species can survive and dominate salt habitats, such as those in the Junggar desert in Xinjiang, northwest China.  相似文献

Changes in ascorbate and glutathione contents and the activities and isoenzyme patterns of enzymes of the ascorbate-glutathione cycle were investigated in embryo axes and cotyledons of germinating lupine (Lupinus luteus L.) seeds. Ascorbate content was not significantly affected over the initial 12 h of imbibition in embryo axes, but afterwards increased, with the most rapid accumulation coinciding with radicle emergence. A somewhat similar trend was observed for glutathione with significant increase in embryo axes shortly before radicle protrusion followed by decline in the next hours. In cotyledons the ascorbate pool rose gradually during germination but the amount of glutathione showed fluctuations during a whole germination period. The activity of ascorbate peroxidase (APX) rose progressively in embryo axes, while activities of dehydroascorbate reductase (DHAR) and glutathione reductase (GR) showed transient increase during germination. New isoforms of APX and GR were synthesized, suggesting that they play a relevant role during germination. All analyzed enzymes were already present in dry seeds which allowed them to be active immediately after imbibition.  相似文献

Following 16, 40 and 64 h exposure to 0.33 M NaCl given after 8 h water imbibition, lentil seeds showed a gradual decrease of germination upon their transfer to water. These salt related changes were accompanied by modifications in the protein patterns of embryo axes as revealed by two-dimensional electrophoresis separation and by the computer image analysis of protein spots. In comparison with 8 h water imbibed seeds, prominent proteins comprised between the 5.1 – 7.6 pH isoelectric point in the first dimension and 75 – 50 kDa molecular mass in the second dimension showed a significant increase in their abundance as salt exposure increased. On transfer to water to complete germination, the content of many of these proteins decreased at 24h in 2 – 3 cm length embryo axes in comparison with the corresponding embryo axes of seeds continuously imbibed in water for 24 h. Some groups of proteins ranging between 15.5 – 17.3 kDa, already present after 8 h water imbibition, were not detectable after 24 h but were expressed in seeds exposed to NaCl and transferred to water for 24 h. Up- and down-regulated proteins in lentil embryo axes, imbibed under non-lethal salt stress conditions, have been tentatively identified by comparison with the protein map of germinating seeds of the model plant Arabidopsis. Perennial ryegrass (Lolium perenne L.) seeds (caryopses) germinateat or near the soil surface, where water potential can fluctuatewidely. This study examined germination of ‘Del Ray’perennial ryegrass seeds when imbibition was interrupted bydehydration prior to radicle emergence. Seeds were hydratedfor 0 to 40 h (26C), dehydrated at atmospheric water potentialsof –4, –40, –100 and/or –150 MPa for4–168 h, then rehydrated. Germination (radicle elongation 1 mm), seedling growth, solute leakage, and endogenous abscisicacid (ABA) levels were measured. Treatment differences in finalgermination percentage, seedling growth, and solute leakagewere generally not significant. However, the onset of radicleemergence was delayed and the rate of germination slowed whendehydration at –150 MPa was initiated after 36 or 40 hhydration. Slowed germination rates were not observed when dehydrationwas initiated before 36 h, when dehydration occurred at –4MPa, or when dehydration at –150 MPa was preceded by dehydrationat –4 MPa for 24 h. Exogenous abscisic acid (ABA) concentrationsabove 10–6 M inhibited germination. However, endogenouswhole seed ABA levels declined during imbibition due to leaching,and did not increase during dehydration treatments that delayedgermination. These results illustrate that rate of late-occurringdehydration treatments is critical in determining subsequentgermination response. We propose that seed response to late-occurringdehydration may be of ecological significance in timing radicleemergence to coincide with adequate soil moisture for seedlingestablishment. Seeds of the empress tree ( Paulownia tomentosa Steud.) were imbibed for two weeks in darkness at constant temperatures (18, 23 or 28°C), and then irradiated with red light for 5 min. Germination was poor if it took place at the same temperature as imbibition, but a high percentage was achieved if the seeds were exposed to higher or lower temperatures before they were irradiated. Maximum germination was obtained when the difference between pretreatment and imbibition was about 10°C. The effect increased with the duration of the pretreatment and was optimal at 24 h. The effect decreased as the time lapse between temperature pretreatment and red light irradiation increased, and it was lost after two days. If pretreatment was shorter than 24 h (12 h). a high percent of germination was obtained by alternating pretreatment and imbibition temperatures. The germination of seeds imbibed in 40% heavy water was also stimulated by temperature pretreatments. Light and temperature also exhibited an interactive effect in the germination of seeds that were imbibed in darkness for only 3 days. For each of the germination phases there was a temperature at which the time needed for 50% germination was the shortest, namely 35°C during imbibition, 37.5°C in the period of Pfr activity. and 32.5°C during radicle protrusion. The data obtained are shortly discussed in relation to the domestication of empress tree in Southern Europe.  相似文献

  • Fruiting season of many Sri Lankan tropical montane species is not synchronised and may not occur when conditions are favourable for seedling establishment. We hypothesised that species with different fruiting seasons have different seed dormancy mechanisms to synchronise timing of germination with a favourable season for establishment. Using six species with different fruiting seasons, we tested this hypothesis.
  • Germination and imbibition of intact and manually scarified seeds were studied. Effect of GA3 on germination was examined. Embryo length:seed length (E:S) ratio of freshly matured seeds and of those with a split seed coat was determined. Time taken for radicle and plumule emergence and morphological changes of the embryos were recorded.
  • The radicle emerged from Ardisia missionis, Bheza nitidissima and Gaetnera walkeri seeds within 30 days, whereas it took >30 days in other species. Embryos grew in seeds of B. nitidissima and G. walkeri prior to radicle emergence but not in Microtropis wallichiana, Nothapodytes nimmoniana and Symplocos cochinchinensis. A considerable delay was observed between radicle and plumule emergence in all six species. Warm stratification and/or GA3 promoted germination of all species.
  • All the tested species have epicotyl dormancy. Seeds of B. nitidissima and G. walkeri have non‐deep simple morphophysiological epicotyl dormancy, and the other four species have non‐deep physiological epicotyl dormancy. Differences in radicle and epicotyl dormancy promote synchronisation of germination to a favourable time for seedling development. Therefore, information on dormancy‐breaking and germination requirements of both radicle and epicotyl are needed to determine the kind of dormancy of a particular species.

Lettuce seeds cvs Hilde, Feltham King and Avoncrisp were subjected, at different phases during imbibition at 22°C, to a high temperature (33°C) inhibitory for germination, for periods ranging from 4 to 144 h, before returning them to 22°C. The results showed, that the first 4h of imbibition and also the phase between the commencement of mitosis and the onset of radicle emergence were more sensitive to the effects of high temperature than other phases in the germination process. Short exposures (8–24 h) to 33°C commencing at the latter phase delayed germination by up to 4 days, and at the earlier by up to 8 days. Percentage germination was unaffectd except after prolonged exposures (> 48 h) from the beginning of imbibition, which reduced it. Seedling emergence from moist sieved soil was both delayed and reduced when imbibing seeds were exposed for a short period from the beginning of imbibition to 33°C compared with seeds imbibing continuously at 19°C. Germination was delayed and not reduced when seed was exposed to 33°C at the phase between commencement of mitosis and the onset of radicle emergence.  相似文献

Germination responses ofMallotus japonicus (Thumb). Muell. Arg. seeds to temperature revealed a gap-detecting mechanism in the seed germination of the species. Among various constant and alternating temperatures examined in the range from 12–40°C, only very limited temperature regimes were found to be favourable for seed germination, specifically, alternating temperatures between 18–32°C and 28–40°C. A single several-hour higher-temperature (32–40°C) treatment could also induce the germination of seeds which had been imbibed for several days at a constant temperature in the range of 20–26°C, suggesting that there is a process requiring higher temperature among the overal germination processes. Seeds located at or near the surface of denuded soil would have a good chance of experiencing such a temperature change when several rainy days are followed by fine weather, while seeds beneath close vegetation would not. On the other hand, the pressence or absence of light or a simulated ‘canopy ligh’ had little effect on the germination. Therefore, it was concluded that the seeds ofM. japonicus have a ‘gapdetecting mechanism’ in the form of a higher-temperature requirement of a certain process involved in the overall germination processes.  相似文献

运用蛋白质组学技术对大豆(Glycinemax)N2899种子萌发0h、8h、36h、60h4个时期蛋白质的差异表达情况进行了研究.结果发现,在考马斯亮蓝染色的双向电泳pH3~10胶上,PDQuest图像分析软件可识别的点约350个,其中表达量变化2.5倍以上的蛋白质点有24个,而绝大部分大豆种子贮藏蛋白在萌发期尚未降解.在萌发的第一阶段,24个差异表达蛋白中有10个蛋白质的丰度发生变化.第二阶段,差异表达蛋白的种类和量增加,其中15个蛋白质是动态变化的,14个蛋白质在胚根突破种皮时表达量达到峰值,表明吸胀后种子内的生命活动越来越强.对这24个蛋白质点进行胶内酶解,用基质辅助激光解析电离飞行时间质谱测定均获得肽质量指纹图谱.搜索大豆的UniGene库初步鉴定出6个蛋白质,分别是核苷二磷酸激酶、热激蛋白、硫氧还蛋白、35ku种子成熟蛋白及种子成熟蛋白PM36.对这些蛋白质在种子萌发过程中可能的作用进行了讨论.  相似文献

Germination responses ofRhus javanica L. seeds to temperature and light were investigated with special reference to their gap-detecting mechanisms in germination, i.e., responses to elevated and/or fluctuating temperatures and sensitivity to leaf-canopy transmitted light. The seeds, which have water-impermeable coats to prevent imbibition, were shown to become permeable and germinable after exposure to higher temperatures of 48–74°C for a brief period depending on the temperature. Once the coat impermeability had been removed by such heat treatment, the seeds became readily germinable over a wide range of temperature and light conditions. The lower and higher temperature limits for germination were around 8° and 36°C, respectively, with an optimal temperature of around 25°C. Simple linear relationships were observed between the temperature and germination rates, i.e., the reciprocals of the time taken by the seed subpopulations to show 10–70% germination in the sub-optimal temperature range, where the required ‘thermal time’ for germination was 2300–3600 Kh. The presence or absence of light or a simulated ‘canopy light’ had little effect on the germination of this species. It was concluded that the seeds ofR. javanica are furnished with a gap-detecting mechanism in the form of a heat requirement for the breakage of water-impermeable seed dormancy, which may be fulfilled by either daytime elevation of the surface temperature of exposed soil, or more effectively by fire.  相似文献

The nitric oxide (NO) donor sodium nitroprusside (SNP) significantly promoted germination of switchgrass (Panicum virgatum L. cv Kanlow) in the light and in the dark at 25°C, across a broad range of concentrations. SNP also promoted seed germination in two other warm-season grasses. A chemical scavenger of NO inhibited germination and blocked SNP stimulation of seed germination. The phenolic (+)-catechin acted synergistically with SNP and nitrite in promoting seed germination. Acidified nitrite, an alternate NO donor also significantly stimulated seed germination. Interestingly, sodium cyanide, potassium ferricyanide and potassium ferrocyanide at 200 μM strongly enhanced seed germination as well, whereas potassium chloride was without effect. Ferrocyanide and cyanide stimulation of seed germination was blocked by an NO scavenger. Incubation of seeds with a fluorescent NO-specific probe provided evidence for NO production in germinating switchgrass seeds. Abscisic acid (ABA) at 10 μM depressed germination, inhibited root elongation and essentially abolished coleoptile emergence. SNP partially overcame ABA effects on radicle emergence but did not overcome the effects of ABA on coleoptile elongation. Light microscopy indicated extension of the radicle and coleoptiles in seeds maintained on water or on SNP after 2 days. In contrast, there was minimal growth of the radicle and coleoptile in ABA-treated seeds even after 3–4 days. These data indicate that seed germination of warm-season grasses is significantly influenced by NO signaling pathways and document that NO could be an endogenous trigger for release from dormancy in these species.  相似文献

The germination of lettuce (Lactuca sativa L.) seeds was greatly reduced when the seeds were heated at 97°C for 30 h prior to imbibition. This dormancy was effectively released when ethylene (1–100 ppm) or benzyladenine (BA) (0.005–0.05 mM) was applied during the imbibition period. Ethylene was not required during the early part of imbibition, but was essential during the period immediately prior to radicle protrusion. Treatment with 1-aminocyclopropane-1-carboxylic acid (ACC) (0.1–10 mM) stimulated germination, but was not as effective as ethylene or cytokinin treatment. During the germination of nondormant lettuce seeds, ethylene production increased rapidly and reached a peak at 24 h, which coincided with the emergence of the radicle, and then declined; the level of ACC increased as ethylene production rate increased, but remained at a high level after radicle protrusion. In heat-pretreated dormant lettuce seeds, the increases in percent germination, ethylene production, and ACC levels were all delayed and lower than those of nondormant seeds, and these increases were accelerated by treatment with ethylene or cytokinin.  相似文献

The effects of long-term seed storage on the physiological properties of phytochrome-mediated germination including water uptake, the temperature and light flunnce dependencies of germination and dark germination were studied. The fluenceresponse relationships of the brief irradiation with monochromatic red (660 nm, 7.5 W m−2) and far-red (750 nm, 6.6 W m−2) light at various times after sowing were also studied. The samples used consisted of three lots of seeds ofLactuca sativa L. cv. MSU-16, which had been harvested in 1976, 1979 and 1985 and stored dry for 9, 6 and 0 years, respectively, in darkness at 23±2 C until the experiments were carried out in July–August, 1985. Seeds with the longer storage periods showed the higher ability to germinate in both continuous darkness and continuous white fluorescent light at 20–30 C. In the seeds stored for 6 or 9 years, red light irradiation for 20 sec given at 15 min or more after sowing at 25 C induced as high a percent germination (85–95%) as those under continuous white fluorescent light. In the freshly harvested seeds, however, germination under continuous white fluorescent light (46%) was considerably lower than the germination induced by the red pulse (97%). Germination of the seeds decreased when the intervals between sowing and a far-red irradiation for 20 sec increased up to 100 min (or 30 min in the freshly harvested seeds). The far-red pulse given later than 100 min (or 6 hr in the freshly harvested seeds) after sowing resulted in an increased germination up to the dark-germination levels with increasing intervals between sowing and the pulse irradiation. Before or at 3 min after sowing, the seeds stored for 6 or 9 years were responsive to the far-red pulse although they were not or hardly responsive to the red pulse, while the freshly harvested seeds were responsive to both the far-red and the red pulses. These data indicate that normal functions of phytochrome completely survived in the dry seeds during storage at 25 C for as long as 6 or 9 years and that these functions are restored into full operation by means of imbibition. The differences in the dependence of germination on the time and fluence of a single pulse of red or far-red light seems to be related to the smaller water content throughout the imbibition in the seeds with the longer storage periods. The greater ability to germinate in the dark indicates the greater amounts of PFR or the greater responsivity to PFR, in the seeds with the longer storage periods.  相似文献
