The effect of substances with different redox potentials in the phytochrome-controlled germination of Paulmtnia tomentosa seeds was examined. Up to 25% of water-imbibed seeds germinated upon irradiation with 5 min red light The seeds irradiated with 5 min red light and incubated in substances with redox potentials equal or higher than E'0= 360 mV [potassium nitrate, potassium hexacyanoferrate (III) or potassium hexachloroiridate (IV)] and sodium nitroprusside germinated up to 80%. The optimal concentration was between 1 and 10 mM. Other electron acceptors such as 2.6-dichlorophenol-indoplhenol, phenazine methosulfate and methylene blue failed to produce and effect. The germination-promoting effect was obtained when the substances were supplied to the incubation medium from the onset of imbibition or at the onset of irradiation. Delay of application of these chemicals until after the red light treatment diminished their effectiveness and the effect was lost after 48 h. The escape from far-red reversibility was evident in the presence of substances which stimulated germination. The results presented support the view that phytochrome-broken dormancy of Paulownia tomentosa seeds is closely connected with redox changes.  相似文献

Summary The qualitative and quantitative composition of secondary metabolites was studied in the shoots and roots of Centaurium pulchellum cultured in vitro. Secoiridoids (gentiopierin, swertiamarin, and sweroside) and xanthones (methylbellidifolin, demethyleustomin, and deccussatin) were isolated. Sweroside was found to be the major secoiridoid compound in the aerial parts of plants growing in nature. while swertiamarin dominated in plants cultured in vitro. In roots of all plants, genciopicrin was the major compound. Xanthone demethyleustomin was the major compound both in the shoots and roots of plants growing in nature and cultured in vitro. Different sugars (glucose, fructose, and sucrose) added in different concentrations in the medium affected the production of secondary compounds.  相似文献

Germination of lettuce seeds cv. May Queen iscompletely prevented either with 10 µM tetcyclacisor with continuous FR illumination. GA3 and the N-substituted phtalimide, AC 94,377, werepartially effective in overcoming tetcyclacis-inducedinhibition but ineffective on photoinhibited seeds. FCcompletely reversed tetcyclacis inhibition and inducedca. 60% germination in continuous FR light. Aninteraction between FC and GA3 (as well asbetween FC and AC 94,377) was evident in stimulationof germination under both inhibitory conditions.Interaction was calculated as a ratio of thepercentage of seeds germinated under the simultaneousaction of stimulators compared to their additiveeffect. This was 2.54 for tetcyclacis- and 2.95 forphotoinhibited seeds. It is concluded that thistype of interaction is promotive synergism. Themagnitude of the interaction was highest if theapplication of FC was delayed after GA3application, and the optimal time lag was 6 h fortetcyclacis-inhibited, or 24 h for photoinhibited seeds.  相似文献

The effect of sucrose on fruiting, seed production, and seed germination of lesser centaury [Centaurium pulchellum (Sw.) Druce] was examined using explants of flowers and flower buds. Sucrose concentrations in the culture medium ranged from 0.003 to 0.3 M. It has been shown that the number of auxiliary buds, capsules dimension, number of viable seeds per capsule and seed dimensions increased with the increase of sucrose concentrations. The highest values were recorded at sucrose concentrations higher than 0.03 M, except for seeds size, which were larger at sucrose concentration ranging from 0.003 to 0.1 M. The germination of in vitro produced seeds was affected by previous culture history: a higher germination percentage was obtained in seeds that were raised from explants originally grown on medium with sucrose concentrations higher than 0.003 M.  相似文献

Hairy roots of Rhamnus fallax Boiss. were induced using Agrobacterium rhizogenes strain A4M70GUS. The culture established on Woody plant media (WPM) showed a typical hairy root phenotype: rapid growth, reduced apical dominance and root plagiotropism. Seven clones of R. fallax were selected on the basis of their differences in colour and the root branching. The growth of hairy root culture, measured through gain in fresh mass, was done under 16-h photoperiod or in the dark. An increase in anthraquinone (AQ) content was obtained in clones with yellow and less branched roots, like clone 1 [16.43 mg g−1(d.m.)] and clone 7 [14.21 mg g−1(d.m.)], compared with other analysed transformed and non-transformed tissue. This study presents the first report of successful transformation of any species from family Rhamnaceae by A. rhizogenes and analysis of AQ production in transformed tissue.  相似文献

Shoots of micropropagated Gentiana acaulis, G. cruciata, G. lutea, and G. purpurea were inoculated with suspensions of Agrobacterium rhizogenes cells, strains ATCC 15834 or A4M70GUS. Adventitious roots appeared at the sites of inoculation in all 4 species. Root tips were excised and cultured on growth regulator-free media for 2-6 years. They exhibited very high branching and plagiotropism. Spontaneous bud initiation occurred in roots of G. cruciata. Roots of G. lutea, G. acaulis and G. purpurea were cultured on media with high kinetin concentration, which induced the formation of friable callus tissues. Only in G. purpurea were these calluses organogenic. Regenerated shoots of G. cruciata and G. purpurea gave rise to plants, that displayed the typical phenotypes of A. rhizogenes-transformed plants: short internodes and rolled leaves. In the roots of G. acaulis and G. cruciata, transformed with A. rhizogenes A4M70GUS, a positive reaction with X-gluc indicated the activity of β-glucuronidase. The DNA extracted from hairy roots and from the roots of transgenic plants hybridized with the appropriate genomic probes in Southern blotting. This is taken as evidence of the stable genetic transformation in the 4 Gentiana species. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Astrocytes can exocytotically release the gliotransmitter glutamate from vesicular compartments. Increased cytosolic Ca(2+) concentration is necessary and sufficient for this process. The predominant source of Ca(2+) for exocytosis in astrocytes resides within the endoplasmic reticulum (ER). Inositol 1,4,5-trisphosphate and ryanodine receptors of the ER provide a conduit for the release of Ca(2+) to the cytosol. The ER store is (re)filled by the store-specific Ca(2+)-ATPase. Ultimately, the depleted ER is replenished by Ca(2+) which enters from the extracellular space to the cytosol via store-operated Ca(2+) entry; the TRPC1 protein has been implicated in this part of the astrocytic exocytotic process. Voltage-gated Ca(2+) channels and plasma membrane Na(+)/Ca(2+) exchangers are additional means for cytosolic Ca(2+) entry. Cytosolic Ca(2+) levels can be modulated by mitochondria, which can take up cytosolic Ca(2+) via the Ca(2+) uniporter and release Ca(2+) into cytosol via the mitochondrial Na(+)/Ca(2+) exchanger, as well as by the formation of the mitochondrial permeability transition pore. The interplay between various Ca(2+) sources generates cytosolic Ca(2+) dynamics that can drive Ca(2+)-dependent exocytotic release of glutamate from astrocytes. An understanding of this process in vivo will reveal some of the astrocytic functions in health and disease of the brain. Summary This study reports the analysis of secondary metabolites of gentiopicrin, swertiamarin, and sweroside in shoot and root cultures of yellow wort (Blackstonia perfoliata), which were initiated from seeds, grown on Murashige and Skoog (MS) medium. Shoot cultures of B. perfoliata were inoculated with suspension of Agrobacterium rhizogenes strain A4M70GUS and hairy roots appeared at the infected sites after 3 wk of inoculation. Tips of adventitious roots of B. perfoliata were grown on hormone-free MS medium and three clones of the transformed roots regenerated shoots spontaneously. Gentiopicrin, swertiamarin, and sweroside were detected in both roots and shoots of B. perfoliata in vitro and in vivo, but gentiopicrin was found to be the major compound. The concentration of growth regulator in the medium affected the production of secoiridoids in B. perfoliata in vitro, where the level of gentiopicrin was higher in plants grown in the presence of indole-3-butyric acid, but the presence of 6-benzylaminopurine was inhibitory to secoiridoid production.  相似文献

The DRB1, DRB3, DRB5, DQA1 and DQB1 allele polymorphisms were analysed in 3 western and 3 eastern villages of the island of Hvar using PCR-SSOP method and 12th International Workshop primers and probes. Three DQB1 alleles (*0304, *0305, *0607) detected in the population of the island of Hvar (HP) have not yet been observed in general Croatian population (GCP). Significant differences were observed between two regions of Hvar for: a) DRB1*0701 allele (p < 0.001), b) DQA1*0201 allele (p < 0.01), and c) DRB1*0101-DQA1*0101-DQB1*0501 haplotypic association (p < 0.05). Two unusual haplotypic associations, which have not yet been described in general Croatian population (GCP), DRB1*0101-DQA1*0102-DQB1*0501 and DRB1*1501-DQA1 *0102-DQB1*0604 were observed in the population from the island of Hvar (HP). Measures of genetic kinship and genetic distances revealed isolation and clusterization which coincides with the known ethnohistorical, as well as biological and biocultural data obtained from a series of previous investigations. The five studied village subpopulations formed two clusters (East-West) to which the far eastern village (with the highest rii of 0.0407) joined later, thus indicating possible impact of historical immigrations from the mainland.  相似文献
